Studies on Constituents of the Higher Fungi of Korea (XLVI) -Antitumor Components Extracted from the Cultured Mycelia of Lyophyllum decastes-

  • To investigate antitumor component of Lyophyllum decastes, the aqueous extract of its shake-cultured mycelia was subjected to antitumor test against sarcoma 180 cells implanted in ICR mice. The extract showed an inhibition ratio of 65.4% and was found to consist of a polysaccharide moiety and a protein moiety. After purification with DEAE-Sephadex A-50 ion exchange chromatography, Fraction D showed the highest inhibition ratio of 75.7%. The antitumor constituent was examined for immunoaccelerating activity and was found to increase macrophage accumulation in the peritoneal cavity and plaque forming cells of the spleen cells. It was named lyophyllan after the genus name.

  • Phase intergrowth in BSCCO thin films have been investigated. It turned out from XRD analyses of these phases that molar fraction of each constituent phase in the intergrowth thin film can be exhibited as a function of substrate temperature and ozone pressure. Superconducting behavior of the intergrowth thin film is also discussed.

A Study on Ion Distribution and Behavior of Acidrain in Mokpo and Yeochon Area (목포, 여천지역 강수의 무기이온 성분농도와 거동에 관한 연구)

  • This study was carried out to investigate the constituents of inorganic ions of precipitation in Mokpo and Yeochon area from Jan. 1996 to Dec. 1997. The volume-weighted mean pH of precipitation was 5.7(4.8~7.8) at Mokpo, and 5.6(5.1~7.4) at Yeochon area, respectively. The non-seasalt(nss) anion concentrations were found in order of $nss-SO^{2-}_4>NO^-_3>nss-Cl^-$ at two areas. The portion of $nss-SO^{2-}_4;and;NO^-_3$ was 71~84% out of anions. The the non-seasalt cation concentrations were found in order of $NH^+_4>nss-Ca^{2-}>nss-Mg^{2+}>nss-K^-$ at two areas. The portion of $NH^+_4;and; nss-Ca^{2+}$ was 85~92% out cations. Compared regionally the year concentraton of $nss-SO^{2-}_4$, the result of Yeochon was 3 times higher than that of Mokpo in 1996, while the results of two regions were almost similar in 1997. The reason was that Yeochon was restricted area of the use of 0.5% sulfur B-C oil, controlled air pollution emission area, controlled semitotal amounts of air pollution emission area and partial operating of factories in 1997. There were no ions having high correlation with $H^+,;but;nss-SO^{2-}_4,;NO^-_3,;NH^+_4;and;nss-Ca^{2+}$ showed high correlation coefficient each other. It seems that these ions have little correlation with $H^+$ because they are washed out on binding state. Factor analysis showed that the first factor was complicated factor containing anthropogenic and soil resource, the second factor was sea-salt resource and the third factor was independent behavior of hydrogen at Mokpo. While, the first factor was complicated factor containing anthropogenic and sea-salt resource, the second factor was $Ca(NO_3)_2$ salt and the third factor was $NH^+_4;and;SO^{2-}_4$ synergied by resource and combination at Yeochon.

Protective Effect Against Hydroxyl Radical-induced DNA Damage and Antioxidant Mechanism of [6]-gingerol: A Chemical Study

  • [6]-Gingerol is known as the major bioactive constituent of ginger. In the study, it was observed to effectively protect against ${\bullet}OH$-induced DNA damage ($IC_{50}$ $328.60{\pm}24.41{\mu}M$). Antioxidant assays indicated that [6]-gingerol could efficiently scavenge various free radicals, including ${\bullet}OH$ radical ($IC_{50}$ $70.39{\pm}1.23{\mu}M$), ${\bullet}O_2{^-}$ radical ($IC_{50}$ $228.40{\pm}9.20{\mu}M$), $DPPH{\bullet}$radical ($IC_{50}$ $27.35{\pm}1.44{\mu}M$), and $ABTS{^+}{\bullet}$radical ($IC_{50}$ $2.53{\pm}0.070{\mu}M$), and reduce $Cu^{2+}$ ion ($IC_{50}$ $11.97{\pm}0.68{\mu}M$). In order to investigate the possible mechanism, the reaction product of [6]-gingerol and $DPPH{\bullet}$ radical was further measured using HPLC combined mass spectrometry. The product showed a molecular ion peak at m/z 316 $[M+Na]^+$, and diagnostic fragment loss (m/z 28) for quinone. On this basis, it can be concluded that: (i) [6]-gingerol can effectively protect against ${\bullet}OH$-induced DNA damage; (ii) a possible mechanism for [6]-gingerol to protect against oxidative damage is ${\bullet}OH$ radical scavenging; (iii) [6]-gingerol scavenges ${\bullet}OH$ radical through hydrogen atom ($H{\bullet}$) transfer (HAT) and sequential electron (e) proton transfer (SEPT) mechanisms; and (iv) both mechanisms make [6]-gingerol be oxidized to semi-quinone or quinone forms.

  • Using solution technique, sulfur in the sulfide ore was indirectly determined by X-ray fluorescence spectrometry. The sample was dissolved with the mixed solution of B$r_2$ and HN$O_3$, and Si$O_2$, a major constituent, was repelled from the solution by HF treatment several times, B$a^{2+}$ solution was added to the solution to precipitate the S$O^4_{2-}$ ion as BaS$O_4$. Measuring the fluorescent X-ray intensity of excess Ba2+ ion in the filtrate, the content of sulfur in the original ore was back-calculated. Comparing the results by this method with the gravimetric method, the mean difference was ${\pm}1.7%$ in the range of 20 to 40% of sulfur content and the method was tolerably reproducible.

Ammonia Flux from Cow Manure in Relation to the Environmental Factors in Livestock Facilities (축산분뇨에서 발생하는 암모니아 플럭스에 대한 환경인자의 영향에 관한 연구)

  • Ammonia is a very important constituent of the atmospheric environment because it is the most dominant gaseous alkaline species present in the atmosphere. Ammonia is known to affect ecosystems at relatively low concentration. Ammonia flux from livestock facilities can be regulated by a number of environmental factors (pH, ammonium ion, temperature, wind speed, etc). The increases in wind speed above manure from 0.0 m/s to 1.0 7m/s resulted in 2.5 times increases in ammonia flux. Wind speed and ammonia flux showed 0.982. A linear relationship with a correlation coefficient (r=0.982). When manure temperature increased from $3^{\circ}C$ to $36^{\circ}C$, the manure pH decreased approximately 0.30 to 0.46. As wind speed above the manure increased from 0.0 m/s to 1.07 m/s, ammonia flux increased approximately 2.5 times. The increasing manure temperature from $3^{\circ}C$ to $10^{\circ}C$, raised ammonia flux from 2.0 to 3.6 times (2.6 times in average similarly). The increases of manure temperature from $3^{\circ}C$ to $25^{\circ}C$ increased ammonia flux from 5.7 to 12.9 times (8.5 times in average). In this study, the correlation coefficient between ammonia flux and manure temperature was found from 0.972 to 0.989. Results of our research showed that ammonium ion concentration, pH of manure and wind speed were important factors in controlling the ammonia flux from manure livestock facilities.

2-D & 3-D Observations on the microstructures of Super Bainitie TRIP Steels using Total Analysis System (TAS (Total Analysis System)를 이용한 SB-TRIP강에서의 2-D & 3-D 미세구조 분석 연구)

  • It has been widely reported that carbide-free bainitic steels or super-bainite WP (SB-TRIP) steels for the automotive industry are a new family of steels offering a unique combination of high strength and ductility. Hence, it is important to exactly evaluate the volume fraction of RA and to identify the 3-D morphology of constituent phases, because it plays a crucial role in mechanical properties. Recently, as electron back-scattered diffraction (EBSD) equipped with focused ion beam (FIB) has been developed, 3-D EBSD technique for materials science are used to these steels. Moreover, newly developed atom probe tomography (APT) technique can provide the exact distribution and chemical concentration of alloying elements in a sub-nm scale. The APT analysis results indicate exactly the distribution and composition of alloying elements in the austenite and bainite phases of SB-TRIP steels with the atomic-scale resolution. And thus, no partitioning of aluminum and manganese atoms was showed between the austenite containing $7.73{\pm}0.39$ at% C and the bainitic ferrite associated with $0.22{\pm}0.06$ at% C in the SB-TRIP steel.

Inhibitory Effect of Chlorogenic Acid on Low-Density Lipoprotein Oxidation Induced by Cu ion

  • Chlorogenic acid, formed of an ester of caffeic acid and quinic acid, which is naturally abundant in many plant species, was used as a model O-dihydoxy phenolic compound. In the previous study, we have reported that the isolated constituent from Apocynum venetum leaves has an inhibitory effect on $Cu^{2+}$-induced oxidative modification of low-density lipoprotein (LDL). Among them, chlorogenic acid showed the most potent anti-LDL oxidative activity than other compounds. For the reason, we investigated the inhibitory effect of the chlorogenic acid on $Cu^{2+}$-induced oxidative modification of LDL, monitored a lag time in the conjugated-diene formation and TBARS formation, and measured TNBS free amino acid group, and form cell formation in vitro system. The TBARS- and diene- formation were strongly inhibited by chlorogenic acid ($0{\sim}100\;{\mu}g/ml$) with dose dependent manner. On the other hand, TNBS reactive lysine amino groups on LDL oxidation were protected by chlorogenic acid- treated cell group. Therefore, chlorogenic acid inhibited to cholesterol accumulation in the isolated peritoneal macrophage.

A review on the medicinal potentials of ginseng and ginsenosides on cardiovascular diseases

  • Ginseng is widely used for its promising healing and restorative properties as well as for its possible tonic effect in traditional medicine. Nowadays, many studies focus on purified individual ginsenoside, an important constituent in ginseng, and study its specific mechanism of action instead of whole-plant extracts on cardiovascular diseases (CVDs). Of the various ginsenosides, purified ginsenosides such as Rb1, Rg1, Rg3, Rh1, Re, and Rd are the most frequently studied. Although there are many reports on the molecular mechanisms and medical applications of ginsenosides in the treatment of CVDs, many concerns exist in their application. This review discusses current works on the countless pharmacological functions and the potential benefits of ginseng in the area of CVDs. Results: Both in vitro and in vivo results indicate that ginseng has potentially positive effects on heart disease through its various properties including antioxidation, reduced platelet adhesion, vasomotor regulation, improving lipid profiles, and influencing various ion channels. To date, approximately 40 ginsenosides have been identified, and each has a different mechanism of action owing to the differences in chemical structure. This review aims to present comprehensive information on the traditional uses, phytochemistry, and pharmacology of ginseng, especially in the control of hypertension and cardiovascular function. In addition, the review also provides an insight into the opportunities for future research and development on the biological activities of ginseng.
