Intake, digestibility, and rumen and metabolic characteristics of cattle fed low-quality tropical forage and supplemented with nitrogen and different levels of starch

  • Objective: Effects of nitrogen supplementation associated with different levels of starch on voluntary intake, digestibility, and rumen and metabolic characteristics of cattle fed low-quality tropical forage (Brachiaria decumbens hay, 7.4% crude protein, CP) were evaluated using ruminal and abomasal cannulated steers. Methods: Five European${\times}$Zebu young bulls (186 kg body weight, BW) were distributed according to a $5{\times}5$ Latin square. The following treatments were evaluated: control, supplementation with 300 g CP/d (0:1), supplementation with 300 g starch/d and 300 g CP/d (1:1), supplementation with 600 g starch/d and 300 g CP/d (2:1), and supplementation with 900 g starch/d and 300 g CP/d (3:1). A mixture of nitrogenous compounds provided 1/3 from true protein (casein) and 2/3 from non-protein nitrogen (mixture of urea and ammonium sulphate, 9:1) was used as the nitrogen supplement. In order to supply energy a unique source of corn starch was used. Results: Supplements increased (p<0.05) dry matter intake, but did not affect (p>0.05) forage intake. There was a cubic effect (p<0.05) of starch on voluntary intake. This was attributed to the highest forage intake (g/kg BW) when using the 2:1 starch:CP ratio. Supplements increased (p<0.05) organic matter (OM) digestibility, but did not affect (p>0.05) neutral detergent fibre corrected for ash and protein (NDFap) digestibility. There was a positive linear effect (p<0.05) of the amount of starch supplemented on OM digestibility. Total NDFap digestibility was not affected (p>0.05) by the amount of supplemental starch. Ruminal ammonia nitrogen concentrations were higher (p<0.05) in supplemented animals, however, a negative linear effect (p<0.05) of amount of starch was observed. Supplements increased (p<0.05) the nitrogen balance (NB) and efficiency of nitrogen utilization. These effects were attributed to increased body anabolism, supported by higher (p<0.05) serum concentration of insulin-like growth factor 1. Increasing the amount of starch tended (p<0.06) to linearly increase the NB. In spite of this, there was a highest NB value for the 2:1 starch:CP ratio amongst the treatments with supplementation. Conclusion: Nitrogen supplementation in cattle fed low-quality tropical forage increases nitrogen retention in the animal's body. An additional supply of starch increases nitrogen retention by increasing energy availability for both rumen and animal metabolism.

MiR-126-3p inhibits apoptosis and promotes proliferation by targeting phosphatidylinositol 3-kinase regulatory subunit 2 in porcine ovarian granulosa cells

  • Objective: Numerous studies have indicated that the apoptosis and proliferation of granulosa cells (GCs) are closely related to the normal growth and development of follicles and ovaries. Previous evidence has suggested that miR-126-3p might get involved in the apoptosis and proliferation of GCs, and phosphatidylinositol 3-kinase regulatory subunit 2 (PIK3R2) gene has been predicted as one target of miR-126-3p. However, the molecular regulation of miR-126-3p on PIK3R2 and the effects of PIK3R2 on porcine GCs apoptosis and proliferation remain virtually unexplored. Methods: In this study, using porcine GCs as a cellular model, luciferase report assay, mutation and deletion were applied to verify the targeting relationship between miR-126-3p and PIK3R2. Annexin-V/PI staining and 5-ethynyl-2'-deoxyuridine assay were applied to explore the effect of PIK3R2 on GCs apoptosis and proliferation, respectively. Real-time quantitative polymerase chain reaction and Western Blot were applied to explore the regulation of miR-126-3p on PIK3R2 expression. Results: We found that miR-126-3p targeted at PIK3R2 and inhibited its mRNA and protein expression. Knockdown of PIK3R2 significantly inhibited the apoptosis and promoted the proliferation of porcine GCs, and significantly down-regulated the mRNA expression of several key genes of PI3K pathway such as insulin-like growth factor 1 receptor (IGF1R), insulin receptor (INSR), pyruvate dehydrogenase kinase 1 (PDK1), and serine/threonine kinase 1 (AKT1). Conclusion: MiR-126-3p might target and inhibit the mRNA and protein expressions of PIK3R2, thereby inhibiting GC apoptosis and promoting GC proliferation by down-regulating several key genes of the PI3K pathway, IGF1R, INSR, PDK1, and AKT1. These findings would provide great insight into further exploring the molecular regulation of miR-126-3p and PIK3R2 on the functions of GCs during the folliculogenesis in female mammals.

Clinical Significance of Serum PDGF-BB and IGF-1 in Coal Workers' Pneumoconiosis (석탄광부 진폐증 환자에서 PDGF-BB, IGF-1의 의의)

  • Background : Coal workers' pneumoconiosis(CWP) is a fibrotic lung disease resulting from the chronic inhalation of coal dust. Various cytokines and growth factors secreted from macrophages and monocytes play a key role in the pathogenesis of pneumoconiosis. The platelet-derived growth factor (PDGF)-BB and the insulin-like growth factor(IGF)-1 secreated from the macrophages and monocytes are believed to stimulate the accumulation of mesenchymal cells and fibrosis of the lower respiratory tract that is observed in fibrotic lung disease. The serum concentraion of PDGF-BB and IGF-1 in 30 CWP patients and 10 healthy controls were measured in order to determine if PDGF-BB and IGF-1 can be used as sensitive biomarkers in CWP. Method : Serum was collected from 30 patients with CWP(13 with simple CWP and 17 with complicated CWP) and 10 healthy controls. The serum concentrations of PDGF-BB and IGF-1 were measured using ELISA (R&D system, Minneapolis, MN). Results : The serum PDGF-BB concentration in patients with complicated CWP($10083.76{\pm}639.07pg/mL$) was significantly higher than in the patients with simple CWP ($8493.88{\pm}848.51pg/mL$) and the healthy controls ($3726.17{\pm}292.20pg/mL$) (p<0.05). Compared to the healthy controls ($413.40{\pm}1.94ng/mL$), there was no significant difference in the serum IGF-1 concentration in patients with simple ($366.77{\pm}183.67ng/mL$) and complicated CWP ($403.18{\pm}15.39ng/mL$) (p>0.05). Conclusion : These results show the important role of the PDGF-BB mediated pathways in the pathogenesis of CWP. These data suggests that the PDGF-BB serum concentration is a useful biomarkers of the fibrotic extent in CWP patients.

Effects of Formula (JR-22) Maybe Containing Traditional Herbs on Maximal Exercise Performance and Antioxidant Meterials in Murine Model (생약조성물 투여가 지구력 향상과 항산화 물질에 미치는 영향)

  • The effects of dietary supplementation of JR-22, formula containing distilled extracts of traditional herbs on maximal exercise performance and endurance were evaluated in mouse and rat model. In acute forced swimming test with 4%∼8% of the body weight attached to the tail, it was shown that dietary JR-22 supplementation increased endurance in exercise performance. There was no change of blood lactic acid, ammonia, inorganic phosphorous ion and creatine kinase activity, however ATP concentration in muscle was increased by JR-22 supplementation. Also, insulin-like growth factor-l (IGF-1) concentration in blood was significantly increased by JR-22 supplementation. In addition, the oxidative damage induced by exercise was reduced by JR-22 supplementation. In these results, we suggested that JR-22 supplementation enhanced maximal endurance exercise performance by the mechanism of increasing ATP and IGF-1 concentration and reducing oxidative damage.

  • Superovulation with exogenous gonadotropins creates a spectrum of pre or periovulatory hormonal changes with subsequent detrimental effects on oocyte quality, fertilization, embryo development, implantation and maintenance of pregnancy. Our recent study determined potential roles for insulin-like growth factor-1 (IGF-1) in uterine environment regulation and preimplant tation in the rat. The evidence indicates that IGF-l may play an important role in the main tenance of a receptive uterine environment for embryonic development and the regulation of decidualization. Embryonic loss and failure of implantations following superovulation may be partially attributed to disturbances in uterine IGF-l action as observed in this study. We investigated the effects of superovulatory doses of gonadotropins on frequency of chromosomal a abnormalities of mouse embryos. Chromosome a analysis of mouse zygotes and 8- to 16-cell stage embryos from spontaneously ovulated, 5, 10, and l 15 lU pregnant mare serum gonadotropin (PMSG) superovulated mice was carried out. Aneuploidy, polyploidy and structural chrom- osomal abnormalities were detected among the four groups. However, only polyploidy was correlated with superovulation. In 10 and 15 IV PMSG treated groups, the rate of polypoidy was 2.9% and 10.5%, respectively.   • A hallmark of cancers is 'leaky' tight junctions (Tjs). TJs mediated paracellular permeability is elevated and TJs maintained cell polarity is frequently lost. Concomitantly, TJs-associated proteins including members of the claudin family of proteins are dysregulated. Recent findings indicate that these TJs changes can contribute to cancer progression. In this study, we examined the effects of ${\beta}-lapachone$, a quinone compound obtained from the bark of the lapacho tree (Tabebuia avellanedae), on the Tjs-associated regulators in human hepatocarcinoma cell lines, HepG2 and Hep3B. ${\beta}-lapachone$ treatment downregulated the levels of insulin-like growth factor 1 receptor (IGF-lR) proteins in both HepG2 and Hep3B cells. But the levels of claudin-3 and -4 proteins were increased in ${\beta}-lapachone$-treated HepG2 and Hep3B cells. And also the zonnula occludens-l (la-I) and p-catenin protein levels by ${\beta}-lapachone$ were increased in a time-dependent manner. However, claudin-3 and -4 mRNA levels were uninhibited by ${\beta}-lapachone$ in HepG2 and Hep3B. The present results suggest that the upregulation of claudin-3 and -4 protein levels by ${\beta}-lapachone$ occurs by a post-transcriptional mechanism and points to a novel mechanism by ${\beta}-lapachone$.

Osteogenic Response of Human Osteoblasts Derived from Mandible and Maxilla: A Preliminary Study (상, 하악골 유래 조골세포의 골형성 능: 일차 연구)

  • Purpose: Maxilla and mandible have different patterns of cortical and trabecular bone and different bone mineral densities, even though both are components of the jaw bone. However, cellular differences between maxilla- and mandible derived osteoblasts (OBs) have rarely been studied. We hypothesize that maxilla- and mandible-derived OBs show different responses to $17{\beta}$-estradiol (E2), which is one of the critical factors for bone formation. This study compares skeletal site-specific cell responses between maxilla- and mandible-derived human OBs to E2. Methods: Maxilla- and mandible-derived OBs derived from an identical donor were separately isolated from a total of five normal healthy subjects aged 18~44 years old, cultured with a treatment of 100 nM estrogen. The responses between maxilla- and mandible-derived OBs to E2 were evaluated and compared using cell proliferation, alkaline phosphatase (ALP) activity and gene expression of osteoprotegerin (OPG), ALP, insulin-like growth factor-1 (IGF-1), and estrogen receptor ${\alpha}$ ($ER{\alpha}$). Results: E2 did not have any distinct effects on the proliferation of both types of OBs. Mandible-derived OBs exhibited higher ALP activity than maxilla-derived OBs in the non-treated condition, which was common in all tested individuals. ALP activities of both types of OBs showed a minor increasing tendency with the treatment of E2, even though there was no statistical significance in some specimens. The gene expression of OB by E2 was diverse, depending on the individuals. There was increased expression of OPG, IGF-1, or $ER{\alpha}$ gene in the part of subjects, which was more repeated in maxilla-derived OBs. In particular, OPG or ALP induction by E appeared less frequently in mandible-derived OBs. Conclusion: Current results revealed that E2 affects maxilla- and mandible-derived OBs into facilitating the osteogenic process despite individual differences. Mandible-derived OBs are less sensitive to bone-forming gene expression by E2.

  • 제2형 당뇨병은 대사성 질환으로 간, 근육 그리고 지방 조직 세포에서 인슐린 작용의 장애로 나타나는 인슐린 저항성으로 혈당의 이용이 감소하여 혈당이 높아짐에도 불구하고 췌장의 베타세포에서 인슐린 분비가 충분하지 못할 때 유발된다. 서구에서는 비만 등으로 인해 인슐린 저항성이 증가하면 인슐린 분비가 높은 고인슐린혈증을 나타내어 당뇨병으로의 진전은 늦다. 하지만 우리나라를 비롯한 아시아의 사람들은 인슐린 저항성이 증가할 때 인슐린 분비가 충분치 못해 혈청 인슐린 농도가 정상인과 비슷하거나 더 낮은 상태에서 당뇨병으로 진전된다. 이러한 차이는 우리나라를 비롯한 아시아 사람들에게서 제2형 당뇨병의 발생이 급격하게 증가할 것이라는 보고되었다. 결국 당뇨병은 간, 근육 및 지방조직에서의 인슐린 작용의 장애와 췌장의 베타세포에서 인슐린 분비의 부족의 복합적인 장애에 의해서 나타나고 이것은 공통적으로 각 조직에서의 인슐린/insulin growth factor (IGF)-1 신호전달의 장애와 관련이 있다. 베타세포에서의 인슐린분비 자체는 인슐린/IGF-1 신호전달과 관계가 없지만 간접적으로 관련이 있다. 인슐린 분비능은 베타세포의 증식과 생존에 의한 베타세포의 양과 밀접한 관련이 있는데 인슐린/IGF-1 신호전달은 베타세포의 증식과 생존을 조절한다. 그러므로 혈당 조절에 관여하는 기능성 식품은 인슐린 작용을 향상시키는 인슐린 민감성 특성을 가지거나, 혈당이 높아질 때 인슐린 분비를 촉진시키는 insulinotropic 작용을 하는 성질을 가지고 있어야 하겠다. 전자의 대표적인 약은 1999년에 미국 FDA에서 승인 받은 peroxisome proliferator-activated receptor $(PPAR)-{\gamma}$ agonist 인 thiazolidinedione 계통의 약물인 troglitazone, pioglitazone, rosiglitazone 등이 있고, 후자는 2007년에 승인 받은 Exenatide는 glucagon like peptide (GLP)-1 agonist이다. 이 두 가지 약은 모두 자연계에 존재하는 동식물에서 유래된 것으로 식품에도 많이 다양한 종류의 인슐린 민감성 물질이나 insulinotropic 작용을 하는 물질이 함유되어 있을 것이다. 이러한 기능 이외에 혈당조절 약이나 식품으로 사용되는 것은 탄수화물의 소화를 방해하는 것으로 탄수화물 소화효소인 a-amylase 또는 maltase의 활성을 억제하여 식후 혈당의 급격한 상승을 방지하는 것이 있다. 우리나라 사람들은 탄수화물의 섭취가 너무 많아서 실제로 이러한 식품이나 약의 효능이 높지 않을 것이다. 혈당을 조절하는 기능성 식품은 이 세 가지 효능 중 일부를 가지고 있는 것이 될 수 있다. 이러한 기능을 스크리닝하기 위해서 3가지 단계를 거쳐야 한다. 먼저 시험관에서 또는 세포 실험을 통해서 앞서 언급한 3가지 기능을 가지고 있는 지 여부를 각각 조사한다. 이중에서 효과가 있는 것은 당뇨 동물 모델을 사용하여 in vivo에서 혈당 강하기능과 혈당 강하기전을 조사하는 실험을 한다. 효과가 있는 식품이 우리가 전통적으로 식품으로 섭취해 왔다면 독성 검사를 거쳐야 할 필요가 없지만 한약재이거나 특수 식품의 경우에는 in vivo 실험 전에 GLP 기관에서 반드시 독성 실험을 거쳐 독성 유무를 확인할 필요가 있다. 동물 실험에서 효과적인 것은 인체 실험을 거쳐 혈당 조절 기능성 식품으로 식약청에서 허가를 받을 수 있겠다. 결론적으로 식품에는 항당뇨 특성을 가진 물질들이 함유되어 있는 것들이 상당히 많다. 혈당 조절기능이 있는 기능성 식품으로 개발할 때 고려해야 할 것은 1) 그 양이 혈당 강하 기능성 식품으로 지정받을 수 있을 정도로 충분히 함유되어 있느냐, 2) 혈당을 강하시키는 기전이 단순히 당의 배설을 촉진시켜서 혈당을 저하시키는 것이 아니라, 인슐린 작용을 촉진시키거나, 포도당 자극에 의한 인슐린 분비를 촉진시키거나 탄수화물의 소화 흡수를 억제시킴으로 혈당을 강하시키는 지 등을 파악하는 것이다. 이러한 조건을 만족시키는 식품은 지속적으로 섭취할 때 당뇨병을 예방하거나 진전을 지연시킬 수 있는 혈당조절기능이 있는 기능성 식품으로 개발 가능성이 있겠다.

Role of the Nuclear Transcription Factor NF-κB Caused by Acute Hypoxia in the Heart (급성 저산소증 상태에서 심장 내 전사인자 NF-κB의 기능)

  • Purpose : Nuclear ($factor-{\kappa}BNF-{\kappa}B$) is now recognized as playing a potential role in programmed cell death and the adaptive response to various stress. Cellular hypoxia is a primary manifestation of many cardiovascular diseases. It seems that vascular endothelial growth factor (VEGF) and insulin like growth factor-I(IGF-I) have a function as a protective molecule in the heart against several stress including hypoxia. In this study, the role of $NF-{\kappa}B$ to the cellular response and regulation of protective molecules against the acute hypoxia in the heart was studied. Methods : To cause acute hypoxic stress to the heart, Sprague Dawley rats were exposed to hypoxic chamer($N_2$ 92% and $O_2$ 8%). After the hypoxic exposure, nuclear proteins, total proteins and mRNA were isolated from heart. Translocation of the transcription factors $NF-{\kappa}B$, NF-ATc, AP-1 and NKX-2.5 were evaluated by electrophoretic mobility shift assay(EMSA). The expression of IGF-I and VEGF were studied before and after the hypoxic stress by competitive-PCR, Northern hybridization and Western hybridization. To confirm the role of the $NF-{\kappa}B$ in the heart, the rats also were pretreated with diethyl-dithiocarbamic acid(DDTC) into peritoneal cavity to block $NF-{\kappa}B$ translocation into nucleus. Results : The expression of $NF-{\kappa}B$, AP-1 and NF-ATc were increased by the hypoxic stress. Increased expression of the VEGF and IGF-I were also observed by the hypoxic stress. However, the blocking of the $NF-{\kappa}B$ translocation reduced those expressions of VEGF and IGF-I. Conclusion : These results suggest that $NF-{\kappa}B$ has a protective role against the acute hypoxia through several gene expression, especially VEGF and IGF-I in heart muscle.

Methylation Pattern of H19 Gene at Various Preimplantation Development Stages of In Vitro Fertilized and Cloned Porcine Embryos

  • Insulin-like growth factor II (IGF2) and H19 genes are mutually imprinted genes which may be responsible for abnormalities in the cloned fetuses and offspring. This study was performed to identify putative differentially methylated regions (DMRs) of porcine H19 locus and to explore its genomic imprinting in in vitro fertilized (IVF) and somatic cell nuclear transferred (SCNT) embryos. Based on mice genomic data, we identified DMRs on H19 and found porcine H19 DMRs that included three CTCF binding sites. Methylation patterns in IVF and SCNT embryos at the 2-, 4-, $8{\sim}16$-cells and blastocyst stages were analyzed by BS (Bisulfite Sequencing)-PCR. The CpGs in CTCF1 was significantly unmethylated in the 2-cell stage IVF embryos. However, the 4- (29.1%) and $8{\sim}16$-cell (68.2%) and blastocyst (48.2%) stages showed higher methylation levels (p<0.01). On the other hand, SCNT embryos were unmethylayted ($0{\sim}2%$) at all stages of development. The CpGs in CTCF2 showed almost unmethylation levels at the 2-,4- and $8{\sim}16$-cell and blastocyst stages of development in both IVF ($0{\sim}14.1%$) and SCNT ($0{\sim}6.4%$) embryos. At all stages of development, CTCF3 was unmethylated in IVF ($0{\sim}17.3%$) and SCNT ($0{\sim}1.2%$) embryos except at the blastocyst stage (54.5%) of IVF embryos. In conclusion, porcine SCNT embryos showed an aberrant methylation pattern comprised to IVF embryos. Therefore, we suggest that the aberrant methylation pattern of H19 loci may be a reason for increased abnormal fetus after embryo transfer of porcine SCNT embryos.
