번식률 향상을 위한 무인 발정발현 관찰시스템 구현 (Implementation of unmanned cow estrus detection system for improving impregnation rate)

  • 논문에서는 무인 발정발현 관찰을 통해 다두 사육 시 발생하는 공태를 줄이고, 정확한 수정적기 판단을 통해 번식률을 향상하고자 하였다. 무리 생활을 하는 소의 특성상 운동량의 증가 유무만으로는 발정 발현의 판단이 어렵기 때문에, 소의 운동량 수집 센서와 개체 정보 종합 관리 프로그램을 개발하여 무인 발정발현 관찰시스템을 구축하고 운동량 정보와 번식 정보를 이용하여 발정 발현 관찰 알고리즘을 개발하였다. 시스템과 알고리즘에 대한 성능 검증은 수정 전 시스템을 통해 수정적기를 관찰하고, 수정하고 21일 후 재발정 여부를 확인, 다음 21일 후 초음파 진단을 통해 수태여부를 확인하여 적절하게 수정적기를 판단하였는지 검증하였고, 이를 규모가 유사한 실제 축산 농가 4곳에서 수행하였다. 각 농가의 총 사육 두수는 87, 81, 93, 82두이며 관찰된 발정 예정인 소는 14, 19, 15, 17두 이다. 이중 미약발정에 해당하는 3, 2, 1, 3두는 관찰에 실패하였으나 정상발정에 해당하는 11, 17, 14, 14두의 발정을 감지하였고, 인공 수정 후 10(91%), 17(100%), 13(93%), 14(100%)두의 소가 수태하여 제안된 무인 발정발현 관찰 시스템이 정상발정 시 수정적기 판단에 유효함을 보였다.

영상처리기법을 이용한 스톨 사육 모돈의 인공수정적기 예측 장치 개발 (Development of a Device for Estimating the Optimal Artificial Insemination Time of Individually Stalled Sows Using Image Processing)

  • The goal of this study was to develop an optimal artificial insemination time estimator(OAITE) for individually stalled sows using image processing and to evaluate the performance of the OAITE through field test. The OAITE consisted of a computer, a multiplexer, three CCD cameras and three LED lamps (950nm wavelength). The computer program used for the OAITE to quantify the lying and non-lying (sitting and standing) rates of sows in stalls was written in LabWindows/CVI. For the purpose of establishing references that would help estimate the optimal artificial insemination(AI) time for sows, the lying rate of the 50 Berkshire⨯Hampshire crossbred sows(parity: 2 to 7) was observed and recorded. The observation was made from the second day after the sows were moved into the stalls when they were artificially inseminated. The results of above process, which compared the lying rates of the day of estrus and the other days, showed that there were no significant differences at the following time bands: 00, 08, 09, 16, and 17(p>0.1). Thus, only the time bands other than these time bands were used to establish the references for determining the onset of the estrus. Based on the lying rates observed and the references established by the procedures above, the study assigned “0” to the lying rate of the non-estrus time band, “0.5” to the lying rate between the non-estrus and estrus time bands and “1” to the lying rate of the estrus time band. The authors of the study assumed that if the OAITE produced “0.5” or above more than 4 times in a row and if the results included “1” at least once, the estrus would have started. In addition, it was assumed that the optimal AI time for sows was between the 26th hour and the 34th hour after the beginning of estrus. The results of sows’ AI of the OAITE group(n=40 sows; AI=1 time) showed that the pregnancy rate was 92.5%, which was the same rate as the control group(n=40 sows; AI=2 times), and that the litter size did not differ between the control and the OAITE group. These data suggest that the OAITE might be effective and economic to estimate the optimal AI time of individually stalled sows.

Estrus synchronization and artificial insemination in Korean black goat (Capra hircus coreanae) using frozen-thawed semen

  • Presently, there is an increased demand for livestock products all over the world which has led to more devotion on improving livestock population. Although goats have been bred for a long time in Korea, but there is not much research conducted on traditional Korean black goat (Capra hircus coreanae) compared to other livestock populations. Mutton consumption has been dramatically changing from medicinal use to edible meat and this trend directs the black goat populations declining and also mutton import quantities are increasing consistently. The present study introduced a new estrus synchronizing technique with subsequent artificial insemination (AI) for Korean black goats to enable crossbreeding with non-native breeds for the small or subsistent farmers. Our data highlighted that, the percentage of motile sperm from the electro-ejaculated samples declined significantly after freezing and melting. In addition, the sperm motility significantly declined with regard to sperm incubation period (0, 5, 60, and 120 min at 37℃) and was negatively correlated (64.2 ± 7.9%, 63.3 ± 5.8%, 49.9 ± 6.3%, and 35.9 ± 7.6%, respectively) in frozen-thawed sperm samples. Moreover, the E2 levels were unchanged even 24 h after controlled internal drug releas (CIDR) withdrawal. But, 48 h and 72 h after CIDR removal, E2 levels increased significantly. These data helps us to consider the two time points for AI; CIDR removal after 24 h, at which E2 decreases, and after 48 h, as the time at which progesterone increases. Additionally, the AI after 48 h of CIDR removal group exhibited significantly higher pregnancy and parturition rates (42.9%) compared to AI after 24 h after CIDR removal 28.6% group. In conclusion, these studies will propose an optimal estrus synchronisation process with subsequent timing of AI and also will promote the Korean black goat breeding industry.

TC-199액내에서 소 미성숙난자의 정자침입 (Sperm Penetration of Bovine Immature Oocytes in TC-199 Medium)

  • Bovine immature oocytes cultured for various times in TC-199 medium were inseminated with frozne-thawed spermatozoa in TC-199 medium supplemented with caffeine(5mM) and heparin(10$\mu\textrm{g}$/ml). Sperm penetraton was possible in oocytes at any stage of maturation, but penetration rates were lower in oocytes inseminated 0~16h (60~76%) than 20h (98%) after culture. Formation of male and female pronuclei were first observed in oocytes inseminated 8h after cultrue. Formation of male and female pronuclei were first observed in oocytes inseminated 8h after culture. The proportions of polyspermy were high(50~76%) in oocytes inseminated at any stage of maturation. Sperm penetration into oocytes at the GV stage started at 8h after insemination and the penetration rates gradually increased as time after insemination proceeds. The proportion(35%) of oocytes matured beyond metaphase-II 20h after sperm-oocytes incubation was low. When oocytes were incubated without spermatozoa in TC-199 medium, maturation rates were significantly higher (P<0.001) in those without(45 and 84% for 16 and 20 h) than with (0 and 36% for 16 and 20 h) caffeine and heparin. These results indicate that TC-199 medium with caffeine and heparin is not suitable for maturation and fertilization of immature oocytes and may inhibit male pronuclear formation in the cytoplasm.

한우 수란우의 임신율에 대한 hCG 영향과 혈장 요소태질소 수준과의 관계 (Relationship of Plasma Urea Nitrogen Level and Influence of hCG on Pregnancy Rates in Hanwoo Recipients)

  • This study was undertaken to test the hypothesis the hypothesis that treatment with hCG (5,000 IU) at the time of embryo transfer would enhance pregnancy rates in recipients, and the concentration of plasma urea nitrogen(PUN) in recipients was related to the effect of hCG on the reproductive performance. Blood samples were taken according to experimental condition for the assessment of the endogenous plasma progesterone concentration and plasma urea nitrogen. Concentrations of progesterone in plasma were higher in cows treated with hCG on day 7(estrus=day 0) than in those untreated during 7∼43 days after insemination. The pregnancy rates were 65.5 and 54.6% for the hCG treated and untreated groups, respectively. In recipient group categorized with PUN concentration of <12 mg/이, the pregnancy rates were 68.8 and 46.7% for the hCG treated and untreated groups, respectively. The results suggest that hCG treatment at 7 days after insemination could be used to increase the pregnancy rate of embryo transfer, and transfer, and only the recipients with PUN concentration of <12 mg/dl were influenced by treatment with hCG.

젖소에서 CIDR 투여에 의한 발정 유도 후 수태율과 다른 인자와의 관계 (Relationship between the Conception Rate after Estrus Induction using CIDR and Other Parameters in Dairy Cows)

  • The purpose of this study was to determine the relationship between conception rate and other parameters (body condition score; BCS, progesterone concentrations and follicle size) before estrus induction with CIDR(intravaginal progesterone-releasing controlled internal drug release). The conception rate in cows with < 2.75, 2.75 to 3.25, and 3.25 <, BCS regardless of AI (artificial insemination) time was 46.6%, 63.3%, and 46.6% at CIDR insertion, respectively. The conception rate regardless of BCS was 54.9% in cows inseminated based on detected estrus, and 48.7% in cows inseminated at 72 to 80 hours (timed artificial insemination, TAI) after removal of CIDR. The conception rate regardless of AI time was 40.0% in cows with low progesterone concentrations (less than 1.0 ng/ml), and 56.6% in cows with high progesterone concentrations (more than 1.0 ng/ml) at CIDR injection. The conception rate regardless of progesterone concentrations was 53.8% in cows inseminated based on detected estrus, and 38.0% in cows of TAI after removal of CIDR. The conception rate regardless of AI time was 43.3% in cows with small follicle (less than 5 mm), 53.3% in cows between 5 mm to 10 mm of follicle, and 63.3% in cows with large folliclc (more than 10 mm) at CIDR injection, respectively. The conception rate regardless of follicle size was 58.4% in cows inseminated based on detected estrus, and 45.9% in cows of TAI after removal of CIDR. These results indicated that if the cows with BCS 2.75 to 3.25, active corpus luteum, and/or large dominant follicle (more than 10 mm) are used for estrus induction, the conception rate will be greater.

  • As part of the study on the development of the setting time estimation system, this study attempted to confirm the change in hardness values for each influencing factor variable and secure its reliability. According to the research results of this paper, the hardness value of the setting time estimation system tended to gradually decrease in the case of the hardness value of the closing time by curing temperature, and the hardness value increased in the concrete state compared to mortar. Therefore, further research on influencing factors will be conducted in consideration of material and statistical factors in the future.

정액의 희석배율 및 주입 정자수가 닭의 수정능력에 미치는 영향

  • 보존온도별(5, 25, 35$^{\circ}C$)로 정액과 희석액의 비율(1:1, 1:2, 1:3)을 달리하여 보존시간(1, 3, 6시간)에 따른 정자운동성을 조사한바, 5$^{\circ}C$ 냉장 온도에서는 희석배율에 따른 보존시간별 정자운동성은 차이가 없었으나, $25^{\circ}C$ 상온에서 6시간, 35$^{\circ}C$ 고온에서 3시간 이상 보존할 때 정액과 희석액의 비율이 1:1보다는 1:2 비율에서 정자운동성이 현저히 높게 유지되었다(P<0.05). 온도가 높을수록 1:1 배율보다는 1:2 이상 희석배율을 높여 주었을 때 정자의 운동성을 높게 유지시킬 수 있었다. Skim milk glucose액을 사용하여 닭 정액을 희석한 후 인공수정 하였을 때, 1회 주입정자수를 0.2, 0.4, 1 및 2억으로 하였을 때 각각 90.67, 94.00, 96.00 및 98.67%의 수정율을 나타냈다. 90%이상의 수정율을 얻기 위해서는 0.2억 이상의 정자 주입이 필요하며, 94%이상 안정적인 수정율을 얻기 위해서는 0.4억의 정자가 필요하다고 판단된다.

Natural vs synchronized estrus: determinants of successful pregnancy in ewes using frozen-thawed Suffolk semen

  • The pregnancy rate in indigenous ewes inseminated with frozen-thawed Suffolk semen following natural and synchronized estrus was determined. The serum Progesterone and Estrogen concentration and vaginal electrical resistance (VER) of ewes at the time of Artificial Insemination (AI) were observed as successful pregnancy determinants. 21 healthy ewes were selected for this experiment during January-April, 2017. 10 ewes were inseminated in natural estrus. Whereas, 11 ewes were inseminated after estrus synchronization using intravaginal sponges containing 60 mg medroxyprogesterone acetate. Trans-cervical Al (TCAI) was performed in all ewes within 12-16 hours of observed heat. Prostaglandin E1 analogue impregnated vaginal sponge was used for cervical relaxation 6-8 hours before insemination. Pregnancy was diagnosed through trans-abdominal ultrasonography after 40 days of AI. The pregnancy rate of ewes in synchronized estrus was higher (54.5%) than in natural estrus (30%). Higher serum Progesterone level (0.90 ± 0.02 ng/mL) and significantly (p < 0.001) lower VER (257.78 ± 10.11 ohm) were observed at the time of AI in ewes becoming pregnant. Results suggest that higher Progesterone concentration and lower VER could be considered as pregnancy indicators. Oestrous synchronization could be implemented to increase the pregnancy rate in ewes.

Effects of sperm insemination on the final meiotic maturation of mouse oocytes arrested at metaphase I after in vitro maturation

  • Objective: The aims of this study were to investigate whether fertilization could induce the resumption of meiosis in mouse oocytes arrested at metaphase I (MI) after in vitro maturation (IVM), and to investigate the effect of $Ca^{2+}$ chelator treatment at the time of fertilization on the transition from MI to metaphase II (MII). Methods: MII-stage and arrested MI-stage mouse oocytes after IVM were fertilized, and then embryonic development was monitored. Blastocysts from each group were transferred into 2.5 days post-coitum pseudo-pregnant ICR mice. MI oocytes after IVM were treated with a $Ca^{2+}$ chelator to investigate the effect of $Ca^{2+}$ oscillations on their maturation. Results: As insemination time increased, the number of oocytes in the MI group that reached the MII stage also increased. The blastocyst rates and total cell numbers in the MII group were significantly higher than in the MI group. No pregnancy occurred in the MI group, but 10 pregnancies were achieved (10 of 12) in the MII group. The proportion of MI oocytes that matured to MII oocytes after fertilization was significantly higher in the non-treated group than in the $Ca^{2+}$ chelator-treated group. Conclusion: The findings that a higher proportion of MI-arrested oocytes progressed to MII after fertilization and that the MI-to-MII transition was blocked by $Ca^{2+}$ chelator treatments before fertilization indicate that the maturation of MI oocytes to MII oocytes is associated with intracellular $Ca^{2+}$ oscillations driven by fertilization.
