Molecular Characterization and Expression Pattern of Gene IGFBP-5 in the Cashmere Goat (Capra hircus)

  • Insulin-like growth factor-binding protein-5 (IGFBP-5) is one of the six members of IGFBP family, important for cell growth, apoptosis and other IGF-stimulated signaling pathways. In order to explore the significance of IGFBP-5 in cells of the Inner Mongolian Cashmere goat (Capra hircus), IGFBP-5 gene complementary DNA (cDNA) was amplified by reverse transcription polymerase chain reaction (RT-PCR) from the animal's fetal fibroblasts and tissue-specific expression analysis was performed by semi-quantitative RT-PCR. The gene is 816 base pairs (bp) in length and includes the complete open reading frame, encoding 271 amino acids (GenBank accession number JF720883). The full cDNA nucleotide sequence has a 99% identity with sheep, 98% with cattle and 95% with human. The amino acids sequence shares identity with 99%, 99% and 99%, respectively. The bioinformatics analysis showed that IGFBP-5 has an insulin growth factor-binding protein homologues (IB) domain and a thyroglobulin type-1 (TY) domain, four protein kinase C phosphorylation sites, five casein kinase II phosphorylation sites, three prenyl group binding sites (CaaX box). The IGFBP-5 gene was expressed in all the tested tissues including testis, brain, liver, lung, mammary gland, spleen, and kidney, suggesting that IGFBP-5 plays an important role in goat cells.

Comparison of Fecal Microbiota of Mongolian and Thoroughbred Horses by High-throughput Sequencing of the V4 Region of the 16S rRNA Gene

  • The hindgut of horses is an anaerobic fermentative chamber for a complex and dynamic microbial population, which plays a critical role in health and energy requirements. Research on the gut microbiota of Mongolian horses has not been reported until now as far as we know. Mongolian horse is a major local breed in China. We performed high-throughput sequencing of the 16S rRNA genes V4 hypervariable regions from gut fecal material to characterize the gut microbiota of Mongolian horses and compare them to the microbiota in Thoroughbred horses. Fourteen Mongolian and 19 Thoroughbred horses were used in the study. A total of 593,678 sequence reads were obtained from 33 samples analyzed, which were found to belong to 16 phyla and 75 genera. The bacterial community compositions were similar for the two breeds. Firmicutes (56% in Mongolian horses and 53% in Thoroughbred horses) and Bacteroidetes (33% and 32% respectively) were the most abundant and predominant phyla followed by Spirochaete, Verrucomicrobia, Proteobacteria, and Fibrobacteres. Of these 16 phyla, five (Synergistetes, Planctomycetes, Proteobacteria, TM7, and Chloroflexi) were significantly different (p<0.05) between the two breeds. At the genus level, Treponema was the most abundant genus (43% in Mongolian horses vs 29% in Thoroughbred horses), followed by Ruminococcus, Roseburia, Pseudobutyrivibrio, and Anaeroplasma, which were detected in higher distribution proportion in Mongolian horses than in Thoroughbred horses. In contrast, Oscillibacter, Fibrobacter, Methanocorpusculum, and Succinivibrio levels were lower in Mongolian horses. Among 75 genera, 30 genera were significantly different (p<0.05) between the two breeds. We found that the environment was one of very important factors that influenced horse gut microbiota. These findings provide novel information about the gut microbiota of Mongolian horses and a foundation for future investigations of gut bacterial factors that may influence the development and progression of gastrointestinal disease in horses.

  • A large-eddy simulation is performed for turbulent flow in a concentric annulus with the inner wall rotation at Re$\sub$Dh/=8900 for three rotation rates N=0.2145, 0.429 and 0.858. Main emphasis is placed on the inner wall rotation effect on near-wall turbulent structures. Near-wall turbulent structures close to the inner wall are scrutinized by computing the lower-order statistics. The anisotropy invariant map for the Reynolds stress tensor and the invariant function are illustrated to reveal the altered anisotropy in turbulent structure. Probability density functions of the splat/anti-splat process are explored to develop a sufficiently complete picture of the contributions of the flow events to turbulent production. The present numerical results show that the altered turbulent structures may be attributed to the centrifugal instability, which leads to the augmentation of sweep and ejection events.

  • This study was performed to find out the usefulness of chestnut powder recovered from the inner layer, which used to be discarded. After the Physicochemical properties of the chestnut powder were examined, breads were prepared with 10-50% of chestnut Powder and the sensory quality was compared by QDA. Bread could be made even using up to 50% of chestnut powder, and in 20-30% of chestnut powder, the sensory quality was acceptable enough compared with wheat bread. Even though the low purity of the chestnut powder affected the flavor and color, the breads contained it were generally acceptable. Therefore, the chestnut powder recovered from the inner layer can be used as the effective food resource as itself, and with the higher purification to remove inner layer, it can be applied to more diverse food.

  • The solar radiation, inner temperature of the tilted box and the amount of condensed water were measured to find the most effective purified water production system. The inner temperature of the tilted box was related with solar radiation that was affected by sunshine and cloud. With analyzing those relationships of the solar radiation, inner box temperature, the angle of tilted boxes and amounts of water condensation optimal collecting system area could be calculated.

  • The goat Capra hircus is one of several economically important livestock in China. Advances in molecular genetics have led to the identification of several single nucleotide variation markers associated with genes affecting economic traits. Validation of single nucleotide variations in a whole-transcriptome sequencing is critical for understanding the information of molecular genetics. In this paper, we aim to develop a large amount of convinced single nucleotide polymorphisms (SNPs) for Cashmere goat through transcriptome sequencing. In this study, the transcriptomes of Cashmere goat skin at four stages were measured using RNA-sequencing and 90% to 92% unique-mapped-reads were obtained from total-mapped-reads. A total of 56,231 putative SNPs distributed among 10,057 genes were identified. The average minor allele frequency of total SNPs was 18%. GO and KEGG pathway analysis were conducted to analyze the genes containing SNPs. Our follow up biological validation revealed that 64% of SNPs were true SNPs. Our results show that RNA-sequencing is a fast and efficient method for identification of a large number of SNPs. This work provides significant genetic resources for further research on Cashmere goats, especially for the high density linkage map construction and genome-wide association studies.

Fabrication of Metallic Sandwich Plates with Inner Dimpled Shell Structure and Static Bending Test (딤플형 내부구조 금속 샌드위치 판재의 제작 및 정적 굽힘 실험)

  • Metallic sandwich plates with various inner cores have important new features with not only ultra-light material characteristics and load bearing function but also multifunctional characteristics. Because of production possibility on the large scale and a good geometric precision, sandwich plates with inner dimpled shell structure from a single material have advantages as compared with other solid sandwich plates. Inner dimpled shell structures can be fabricated with press or roll forming process, and then bonded with two face sheets by multi-point resistance welding or adhesive bonding. Elasto-plastic bending behavior of sandwich plates have been predicted analytically and measured. The measurements have shown that elastic perfectly plastic approximation can be conveniently employed with less than 10% error in elastic stiffness, collapse load, and energy absorption. The dominant collapse modes are face buckling and bonding failure after yielding. Sandwich plates with inner dimpled shell structure can absorb more energy than other types of sandwich plates during the bending behavior.

Inhibitory Effects of Chestnut Inner Shell Cytokine Production from Human Mast Cell Line (율피추출물의 사람 비만세포주 사이토카인 발현 억제 효과)

  • Objectives : Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by eczematous inflammtion of the skin. The chestnut inner shell extracts (CI) has been used as a cosmetic material for a long time in Korea. However, the precise anti-allergy effects of CI have yet to be clearly elucidated. In this study, we attempted to evaluate the effect of CI on mast cell-mediated allergy inflammation. Methods : To find the anti-allergy and inflammatory effect of CI, we investigated the inhibitory effect of CI on the production of inflammatory mediators using by enzyme-linked immunosorbent assay in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore (A23187) stimulated-human mast cell (HMC-1). Results : In this study, we found that CI did not show cytotoxic effect at up to 10 ug/ml on HMC-1. CI inhibited the production of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-6 and IL-8 in stimulated HMC-1. Maximal rate of TNF-${\alpha}$, IL-6 and IL-8 inhibition by CI (10 ug/ml) were about 47.6%, 44.1% and 22.5% respectively. In addition, we showed that Fr.3 isolated from n-Butyl alcohol layer of CI attenuated the production of TNF-${\alpha}$, IL-6 and IL-8 in HMC-1. Conclusion : Taken together, the findings of this study provide us with a novel insight action of CI as a potential molecule for use in the treatment of allergic inflammation diseases.

Computed tomography investigation of the three-dimensional structure and production method of White Porcelain Water Dropper with Openwork Lotus Scroll Design and Eight Trigram Design in Cobalt-blue Underglaze (CT 조사를 통한 청화백자투각연당 초팔괘문연적의 3차원적 구조와 제작방법에 대한 고찰)

  • This study investigated White Porcelain Water Dropper with Openwork Lotus Scroll Design and Eight Trigram Design in Cobalt-blue Underglaze (hereinafter, the "water dropper") in the collection of the National Museum of Korea using computed tomography (CT). A replica was produced to examine both the structure and its original production method. The CT scanning identified no joint lines or pores in the clay, which suggests that the body (the lower part of the water dropper) was shaped in a single piece using a mold and was then matched with a mold-formed lid (the upper part of the water dropper). The inner container of the body portion was roughly trimmed with a bamboo knife so that its upper surface could be securely attached to the bottom of the lid and prevent any leakage in the joined surface. It appears that the inner container for storing water was made first in a cylindrical shape that met the unit of quantity used at the time and could be easily formed by molding. It was transformed into a trapezoid shape during the process of combining it with the lid. A cylindrical inner container was reproduced using silicon 3D printing to compare its capacity with that of the original inner container. The comparison revealed that the reproduced container had a capacity of 152.5㎖, whereas the original container holds approximately 168.6㎖, a figure similar to three hop (around 174㎖) in Joseon-period units of quantity. Since the capacity of the cylindrical inner container corresponds to a known measure from the late Joseon dynasty, it is likely that the water dropper was originally produced to contain a cylindrical inner container.

  • Tool wear in the shearing process such as blanking, piercing and trimming is very important, because it has great effects on the dimensional accuracy, working efficiency and economy. Most of tools in the shearing process have the coated layer at surface fur good wear and corrosion resistance. When the surface of tool is teated, the wear Phenomena of coated surface layer and inner layer may be different. This paper describes a computer modelling technique by the finite element method in order to investigate the wear mechanism and to predict the wear profile of Ti-N coated tool in piercing process according to the volume of Production. Wear coefficients of the coated layer and inner layer are obtained through Pin-on-Disk wear test, respectively. To verify the effectiveness of the suggested technique, the technique is applied to wear analysis in piercing recess of piston pin and simulation results are compared with experimental ones.
