Investigation of Agrobacterium-mediated Transient dsRNA Expression in Tobacco

  • The Agrobacterium tumefaciens mediated gene transfer is widely used to generate genetic transformation of plants and transient assay of temporal exogenous gene expression. Syringe infiltration system into tobacco (Nicotiana benthamiana) leaves is a powerful tool for transient expression of target protein to study protein localization, protein-protein binding and protein production. However, the protocol and technical information of transient gene expression, especially double strand RNA (dsRNA), in tobacco using Agrobacterium is not well known. Recently, dsRNA is crucial for insecticidal effect on destructive agronomic pest such as Corn rootworm. In this study, we investigated the factor influencing the dsRNA expression efficiency of syringe agro-infiltration in tobacco. To search the best combination for dsRNA transient expression in tobacco, applied two Agrobacterium cell lines and three plant vector systems. The efficiency of dsRNA expression has estimated by real-time PCR and digital PCR. As a result, pHellsgate12 vector constructs showed the most effective accumulation of dsRNA in the cell. These results indicated that the efficiency of dsRNA expression was depending on the kind of vector rather than Agrobacterium cells. In summary, the optimized combination of transient dsRNA expression system in tobacco might be useful to in vivo dsRNA expression for functional study and risk assessment of dsRNA.

Study on Development of Novel Biopesticides Using Entomopathogenic Bacterial Culture Broth of Xenorhabdus and Photorhabdus (Xenorhabdus 및 Photorhabdus 세균 배양액을 이용한 생물농약 개발에 관한 연구)

  • Two groups of entomopathogenic bacteria, Xenorhabdus and Photorhabdus, are known to suppress insect immune responses by inhibiting eicosanoid biosynthesis. This study used these bacterial culture broths to develop novel biochemical insecticides against the diamondback moth, Plutella xylostella. Though the bacterial culture broths alone showed little insecticidal activity, they significantly enhanced pathogenicity of Bacillus thuringiensis against the fourth instar larvae of P. xylostella. Sterilization of the bacterial culture broth by autoclaving or $0.2\;{\mu}m$ membrane filtering did not influence the synergistic effect on the pathogenicity of B. thuringiensis. Three metablites identified in the culture broth of X. nematophila also showed similar synergistic effects. In field test, both entomopathogenic bacterial culture broth also enhanced the control efficacy of B. thuringiensis against P. xylostella.

Ex vivo Cytotoxicity of the Bacillus thuringiensis Cry4B δ-Endotoxin to Isolated Midguts of Aedes aegypti Larvae

  • The pathological effect of the Bacillus thuringiensis Cry $\delta$-endotoxins on susceptible insect larvae had extensive damage on the midgut epithelial cells. In this study, an ex vivo assay was devised for assessing the insecticidal potency of the cloned Cry4B mosquito-larvicidal protein that is expressed in Escherichia coli. Determination of toxicity was carried out by using a cell viability assay on the midguts that were dissected from 5-day old Aedes aegypti mosquito larvae. After incubation with the toxin proteins, the number of viable epithelial cells was determined photometrically by monitoring the quantity of the bioreduced formazan product at 490 nm. The results showed that the 65-kDa trypsin-activated Cry4B toxin exhibited toxic potency ca. 3.5 times higher than the 130-kDa Cry4B protoxin. However, the trypsin-treated products of the non-bioactive Cry4B mutant (R158A) and the lepidopteran-specific Cry1Aa toxin displayed relatively no ex vivo activity on the mosquito-larval midguts. The ex vivo cytotoxicity studies presented here confirms data that was obtained in bioassays.

  • The winter cherry bug (Acanthocoris sordidus Thunberg, Coreidae, Hemiptera) is a insect pest that have a piercing sucking type mouthpart, and it is injured to solanaceous plants including pepper plant. For the control of the winter cherry bug, 20 organic synthetic insecticides and 7 environmental friendly agricultural materials (EFAMs) were selected with single formulation. Fenitrothion, fenthion and phenthoate were significantly higher mortality to adults and 3rd instar nymphs of winter cherry bug as over 95 and 100 % after 72 hours with treatment. Otherwise, in case of EFAMs, there is no significantly higher effective materials to adults and 3rd instar nymphs of winter cherry bug. Eighty percentage of Sophora flavescens was showed only around 50% mortality with the high variation after 72 hours with treatment against adult of the winter cherry bug.

  • This study was conducted to establish a nce insect pest control system, ~ es.ys tematic application of insecticides using carbohran and buproiezin, in Korea. The effects oi various dates of application and rates of buprofezin (25% WP) after carbofuran (3G) soil incorpombon in late May on the pop~~lation densities of the brown planthoppa (BPH) immigrating in July were investigated Appropriate application tune of buprofezin for the BPH that had evaded insecticidal effect of 5011 incorporated carbofuran was late July-early August. Application rate of buprofezln at 7.0g a.i/lOa was enough to suppress the BPH density. Buprofezin treatment after carbofumn soil incorporation could also suppress the whlte backed planthopper population but did not affect the densities oi the paddy rice spiders. Considering the charactenstics of occurring patterns of the nce insect pests in Korea. buprofezin treatment m late July or early August after carbofuran soil incorporation in late May can be a useful application system of ir~sectic~deins controlling early season Insect pests and migmtoly planthoppers on rice.

Production of Microbial Insecticide Using Bacillus thuringiensis BT17 for the Control of Lepidopteran Larvae (Bacillus thuringiensis BT17 균주를 이용한 인시목 유충 방제용 미생물 살충제 생산)

  • Insecticidal crystalline toxin producing Bacillus thuringiensis BT17 strain was isolated and identified as B. thuringiensis serovar colmeri by 16S rRNA analysis. BT17 strain produced crystalline ${\delta}$-endotoxin against to Lepidopteran larvae effectively on the culture broth of soybean meal and skim milk, $30^{\circ}C$ and 36 h shaking culture of 280 rpm. The maximum colony forming unit achieved when the culture was continued for 24 h, but the number of crystals increased until 36 h in the 200 L fermentor. Liquid type of biological insecticide product was made, and after 3 months storage in $20^{\circ}C$ the number of crystals was increased up to twice than beginning. Biocontrol effect of BT17 insecticide product was better in Plutella xylostella than in Spodoptera exigua, and the toxicity to animals was negligible.

  • Paulownia coreana has various advantages as a convenient workability, aesthetic outlook, beautiful patterns, low weight and high strength, and its permeability. P. coreana has been widely used for storage but there are no empirical researches proving its functionality in a field of conservation science until now. In this study, the seasonal and daily temperature and relative humidity control, and microbe and insect repellent activity were evaluated under the controlled and uncontrolled circumstances from 2015 to 2016. The results showed to be mainly excellent in relative humidity control and the buffering effect was good to adjust the average daily relative humidity range from the outside. With respect to the antimicrobial properties of P. coreana, we observed that its water-soluble extract produced visible zones of inhibition against five bacteria. However, it was difficult to predict the antimicrobial and/or insecticidal properties.

Toxicity Evaluation of Agricultural Insecticides on Workers of Pharaoh Ant, Monomorium pharaonis (Hyme-noptera : Formicidae) (애집개미 (Monomorium pharaonis) 일개미에 대한 농업용 살충제의 독성평가)

  • Toxicities of 34 agricultural insecticides was tested against Monomorium pharaonis workers by diet dipping method. Ten insecticides among them, bifenthrin, chlorpyrifos, chlorpyrifos-methyl, cypermethrin, dichlorvos, fenthion, fenitrothion, methidathion, pirimifos-methyl, and phenthoate showed 100% rapid mortality. $LC_{50}$ (ppm) values of selected insecticides were appeared on the order of pirimifos-methyl (0.33), chlorpyrifos-methyl (0.76), phenthoate (1.70), bifenthrin (1.78), dichlorvos (2.50), cypermethrin (9.92), chlorpyrifos (22.21), fenitrothion (36.58), fenthion (40.96), and methidathion (64.34). $LT_{50}$ (day) values by diet dipping method showed that dichlorvos, benfuracarb and cypermethrin acted more rapid than boric acid and hydramethylnon. The values of the former three were 0.25, 0.38 and 0.27 days, and those of the latter two were 3.4 and 2.6 days, respectively. In persistance effect tests, chlorpyrifos-methyl, fenthion and methidathion showed over 90% insecticidal activity for 13 days.

Mortality of Cigarette Beetle, Lasioderma serricorne, Fabricius (Coleoptera : Anobiidae) and Corrosion of Metals as Influenced by Sulfuryl fluoride ($SO_2F_2$) (훈증제 sulfuryl fluoride ($SO_2F_2$)의 궐련벌레(Lasioderma serricorne, Fabricius)에 대한 방제효과 및 몇 가지 금속류 성상에 미치는 영향)

  • Insecticidal effect or sulfuryl fluoride ($SO_2F_2$) to cigarette beetle, Lasiodema serricorne (F.), was studied in two different containers, one was 1m$^3$(without cardboard block) and the other was 0.5m$^3$(with cardboard block). Adults and larvae were transferred into a small metal can before placed in the containers. Each can was held for 8, 24, 48, 72, and 96 hours after $SO_2F_2$(10, 20, 30, and 40 g/$\ell$) treatment. All adults were killed in an 1m$^3$container. Larval mortality was 99 to 100% when the dosage of SO$_2$F$_2$was 10 and 20 g/$\ell$. Cardboard was attached between two 0.5m$^3$ containers ; one was fumigation area and the other was insect area. SO$_2$F$_2$ penetrated cardboard within 24 hours in most trials. $SO_2F_2$could affect all adults and larvae regardless of their susceptibility. All adults were killed at 8 hours in a 0.5m$^3$ container. But low mortality of larvae was recorded at 8 hours when small dosage was treated (30% in 10 g/$\ell$, 87.2% in 20 g/$\ell$). The mortality was increased as SO$_2$F$_2$ dosage increased. Six different metals (stainless, copper, brass, aluminum, iron, and zinc) were also tested to study metal corrosion and discoloration. No corrosion and discoloration was observed in most metals treated with $SO_2F_2$.

Insecticidal Activity and Ovipositional Repellency of Monoterpenoids against Moechotypa diphysis Adults (Coleoptera: Cerambycidae) (털두꺼비하늘소 성충에 대한 monoterpenoids의 살충활성과 산란기피)

  • These studies were carried out to investigate the fumigant toxicity, the contact toxicity and the ovipositional repellency of 25 monoterpenoids against oak longicorn beetle adults, Moechotypa diphysis. Monoterpenoids with 100% of fumigant toxicity were 1, 8-cinehone, fenchone, pulegone and (equation omitted)-terpinene at 20 (equation omitted)/954 ml (air) concentration, pulegone and (equation omitted)-terpinen at 10 (equation omitted)/954 ml (air) concentration, pulegone at 5 (equation omitted)/954 ml (air) concentration. Most monoterpenoids showed low or no contact toxicity but only pulegone showed about 70% mortality. Some monoterpenoids which had repellency to female adults with Y-tube olfactometer were bornylacetate, carvacrol, 1, 8-cineole and menthol at 1 (equation omitted) dose, while citronellol showed attractant response. Carveol, geraniol and perillyl alcohol of 25 monoterpenoids showed ovipoistional repellency of 82.1%, 78.3%, 87.5%, respectively, at the concentration of 1,000 ppm in the laboratory condition. In the field, the result tested with three monoterpenoids indicated that geraniol was the most effective one at the concentration of 10,000ppm and 1,000 ppm, but residual effect wasn't found.
