Biological Methanol Production by a Type II Methanotroph Methylocystis bryophila

  • Methane (CH4) is the most abundant component in natural gas. To reduce its harmful environmental effect as a greenhouse gas, CH4 can be utilized as a low-cost feed for the synthesis of methanol by methanotrophs. In this study, several methanotrophs were examined for their ability to produce methanol from CH4; including Methylocella silvestris, Methylocystis bryophila, Methyloferula stellata, and Methylomonas methanica. Among these methanotrophs, M. bryophila exhibited the highest methanol production. The optimum process parameters aided in significant enhancement of methanol production up to 4.63 mM. Maximum methanol production was observed at pH 6.8, 30℃, 175 rpm, 100 mM phosphate buffer, 50 mM MgCl2 as a methanol dehydrogenase inhibitor, 50% CH4 concentration, 24 h of incubation, and 9 mg of dry cell mass ml-1 inoculum load, respectively. Optimization of the process parameters, screening of methanol dehydrogenase inhibitors, and supplementation with formate resulted in significant improvements in methanol production using M. bryophila. This report suggests, for the first time, the potential of using M. bryophila for industrial methanol production from CH4.

Effect of Cellulose Degrading Bacteria Isolated from Wild and Domestic Ruminants on In vitro Dry Matter Digestibility of Feed and Enzyme Production

  • Cellulolytic bacterial strains have been isolated from the faeces of wild (blackbuck, Antilope cervicapra; nilgai, Baselophus tragocamelus chinkara, Gazella gazella spotted deer, Axis axis and hog deer, Cervus porcinus) and rumen liquor of domestic (sheep, Ovis aries) ruminants. Five best cellulose degrading bacterial isolates (Ruminococcus sp.) were used as microbial feed additive along with buffalo rumen liquor as inoculum to study their effect on digestibility of feed and enzyme production in in vitro conditions. The bacterial isolate from chinkara (CHI-2) showed the highest per cent apparent dry matter (DM) digestibility ($35.40{\pm}0.60$), true dry matter digestibility ($40.80{\pm}0.69$) and NDF ($26.38{\pm}0.83$) digestibility (p<0.05) compared to control ($32.73{\pm}0.56$, $36.64{\pm}0.71$ and $21.16{\pm}0.89$, respectively) and other isolates at 24 h of incubation with lignocellulosic feeds (wheat straw and wheat bran, 80:20). The same isolate also exhibited the highest activities of fibre degrading enzymes like carboxymethylcellulase, xylanase, ${\beta}$-glucosidase and acetyl esterase. The bacterial isolate from chinkara (Gazella gazella) appears to have a potential to be used as feed additive in the diet of ruminants for improving utilization of nutrients from lignocellulosic feeds.

다른 기주 탄저병균의 딸기에 대한 병원성 (Pathogenesity of Colletotrichum gloeosporioides from Other Hosts on Strawberry)

  • 다른 기주에 발생하는 탄저병(C. gloeosporioides)이 딸기에 전염되는지를 알아보기 위해 사과, 고추, 대추, 감의 이병된 과실에서 탄저병균을 분리동정하고 딸기에서의 병원성을 비교하였다. 각 기주별로 분리된 C. gloeosporioides의 배양적 특성과 포자의 크기는 서로 유사하였으나 말기에 접종을 하였을 경우에는 감에서 분리된 균주가 병원성이 가장 강하였고, 대추에서 분리된 균주는 약한 병원성을 보였다. 각 기주별로 병원성을 나타내는 균주를 선발하여 포장에서 병원성을 검정하였을 때 모두 탄저병이 발병되었으므로 다른 기주에서 발생한 탄저병도 자연상태에서 딸기로 전염이 가능한 것을 확인할 수 있었다.

Candida mogii ATCC 18364를 이용한 D-Xylose로부터 Xylitol 생산 (Xylitol Production from D-Xylose by Candida mogii ATCC 18364)

  • 볏짚 가수분해물로부터 xylitol 생산능력이 가장 우수하다고 알려진 Candida mogii ATCC 18364의 xylose 발효특성에 관한 기초연구를 수행하였다. 상업적인 자일로즈로부터 C. mogii게 의해 xylitol을 생산할 때 수율과 속도에 미치는 여러 영향을 검토한 결과, 포도당을 함유한 기질에서 12시간 배양한 종균을 3.0 g/L의 농도로 접종하여 발효시켰을 때 가장 좋은 결과를 얻었다. 또한 xylose 발효에서 가장 중요한 요인인 용존산소 농도의 영향을 알아보기 위해, 2 L 발효조에서 통기량과 교반속도를 조절해 가면서 xylitol 생산을 비교 검토하였다. 농축균과 10% 접종균을 사용했을 경우 모두, 1 vvm의 통기량과 200 rpm의 교반속도에서 xylitol 생산이 가장 우수하였다.

Molecular identification of dye degrading bacterial isolates and FT-IR analysis of degraded products

  • In the present study, dye decolorizing bacteria were isolated from water and soil samples, collected from textile industries in Jodhpur province, India. Two bacterial species namely, Bacillus pumilis and Paenibacillus thiaminolyticus were screened and identified based on biochemical characterization. The degradation efficiency of these two microorganisms was compared through optimization of pH, incubation time, initial dye concentration and inoculum size. B. pumilis and P. thiominolyticus were able to degrade 61% and 67% Red HE3B, 81% and 75% Orange F2R, 49.7% and 44.2% Yellow ME4GL and 61.6% and 59.5% Blue RC CT dyes of 800mg/l concentration respectively. The optimum pH and time were found to be 8 within 24 hours. The FT-IR analysis confirmed that microorganisms were able to degrade toxic azo dyes into a non-toxic product as proved through structural modifications to analyze chemical functions in materials by detecting the vibrations that characterize chemical bonds. It is based on the absorption of infrared radiation by the microbial product. Therefore, Bacillus pumilis and Paenibacillus thiaminolyticus are a promising tool for decolorization of dyes due to its potential to effectively decolorize higher azo dye concentrations (10-800 mg/L) and can be exploited for bioremediation.

선충 기생 전적 진균의 접종원 농도와 온도조건에 따른 성충감염 및 집단 감소효과 (Decrease of Nematode Population by Introduction of Nematophagous Fungi into The Soil as Affected by Inoculum Concentration and Temperature in Vitro)

  • Five nematophagous fungi were evaluated for their nematicidal effect in vitro on Rhabditis sp. and Meloidogyne hapla in soil. Inocula of Arthrobotrys arthrobotryoides, A. conoides, A. oligospora, Dactylella lobata, and Fusarium oxyaporum were grown in moistened corn-sandy soil and chopped potato-sandy soil media, and incubated at 26$^{\circ}C$ for one week. The prepared inocula were incorporated in autoclaved sandy soil, mixing thoroughly at rates equ-invalent to 1:50, 1:100, 1:200, and 1:400, repectively, before 80g of the mixture carrying 100 Rhabditis sp. was put into petri plates. Nematophagous fungi effectively teduced the popuation of Rhabditis sp. in soil in a week or two following treatment of the incula at concentration of 1:50 and 1:100. The optimum was at $25^{\circ}C$ for nematicidial effect as high as 80-100%. The at the rate of 1:100 prepared incula were incorporated in auto-claved soil, where 100 Juveniles M. hapla were introduced per 80% soil. All fungi infected the M. hapla effectively in soil, caysing more than 90% mortality within one week. This result indicated the potential value of these fungi as promising biocontrol agents.

Enhancing anaerobic digestion of vegetable waste and cellulose by bioaugmentation with rumen culture

  • Anaerobic digestion (AD) has been widely used to valorize food waste (FW) because of its ability to convert organic carbon into $CH_4$ and $CO_2$. Korean FW has a high content of fruits and vegetables, and efficient hydrolysis of less biodegradable fibers is critical for its complete stabilization by AD. This study examined the digestates from different anaerobic digesters, namely Rs, Rr, and Rm, as the inocula for the AD of vegetable waste (VW) and cellulose (CL): Rs inoculated with anaerobic sludge from an AD plant, Rr inoculated with rumen fluid, and Rm inoculated with anaerobic sludge and augmented with rumen fluid. A total of six conditions ($3\;inocula{\times}2\;substrates$) were tested in serial subcultures. Biogas yield was higher in the runs inoculated with Rm than in the other runs for both VW (up to 1.10 L/g VS added) and CL (up to 1.05 L/g VS added), and so was biogas production rate. The inocula had different microbial community structures, and both substrate type and inoculum source had a significant effect on the formation and development of microbial community structures in the subcultures. The overall results suggest that the bioaugmentation with rumen microbial consortium has good potential to enhance the anaerobic biodegradability of VW, and thereby can help more efficiently digest high fiber-content Korean FW.

A Review of the Efficacy of Ultraviolet C Irradiation for Decontamination of Pathogenic and Spoilage Microorganisms in Fruit Juices

  • Ultraviolet C (UV-C, 200-280 nm) light has germicidal properties that inactivate a wide range of pathogenic and spoilage microorganisms. UV-C has been extensively studied as an alternative to thermal decontamination of fruit juices. Recent studies suggest that the efficacy of UV-C irradiation in reducing microorganisms in fruit juices is greatly dependent on the characteristics of the target microorganisms, juice matrices, and parameters of the UV-C treatment procedure, such as equipment and processing. Based on evidence from recent studies, this review describes how the characteristics of target microorganisms (e.g., type of microorganism/strain, acid adaptation, physiological states, single/composite inoculum, spore, etc.) and fruit juice matrices (e.g., UV absorbance, UV transmittance, turbidity, soluble solid content, pH, color, etc.) affect the efficacy of UV-C. We also discuss the influences on UV-C treatment efficacy of parameters, including UV-C light source, reactor conditions (e.g., continuous/batch, size, thickness, volume, diameter, outer case, configuration/arrangement), pumping/flow system conditions (e.g., sample flow rate and pattern, sample residence time, number of cycles), homogenization conditions (e.g., continuous flow/recirculation, stirring, mixing), and cleaning capability of the reactor. The collective facts indicate the immense potential of UV-C irradiation in the fruit juice industry. Existing drawbacks need to be addressed in future studies before the technique is applicable at the industrial scale.

Effects of arbuscular mycorrhizal fungi on enhancing growth, fruit quality, and functional substances in tomato fruits (Lycopersicon esculentum Mill.)

  • This study aimed to investigate the efficiency of arbuscular mycorrhizal fungi (AMF) in enhancing plant performance and bioactive compound concentrations in tomatoes (Lycopersicon esculentum Mill.). This factorial pot experiment included nine replications over 120 days of cultivation. Three AMF species (Rhizophagus prolifer, Claroideoglomus etunicatum, and Acaulospora mellea) were utilized as inoculum, while non-mycorrhizal controls with or without synthetic NPK fertilizer were compared. Interestingly, C. etunicatum KS-02 inoculations effectuated the best fruit growth and weight, which were statistically higher than those of the control without AMF. However, only fruit fresh weight was higher in plants inoculated with C. etunicatum KS-02 than those treated with the synthetic NPK fertilizer. In addition, C. etunicatum KS-02 inoculations induced a ≥ 11% increase in DDPH (1,1-diphenyl-2-picrylhydrazyl) activity, lycopene content, and carotenoid content compared to the control. This study is the first to report Claroideoglomus species' effectiveness in promoting growth, fruit yield, and bioactive compound production in L. esculentum Mill. These findings substantiate the significant potential of C. etunicatum KS-02 for tomato cultivation without the adverse effects of excessive synthetic fertilizer use.

Glucose와 Yeast Extract를 이용하여 배양된 유산균을 이용한 하수 일차 슬러지의 가용화 (An Investigation of the Solubilization of Primary Sewage Sludge using Lactic Acid Bacteria Cultured in a Glucose and Yeast Extract Medium)

  • The intention of this research was to investigate the solubilization of primary sewage sludge using lactic acid bacteria cultured in a glucose and yeast extract medium. Glucose as the carbon source and yeast extract as the source of nitrogen were chosen as an economic medium with the potential for the mass production of lactic acid bacteria. The optimal concentrations of the medium were 3% (w/v) glucose and 2% (w/v) yeast extract. In this study, in order to improve field applications for the solubilization of sludge at sewage treatment plants, a powdered form of lactic acid bacteria was produced. The optimal inoculum of the powder for the maximum efficiency of solubilization was 1% (w/v). In that condition, the SCOD value increased from 8600 (mg/L) at the beginning of experiment to 10290 (mg/L) at 96 h, with the highest solubilization rate (20.6% DDCOD) and 11.2% (SCOD). Also, the TVFAs of the lactic acid bacteria inoculation group were produced more than that of the control group. In particular, acetic acid was produced 5 times more in the experimental group than in the control group. In this research, the potential of lactic acid bacteria in the pretreatment of primary sewage sludge as a solubilizer, and as an energy source producer for microbial fuel cells was revealed.
