  • A morphometric analysis is used to clarify the interspecific relationship between 4 species of the subgenus Parapagaronia in Pagaronia. The quantitative distinctions are confirmed at 87.5% of correct assignment in male and 96.25% in female.

Morphological Relationship between Drosophila melanogaster, D. simulans and their Hybrids (Drosophila melanogaster와 D. simulans 및 그들간 잡종의 형태학적 유연관계)

  • The morphological relationship between Drosophila melanogaster and D. simulans, and their hybrids were investigated using comparisons of internal reproductive organs. male sex comb tooth number and male genital discs. InterspecIfic hybrids of all crosses were completely sterile, and had rudImentary gonads. Dysgenic ovaries and testes were morphologically similar to those of gonadal dysgenesis stenlity associated with the PM system in D. melal1ogaster. The mean number of sex comb tooth in D. melrmogaster, D. simulans and their male hybrids were 10.73, 8.35 and 9.97, respectively. The general aspects of the genital disc of the malo hybrid were a mosaic-like structure between D. melanogaster and D. simulans.

  • Two infectious species of Streptococcosis pathogens were detected by multiplex PCR assay. Detection rates of Streptococcus iniae and S. parauberis could reach 44.9% and 55.1% respectively for one year during 2004 to 2005 in Jeju island. These findings showed that S. parauberis strains were important pathogen with streptococcosis of olive flounder in Jeju island. These findings showed that S. parauberis strains were important pathogen with streptococcosis of olive flounder in Jeiu island. In the present study we have investigated the interspecific relationship of all Jeju area of S. parauberis by RAPD analysis. Represent strains divided to four groups by RAPD fingerprints. The important differences observed between the olive flounder isolates suggest that they could constitute a well-differentiated group or a separate clonal line within this bacterial species. Though, serological research of S. parauberis strains in Jeju island not exist yet. These strains doing the serological evolution.

First Description of Petalonia zosterifolia and Scytosiphon gracilis (Scytosiphonaceae, Phaeophyceae) from Korea with Special Reference to nrDNA ITS Sequence Comparisons

  • Scytosiphonaceae is an acetocarpalean brown algal family, that is a recent focus of synstematics and marine biodiversity. We describe Petalonia zosterifolia and Scytosiphon gracilis from Korea for the first time. P. zosterifolia occurred on the East coast, and had flat, linear and solid thalli. S. gracilis was found in Jeju, and had cylindircal to flat and hol-low thalli. However, these two species are so similar that it is difficult to identify by morphology alone. In order to determine if the nuclear DNA reveals the distinctness of both species and to know their phylogenies, the ITS region sequences were newly detrmined in 22 samples of P. zosterifolia, Scytosiphon gracilis, and other three members of the genera from Korea. We found 0.12% variation among samples of P. zosterifolia from different locations, and no variation between S. gracilis samples from diferent years, but extensive interspecific divergences (13.62-22.83%) of each species to other members in Petalonia and Scytosiphon . The ITS sequence dta consistently showed a close relationship between P. zosterifolia and S. gracilis. This result is congruent with morphology and with the published data of plastid rbc and partial nrDNA large subunit gene sequences, and suggests that P. zosterifolia and S. gracilis might have diverged from the most recent common ancestor.

The NILs from an interspecific cross show enhanced plant height and antioxidant activity

  • A high-resolution physical map targeting a cluster of yield-related QTLs on the long arm of rice chromosome 9 was constructed across a 35.5kb region containing the six predicted genes including the probable ascorbate peroxidase (OsApx). The $BC_3F_6$ near isogenic lines (NILs) were derived from a cross between the Oryza sativa Hwaseong and O. rufipogon. The plant height and length of internodes were compared between Hwaseong and NILs. There were significant differences in plant height between Hwaseong and NILs. The NILs internodes were longer than Hwaseong, showing dramatic elongation in the first and fourth internodes; thereby, leading to increased plant height. The antioxidant activity of Hwaseong and NILs was also analyzed by 3,3-diaminobenzidine (DAB) staining and 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. In order to understand whether or not OsApx gene is important in scavenging $H_2O_2$ in rice, DAB staining was used. Intense dark-brown coloration was observed in Hwaseong than NILs. In addition, DPPH scavenging ability of Hwaseong showed lower value than NILs. These results indicated that the internode elongation and antioxidant activity might possibly be controlled by OsApx. To know the causative relationship of the gene and phenotype, we will further analyze the gene expression and use it for functional studies by complementation transgenic approach.

Taxonomic Review of the Genus Echinochloa in Korea (II): Inferred from Simple Sequence Repeats

  • Echinochloa (L.) P. Beauv. includes some of the noxious weeds, causing a serious yield loss when they are dominant in the fields. Identification of the Echinochloa is very difficult because many interspecific and intraspecific forms of the species are found. However, it is important to identify the species exactly and to know the genetic diversity of the species for effective weed management. This study was conducted to identify and summarize the Echinochloa species by comparing the genetic variation and relationship among Korean Echinochloa species using SSR. The genetic diversity of 107 individuals, including seven species were assessed using five SSR markers. UPGMA dendrogram generated two clades (I and II) and clade II divided again into two subclades (II-1 and II-2) whereas the model based genetic structure proposed four subpopulations. The two subpopulations were corresponded to clades I and II-1 and the other two were arranged to clade II-2 of the UPGMA dendrogram. We have concluded that E. colona and E. glabrescens might have not distributed in Korea. The biological varieties, praticola and echinata, of E. crus-galli should be treated as E. crus-galli. Korean Echinochloa should be summarized with four species, i.e., E. oryzicola, E. crus-galli, E. esculenta, and E. oryzoides.

  • The genus Echinochloa (L.) P. Beauv. comprised of approximately 30-40 species in the tropical and warm temperate regions of the world, including numerous interspecific and intraspecific types which make the genus difficult to identify. As an attempt to identify the species within the genus easier, the taxonomy of the genus Echinochloa, Poaceae in Korea was reviewed on the basis of sequencing data derived from nuclear ribosomal DNA internal transcribed spacer (ITS) and external transcribe spacer and chloroplast DNA trnL intron, trnL-F intergenic spacer and matK regions using a total of 46 accessions representing all the species in Korea. The results of maximum parsimony found separate lineage comprised of E. colona and E. frumentaceae which are not Korean species, but no resolution within Korean Echinochloa species, supporting the suggestion of Yamaguchi group that E. crus-galli, E. oryzoides, and E. esculenta should be considered to belong to the same species. However, the relationship between these three species and the other species, i.e. E. oryzicola should be better understood with more detail studies.

Ecological Niche and Interspecific Competition of Two Frog Species (Pelophylax nigromaculatus and P. chosenicus) in South Korea using the Geographic Information System (지리정보시스템을 이용한 한국산 참개구리와 금개구리의 생태적 지위와 종간 경쟁에 대한 연구)

  • An ecological niche is defined as the specific role of a species influenced by time, space, and other resources. By investigating overlaps between ecological niches of different species, we could estimate the degrees of interspecific competition. Such studies often use geographic information systems (GIS) to discover niche overlaps between species. In this study, we used GIS to estimate the spatial niches of two Korean frog species(Pelophylax nigromaculatus and P. chosenicus). This enabled us to predict their geographic distributions in order to identify their coexistence regions and distribution patterns. The results confirmed that altitude was an important variable for predicting their distribution, with a correlation with their climatic range. Spatial distributions of the two frog species were highly overlapped, as the distribution range for P. nigromaculatus included most of the range of P. chosenicus, showing a sympatric distribution pattern. Within the coexisting regions, however, the presence sites for the two species did not overlap, implying weak competition. To confirm the principal factors influencing their competitive relationship and reasons for their sympatric distribution pattern, we need more detailed in-depth studies on the diverse environmental variables within the regions where the two species coexist. By doing so, we would be able to identify various mechanisms for avoiding competition in sympatric frog species.

Mapping Grain Weight QTL using Near Isogenic Lines from an Interspecific Cross (벼 종간잡종 유래 근동질 유전자계통 이용 종자중 관여 유전자 분석)

  • In previous studies, we reported QTLs for grain weight (GW), qGW3 and for spikelets per panicle (SPP), qSPP3 linked to RM60 on chromosome 3 using advanced backcross lines derived from a cross between Oryza sativa ssp. Indica cv. Milyang 23 and O. glaberrima. The O. glaberrima alleles at this locus increased GW and spikelets per panicle in the Milyang 23 background. To further confirm and narrow down the position of the QTLs on chromosome 3, substitution mapping was performed using five lines containing the target O. glaberrima segment on chromosome 3. The size and position of the O. glaberrima segment on chromosome 3 were different in each line. These lines possessed 3-10 non-target O. glaberrima introgressions in the Milyang 23 background. These five lines were evaluated for seven agronomic traits including 1,000 grain weight and spikelets per panicle and also genotyped with seven SSR markers. Four lines were informative in delimiting the position of QTLs, qGW3 and qSPP3. Two lines with the O. glaberrima segment flanked by SSR markers, RM60 and RM523 displayed significantly higher values than Milyang 23 in GW and SPP whereas two lines without that O. glaberrima segment displayed no difference in GW and SPP compared to Milyang 23. The result indicates that two QTL, qGW3 and qSPP3 are located in the interval between RM60 and RM523 which are 1.2-Mb apart. Introgression lines having QTLs, qGW3 and qSPP3 would be useful materials not only to indentify the relationship between these two yield QTLs, but also to develop high yielding variety via marker-aided selection technology.

Cell Surface Antigenic Relationship of Pathogenic Mycobacteria (병원성 Mycobacteria의 세포표면항원간의 항원적 상관 관계)

  • Cell surface antigenic relationships between pathogenic mycobacteria have been investigated by the enzyme-linked immunosorbent assay using phenolkilled cells and their rabbits antisera. Homologous and heterologous reactions of Mycobacterium avium-intracellulare antisera before and after homologous and heterologous absorption revealed a close antigenic relationship between strains of the same species and between species if they were members of M. avium(MA)-intracellulare(MI)-scrofulaceum(MG) complex. MAI sera showed a considerable reaction with M. kansasii(MK) and tuberculosis(MTB), but not with the other species. MA(K40004) antiserum reacted with other mycobacteria except few strains of MI and 50~89% of homologous reaction was reduced by heterologous absorption with cells of MI or MS. Intraspecific reaction of MI antisera was natural1y stronger than interspecific reaction and different in extent due to a magnitude of antigenic sharing. Antigenic relationships between N-260D, N-260R, N-260T, and K41014 was somewhat closer than that with N-242D, N-257T, N-28ID, and N-275T. M. nonchromogenicum(MNC) antisera showed a strong interspecific reaction with exception of M. chelonei(MC) and triviale(MTV) to which they reacted weakly or none. Antigenic sharing with M. terrae(MTR) and MG(K30003) was next to intraspecific sharing. NC-3 shared antigens considerably with MA, MC, and M. fortuitum(MF) while NC-11 did not. MTR antisera showed a strong cross-reaction with MI but their homologous reaction was not reduced by MI absorption indicating a paucity of shared antigen of MTR surface. Intraspecific antigenic sharing of course was large with on exception between T-8 and T-13. A considerable amount of antigenic sharing was also found with MNC, MC and MF. Unlike T-8 serum, T-13 antiserum strongly cross-reacted with MA, MG, MK, and MTB. In general, antigenic relationships of mycobacteria, that have been elucidated in this study, well conformed to taxons delineated by the various biological and biochemical means.
