Conversion of water-insoluble components of the basidiocarps of ganoderma lucidum to water-soluble components by hydrolyzing with chitinase

  • We investigated the optimum conditions for conversion of water-insoluble components of basidiocarps of Ganoderma lucidum to water-soluble components by hydrolyzing with chitinase. We also tried it with Ganoderma luciclum residue remaining after extracting hot water-soluble components of Ganoderma lucidum. After hydrolyzing under optimum conditions (20 ppm chitinase, 2% Ganoderma lucidum or 6% Ganoderma lucidum residue, at pH 3 and at $ 35^{\circ}C$), the contents of total water-soluble components (polysaccharide or protein) were measured, and it was found that the contents of water-soluble components increased to 1.5-2.7 fold. Michaelis constant, $K_m$ and maximum rate, $V_max$ calculated by Lineweaver-Burk plot for hydrolysis of Ganoderma lucidum were 1.75% and 0.02%/min respectively and those for hydrolysis of Ganoderma lucidum residue were 53.15% and 0.53%/min respectively The protein-bound polysaccharide was isolated after hydrolysis and molecular weights were measured by Sepharose CL-4B gel filtration and compared with the molecular weights of polysaccharide before hydrolysis.

국내산 대두의 불용성 물질을 포함한 두유 요구르트의 발효 및 품질 특성 (Fermentation and Quality Characteristics of Soy Yogurt incorporating Insoluble Components of Domestic Soybeans)

  • The objective of this research was to determine the fermentation and quality properties of soy yogurt incorporating insoluble components fermented with Streptococcus thermophilus (S. thermophilus) and Lactobacillus bulgaricus (L. bulgaricus) for 24h. Boiled soybeans were ground and reconstituted into equivalent total solids (18%). After soymilk was homogenized with sugar, it was pasteurized in a water bath at $90^{\circ}C$ for 30 min. Two kinds of lactic acid bacteria were inoculated into two types of soymilk after cooling down $40{\sim}45^{\circ}C$ and fermentation at $37^{\circ}C$ for 24h. Titrable acidity, pH, viscosity, sugar content, and number of viable cells were determined in triplicate. Soy yogurt fermented with L. bulgaricus did not reach pH 4.5, where as S. thermophilus was considered good for achieving pH 4.08 and titratable acidity of 2.27% for 24 h. Soy yogurt fermented with S. thermophilus had a higher viscosity and lower sugar contents than that fermented with L. bulgaricusas incubation time increased. Total viable count was $1.80{\times}10^{10}CFU/mL$ on S. thermophilus and $2.16{\times}10^8CFU/mL$ on L. bulgaricus after 24 h at $37^{\circ}C$. However, there was no significant difference in sensory intensities and preference between the two samples. As a result, S. thermophilus was identified as a better culture than L. bulgaricus for the manufacture of soy yogurt incorporating insoluble components.

부산지역 강하먼지와 불용성 성분의 침적량에 관한 연구 (A Study on Bulk Deposition Flux of Dustfall and Insoluble Components in Pusan, Korea)

  • Dustfall particles were collected by the modified American dust jar (wide inlet bottle type) at 6 sampling sites in Pusan area from March, 1999 to February, 2000. Thirteen chemical species (Al, Ca, Cd, Cr, Cu, Fe, K, Mg, Mn, Ni, Pb, Si, and Zn) were analyzed by AAS and ICP. The purposes of this study were to estimate qualitatively various bulk deposition flux of dustfall and insoluble components by applying regional and seasonal distribution. Dustfall amount of regional variations were found in order of coastal zone, industrial zone, commercial zone, agricultural zone and residential zone, and seasonal total dustfall had higher concentrations during spring for 6.741 ton/${km}^2$/season, lower concentrations during summer for 1.989 ton/${km}^2$/season, and annual total concentration was 17.742 ton/${km}^2$/year. The regional distributions of enrichment factor show well-defined anthropogenic metals (Cd, Cu, Pb, and Zn) at industrial and agricultural zone, and contribution rate of soil particles were found in order of summer, fall, winter and spring. Factor loading effects of chemical composition of dustfall were found in order of road traffic emission source and combustion processed source, industrial activity source, soil source and marine source.

녹용의 약효 성분에 관한 연구(VI) 녹용의 지용성 성분 및 Pantocrin이 흰쥐 척수 신경의 Aldolase 활성에 미치는 영향 (Biochemical Studies on Antler (Cervus nippon taiouanus) (VI) Comparative Study on the Effect of Lipid Soluble Fractions of Antler Sponge and Velvet Layers and Pantocrin on the Aldolase Activity in the Rat Spinal Nerves)

  • In the present study, in attempt was made to observe the effect of lipid components in pantocrin and antler velvet and sponge layers extracts on the aldolase activity in the rat spinal nerves. The antler chloroform-methanol (2:1) extracts are known to be composed of neutral lipids, cholesterol, cerebrosides, spingomyelin, phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl serine and gangliosides. The antler extracts were studied on the aldolase activity in the rat spinal nerves. The aldolase activity was measured by the method of Lehninger. To investigate the components of tile extracts which affect the enzyme activity, the chloroform-methanol (2:1) extracts of antler were fractionated into petroleum ether soluble and insoluble fractions and their effects on the enzyme activity were compared. It was found that the chloroform-methanol (2:1) extracts of the antler sponge and velvet layers as well as those petroleum ether soluble fractions obtained from the chloroform-methanol (2:1) extracts, decrease the aldolase activity by 18-23%. However, pantocrin showed increasing effect on the aldolase activity by 42.6%. The petroleum ether insoluble fraction of the chloroform-methanol (2:1) -extract showed no significant increasing activity (about 13%) on the spinal nerves aldolase. The components of pantocrin ind the petroleum ether insoluble fractions were attempted to analysize by thin layer (silica gel) and gas liquid chromatography.

바람의 강도에 따른 강하먼지와 불용성 성분의 조성특성 (A study on bulk deposition flux of dustfall and insoluble components by the wind intensity in Busan, Korea)

  • Weather elements were observed by the AWS (Automatic Weather System) and dustfall particles were collected by the modified American dust jar (wide inlet bottle type) at 4 sampling sites in Busan area from March. 1999 to February, 2000. Thirteen chemical species (Al, Ca, Cd, Cr, Cu, Fe, K, Mg, Mn, Ni, Pb, Si, and Zn) were analyzed by AAS and ICP. The purposes of this study were to estimate qualitatively various bulk deposition flux of dustfall and insoluble components by applying regional and seasonal wind intensity. Frequency of wind speed were found in order of low(1-3m/s), very low(<1m/s), medium(3-8m/s) and high(>8m/s), and annual mean had higher range at low(1-3m/s) for 56.3%. Strong negative linear correlation were observed between dustfall and wind direction (northeastern and eastern), but strong positive linear correlation were observed between dustfall and wind direction (western and northwestern) at industrial, commercial and coastal zone(p<0.05). While a negative correlation were observed between wind speed frequency of very low(<1 m/s) and dustfall, and positive correlation were observed between wind speed frequency of low(1-3m/s) and dustfall in coastal zone(p<0.05). The correlation coefficient was observed 0.556 between wind speed frequency of low(1-3m/s) and Ni by commercial zone(p<0.05). The correlation coeffcient show well-defined insoluble trace metals (Al, Ca, Cr, Cu, Fe, Pb, and Zn) and wind speed frequency of low(1-3m/s) at coastal zone, which was found significant difference(p<0.01).

강수에 의한 대기 에어로졸 입자의 세정특성 (Characterization of Atmospheric Aerosols Scavenged by Rain Water)

  • In order to study the scavenging mechanism as a final goal, the characteristics of chemical components in the rain water were examined as a function of the amount of rainfall. The rain drops were collected sequentially with a rainfall interval amount of each 0.1 .sim. 0.5 mm from the beginning of rain. Rain water was separated into the soluble and insoluble components and the concentrations of 15 elements in both components were determined by the PIXE analysis. The elemental cencentrations decreased quickly till about 0.3 .sim. 0.5 mm of rainfall was obtained and then decreased gradually afterward. Fe, Ti, and Si in the aerosol particles caught in rain water were in high insoluble state. In contrast, almost whole of S and Cl were dissolved in rain water.

Experimental Studies on Wet Scavenging of Atmospheric Aerosols by Rain Drops

  • Wet scavenging by rain drops is a most important removal process of air pollutants. In order to study the scavenging mechanisms of aerosol particles, the characteristics of chemical components in the rain water were examined as a function of the amount of rainfall. Rain water were collected continuously and separated into the soluble and insoluble components. The elemental concentrations in both components were determined by a PIXE analysis. The physical and chemical characteristics of atmospheric aerosols during the rainfall events were measured simultaneously. The elemental concentrations in rain water decreased substantially just after rain started and then gradually declined in subsequential rain fall exceeding 1.0 mm. The large particles were scavenged more easily than the fine particles. Fe, Ti and Si in rain water were in high insoluble state. Contrarily, almost whole of S was dissolved in rain water.

조리방법에 따른 채소의 불용성 식이섬유 함량 변화에 관한 연구 (Effect of Gooking on Water Insoluble Dietary Fiber in Vegetables)

  • Recently, interests of dietary fiber associated with critical physiological effects have been rising in Korea physiological effect in the body. In the present study, the contents of fiber components in 15 kinds of vegetables being consumed commonly in Korea were investigated, and the effects of various treatments (cooking and Kimchi fermentation) on fiber were studied. The results are summarized as follows. Fiber contents of vegetables were 11.8∼31.9% of neutral detergent fiber(NDF), total insoluble dietary fiber, 10.9∼25.4% of acid detergent fiber(ADF), 8.8∼23.8% of cellulose, 0.6∼10.6% of hemicellulose and 1.0∼5.2% of lignin, on dry weight basis. Especially, peppers had higher contents of NDF than the other vegetables. In the vegetables used in the present study, it was found that a great portion of NDF, total insoluble dietary fiber, was composed of cellulose because cellulose covered 63% of NDF. 'Cooking increased the NDF, ADF and cellulose contents, and most change was due to the change of cellulose. The values of hemicellulose and lignin showed an Irregular pattern upon cooking. Fermentation slightly increased NDF, ADF and cellulose, while hemicellulose and lignin showed irregular pattern.

  • Because yeast is rich in vitamins, nucleotides and essential amino acids, it has been used as a nutrient and dietary supplements. Particularly yeast extract functions as a seasoning, flavor enhancer in meats. In this study, naturally occurring insoluble components during yeast extract process by autolysis were microanalyses by using SEM, XRD, HPLC, GC, FT-lR and DSC. On the basis of useful datas, authors have characterized the analytical points for the purpose of improving products in the preparation of yeast extract.

  • A study on the effects of the root components of Angelica koreana Max. on voluntary activity in mice has confirmed that: 1. MeOH soluble Ex., MeOH insoluble Ex., oxypucedanine and isoimperatorin were antagonized against the voluntary activity in mice pretreated with a dose of C.N.B. 30mg/kg. 2. MeOH insoluble Ex. showed higher depressant effect than MeOH soluble Ex.. 3. the depressant activity of oxypucedanine was significantly more effective than that of isoimperatorin.
