Characterization of Levan Hydrolysis Activity of Levansucrase from Zymomonas mobilis ATCC 10988 and Rahnella aquatilis ATCC 33071

  • To investigate production and hydrolysis of levan, the levansucrase enzymes from Zymomonas mobilis ATCC 10988 and Rahnella aquatilis ATCC 33071 were used. The optimum temperature of R. aquatilis levansucrase for levan formation was $37^{\circ}C$, whereas that of Z. mobilis was $4^{\circ}C$, under the experimental conditions. Both levansucrases also catalyzed the reverse levan hydrolysis reaction. Levan hydrolysis reactions from both levansucrases were temperature dependent; high temperature ($20^{\circ}C$) was more favorable than low temperature ($4^{\circ}C$) by 4 times. Fructose was the only product from hydrolysis reaction by both levansucrases, showing that both levansucrases mediated the hydrolysis reaction of exo-enzyme acting. In both enzymes, initial levan hydrolysis activity was almost accounted to 1% of initial levan formation activity. The results allow the estimation of the fructose release rate in enzyme processing conditions.

The Influence of Collision Energy on the Reaction H+HS→H2+S

  • Quasi-classical trajectory calculations have been carried out for the reaction H+HS by using the newest triplet 3A" potential energy surface (PES). The effects of the collision energy and reagent initial rotational excitation are studied. The cross sections and thermal rate constants for the title reaction are calculated. The results indicate that the integral cross sections (ICSs) are sensitive to the collision energy and almost independent to the initial rotational states. The ro-vibrational distributions for the product $H_2$ at different collision energies are presented. The investigations on the vector correlations are also performed. It is found that the collision energies play a postive role on the forward scatter of the product molecules. There is a negative influence on both the alignment and orientation of the product angular momentum for low collision energy at low energy region. Whereas the influence of collision energy is not obvious at high energy region.

  • For the mass production of DFAIII and for the development of techniques of separation and purification of it, the methods of production of DFAIII and its recovery was investigated by fermentation with the strain of Arthrobacter ureafaciens KCTC 3387 and by enzyme reaction. In the first method, DFAIII was produced by fermentation with the strain of Arthrobacter ureafaciens KCTC 3387 and recovered from culture supernatant with silica gel gy filtration, in the second method, it was produced by enzyme reaction and recoverd with the same method of the first, and in third method it was produced by fermentation and recovered by addition of ethanol to the culture supernatnat.Against 25g/L of initial concentration of inulin, 1.57, 4.40, 0.34 g/L of powder of DFAIII was recovered respectively and the rate of recovery was 6.3, 17.6 1.4% and the purity was estimated at 81, 97, 87% respectively. For the production of DFAIII and its recovery, enzyme reaction method was the highest in the rate of recovery and its purity. By fermentation method, DFAIII was produced with 50% fo initial concentration of substrate but th rate of recovery was lower than enzyme reaction method and purity was lowest among the three methods. Ethanol pricipitation method showed the lowest rate of recovery.

  • Reaction characteristics and kinetics of a oxygen carrier (OCN717-R1SU) for chemical looping combustion (CLC) have been investigated using TGA by changing gas concentration (10-30 vol.% $CH_4$) and reaction temperature ($825-900^{\circ}C$). Reaction rate of OCN717-R1SU increased as temperature increased and it was found that reaction is delayed at the initial reaction regime. Johnson-Mehl-Avrami (JMA) model was adopted to explain the reaction phenomenon. The activation energy (E) determined by JMA model in reduction reaction of OCN717-R1SU is $151.7{\pm}2.03kJ/mol$ and pre-exponential factor and JMA exponent were also obtained. The parameters calculated in this study will be applied in design of the reactor and operation conditions for CLC process.

The extinction of unsteady counterflow diffusion flame without the retardation effect of a mixing layer (혼합층의 지연효과를 배제한 비정상 대향류 확산 화염의 소화)

  • The extinction of unsteady diffusion flame was experimentally studied in an opposing jet counterflow burner using diluted methane. The stabilized flame was perturbed by linearly varying velocity change that was generated by pistons installed on both sides of the air and fuel stream. As the results, the extinction of unsteady flame is dependent not only on the history of unsteadiness, but also on the initial condition. We found that there are several unsteady effects on the flame extinction. First, the extinction strain rates of unsteady cases are extended well beyond steady state extinction limits. Second, as the slope of the strain rate change increases, the unsteady extinction strain rate becomes larger. Third, the extension of unsteady extinction strain rate becomes smaller as the initial strain rate increases. We also found that the extension of the extinction limit mainly results from the unsteady response of the reaction zone because there is no retardation effect of a mixing layer for our experimental condition.

A Study on the Decomposition Rate of Phenol in the Batch Type Ozonation (회분식 오존 공정에서 페놀의 분해 속도에 관한 연구)

  • The characteristics of the ozone treatments of phenol were studied in a laboratory scale wastewater treatment system. The ozone treatment of wastewater was carried out in a batch-type reactor. The initial pH of wastewater(7-10), volumetric flow rate(1-2l/min) and ozone concentration(20~30 mg/l) of aerating gas were considereal as experimental variables in the ozone treatment. Phenol was decomposed easily by the ozone in a batch treatment, where the rate determining step was the COD removal that is decomposition of intermediates formed by the ozonation of phenol. Phenol decomposition and COD removal could be expressed by the first order reaction for the phenol concentration and COD, respectively. Rate constants of phenol decomposition and COD removal increased with the initial pH, volumetric flow rate and ozone concentration of aeration gas. Under the present experimental condition, their relationships could be given by for the phenol decomposition $k'=4.46\times 10^{-9}[pH]_o ^{3.94}[O_3]^{1.42}Q_{O3}^{1.57}$ for the COD removal $k=2.46\times 10^{-10}[pH]_o ^{5.19}[O_3]^{1.15}Q_{O3}^{1.19}$

Kinetics of Initial Water Vapor Adsorption by Inonotus obliquus Mushroom Powders

  • Water vapor adsorption kinetics of Inonotus mushroom powders were investigated in temperature and water activity ranges of 20 to 40$^{\circ}C$ and 0.30 to 0.81, respectively. Initial water vapor adsorption rate of mushroom powders increased with increases in temperature and water activity. The temperature dependency of water activity followed the Clausius-Clapeyron equation. The net isosteric heat of sorption increased with an increase in water activity. Water vapor adsorption kinetics of the mushroom powders can be well described by a simple empirical model. Temperature dependency of the reaction rate constant followed the Arrhenius relationship. The activation energy ranged from 56.86 to 91.35 kJ/mol depending on water activity. Kinetic compensation relationship was observed between k$_o$ and E$_a$ with the isokinetic temperature of 790.27 K.

  • The synthesis of lauryl palmitate from palmitic acid and lauryl alcohol was investigated in organic solvents using lipase. Water-immiscible organic solvent such as hexane, toluenem cyclohexane, and isooctane were found to be suitable of ester synthesis . The effect of water content on the initial rate of conversion was examined . As the content increased, the reaction rate increased. But addition of water in organic solvent decreased therostability of enzyme . The best lauryl palmitate synthesis was achieved with water content of 0.2-0.4% reaction temperature of 4$0^{\circ}C$ and 45$^{\circ}C$ for Candida cylindracea lipase porcine, pancreatic lipase, respectively. when ester synthesis was carried out under the optimum conditions, the conversion yield of palmitate into lauryl palmitate after 70hrs reached 85% and 69 % for the Candida cylindracea lipase and porcine opancreatic lipase, respectivley.

Decolorization of a Rhodamine B Using Ru-graphite Electrode (Ru-흑연 전극을 이용한 Rhodamine B의 색 제거)

  • For the RhB removal from the wastewater, electrochemical method was adapted to this study. Three dimensionally stable anode (Pt, Ir and Ru) and graphite and Ru cathode were used. In order to identify decolorization, the effects of electrode, current density, electrolyte and air flow rate were investigated. The effects of electrode material, current, electrolyte concentration and air flow rate were investigated on the decolorization of RhB. Electro-Fenton's reaction was evaluated by added $Fe^{2+}$ and $H_2O_2$ generated by the graphite cathode. Performance for RhB decolorization of the four electrode systems lay in: Ru-graphite > Ru-Ru > Ir-graphite > Pt-graphite. A complete color removal was obtained for RhB (30 mg/L) at the end of 30 min of electrolysis under optimum operations of 2 g/L NaCl concentration and 2 A current. $Fe^{2+}$ addition increased initial reaction and decreased final RhB concentration. However the effect was not high.

  • This paper deals with a degenerate parabolic system with coupled nonlinear localized sources subject to weighted nonlocal Dirichlet boundary conditions. We obtain the conditions for global and blow-up solutions. It is interesting to observe that the weight functions for the nonlocal Dirichlet boundary conditions play substantial roles in determining not only whether the solutions are global or blow-up, but also whether the blowing up occurs for any positive initial data or just for large ones. Moreover, we establish the precise blow-up rate.
