정서화담강기탕(定瑞化痰降氣湯)이 Xylene으로 유발된 마우스의 급성 염증에 미치는 영향 (Anti-Inflammatory Effect of 'Jungcheonhwadamgangki-tang'in Xylene-Application Mouse Ear Acute Inflammation Model)

  • In the present study, the anti-inflammatory effect of 'Jungcheonhwadamgangki -tang' water extract was tested in Xylene-Application mouse ear acute inflammation model. The test articles were once dosed before Xylene-Application, and the changes on body weight and ear weights and histopathological observation of induced ear were conducted with ear histomorphometry. The increases of absolute and relative ear weight detected in vehicle control compared to that of sham , were significantly and dose-dependently inhibited by Jungcheonhwadarn -gangki-tang in the present study. A classic acute inflammatory histological changes such as subcutaneous edema, hypertrophy and infiltration of inflammatory cells, was detected in vehicle control. However, these histological changes were significantly and dose-dependently inhibited by Jungcheonhwadam-gangki-tang. In addition, the increases of ear thickness half and thickness full detected in the vehicle control, were also dose-dependently decreased in the all Jungcheonhwadamgangki-tang-dosing groups. Base on these results, it is concluded that Jungcheonhwadamgangki-tang water extracts have clear anti-inflammatory effect on the acute inflammation, and about 500 mg/kg of Jungcheonhwadamgangki-tang water extracts have similar effect compared to that of Dicrofenac 15 mg/kg.

마우스를 이용한 십전대보탕(十全大補湯) 발효물의 단회투여 독성 연구 (Single Oral Dose Toxicity Test of Fermented Sipjeondaebo-tang Extracts in Mice)

  • Objectives : Sipjeondaebo-tang is a medicine traditionally prescribed as a restorative. The aim of this study was to investigate the single oral dose toxicity and safety of extract of fermented Sipjeondaebo-tang in ICR mice. Methods : In single oral dose toxicity study, non-fermented or fermented Sipjeondaebo-tang were administered by oral gavage to ICR mice (5 males, 5 females) at single doses of varying concentrations: 1250, 2500 and 5000 mg/kg. Changes of body weight, general behavior, adverse effects and mortality were determined throughout the experimental period. Hematological parameters, organ weights and necropsy findings were evaluated at the end of the experiment. Results : There were no mortality or signs of toxicity in single oral dose toxicity studies. There were also no significant differences in body weight, organ weight, or hematological parameters between the treatment and control groups. Conclusions : Fermented Sipjeondaebo-tang did not cause remarkable adverse effects in ICR mice. The oral lethal dose of fermented Sipjeondaebo-tang is more than 5000 mg/kg and no-observed-adverse-effect level (NOAEL) of the extract for both male and female mice is 5000 mg/kg.

Determination of indoor doses and excess lifetime cancer risks caused by building materials containing natural radionuclides in Malaysia

  • The activity concentrations of $^{226}Ra$, $^{232}Th$, and $^{40}K$ from 102 building materials samples were determined using a high-purity germanium (HPGe) detector. The activity concentrations were evaluated for possible radiological hazards to the human health. The excess lifetime cancer risks (ELCR) were also estimated, and the average values were recorded as $0.42{\pm}0.24{\times}10^{-3}$, $3.22{\pm}1.83{\times}10^{-3}$, and $3.65{\pm}1.85{\times}10^{-3}$ for outdoor, indoor, and total ELCR respectively. The activity concentrations were further subjected to RESRAD-BUILD computer code to evaluate the long-term radiation exposure to a dweller. The indoor doses were assessed from zero up to 70 years. The simulation results were $92{\pm}59$, $689{\pm}566$, and $782{\pm}569{\mu}Sv\;y^{-1}$ for indoor external, internal, and total effective dose equivalent (TEDE) respectively. The results reported were all below the recommended maximum values. Therefore, the radiological hazards attributed to building materials under study are negligible.

간경변 환자에서 콜히친 사용에 의한 호중구성 백혈구감소증 1예 (A Case of Colchicine-Induced Neutropenia in Liver Cirrhosis Patient)

  • Colchicine is an alkaloid that has been used for treating acute gouty arthritis, psoriasis, scleroderma and Behcet's syndrome. Colchicine decreased liver fibrosis in rats with carbon tetrachloride induced cirrhosis and in patients with many liver diseases. Therapeutic oral doses of colchicine may cause nausea, vomiting, abdominal pain and diarrhea. The adverse effect of colchicine associated with the dose is bone marrow suppression, and especially neutopenia. Neutropenia has often been reported in patients have taken an overdose of colchicine. We describe a 64-year-old female liver cirrhosis patient with neutropenia that was induced by a therapeutic dose of colchicine.

보중익기탕가미방(補中益氣湯加味方)에 의한 비만(肥滿) 세포(細胞) 매개성(媒介性) 즉각형(卽刻型) 알레르기 반응(反應)의 억제(抑制) (Inhibition of mast cell-mediated immediate-type allergic reactions by Bojungikgitanggamibang)

  • Objective : Mast cells are a potent source of mediators that regulate inflammatory response in allergies and asthma. The author studied the effect of Bojungikgitanggamibang(BITB) on mast cell-mediated anaphylactic reaction. Method : When BITB was given as pre-treatment at concentrations ranging from 0.01 to 1 mg/ml, the histamine release from rat peritoneal mast cells induced by compound 48/80 was reduced in a dose-dependent manner. Result : BITB dose-dependently inhibited compound 48/80-induced systemic anaphylactic shock. BITB also inhibited passive cutaneous anaphylaxis activated by anti-dinitrophenyl IgE. In addition, BITB inhibited phorbol 12-myristate 13-acetate and A23187-induced interleukin-6 secretion from human mast cell line HMC-1 cells. Conclusion : These results indicate that BITB may be actively anti-allergic.

호흡에 의해 내부 움직임을 갖는 장기에 전달되는 선량에서 Time Gating Threshold(TGT)의 효과 (An effect of time gating threshold (TGT) on a delivered dose in internal organ with movement due to respiration)

  • 본 연구는 호흡시에 내부 장기가 움직일 때 시간에 따른 threshold 값을 주어졌을 때 선량분포에 대한 연구를 수행한 것이다. 이전 연구에서 보고 된 것처럼 일정 시간에 따라 움직이는 내부 장기의 움직임은 Rujan 등에 의해 보고된 3차원적 수학적 계산방법에 의해 장기의 위치를 나타내었다. 그 결과 처음exhale에서 1초동안 간의 움직임은 2mm이내에 위치하는 것을 알게 되었다. 그래서 이 연구에서 TGT는 간의 움직임이 가장 적은 처음 exhale에서 1초동안 움직일 때의 선량분포를 평가하였다. TGT 값을 주었을 때 선량분포를 비교하기 위해 다음 조건으로 방사선을 조사하였다. 1) threshold 범위에서 target이 움직일 때(1초, 1.5호), 2) threshold 없이 target이 움직일 때, 3) target이 움직이지 않을때. 각각 조건의 선량분포를 비교 평가하였다

Evaluating internal exposure due to intake of 131I at a nuclear medicine centre of Dhaka using bioassay methods

  • Handling of radioisotopes may cause external and internal contamination to occupational workers while using radiation for medical purposes. This research aims to monitor the internal hazard of occupational workers who handle 131I. Two methods are used: in vivo or direct method and in vitro or indirect method. The in vivo or direct method was performed by assessing thyroid intake with a thyroid uptake monitoring machine. The in vitro or indirect method was performed by assessing urine samples with the help of a gamma-ray spectroscopy practice using a High-Purity Germanium (HPGe) Detector. In this study, fifty-nine thyroid counts and fifty-nine urine samples were collected from seven occupational workers who were in charge of 131I at the National Institute of Nuclear Medicine and Allied Sciences (NINMAS), Dhaka. The result showed that the average annual effective dose of seven workforces from thyroid counts were 0.0208 mSv/y, 0.0180 mSv/y, 0.0135 mSv/y, 0.0169 m Sv/y, 0.0072 mSv/y, 0.0181 mSv/y, 0.0164 mSv/y and in urine samples 0.0832 mSv/y, 0.0770 mSv/y, 0.0732 mSv/y, 0.0693 mSv/y, 0.0715 mSv/y, 0.0662 mSv/y, 0.0708 mSv/y.The total annual effective dose (in vivo and in vitro method) was found among seven workers in average 0.1039 mSv/y, 0.0950 mSv/y, 0.0868 mSv/y, 0.0862 mSv/y, 0.0787 mSv/y, 0.0843 mSv/y, 0.0872 mSv/y. Following the rules of the International Commission on Radiological Protection (ICRP), the annual limit of effective dose for occupational exposure is 20 mSv per year and the finding values from this research work are lesser than this safety boundary.

행인(杏仁)의 랫트 단회 경구 투여 독성시험 (Rat Single Oral Dose Toxicity Test of Armeniacae Semen (Including Endocarp))

  • Objectives : The object of this study was to obtain acute toxicity information (single oral dose toxicity) of Armeniacae Semen (AS), a medicinal herb used for treating constipation and various respiratory diseases, in rats. Methods : In order to observe the $LD_{50}$ (50% lethal dose), approximate lethal dosage (ALD) and target organs, AS aqueous extracts were orally administered once to female and male Sprague Dawley rats at dose levels of 2,000, 1,000, 500 and 0 (control) mg/kg (body weight). The mortality, changes in the body weight, clinical signs and gross observation were monitored for 14 days after single oral treatment of AS extracts, and the organ weights and histopathological findings of principle organs were observed after sacrifice. Results : After single oral treatment of AS 2,000 and 1,000 mg/kg, all (5/5; 100%) female and male rats died within 30 minutes after treatment, while no mortalities were observed in the female and male rats treated with 500 mg/kg of AS extract. Therefore, $LD_{50}$ in female and male rats was calculated as 741.95 mg/kg. Seizure, loss of locomotion, salivation, increases of respiration and heart-beat were observed after AS extract treatment, which were observed in all rats including the lowest dosage group, 500 mg/kg in the present study. In addition, lung congestion was visible in all mortal rats with AS 2,000 and 1,000 mg/kg, respectively. Conclusions : The results obtained in this study suggest that AS extract ranges in Class III, because the $LD_{50}$ and ALD in both female and male rats were calculated as 741.95 mg/kg and 500~1,000 mg/kg, respectively. However, AS extract should be carefully treated at clinical applications, because salivation, increase of respiration and heart-beat were also observed in the lowest dosage group, 500 mg/kg in the present study.

Administrative dose control for occupationally-exposed workers in Korean nuclear power plants

  • Korean nuclear power plants (NPPs) have various radiation protection programs to attain radiation exposure as low as reasonably achievable (ALARA). In terms of ALARA, this paper provides a comprehensive overview of administrative dose control for occupationally-exposed workers in Korean NPPs. In addition to dose limits, administrative dose constraints are implemented to resolve an inequity of radiation exposure in which some individuals in NPPs receive relatively higher doses than others. Occupational dose constraints in Korean NPPs are presented in this paper with the background of how those values were determined. For pressurized water reactors, 80% and 90% of the annual average limit for an effective dose, 20 mSv/y, are set as the primary and secondary dose constraints, respectively. Pressurized heavy water reactors (PHWRs) have also established the primary and secondary dose constraints corresponding to 70% and 80% of the effective dose limit, and additional constraints for tritium concentration are provided to control internal exposure in PHWRs. Follow-up measures for exceeding these administrative dose constraints are also introduced compared to exceeding the dose limits. Finally, analysis results of dose distributions show how the implementation of administrative dose constraints impacted the occupational dose distributions in Korean NPPs during the years 2009-2018.
