Genetic Identification of the Kimchi Strain Using PCR-based PepN and 16S rRNA Gene Sequence (PepN과 16S rRNA Gene Sequence 및 PCR 방법을 이용한 김치 젖산균의 동정)

  • The WL6 strain isolated from Kimchi could not be made scientific name because it was identified as three species, i.e., Leuconostoc mesenternides ssp cremoris, Leu. mesenteroides ssp. dextranicum or Lactobacillus bifermentans when it was tested by API kit or Biolog system methods. The unidentifiable WL6 strain was finally reclassified as Lactobacillus bifermentans by genetic identification using two PCR-based specific sequence primer sets which were originated from homologous pepN and 16S rRNA genes.

Recognizing Hand Digit Gestures Using Stochastic Models

  • A simple efficient method of spotting and recognizing hand gestures in video is presented using a network of hidden Markov models and dynamic programming search algorithm. The description starts from designing a set of isolated trajectory models which are stochastic and robust enough to characterize highly variable patterns like human motion, handwriting, and speech. Those models are interconnected to form a single big network termed a spotting network or a spotter that models a continuous stream of gestures and non-gestures as well. The inference over the model is based on dynamic programming. The proposed model is highly efficient and can readily be extended to a variety of recurrent pattern recognition tasks. The test result without any engineering has shown the potential for practical application. At the end of the paper we add some related experimental result that has been obtained using a different model - dynamic Bayesian network - which is also a type of stochastic model.

  • This study evaluates the performance of a coil spring-viscous damper system for the vibration and seismic isolation of an Emergency Diesel Generator (EDG) by measuring its operational vibration and seismic responses. The vibration performance of a coil spring-viscous damper system was evaluated by the vibration measurements for an identical EDG set with different base systems - one with an anchor bolt system and the other with a coil spring-viscous damper system. The seismic performance of the coil spring-viscous damper system was evaluated by seismic tests with a scaled model of a base-isolated EDG on a shaking table. The effects of EDG base isolation on the fragility curve and core damage frequency in a nuclear power plant were also investigated through a case study.

PRIMITIVE CIRCLE ACTIONS ON ALMOST COMPLEX MANIFOLDS WITH ISOLATED FIXED POINTS

  • Let the circle act on a compact almost complex manifold M with a non-empty discrete fixed point set. To each fixed point, there are associated non-zero integers called weights. A positive weight w is called primitive if it cannot be written as the sum of positive weights, other than w itself. In this paper, we show that if every weight is primitive, then the Todd genus Todd(M) of M is positive and there are $Todd(M){\cdot}2^n$ fixed points, where dim M = 2n. This generalizes the result for symplectic semi-free actions by Tolman and Weitsman [8], the result for semi-free actions on almost complex manifolds by the author [6], and the result for certain symplectic actions by Godinho [1].

  • Let G be a graph with no isolated vertex. A total dominating set, abbreviated TDS of G is a subset S of vertices of G such that every vertex of G is adjacent to a vertex in S. The total domination number of G is the minimum cardinality of a TDS of G. In this paper, we study the total domination number of central graphs. Indeed, we obtain some tight bounds for the total domination number of a central graph C(G) in terms of some invariants of the graph G. Also we characterize the total domination number of the central graph of some families of graphs such as path graphs, cycle graphs, wheel graphs, complete graphs and complete multipartite graphs, explicitly. Moreover, some Nordhaus-Gaddum-like relations are presented for the total domination number of central graphs.

Seven Undescribed Aspergillus Species from Different Niches in Korea

  • An investigation of species of the genus Aspergillus present in arthropod, freshwater, and soil led to the discovery of seven undescribed species in Korea. Based on their morphological characteristics and molecular phylogeny analyses using a combined data set of β-tubulin (BenA) and calmodulin (CaM) sequences, the isolated strains CNUFC IGS2-5, CNUFC YJ1-19, CNUFC WD27, CNUFC U8-70, CNUFC AS2-24, CNUFC S32-1, and CNUFC U7-48, were identified as Aspergillus brunneoviolaceus, A. capensis, A. floccosus, A. inflatus, A. parvulus, A. polyporicola, and A. spelaeus, respectively. In the present study, the detailed morphological descriptions and phylogenetic relationships of these species are provided.

  • Modern OSs, including Linux, show high scalability by adopting a monolithic kernel design, but have weak security because they share all memory space. This study presents a kernel module that are isolated inside the kernel using WebAssembly. WebAssembly provides a high-performance virtual machine by defining a low-level instruction set while guaranteeing memory safety. In this paper, the WebAssembly execution environment is implemented inside the kernel, allowing developers to control the operation of kernel modules and achieving higher security.

Development of Broad-range and Specific 16S rRNA PCR for Use in Routine Diagnostic Clinical Microbiology (임상미생물 검출을 위한 광대한 범위와 특이도를 가지는 16S rRNA PCR법 개발)

  • Broad-range and specific 16S rRNA gene PCR is used for detection and identification of bacterial pathogens in clinical specimens from patients with a high suspicion for infection. We describe the development of a broad-range and specific PCR primer, based on bacterial 16S rRNA, for use in routine diagnostic clinical microbiology services. The primers were designed by using conservative regions of 16S rRNA sequences from 10 strains. Ninety-eight clinical strains were isolated from clinical patient specimens. A total of 98 strains of bacteria were identified by phenotypic methods; PCR with newly designed primers and universal primers. All purified PCR products were sequenced using both forward and reverse primers on an automated DNA analyzer. In this study, we evaluated the usefulness of the newly designed primers and the universal primers for the detection of bacteria, and both these techniques were compared with phenotypic methods for bacteria detection. When we also tested 98 strains of clinical isolates with newly designed primers, about 778 bp DNA fragments were amplified and identified from all strains. Of the 98 strains, 94 strains (95.9%) correspond in comparison with phenotypic methods. The newly designed primers showed that the identities of 98 (100%) strains were the same as those obtained by universal PCR primers. The overall agreement between the newly designed primers and universal primers was 100%. The primer set was designed for rapid, accurate, and cheap identification of bacterial pathogens. We think the newly designed primer set is useful for the identification of pathogenic bacteria.

Electron Microscopic Study on the Role of Actin Filaments during the Formation of Bile Canaliculi in Isolated Rat Hepatocyte Culture System (흰쥐에서 분리 배양한 간세포의 담세관 형성에 있어서 액틴미세섬유의 역할에 관한 전자현미경적 연구)

  • Bile canaliculi are the structure delivering bile secreted by hepatocytes into the bile passage. Bile secretion is mainly controlled by the cytoskeletal elements, mainly of actin in the microvilli, pericanalicular web. Most studies on the bile secretion have been done in viva situation, however, to control the various parameters in vitro culture system seem to be more useful. To set up an in vitro experimental system, the investigator isolated hepatocytes with an enzymatic method using a mixture of collagenase and hyaluronidase from normal Sprague-Dawley rat liver and cultured. Isolated hepatocytes were round and formed cords in culture. Microvilli covered the whole surface of hepatocytes. Bile canaliculi were formed between hepatocytes and were characterized by the presence of microvilli of various lengths and shapes mainly arising from small surface mounds. Actin filament core in the microvilli and pericanalicular actin web were incomplete. After cytochalasin D treatment, cultured hepatocytes were round but the surface were irregular with surfacen blebs, folds and grooves. Microvilli on the surface were scarce. Bile canaliculi were markedly dilated often with the detached junctional complexes. Bile canaliculi lacks microvilli almost completely and extended into the pericanalirular cytoplasm showing complex vacuolar and tubular structures by transmission electron mciroscopy. Pericanalicular actin web, intermediate filaments were hardly identified. Subsurface actin filaments were scattered scarcely under the cell membranes. These results suggest that hepatocytes isolated from rats can survive and form bile canaliculi in culture and the actin filaments are involved in the formation and/or maintenance of the bile canaliculi.

Establishment and Application of a Multibead Serotyping Assay for Pneumococci in Korea (다중구슬 폐구균 혈청형 분석법의 국내 확립과 적용 연구)

  • Purpose: Serotyping pneumococcal isolates is important to monitor efficacy of pneumococcal vaccines. Because of difficulties of typing pnueumocci, a multiplex bead-based (multibead) serotyping assay was recently introduced. The aim of this study is to establish a new multibead serotyping assay and to apply this method to analyze clinical isolates of pneumococci in Korea. Methods: To establish the multibead serotyping assay, six key reagents were transferred from University of Alabama at Birmingham (UAB) to Ewha Center for Vaccine Evaluation and Study (ECVES): bead set coated with polysaccharide and monoclonal antibody pool were used in one multiplex inhibition-type immunoassay and 2 bead sets coated DNA probe and 2 primer pools were used in two multiplex PCR-based assays. After multibead serotyping assay was set up, 75 test samples of pneumococci were analyzed whether ECVES is able to identify serotype correctly. After confirming the performance, serotyping assay was applied to identify serotypes of 528 clinical isolates of pneumococci collected from 3 different hospitals. Results: After establishment of the multibead pneumococcal serotyping assay system at ECVES, 75 test samples were analyzed. There was no discrepancy of serotypes of 75 test samples between the results assigned at UAB and those at ECVES. The serotypes of 528 pneumococci isolated from patients or healthy subjects were determined in 94.3% of isolates (498/528). Conclusions: The multibead pneumococcal serotyping assay can be successfully established in Korea. With this method, surveillance of serotypes of pneumococci isolated from patients as well as healthy subjects could be studied.
