Characterization and Purification of Anti-Complement Polysaccharide from Spirodela polyrhiza

  • We purified and characterized a crude polysaccharide from Spirodela polyrhiza with anti-complement activities. The crude polysaccharide fraction (SP-0) which had potential anti-complement activity was extracted in hot water for 4 hrs at 10$0^{\circ}C$. The ethanol-precipitate, the crude polysaccharide traction (SP-1), showed a potent anti-complement activity. Further purification of the crude polysaccharide (SP-1) was carried out by cetavlon, ion exchange chromatography and gel column chromatography. Among cetavlon fractions, SP-4 showed the most potent anti-complement activity. When 100 $\mu\textrm{g}$/mL of SP-4 was incubated with an equal volume of normal human serum (NHS), the TCH$_{50}$ was reduced by about 78%. When the SP-4 fraction was further purified by DEAE-Sepharose (Cl$^{[-10]}$ ), the SP-4IIa, SP-4IIb and SP-4IIc, absorbed fractions, were almost the same as the anti-complement activities of SP-4. SP-4IIc, having the greatest potential activation and the highest yield by ion exchange chromatography, was further purified by gel column chromatography on a Sepharose CL-6B column. Four polysaccharide fractions of SP-4IIc-1, SP-4IIc-2, SP-4IIc-3 and SP-4IIc-4 were obtained, consisting mainly of arabinose, rhamnose, galactose and glucose, with approximate molecular weights of about 305,000, 132,000, 64,000 and 12,000, respectively. Among these subfractions, SP-4IIc-1 had the most potent anti-complement activity. When the SP-4IIc-1 aggregate was applied to a gel column chromatography in 10 mM and 50 mM NaCl solution, the position of the peak fractions shifted to a low molecular weight region, and the molecular weight of SP-4IIc-1 decreased with increased NaCl concentration in the gel column chromatography. It was found that the self-aggregation formed spontaneously in void volume by gel column chromatography using Sepharose CL-6B in water and the self-aggregation significantly affected the anti-complement function.

  • Pleurotus ostreatus로부터 glyoxalase I(S-lactoyl-glutathione methylglyoxal lyase, EC 4.4.1.5)이 S-hexylglutathione affinity chromatography, Sephadex G-150 gel permeation chromatography, DEA-sepharose A-50 CL-6B ion exchange chromatography를 통해 순수 분리되었다. 이 결과, 전체 활성도의 21.7% fmf 수확하였으며, 분리 배수는 2,294 배 이었다. Gel filtration chromatography로 측정한 효소의 분자량은 34 kDa이며, SDS-PAGE 결과 본 효소는 분자량 17 kDa인 동일한 소단위체 두 개로 구성된 이합체라고 생각된다. Methylglyoxal과 phenylglyoxal에 대한 $K_m$ 값은 각각 0.39 mM 과 0.22 mM 이며 L-xylosone과 hydroxypyruvaldehyde에 대해서도 강한 친화력을 보여주었고, pH 6.5~7.5, $35~45^{\circ}C$에서 활성도가 가장 높았다. 이 효소의 반응 과정을 핵자기공 명분광법으로 분석한 결과, 분자내의 양성자 전달과정이 뚜렷이 관찰되었다.

우라늄의 분리 및 회수에 관한 연구 (A study on the separation and recovery of uranium)

  • The anionic exchange resins with the Dowex-1 and Amberlite CG-400 form were transformed into resin of sulfate and acetate acid form, respectively. The uranyl complex ions with SO$_{4}$$^{2-}$ and CH$_{3}$COO$^{-}$ were adsorbed on the anion exchange resion mentioned above, and these complex ions were eluted as mixture eluents of 0.7M HNO$_{3}$ - 0.5M NH$_{4}$NO$_{3}$ by anion exchange chromatography. The optimum adsorption conditions of uranyl anion complex ions adsorbed on the upper of the resin colmun were 1.5-2.0 ml/min of flow rates at pH 2.0 and adsorptive power of uranyl complex ion of sulfuric acid type were nearly consistent with the Caussion normal distribution curve, whereas the elution state of UO$_{2}$(Ac)$_{2}$$^{4-}$ with acetic acid type was departed. The weighing form obtained from resin of sulfuric acid and aceric acid type was U$_{3}$O$_{8}$ whose recovery was 91.7%. The possibility of recovering uranium from the monazite sulfate solution using a strong base anion resin, Amberlite CG-400(sulfate form), was successfully recovered more than 90%.

  • 우렁쉥이 껍질에서 추출한 autoclave 추출구와 neutrase 추출구는 지방흡수력의 경우 대조시료인 chitin과 chitsan에 비해 떨어지는 대략 250$\~$$300\%$ 정도이었으나, 포말성은 각각 0.70과 0.67로, 포말안정성은 0.50과 0.38로 우수하였다 Autoclave 추출구 및 neutrase 추출구의 유화능은 각각 $49.2\%$$47.1\%$, 유화안정성의 경우 각각 $48.3\%$$44.8\%$ 정도였으며 특히, 색소흡착능은 각각 $85.2\%$$85.6\%$로 대조시료에 비해 다소 우수하였다 항혈액응고능은 모든 시료에서 hPn 저 지효과는 나타났으나 PT값은 약 14.5초로 일정하여 혈액응고저지 효과가 거의 없었다. 특히 autoclave 처리구를 DEAE-cellulose ion exchange chromatography하여 얻은 F4 (획분4)는 APTT가 75초로 가장 효과적 이 었으며, Sephadex G-100의 $F_2$가 70초 정도였다. ACE저해능은 모든 시료에서 ACE 저해효과가 있었으며, 채택된 실험방법에 따라 동일시료 경우에 있어서도 저해효과 값이 차이가 났으며, 특히 DEAE-cellulose 이온교환 처리한 $ F_4$ (획분4)가 $35.7\%$로 가장 효과적이었다.

Purification and Characterization of Farnesyl Protein Transferase from Bovine Testis

  • Famesyl protein transferase involved in the first step of post-translational modification of $p21^{ras}$ proteins transfers the famesyl moiety from famesyl pyrophosphate to a cysteine residue in $p21^{ras}$ proteins. The enzyme was first purified 30,000-fold from bovine testis by use of 30~50% ammonium sulfate fractionation, DEAE-Sephacel ion exchange chromatography, Sephacryl S-300 gel filtration chromatography, Sephacryl S-200 gel filtration chromatography, and hexapeptide (Lys-Lys-Cys-Val-Ile-Met) affinity chromatography. The molecular weight of the purified enzyme was estimated to be ~100 kDa by gel filtration and SDS-polyacrylamide gels showed two closely spaced bands of ~50 kDa protein. These indicate that the enzyme consists of two nonidentical subunits, a and 13, which have slightly different molecular weights. The enzyme was inhibited by hexapeptide (Lys-Lys-Cys-Val-Ile-Met), which acted as an alternative substrate that competed for famesylation. Kinetic analysis by measuring initial velocities showed that famesyl protein transferase is a very slow enzyme. EDTA-treated famesyl protein transferase showed little activity with $Mg^{2+}$ or $Zn^{2+}$ alone, but required both $Mg^{2+}$ and $Zn^{2+}$ for the catalytic activity.

  • 바이러스는 생물 의약 산업에서 다양한 응용 분야를 가지고 있다. 그들은 살충제 생산, 백신 생산, 유전자 전달, 암 치료제 등에 사용된다. 바이러스의 하류 처리는 그들의 생물학적 및 의약적 응용을 위한 필수 단계이다. 다양한 과정 중에서 바이러스의 정제는 매우 중요하다. 막 크로마토그래피는 이 과정에서 중요한 역할을 한다. 이온 교환 막 크로마토그래피는 주로 사용되는 방법이지만 크기 배제 및 불충분한 정제에 관한 다양한 제한을 가지고 있다. 또한, 이는 인플루엔자와 같은 빠르게 변화하는 바이러스의 균주에 적용될 수 없다. 이 검토는 막 크로마토그래피의 다양한 개선된 방법 또는 대안을 검토한다. 이는 정제, 바이러스 회수율 및 방법의 확장성에 초점을 맞추고 있다.

펙틴의 정제 및 분석 (Purification and Analysis of Pectins)

  • Pectins present in the primary cell walls and middle lamellae of plant cell walls are extracted by water, cheating agents, acid or alkali solutions. However, some neutral contaminating components are extracted in conjunction with pectins during the extraction process. Thus, the accurate characterization of physi-cochemical properties of pectins necessitates to get rid of the impurities. In this review, dialysis, alcohol precipitation, ion exchange chromatography and metal precipitation were compared as procedures to purify the pectin extracts. In addition, the chemical methods to analyze pectins are discussed in terms of three major chemical constituents, i.e., anhydrogalacturonic acid, methoxyl groups and neutral sugars.

맹꽁이 조직내 유리 아미노산의 정성적 분석 (Qualitative Analysis of the Free Amino Acids in Gallula tornieri Tissues)

  • The present work deals with the patterns of the free amino acids in several tissues of Gallula tornieri VOGT analysed by means of paper chromatography and ion-exchange resins. The tissues used were heart, stomach , muscle, brain , testis , skin , and liver. 14 amino acid was detected in heart, 11 in muscle , 15 in stomach , 9 , in brain , 10 in skin in liver , and 10 in testis. It was found that there were distinguishable differences in the patterns of the free amino acids among the different tissues.

  • Cytolysin produced by Vibrio vulnificus ATCC 27562 was partially purified by sequential ammonium sulfate precipitation, gel filtration with Sephadex G-200, and ion exchange chromatography with DEAE-Sephadex. The partially purified cytolysin was inactivated by cholesterol. More than one molecule of the cytolysin was required to lyse a single erythrocyts. The antiserum against cytolysin enhanced the survival ratio of mice infected with low dose of V. vulnificus.

해조류 추출물로부터 홍합(Mytilus edulis)의 부착 저해물질 정제

  • 연안 해역에서 해조류와 부착패류는 양식장 시설 뿐만아니라 수취구 어선바닥 등에서 그 피해는 심각한 실정이다. 해조류에 의한 부착은 잎파래 가시파래 등이 주가 되고 패류로는 홍합과 따개비가 주를 이룬다 이들의 부착방지를 위해서 염소살균, TBTO (Tributyltin oxide), CuSO$_4$등의 처리를 하고 있지만 이러한 처리에 의해서는 또다른 환경 문제가 야기되고 있다. 이같은 부착방지 물질 탐색의 일환으로 흥합을 탐색 대상종으로 선택하여 각종 해조류 추출물들로부터 부착 방지 효과를 조사하였다. 그 결과 고리매(Sytosimphon lomentaria)로부터 가장 높은 활성을 확인 하였고, TLC, silica-gel chromatography, Ion-exchange, HPLC를 통해 정제 하였다.
