Structure of Egg Envelope and Oogenesis of the Korean Endangered Fish Gobiobotia brevibarba (Pisces: Cyprinidae) (멸종위기 어류 돌상어 Gobiobotia brevibarba의 난자형성과정과 난막의 형태)

  • A histological study on the egg envelope and oogenesis of Gobiobotia brevibarba (Pisces, Cyprinidae) was carried out by a light microscope and a scanning electron microscope. Various developmental cells appeared in the ovary caught during May 2014, spawning season. For the relative area of oocyte, the ovary consisted of mature stage (74.5%), a vitellogenic stage (yolk granule stage, 16.6% and yolk vesicle stage, 6.6%) and previtellogenic stage (perinucleolus stage 2.2%), which means its spawning season. The cytoplasm of the perinucleolus oocyte is acidic and many nucleoli are located at the inner side of the nuclear membrane. The yolk vesicles, an early vitellogenic stage, has a follicular layer and a zona radiata clearly. Numerous villi, called an egg envelope, begin to form on the zona radiata. The yolk granules, an another vitellogenic stage, proceeds and they show a strong eosinophilic nature. Such yolk granules appeared between the yolk vesicles occupying most cytoplasm, and as the stage proceeds, there are some yolk masses fused with each other. Egg envelope is covered with plenty of villi ($2{\sim}3{\mu}m$ in the length) over the entire egg surface.

Development and Application of Cone Penetrometer with Impact Penetration Rod for Evaluation of Track Substructure (철도궤도 하부구조물 평가를 위한 타격식 관입 롯드가 체결된 콘 관입기의 개발 및 적용)

  • To minimize the cost of maintenance, repair and over-design of track substructure, an accurate evaluation of strength and stiffness of the track substructure is necessary. In this study, a cone penetrometer with impact penetration rod (CPI) is developed for the evaluation of track substructure. For applicability test, the chamber and field tests were performed. As the experimental results of the CPI, dynamic cone penetration endex (DCPI), cone tip resistance ($q_c$), friction resistance ($f_s$) and friction ratio (Fr) were obtained. In the chamber test, the experimental results show reasonable values for the simulated track substructure. In the field test, the CPI clearly detects the interface between the ballast and the subgrade. Also, discontinuous layers are detected in the subgrade. It is expected that the developed CPI may be an effective tool for the evaluation of track substructure by evaluating the ballast layer by dynamic penetration and the subgrade by static penetration of the inner rod.

Characteristics of Water Temperature and Salinity in the Bottol Bada, Yeosu during Summer in 2010 (2010년 하계 보돌바다의 수온과 염분 특성)

  • This study was determined to analysis the characteristics of water mass in the Bottol Bada, Yeosu in August, 2010 based on the data from the distribution of water temperature and salinity. Sampling was carried out a total of three times (i.e. July 29, August 13, and August 30, 2010) and performed at three stations. Observation was done during the period of time 10:00-15:00, indicating the decreasing tidal height and turn of tide. In July 29, thermocline was found at 4 m in St. 1, but the stratification did not observe in August 13 and August 30. The remarkable water temperature between surface and bottom was found in St. 2 and St. 3, whereas St. 1 did not find. A particular finding during this study showed a cold water mass at bottom layer from St. 2 and St. 3, which was first occurred in July 29 and persisted in August 30 without any of destruction. Water temperature had a remarkable fluctuation between surface and bottom, whereas salinity had a unique in St. 1. St. 2 and St. 3 showed the increasing salinity according to water depth in August 13 and August 30. Transparency had considerable fluctuations in St. 1 and St. 3 depending to sampling date, but St. 2 did not fluctuate. Consequently, the Bottol Bada had a significantly different water mass between inner and outer waters. Furthermore, strong irradiance and weak wind play an important role in developing the stratification between surface and bottom, in particular the introduction of offshore waters contribute to highly developing the stratification in the Bottol Bada during the period of August in 2010.

  • These studies were carried out to examine the anatomical, karyological characteristics, the variation of isozymes in needle and the frost damage of tissue for the purpose of investigating the relationships among the three natural Torreya nucifera populations of Cheju-do, Namhae, and Mt. Naejang in Korea. The results obtained can be summed up as follows : 1. The numbers of endodermal cells and inner layer cells of mucilage canal of needle conductive tissue were different in each population. The number of those of Cheju-do population was the largest. 2. In somatic chromosome, Namhae population showed one more secondary constriction. The values of $b^{arm}/a^{arm}$ were the same in all the three populations, but Cheju-do population particularly showed a different minimum value. And the karyotype formulas of each population showed difference. 3. During the meiosis, each population showed no significant difference in the irregular phenomena of chromosome. 4. In isoperoxidase and esterase variations of needle, each population showed its particular number and variation of bands. Cheju-do population showed the largest number and greatest variation of bands. 5. Under the same freeging conditions, the frost damage of Mt. Naejang population was comparatively slight, and Cheju-do population suffered from a greater frost damage than the others.

The Expression Pattern of the Tight Junction Protein Occludin in the Epidermal Context When Comparing Various Physical Samples (신체 부위별 표피에서 밀착연접 단백질 중 오클루딘의 발현도 연구)

  • 'Tight junctions (TJ)' have recently been identified in the granular cell layer of the human epidermis, where they contribute to the normal adhesion between keratinocytes and to the physiologic barrier function of the epidermis. Among the TJ proteins in the epidermis, occludin is an important transmembrane protein, which is considered as a major component. The purpose of this study is to investigate whether regional variation exists in the expression of the tight junction protein occludin in normal human epidermis. Indirect immunofluorescence staining for occludin was performed with specimens taken from different areas of normal skin (4 from each of 7 different anatomical sites, including the scalp, face, posterior neck, upper arm, abdomen, lower back, and inner thigh). The degrees of the expression-intensity in each specimen were estimated with the reciprocals of positive end-point titer of occludin in an indirect immunofluorescence study. The highest degree expression-intensity of the TJ protein occludin among the different areas of normal epidermis was observed on the face and abdomen with a titer of 600 (p=0.001). The lowest intensity of expression of occludin was seen in the epidermis from the upper arm. Skin specimens from the scalp, neck, back, and leg demonstrated intermediate degrees of the expression in intensity. The expression of occludin in the skin samples obtained from different locations of the body showed a statistically significant variation. This suggests that there is a certain degree of regional variation in the expression-intensity of TJ protein 'occludin' in the human epidermis.

Distribution Pattern of dominant Benthic Diatoms on the Mangyung-Dongjin Tidal Flat, West Coast of Korea (서해 만경-동진 조간대의 주요 우점 저서 규조류의 분포)

  • Marine benthic diatoms and environmental factors were studied at 60 sites on the Mangyung-Dongjin tidal flat of the west coast of Korea. Sediment samples were taken quantitatively from the upper 5 mm layer to obtain a representative estimate of the epipelic and epipsammic cell concentration. Surface sediments taken simultaneously with the quantitative diatom samples were analysed for the grain size. Exposure duration of study sites were calculated by the tide data recorded at Kunsan Outer-Harbour. Coarse sediments dominated mainly on the offshore coastal and lower tidal flat, whereas fine sediments occurred on inner and higher tidal flat. Total 371 diatom taxa were collected and the genera represented by a great number were Navicula and Nizschia. The 16 abundant species occupying more than 1% of total cell number are of the following: Paralia sulcata, Navicula sp. 1, Navicula arenaria, Cymatosira belgica, Amplora holsatica, Amphora coffeaeformis, Achnanthes hauckiana, Rhaphoneis amphiceros, Thalassionema nitzschioides. Navicula sp. 2, Dimeregramma minor, Amphora sp. 1, Cyclotella atomus, C, striata, Nitzschia kuetzingiana, Stephanodiscus sp. 1. The distribution pattern of these dominant species are described in relation to the habitat condition. Most of these species showed high densities in fine sediments. However, they occurred even silty sand and sandy sediments in low abundance. The epipsammic forms belonging to the Araphidineae and Monoraphidineae were restricted on the lower tidal flat. The typical species found in coarse sediments were: Cocconeis sp. 1, Opephora martyi, Amphora sabyii, Dimeregramma minor var. nana, Fragilaria virescens var. oblongella, F. virescens, Cocconeis grata. The higher tidal flat consisting of fine sediments showed relatively higher cell numbers than the lower tidal flat. River mouth region was the highest in abundance.

General characteristics of Antheraea yamamai silkworm cocoon cultured in Korea (천잠 누에고치의 일반 성상)

  • Antheraea yamamai silkworm cocoon is considered as a natural biocompatible materials, but can not be used as medical resources due to its difficult processability. In this study, we examined the general characteristics including cocoon shell weight and structural and thermal properties of Antheraea yamamai silkworm cocoon. The cocoon shell weight and thickness of wild silkworm cocoon was 0.528 g and 0.424 mm, respectively. The cocoon has yellow-green color in outside cocoon but white in inner layer cocoon. Amino acid analysis showed that the main amino acid of Antheraea yamamai cocoon is alanine, glycine, serine, aspartic acid, tyrosine and arginine. X-ray diffractometry showed that strong diffraction peaks at $2{\theta}=16.8^{\circ}$, $20.4^{\circ}$, corresponded to ${\beta}$-sheet structure and sharp diffraction peaks at $2{\theta}=15.0^{\circ}$, $24.3^{\circ}$, $30.0^{\circ}$ due to the presence of calcium oxalate on cocoon surface. Antheraea yamamai cocoon was degraded at $370^{\circ}C$. These results might be used as basic information for development of non-textile materials using Anthereae yamamai silkworm cocoon.

Review of Failure Mechanisms on the Semiconductor Devices under Electromagnetic Pulses (고출력전자기파에 의한 반도체부품의 고장메커니즘 고찰)

  • This review investigates the basic principle of physical interactions and failure mechanisms introduced in the materials and inner parts of semiconducting components under electromagnetic pulses (EMPs). The transfer process of EMPs at the semiconducting component level can be explained based on three layer structures (air, dielectric, and conductor layers). The theoretically absorbed energy can be predicted by the complex reflection coefficient. The main failure mechanisms of semiconductor components are also described based on the Joule heating energy generated by the coupling between materials and the applied EMPs. Breakdown of the P-N junction, burnout of the circuit pattern in the semiconductor chip, and damage to connecting wires between the lead frame and semiconducting chips can result from dielectric heating and eddy current loss due to electric and magnetic fields. To summarize, the EMPs transferred to the semiconductor components interact with the chip material in a semiconductor, and dipolar polarization and ionic conduction happen at the same time. Destruction of the P-N junction can result from excessive reverse voltage. Further EMP research at the semiconducting component level is needed to improve the reliability and susceptibility of electric and electronic systems.

Form and Material Analysis of Yuso Used in Joseon Period Scroll Paintings (조선시대 족자 장황에 사용된 유소 형태 및 재질분석)

  • Yuso is the term for decorative tassels of a braided string which hangs a scroll painting. This study, drawing on extant research concerning the yuso made for Joseon period portrait scrolls of kings and meritorious retainers, focuses on the yuso created to hang literati portraits. Concretely, It examines yuso of seven portraits in the collection of the National Museum of Korea in order to characterize their appearance and determine their material composition. The study found that most of the yuso are sixteen-strand strings braided into a rounded cross-section(dongdahoe). The seven yuso, of which six are red and one indigo-blue, reflect the popular style associated with Joseon period literati portraits. The yuso for the portrait of Yun Geup(duksu 3503) is made from gilded paper. Analysis showed Fe particles present in a red pigment underlying the gold layer, suggesting the presence of red ochre(seokganju), an iron oxide mineral. The yuso of the portrait of Shin Im(duksu 4846) is used a paper which contains gold as well as traces of Pb, Hg and Ag. The paper in the yuso for the portrait of Yi Seongwon(bongwan 10122) mainly consisted of Ag, indicating silver paper having been used in its fabrication. The inner paper in the yuso of the portrait of Yi Seogu(sinsu 1065) is a leather combined with Ag, Fe, and Br, according to chemical analysis. The FTIR of the leather sample reveals that the spectrum in the fingerprint region is nearly identical to that of sheepskin, indicating the yuso was made from gold-coated sheepskin.

Experimental Studies on Pathogenicities and Developmental Stages of Nosema apis(Zander, 1909) (꿀벌에 기생(寄生)하는 Nosema apis의 병원성(病原性)과 발육단계(發育段階)에 관한 연구(硏究))

  • Studies on pathogenicities and developmental stages of Nosema apis (Zander, 1909) were carried out through artificial infection to Nosema free honey bees with various levels of spores isolated from local honey bee colony. The results obtained were summarized as follows: 1. The clinical symptoms were observed as dysentery, enteritis of mid-gut (enlargement and decoloration), crawling posture and shortening of the longevity of worker bees in the rearing honey bee colony inoculated with the spores. 2. Number of spores harvested from laboratory rearing honey bees were progresively increased to 4 weeks after inoculation. The regression equations and coefficients of correlations to various spore levels were as follows in each treatment colony. Colony 1. ($$1,000{\times}10^4spores/ml$$) $$y_{c1}=471{\times}10^{4}x+454{\times}10^4(r=0.65^*$$) Colony 2. ($$500{\times}10^4spores/ml$$) $$y_{c2}=340{\times}10^{4}x+207.8{\times}10^4(r=0.99^{**}$$) Colony 3. ($$100{\times}10^4spores/ml$$) $$y_{c3}=150{\times}10^{4}x+84.2{\times}10^4(r=0.99^{**}$$) Colony 4. ($$10{\times}10^4spores/ml$$) $$y_{c4}=13.8{\times}10^{4}x+13{\times}10^4(r=0.98^{**}$$) 3. Average longevity of worker bees artificially infected with Nosema apis was shortened as 21.7~43.8% compare to the control. (p<.05, p<.01) 4. The spores which were isolated from honey bee colony infected with Nosema disease were ovoid or spherical form, and measured, as a rule, from $4.7{\mu}m$ to $6.1{\mu}m$ (mean $5.3{\mu}m$) in length and from $2.4{\mu}m$ to $3.2{\mu}m$ (mean $2.9{\mu}m$) in width. 5. In the mid-gut of honey bees, the spore was progresively germinated and became trophozoite stage. The trophozoites were grown to meronts and their binary fission were begun. The divided two sporoblasts were developed to the spores which had elastic membrane. The new spores were shed in excreta of honey bees 10~15 day after inoculation at $25{\pm}2$ centigrade. 6. The ultrastructure of spore membrane consisted of three layers, such as, outer, middle and inner layer. The sporoplasm consisting lamellar structure occupied only anterior part of the spore and was often extended to posterior direction where definite vacuoles and a polar filament was able to detect.
