Prevalence of virulence and cytolethal distending toxin (CDT) genes in thermophilic Campylobacter spp. from dogs and humans in Gyeongnam and Busan, Korea

  • The prevalence of thermophilic Campylobacter (C.) spp. in stray, breeding, and household dogs was 25.2, 12.0, and 8.8%, respectively. C. jejuni and C. upsaliensis were the predominant Campylobacter spp. from household dogs. cdtA, cdtB, and cdtC were detected by PCR in all isolates. Despite the high cytolethal distending toxin (CDT) gene prevalence, only 26 (31%) C. jejuni strains and one (15.3%) C. coli strain showed evidence of CDT production in HEp-2 cell cytotoxicity assays. Virulence-associated genes detected in the C. jejuni and C. coli isolates were cadF, dnaJ, flaA, racR, ciaB, iamA, pldA, virB11, ceuE, and docC. cadF, dnaJ, flaA, and ceuE were found in all C. jejuni and C. coli isolates. When detecting Guillain-Barr$\acute{e}$ syndrome-associated genes (galE, cgtB, and wlaN), galE was identified in all isolates. However, cgtB and wlaN were more prevalent in C. jejuni isolates from humans than those from dogs. Adherence and invasion abilities of the C. jejuni and C. coli strains were tested in INT-407 cells. A considerable correlation (adjusted $R^2$= 0.678) existed between adherence and invasion activities of the Campylobacter spp. isolates.

Histological and Cytological Changes Associated with Susceptible and Resistant Responses of Chili Pepper Root and Stem to Phytophthora capsici Infection

  • Microscopic study of chili pepper (Capsicum annuum L.) infected with Phytophthora capsici, causing Phytophthora blight of chili pepper, was conducted to compare histological and cytological characteristics in the root and stem of susceptible (C. annuum cv. Bugang) and resistant (C. annuum cv. CM334) pepper cultivars. The susceptible pepper roots and stems were extensively penetrated and invaded by the pathogen initially into epidermal cells and later cortical and vascular cells. Host cell walls adjacent to and invaded by the infecting hyphae were partially dissolved and structurally loosened with fine fibrillar materials probably by cell wall-degrading enzymes of the pathogen. In the resistant pepper, the pathogen remained on root epidermal surface at one day after inoculation, embedded and captured in root exudation materials composed of proteins and polysaccharides. Also the pathogen appeared to be blocked in its progression at the early infection stages by thickened middle lamellae. At 3 days after inoculation, the oomycete hyphae were still confined to epidermal cells of the root and at most outer peripheral cortical cells of the stem, resulting from their invasion blocked by wound periderms formed underneath the infection sites and/or cell wall appositions bounding the hyphal protrusions. All of these aspects suggest that limitation of disease development in the resistant pepper may be due to the inhibition of the pathogen penetration, infection, invasion, and colonization by the defense structures such as root exudation materials, thickened middle lamellae, wound peridems and cell wall appositions.

Predicting the Invasion Potential of Pink Muhly (Muhlenbergia capillaris) in South Korea

  • Predictions of suitable habitat areas can provide important information pertaining to the risk assessment and management of alien plants at early stage of their establishment. Here, we predict the invasion potential of Muhlenbergia capillaris (pink muhly) in South Korea using five bioclimatic variables. We adopt four models (generalized linear model, generalized additive model, random forest (RF), and artificial neural network) for projection based on 630 presence and 600 pseudo-absence data points. The RF model yielded the highest performance. The presence probability of M. capillaris was highest within an annual temperature range of 12 to 24℃ and with precipitation from 800 to 1,300 mm. The occurrence of M. capillaris was positively associated with the precipitation of the driest quarter. The projection map showed that suitable areas for M. capillaris are mainly concentrated in the southern coastal regions of South Korea, where temperatures and precipitation are higher than in other regions, especially in the winter season. We can conclude that M. capillaris is not considered to be invasive based on a habitat suitability map. However, there is a possibility that rising temperatures and increasing precipitation levels in winter can accelerate the expansion of this plant on the Korean Peninsula.

Connection the Rhizomicrobiome and Plant MAPK Gene Expression Response to Pathogenic Fusarium oxysporum in Wild and Cultivated Soybean

  • Little known the connections between soybeans mitogen-activated protein kinase (MAPK) gene expression and the rhizomicrobiome upon invasion of the root pathogen Fusarium oxysporum. To address this lack of knowledge, we assessed the rhizomicrobiome and root transcriptome sequencing of wild and cultivated soybean during the invasion of F. oxysporum. Results indicated F. oxysporum infection enriched Bradyrhizobium spp. and Glomus spp. and induced the expression of more MAPKs in the wild soybean than cultivated soybean. MAPK gene expression was positively correlated with Pseudomonadaceae but negatively correlated with Sphingomonadaceae and Glomeraceae in both cultivated and wild soybean. Specifically, correlation profiles revealed that Pseudomonadaceae was especially correlated with the induced expression of GmMAKKK13-2 (Glyma.14G195300) and GmMAPK3-2 (Glyma.12G073000) in wild and cultivated soybean during F. oxysporum invasion. Main fungal group Glomeraceae was positively correlated with GmMAPKKK14-1 (Glyma.18G060900) and negatively correlated with GmRaf6-4 (Glyma.02G215300) in the wild soybean response to pathogen infection; while there were positive correlations between Hypocreaceae and GmMAPK3-2 (Glyma.12G073000) and between Glomeraceae and GmRaf49-3 (Glyma.06G055300) in the wild soybean response, these correlations were strongly negative in the response of cultivated soybean to F. oxysporum. Taken together, MAPKs correlated with different rhizomicrobiomes indicating the host plant modulated by the host self-immune systems in response to F. oxysporum.

화엄늪 식생의 분포특성에 따른 습지 관리방안 (Wetland Management Plan on Distributional Characteristics of Vegetation in Hwaeom Wetland)

  • 본 연구는 화엄늪 습지보호지역에서 식생의 공간적 분포특성을 파악하고 올바른 식생학적 보전 및 복원 방안을 제시하고자 시행하였다. 식생자료는 2008년과 2013년에 Braun-Branquet의 방법을 적용하여 총 117종(미동정종 포함)의 식물종이 포함된 29개가 획득되었다. 식물군락은 총 8개로 구분되었으며, 수분조건에 따라 건생형(신갈나무-애기감둥사초군락, 산철쭉군락-전형하위군락, 참억새하위군락, 미역줄나무군락)과 습생형(진퍼리새군락-전형하위군락, 왕미꾸리꽝이하위군락, 참억새하위군락, 바늘골-끈끈이주걱군락)으로 나누어졌다. 습생입지에 발달하는 습생형은 습지보호지역 내에서 보호의 핵심 대상으로 습지의 함몰된 공간 또는 물골 주변에 주로 발달하였다. 면적은 습생형이 전체의 약 3.8%에 불과하였으며, 습지보호지역 안에서 참억새가 우점하는 건생형이 약 51.5%로 가장 넓게 분포하였다. 습지보호지역 내에 다양한 수목들이 침투해 있었다. 2008년에는 교목성 수종 8종(총 228개체)과 관목성 수종 6종(총 51개체)을 포함하여 총 14종의 침투식물들이 관찰되었으며, 2013년에는 오리나무 2개체가 생육하는 것이 확인되었다. 화엄늪의 지속적인 보전과 관리를 위해서 식생유형, 공간분포 변화 및 침투식물에 대한 지속적인 모니터링, 핵심 완충 전이지역의 구획화 관리 등을 통한 능동적이고 가변적인 설정방안 모색을 제안하였다.

Erwinia rhapontici가 기주식물 조직에서 생산한 Pectate Lyase의 특성 (Characterization of Pectate Lyase Produced by Erwinia rhapontici During Growth in Host Plant Tissue)

  • Erwinia rhapontici causes soft-rot disease in a number of plants such as rhubarb, onion, hyacinth and garlic. Pectate lyase (Pel) depolymerizes pectin and other polygalacturonates, which is though to play a role in bacterial invasion of plants. Pel activity was not detected in E. rhapontici cultured in a minimal salts medium containing glycerol, polygalacturonate, or citrus pectin as a carbon source. However, when sterilized potato tuber and Chinese cabbage slices were added to minimal salts polygalacturonate (0.5%) medium, E. rhapontici produced pectate lyase enzyme. Also Pel activity was consistently detected from macerated potato tubers, Chinese cabbage leaves, lettuce leaves and celery petioles tissue. Pel in the extract of macerated Chinese cabbage caused by E. rhapontici strain 1, resulted in electrolyte loss, tissue maceration and cell death of potato tuber tissue. These results indicate that E. rhapontici produces pectate lyase only in the presence of non-diffusible plant components, and that this enzyme probably contributes to its pathogenicity.

묵밭에서 교란처리구배에 따른 중간교란가설의 검증 (Test of Intermediate Disturbance Hypothesis by Experimental disturbance Gradient in Old-Field Plant Community)

  • In order to test of intermediate disturbance hypothesis that an intermediate level of disturbance maximize biodiversity and to elucidate the effect of disturbance during the early old-field succession, the response of plant community along an experimental disturbance gradient was investigated in a five-year old-field. Response of plant community along an experimental disturbance gradient was connected with light condition because artificial disturbance gradient had been treat by clipping of plants and removing of litter. Artificial disturbance in five-year old-field plant community retarded rate of succession by increasing invasion and performance of the earlier successional species in the initial and middle stage of disturbance treatment. The species richness in the blocks of intermediate disturbance level showed the peak in early and mid-summer. This result did correspond with the prediction of intermediate disturbance hypothesis.

Overcoming Encouragement of Dragon Fruit Plant (Hylocereus undatus) against Stem Brown Spot Disease Caused by Neoscytalidium dimidiatum Using Bacillus subtilis Combined with Sodium Bicarbonate

  • The use of the supernatant from a Bacillus subtilis culture mixed with sodium bicarbonate was explored as a means of controlling stem brown spot disease in dragon fruit plants. In in vitro experiments, the B. subtilis supernatant used with sodium bicarbonate showed a strong inhibition effect on the growth of the fungus, Neoscytalidium dimidiatum, the agent causing stem brown spot disease and was notably effective in preventing fungal invasion of dragon fruit plant. This combination not only directly suppressed the growth of N. dimidiatum, but also indirectly affected the development of the disease by eliciting the dragon-fruit plant's defense response. Substantial levels of the pathogenesis-related proteins, chitinase and glucanase, and the phenylpropanoid biosynthetic pathway enzymes, peroxidase and phenyl alanine ammonia-lyase, were triggered. Significant lignin deposition was also detected in treated cladodes of injured dragon fruit plants in in vivo experiments. In summary, B. subtilis supernatant combined with sodium bicarbonate protected dragon fruit plant loss through stem brown spot disease during plant development in the field through pathogenic fungal inhibition and the induction of defense response mechanisms.

Anti-Invasive and Anti-Angiogenic Effects of Xanthohumol and Its Synthetic Derivatives

  • Invasion and metastasis is the main cause of cancer mortality. Angiogenesis is a prerequisite for the tumor growth and metastasis. Matrix metalloproteinases (MMPs) are the key enzymes playing in the invasive growth and metastasis of cancer as well as angiogenesis. Xanthohumol, a prenylated chalcone of the Hop plant (Humulus lupulus L), has been reported to suppress cancer invasion and angiogenesis. In the present study, we investigated the antiinvasive effects of xanthohumol (1) and its synthetic derivatives, 4'-O-methylxanthohumol SEM ether (2), xanthohumol C (3), and xanthohumol C MOM ether (4) in relation to MMP expression in HT-1080 human fibrosarcoma cells. The compound 1 and its derivative, 3 and 4, significantly inhibited serum-induced HT-1080 cell invasion, and 12-O-tetradecanoylphorbol-13-acetate (TPA)-enhanced activity and expression level of MMP-2 and MMP-9 in a concentration-dependant manner. In addition, they inhibited TPA-enhanced expression of MT1-MMP with relatively weak inhibition in tissue inhibitor of metalloproteinase (TIMP)-1 and TIMP-2 level. The compound 1 significantly decreased the cell viability, whereas the derivatives, 2 and 3 showed no cytotoxicity, and compound 4 showed slight cytotoxicity in the cells. Furthermore, in a chick chorioallantoic membrane (CAM) assay, the derivatives 3 and 4 dose-dependently suppressed vascular endothelial growth factor (VEGF)-induced angiogenesis, which is similar to that of compound 1. Taken together, the results indicate that compounds 3 and 4 may be valuable anti-angiogenic agents in the treatment of chronic diseases such as cancer and inflammation working through suppression of MMP-2 and MMP-9.

경인 아라뱃길의 외래식물 분포 현황 (Distribution on the Alien Plants in the Gyeong-in Ara Waterway, Korea)

  • This study was carried out to investigate distribution on the alien plants in the Gyeong-in Ara Waterway. The alien plants were a total of 82 taxa: 17 families, 63 genera, 80 species, and 2 varieties. This number corresponds to 25.5% of alien plants identified in Korea. The proportion of alien plants in every year was increased from upstream to downstream. As the result of the analysis on vegetation stratification, bank of waterside was covered with artificial materials preventing existence of vegetation, and had step-type cross section. Floodplain was composed of waterfront area. An array of vegetation was not typical dispersion, and terrestrial and alien plants were dominated the Gyeong-in Ara Waterway. Evaluation of naturalness based on the vegetation stratification showed grade 3 or 4. In order to solve a problem, method and level of restoration should be decided based on the result of diagnostic assessment. Therefore, we need to restore the step-type cross section as pool type one. From waterside to bank in this waterway, we recommend to introduce natural plants by imitating reference species composition. Since, an invasion of alien plants is expected to be accelerated due to the continuous artificial disturbance, we recommend to quantitative investigation on the invasion of alien plants and monitoring on the change of distribution.
