In this study, pooled whole semen and diluted semen with skim milk lactose solution and yolk skim milk lactose solution were inseminated at 6 and 7 days interval during 90 days. Egg production, fertility and hatchability were investigated. The results obtained from this study are summarized as following: 1. The average fertility of 57.8% for whole semen was clearly higher than that of 35.2% for diluted semen during 90 days insemination trial. 2. The highest fertility was a, pp.ared from 35 to 99 days after insemination for the whole semen, but from 28 to 60 days for the diluted semen during 90 days insemination trial. 3. In case of 7 days insemination interval, highest fertility of 86.6 and 70.0% for the whole semen and the diluted semen was a, pp.ared on 2 days after insemination and thereafter the fertility was gradually decreased according to passage of insemination. The lowest fertility of 35.0 and 0.0% for the whole semen and diluted semen was a, pp.ared on 1 day after insemination. 4. In case of 6 days insemination interval, highest fertility of 80.0 and 55.8% for the whole semen and the diluted semen was also a, pp.ared on 2 days after insemination and thereafter the fertility was slowly decreased according to passage of insemination. However, lowest fertility of 25.0 and 20.0% for the whole semen and the diluted semen was a, pp.ared on 0 day after insemination. 5 It suggests that there was no difference in fertility between the skim milk lactose and the yolk skim milk lactose dilutors. 6. In case of whole semen, average fertility of 7 days insemnaition interval was a, pp.rently lower than that of 6 days, however there was no difference in fertility between 6 and 7 days insemination interval. 7. Insemination interval of 6 and 7 days and passage day after insemination did not alter egg production and hatchability of fertilized egg production and hatchability of fertilized egg in both whole and diluted semen.

Objective: Artificial insemination plays an important role in genetic improvement in the goat farming system. The aim of this study was to investigate the effect of cervical application of hyaluronan (HA) on the fertility in goats after cervical artificial insemination using frozen-thawed (F-T) semen. Methods: After oestrous synchronisation with progesterone sponges and pregnant mare serum gonadotropin injection, both nulli- and multi-parous goats, were randomly allocated to 2 groups, and were inseminated with 0.25 mL of F-T semen (150×106 spermatozoa) twice at 52 h and 56 h after sponge removal. Prior to the insemination, goats in Group 1 only were given topical cervical HA application at 48 h after sponge removal. Site of insemination was recorded as os-cervix or intra-cervix or intra-uterus. Pregnancy was tested ultrasonographically 42 days after insemination. The data on pregnancy rates and percentage of animals according to the site of semen deposition were compared by Chi-square analysis. Results: The overall pregnancy rate was significantly (p<0.004) higher in goats with prior application to the cervix with HA (63.3%) than without (36.0%). Same pattern was observed in the pregnancy rates of nulli- and multi-parous goats in both the groups. Percentage of nulliparous goats according to the site of insemination in the HA group did not differ between first and the second insemination. However, in multiparous goats the percentage of animals inseminated intra-cervically was significantly increased (p≤0.05) between the first and the second inseminations. Conclusion: The results suggest that significantly higher fertility rate in the "HA goats" compared to the "without HA" group was because of deeper insemination facilitated by topical cervical application of HA. The deeper insemination into the cervical canal increase the rate of fertilisation when the cervical artificial insemination is performed.
The cattle follicular oocytes matured for 26~28h in culture condition were examined at 4, 5, 6, 7, 8, 10, 12, 14, 16 and 18h after insemination with bull spermatozoa preincubated for 4.5h in the uter isolated from estrous hamsters. After further culture with spermatozoa for 4~18 h, 73~89% of the total oocytes had matured to the second metaphase. None of the follicular oocytes matured in culture, were fertilized 5h after insemination. But when the oocytes were examined at 6, 8, 10, 14 and 18h after insemination, 60, 73, 82, 80 and 87% of oocytes were fertilized, respectively. The majority of the fertilized oocytes had enlarged sperm head at 6h after insemination and a part of the fertilized oocytes begun to develop from enlarged sperm head to male pronuclear stage at 8h after insemination, and most of them developed to male and female pronuclear stage at 10h after insemination. The results suggest that the penetration of spermatozoa into the oocytes may occur earlier than 6h after insemination and development of their pronuclear stage may occur at 8h after insemination.
Objective: The aim of this study was to compare the effects of conventional insemination (in vitro fertilization [IVF]) and intracytoplasmic sperm injection (ICSI) on the fertilization, developmental competence, implantation potential, and clinical pregnancy rate of embryos derived from in vitro matured oocytes of patients with polycystic ovary syndrome (PCOS). Methods: A prospective study was carried out among 38 PCOS patients who had undergone In vitro maturation (IVM) treatment. In total, 828 immature oocytes were collected from 42 cycles and randomly assigned for insemination by IVF (416 oocytes) or ICSI (412 oocytes). After fertilization, the embryos were cultured until the blastocyst stage and single embryos were transferred after endometrial preparation and under ultrasound guidance. Results: No significant differences were found in the maturation rate (78.1% vs. 72.6% for IVF and ICSI insemination, respectively; p= 0.076), fertilization rate (59.4% vs. 66.9% for IVF and ICSI insemination, respectively; p= 0.063), or the formation of good-quality blastocysts (40.9% vs. 46.5% for IVF and ICSI insemination, respectively; p= 0.314). Implantation and clinical pregnancy also did not show significant differences. Conclusion: There was a comparable yield of in vitro matured oocytes derived from PCOS patients in terms of fertilization, blastocyst formation, implantation rate, and clinical pregnancy between IVF and ICSI insemination. These findings provide valuable insights for choosing assisted reproductive treatment in women with PCOS, as IVM offers promising outcomes and is less invasive and less costly.
This experiment was carried out to study effects fo dosage and insemination interval on fertility and hatchability of whole semen and diluted semen with yolk skim milk and skim milk dilutors. The results obtained are as follows: 1. Whole semen showed higher fertility than diluted semen with yolk skim milk and skim milk dilutors. In case of diluted semen, the fertility was higher in 0.04$m\ell$ dosage than 0.02$m\ell$ and in skim milk than yolk skim milk dilutor. 2. The average fertility in inseminational intervals of 6, 5, 4 and 3 days was 52.4, 35.5, 48.7 and 44.2% in whole semen and 40.6, 17.2, 13.9 and 20.5% in 0.04$m\ell$ diluted semen with yolk skim milk dilutor. The fertility was not improved by shortening of insemination interval. 3. There was no considerable difference in hatchability of fertilized egg among the dosage of 0.02$m\ell$ of whole semen, 0.02$m\ell$ and 0.04$m\ell$ of diluted semen with yolk skim milk, and among the insemination intervals of 6, 5, 4 and 3 days. 4. Some differences in fertility among the passed days after insemination were decreased in the whole semen by shortening insemination interval from 6 and 5 days to 4 and 3days and also decreased in the diluted semen by shortening it to 3 days and by increasing dosage from 0.02$m\ell$ to 0.04$m\ell$. 5. Hatchability of fertilized egg showed no difference among the passed days during 6 days insemination interval both in the whole semen and the diluted semen. 6. The whole semen and the diluted semen with skim milk had not considerable difference in fertility among the passed days during 3 and 4 days insemination intervals, but the diluted semen with yolk skim milk had. 7. Hatchability of fertilized egg from the whole semen diluted semen with yolk skim milk and skim milk dilutors showed no difference among the passed days during 3 and 4 days insemination intervals.
Semen were collected from 9 male dogs and frozen by liquid nitrogengas. Frozen semen were thawed at 7$0^{\circ}C$ for 8 seconds. About $2{\timss}10^8$ sperm per insemination were inseminated to 10 bitches (3 Retrievers, 4 Chihuahuas, 1 Yorkshire Terriers, 1 Maltese, and 1 Poodle) at three and six days after the estimated peak of luteinizing hormone. For small breed dogs, uretero-renoscope (Kahl Storz, Germany, 12.5 Fr) was used for trans-cervical insemination, whereas cystoscope(Kahl Storz, Germany, 22Fr) was used for large breeds (Retrievers). Pregnancy was diagnosed by ultrasonography at 30 days after insemination. All of 3 Retrievers (100.0%) and 3 bitches of 7 small breed dogs (42.9%) were conceived (60.0% in total). This result indicated that trans-cervical insemination using endoscope is an effective method for AI with frozen semen not only for large breed dogs such as Retriever but also for small breeds.
This study was conducted to determine the optimum time and the insemination site of frozen in Cheju native cows, and 660 cows first service inseminated with rectovaginal methods by 3 skillful technicians as 3 districts near Cheju City and Sogwipo City areas obtained the following results. Optimum time for insemination was 16-20 hours after the onset of estrus and 80.6% of 67 conceived to first insemination. In the rectovaginal technique used widely at present conception rate for the palpation of ovaries showed 6.6% (index) higher than that of non-palpation. Conception rate of the insemination site; the intra-uterine horn, intra-uterus, just inside the cervix and depositing in mid-cervix were 62.5, 78.4, 74.6, and 80.9% respectively, however, there was no difference in conception rate between cervical and uterine deposition of forzon semen except for the intra-uterine horn.
Objective: This study investigated the effect of adding seminal plasma to frozen-thawed semen on the quality of sperm and pregnancy following insemination in dromedary camels. Methods: In experiment 1, the frozen-thawed semen from 9 collections (3 bulls) was further diluted with either the base extender or homologous seminal plasma (HSP). In the second experiment, a pooled sample of frozen-thawed semen was diluted with either seminal plasma from another three bulls. Live percentage, total and progressive motility, functional and acrosome integrity, and sperm kinematics were evaluated at 15, 60, and 120 minutes post-thawing and compared to the non-treated control. In experiment 3, frozen semen was used to inseminate camels in the following experimental groups: 1-Single insemination with double dose undiluted frozen semen (n = 9); 2-Re-insemination in 6 hours with undiluted semen (n = 13); 3-Single insemination with HSP treated sperm (n = 14). Results: Frozen-thawed sperm diluted in HSP or the non-homologous seminal plasma from Bull C indicated an improvement in all parameters after 1 hour post-thawing incubation (p<0.05). The proportion of total and progressively motile sperm did not drop significantly at 60 minutes post-thawing when diluted with the seminal plasma of Bull C (p>0.05). Double insemination with nontreated sperm and single insemination with HSP-treated sperm resulted in similar pregnancy rates (15.3% vs 21.4%, p>0.05). None of the camels conceived with double-dose single insemination of nontreated sperm. Conclusion: Seminal plasma improves sperm longevity and motility after thawing in dromedary camel with a significant between-bull variation in effect. Low post-thaw sperm longevity might be the cause behind the low pregnancy rates in frozen semen insemination of dromedary camels.

Background: Estrus in cows can be detected through vaginal electrical resistance or conductivity. However, there are no studies measuring vaginal electrical resistance in Hanwoo cows. This study aims to measure the vaginal electrical resistance value in Hanwoo cows and compare it with estrus and ovulation. Methods: Vaginal electrical resistance values of 73 Hanwoo cows were measured before and after estrus at the Gyeongsangbuk-do Livestock Research Institute. Measurements were taken on days -6, -3, -2, -1, 0, 1, 2, 3, and 6 of artificial insemination. Large follicles and ovulation were confirmed using transvaginal ultrasonography. Results: The vaginal electrical resistance averaged 225.6 ± 6.3 Ω days before the artificial insemination date, decreasing until the day of artificial insemination. The average vaginal electrical resistance was 163.7 ± 4.6 Ω on the date of artificial insemination, and 188.8 ± 4.3 Ω one day after artificial insemination, when large follicles were observed. In addition, on the 6th day after artificial insemination, the vaginal electrical resistance averaged 231.4 ± 5.5, which was similar to the 6th day before artificial insemination (222.5 ± 6.3). Transvaginal ultrasonography showed that most of the cows ovulated one day after artificial insemination. Conclusions: The accuracy of estrus is high if the vaginal electrical resistance is measured for cows with confirmed estrus, making is a potentially useful for determining the timing of artificial insemination.
A device far estimating the optimal artificial insemination time consisted of a computer, RS module, and six direct-reflex photo-sensors. Program was written in LabWindows CVI. In order to establish references for estimating optimal artificial insemination time, lying rate of the thirty three $Berkshire{\times}Hampshire crossbred sows (parity 5 to 6) was recorded from 2 days after moving into stalls to artificial insemination using a time lapse VCR and was measured every hour with one minute interval using the scan point sampling method. Twenty low hours in a day were divided into three comparing periods : 21:00 to 06:59, 07:00 to 13:59, and 14:00 to 20:59. If sum of the percentages of tying during a comparing period was less than the reference value, the starting hour of the comparing period was considered as onset of estrus and the optimal artificial insemination time was estimated at 20 to 24 hours after onset of estrus. The experimental device was evaluated with twenty five $Berkshire{\times}Hampshire$ crossbred sows (parity 2 to 7) and 23 of 25 sows $(92\%)$ were pregnant. This result suggests that the experimental device is excellent in performance for estimating optimal artificial insemination time.
