Chromosome Analysis Using GISH and FISH of Interspecific Hybrids between Allium cepa L. and A. fistulosum L. (GISH와 FISH를 이용한 양파와 파간 종간교잡계통의 염색체 분석)

  • Fluorescence in situ hybridization (FISH) and genomic in situ hybridization (GISH) were used for chromosome analysis of hybrids (2n=16) between onion (Allium cepa L., 2n=2X=16) and welsh onion (A. fistulosum L., 2n=2X=16). 5S rDNA, 45S rDNA, and tandemly repeated DNA (TSD) sequence were used as probes for FISH analysis. A. fistulosum specific DNA probe of telomeric repeats and A. fistulosum DNA were used for GISH analysis. In the analysis of meiotic chromosome GISH revealed that hybrids have 7 bivalants and 2 univalents chromosome and 2 univalents were derived from A. fistulosum chromosomes. In somatic chromosomes of hybrid each 8 chromosomes were derived from A. cepa and A. fistulosum, respectively. FISH signal of 45S rDNA probe in A. fistulosum was detected at secondary constriction of chromosomes, while FISH signal in A. cepa was observed in both secondary constriction and telomere of chromosomes. TDS signals in A. fistulosum chromosomes were detected at all subtelomeric of 8 chromosomes and also in 2 pericentromeric of the chromosomes, whereas TDS signals in A. cepa were observed only in subtelomeric in all chromosomes. The pattern of TDS signal in hybrid chromosomes was similar to those of A. fistulosum chromosomes.

  • Interspecific crossing was conducted to obtain hybrid lilly with better quality of adapting to the unfavorable environment using oriental Lilium spp. as a female parent. Average interspecific crossing rate between oriental and asiatic lines by cut-style pollination was as low as 29%. Although the average germination rate of zygotic hybrid embryo between oriental 'Rodolfa' ${\times}$ asiatic 'Toronto' was increased from 76.6% to 78.3%. on 114 or 1/6 strength MS medium compared to the control, the rate of zygotic embryos formation between oriental 'Farolito' ${\times}$ asiatic 'Buff Pixie' was significantly enhanced up about 86~90% on 1/2 strength MS medium. Meanwhile, germination rate of interspecific hybrid embryo between oriental 'Snow Cristal' ${\times}$ asiatic 'Royal Trinity' showed up 100%. Germination rate of interspecific hybrids slightly increased by addition of $0.5-1.0mg{\cdot}L^{-1}$ GA. Germination rate of zygotic embryos between oriental 'Farolito' ${\times}$ asiatic 'Buff Pixie' and oriental 'Belcanto' ${\times}$ L. callosum was highest on the MS medium containing $0.01mg{\cdot}L^{-1}$ NAA and $0.1mg{\cdot}L^{-1}$ kinetin, as 66.6% and 45.2% respectively.

Characterization of Xylanase from an Hybird between Aspergillus oryzae var. oryzae and Aspergillus Nidulans 514 by Nuclear Transfer (핵전이에 의한 Aspergillus oryzae var. Oryzae와 Aspergillus nidulans 514의 잡종으로부터 생산된 Xylanase의 특성)

  • Interspecific hybrids between Aspergillus oryzae var oryzae and A. nidulans 514 were obtained by nuclear transfer technique. Several autotrophic mutants isolated from conidiospores of the two strains were mutagenized with ultraviolet and N-methyl-N-nitrosoguanidine. Optimal conditions for formation of intergeneric hybrids were investigated. Frequencies of hybrid formation by nuclear transfer were $3{\times}10^{-5}{\sim}1{\times}10^{-5}$. From observation of genetic stability, conidial size, DNA content, and nuclear stain, it was suggested that their karyptypes are aneuploid. The hybrids showed 1.1~1.4 fold higher xylanase activities than parental strains did. The xylanase of Aspergiilus sp. TAVD514-3 was purified and some of it's enzymatc characteristics were investigated. The enzyme was purified about 85 fold with an overall yield of 17% from the culture medium by ammonium sulfate fractionation, Sephadex G-75 gel permeation chromatography, and CM-sephadex A-50 ion exchange chromatography. The purified enzyme functions optimally at pH 9.0 and 80$^{\circ}C$. The enzymatic activity was increased by the presence of $Mg^{2+}$ and $Mn^2$ ions.

Studies on the Interspecific and Intergeneric Hybridization in Herbage Grasses II. Morphological physiological charateristics of Italian ryegrass X tall fescue hybrid (화본과목초의 종.속간 잡종에 관한 연구 II. Italian ryegrass X tall fescue F1 잡종의 형태 및 생리적 특성)

  • Tall fescue(Festuca amndinacea Schreb.) is a productive pasture grass that is widely adapted to the soil and climate of Korea. It is regarded, however, as relatively low in palatability and nutritive value. On the other hand, Italian ryegrass(lo1ium rnultz$om Lam.) is highly nutritious and palatable, but is essentially weak in cold tolerance. Numerous investigators have succeeded in producing hybrids between Lolium spp. and Festuca spp. but no improved varieties have resulted. Therefore, intergeneric hybridization of Italian ryegrass with tall fescue is employed in 1984 to improve the adaptive and cold resistance quality of Italian ryegrass. In the mean time, F1-hybrid plants obtained by crossing Italian ryegrass x tall fescue were observed in open field. The results are summarized as follows: 1. Leaf-size and silica cell hair were intermediate between the types of parents but the shape of auricle tended to show Italian ryegrass and auricle-cilia to show tall fescue. 2. Roots of $F_1$-hybrid plants were mainly distributed at top soil and did not produced florescence material. 3. Head type of hybrids was intermediate between the types of parents but ray length was somewhat less than the length of tall fescue ray. Typical Italian ryegrass or tall fescue-like phenotypes were not observed. 4. Anthers of $F_1$-hybrid plants were formed but pollen were not formed. Even though pollen were formed, they were all empty. 5. Hybrid plants were more winterhardy and summer persistency than Italian ryegrass. They showed a equivalent growth to Italian ryegrass in spring and to tall fescue in summer. 6. Chromosome of $F_1$-hybrids was allopolyploid(2n=4~=28) and all $F_1$-hybrid plants were sterile.

Development of Lipase Hyper-producing Strain from Hybrids between Aspergillus niger and Penicillium notatum by Nuclear Transfer (핵전이에 의한 Aspergillus niger와 Penicillium notatum 잡종에서의 lipase 고생산 균주의 개발)

  • Interspecific hybrids between Aspergillus niger and Penicillium notatum (Tyr-), hyperlipolytic enzyme-producing fungi, were obtained by nuclear transfer technique. Optimal conditions for formation of intergeneric hybrids were investigated. Maximum production of protoplasts was obtained by 1% Novozyme 234 at $30^{\circ}C$ for 3 hrs and the most effective osmotic stabilizers for the isolation of protoplasts were 0.6 M KCl. Frequencies of hybrid formation by nuclear transfer were $3.8{\times}10^{-3}{\sim}1.3{\times}10^{-3}$. From the observation of genetic stability, conidial size, DNA content, and nuclear stain, it was suggested that their karyotypes are aneuploid. The hybrids showed $1.2{\sim}1.7$ fold higher lipase activities than parental strains. It was strongly supported by results of this study that nuclear transfer technique is much more efficient in the formation of intergeneric hybrids than protoplast fusion and is very useful for the improvement of strains.

  • The green marine algae, Capsosiphon fulvescens has been cultivated in south coast of southern Korea for many years on a commercial scale. This species is very popular in Korean as a food supplement because of its attractive flavor and flexcible taste. It is, therefore, necessary to isolate and utilize qualified germplasms for mass production of this economic seaweed. Several reports have been published on phycological applications of RAPDs including the characterization of interspecific genetic variation, the identification of isolates and hybrids, and the study of phylogenetic relationships. However few authors have used RAPDs to assess the genetic variability among populations of a seaweed species(van Oppen et al., 1994; Alberto et al., 1997). The present study was undertaken for characterizing the identities of Capsosiphon fulvescens populations cultivating in Korea through the analysis of PCR based random amplified polymorphic DNAs (Welsh and MacClelland, 1990; Willams et al., 1990) and chemical composition aimed to isolate the useful strains for aquaculture. Chemotaxonomic Studies on the Citrus Plants cultivated in Je Ju Island (제주도산 감귤속 식물의 성분 분류학적 연구)

  • A thin-layer chromatographic study was made of the chloroform-soluble and flavonoid fractions from the fruit peels of 16 species, 2 varieties and 5 formas of the Citrus plants cultivated in Je Ju Island for their interspecific relatinships. In addition, 3 hybrids and 9 native plants were also studied for their taxonomic position. Three phenograms were developed from these chromatographic data after cluster analysis via the unweighted paired group method using rithmatic average by Sneath and Sokal. These plants were grouped into 5 alliences based on the phenogram obtained from the chloroform-soluble fracitons, which were nearly identical to the subgenus rank by Tanaka, and rutinoside and neohesperidoside groups by Horowitz. Those from the flavonoid and methanol-soluble fractions were not able to evaluate the morphological classification except for a few cases.

Early Life History Characteristics of an Induced Hybrid of Rhodeus ocellatus and Acheilognathus yamatsutae (흰줄납줄개(Rhodeus ocellatus)와 줄납자루(Acheilognathus yamatsutae) 잡종의 초기생활사 특징)

  • This study was conducted to identify the taxonomic differences in the characteristics of Rhodeus ocellatus and Acheilognathus yamatsutae during their early life history via an interspecific hybridization experiment. Results showed that the hatching rates of the cross-bred eggs were 60% for cross OY (R. ocellatus ♀×A. yamatsutae ♂) and 70% for cross YO (A. yamatsutae ♀×R. ocellatus ♂). The dorsal fin rays of YO were similar to those of R. ocellatus. The hybridization test of A. yamatsutae and R. ocellatus living in the same water stream revealed that no interspecies reproductive isolation occurred. OY and YO showed similar maternal and paternal characteristic. The juvenile of OY did not resemble both paternal and maternal fin rays, indicating a difference. The hatching larvae of the hybrids showed the morphological characteristics of their parents but varied in terms of the shapes of their parts.

  • This study aimed to investigate taxonomic differences in the early life history characteristics of Rhodeus ocellatus and Rhodeus notatus through an interspecific hybridization experiment. The hatching rates of the cross-bred eggs were 50% for the ON cross (R. ocellatus ♀×R. notatus♂) and 67% for the NO cross (R. notatus♀×R. ocellatus♂). Interestingly, the hatching time of the hybrids was longer than that of their parents. The newly hatched larvae exhibited distinct differences in their physical characteristics. The head of the adult fish protruded forward more than that of the ON cross. The pterygoid process was 45° for the ON cross and 90° for the NO cross. Furthermore, the tail shape of the ON and NO crosses closely resembled that of R. notatus.

Variation of Nuclear DNA Content in Interspecific Allium cepa L.×A. fistulosum L. hybrids and Their Successive Backcross Lines (양파와 파간의 종간잡종 F1과 여교잡계통의 핵 DNA 함량)

  • Interspecific hybrid plants between Allium cepa L. (2n=2X=16) and A. fistulosum L. (2n=2X=16)and their backcross lines were developed by artificial pollination in order to introduce new desirable characters of A, cepa to A. fistulosum. The 2C nuclear DNA content has been estimated by flow cytometry in 5 Allium fistulosum inbreed lines, 2 interspecific hybrid lines of A. cepa${\times}$A. fistulosum and 34 their backcross lines $BC_1F_1$ to $BC_2F_2$, using propidium iodide (PI) as a fluorescence dye. Estimated 2C DNA values ranged from 22.2 pg to 23.7 pg in 5 A. fistulosum inbreed lines, 37.9 pg in F1 hybrid between A. cepa and A. fistulosum, 24.3 pg to 27.3 pg in 7 backcross lines in $BC_1F_1$, 21.9 pg to 24.4 pg in 9 $BC_1F_2$, 22.9 pg to 25.1 pg in 14 $BC_2F_1$, 22.6 pg to 23.4 pg in 4 $BC_2F_2$. This study showed mean 2C nuclear DNA content of $F_1$ hybrid was higher than their backcross progeny lines, while it was lower than female parental line, A. cepa (2C DNA=33.2 pg). Mean 2C DNA content of backcross lines, $BC_1F_1$ to $BC_2F_2$ was not significantly different but their 2C DNA contents in the more progress generation from $BC_1F_1$ to $BC_2F_2$ were reduced.
