Streptococcus salivarius subsp. thermophilus가 생산하는 Bacteriocin의 특성 (Characteristics of the Bacteriocin Produced from Streptococcus salivarius subsp. thermophilus)

  • One bacterial strain, that had made the largest inhibition zone at the antagonism assay and also that lost the inhibition activity by the protease treatment, was isolated from raw milk. That strain was identified as Streptococcus salivarius subsp. thermophilus. The specific growht rate of this strain was maximum at 45$\circ $C. However, at this temperature the strain produced no bacteriocin. The bacteriocin activity was quite stable even at high temperature. Moreover, the activity of the vacteriocin was sensitive to proteases. but not to $\alpha $-amylase, DNase I, or RNase.

  • Attempts were made to investigate the antagonistic activity of soil borne microorganisms Pseudomonas spp. and Trichoderma spp. against to the pathogens of turf diseases Rhizoctionia solani spp. and Pythiom spp. in vitro by a dual culture bioassay. Inhibition zone between the edge of the my-celium and the margin of each antagonistic bacteria, Pocudontonas, on potato dextrose agar was measured 3 days after incubation at 28˚C. Psudomonas spp. showed relatively high inhibition of mycelium growth of R. solani AG-i and Pythium spp. which cause brown patch and pythium blight, respectively. Antagonistic fungi Trichodenma spp. also showed effective inhibition against mycelium growth of both pathogens, more proper methods of measuring the inhibition effects were required because of fast growth of Trichodenna hypae. Brown patch and pythium blight both, re-quire most higher rate of fungicide use to control in golf curses in Korea. Application of antagon-istic microorganisms are useful as biological resources an approach to sole environmental contamination.

간질에서의 핵의학 영상 (Nuclear Imaging in Epilepsy)

  • Correct localization of epileptogenic zone is important for the successful epilepsy surgery. Both ictal perfusion single photon emission computed tomography (SPECT) and interictal F-18 fluorodeoxyglucose positron emission tomography (FDG-PET) can provide useful information in the presurgical localization of intractable partial epilepsy. These imaging modalities have excellent diagnostic sensitivity in medial temporal lobe epilepsy and provide good presurgical information in neocortical epilepsy. Also provide functional information about cellular functions to better understand the neurobiology of epilepsy and to better define the ictal onset zone, symptomatogenic zone, propagation pathways, functional deficit zone and surround inhibition zones. Multimodality imaging and developments in analysis methods of ictal perfusion SPECT and new PET ligand other than FDG help to better define the localization.

생약추출물로부터 생리활성의 검색 (Screening for Biological Activity of Crude Extracts from Medicinal plants)

  • 뉴질랜드의 자생식물인 Brachyglottis monroi와 Trichocolea hatcheri의 에탄올추출물에 대한 생리활성을 조사한 결과, 그람음성균인 Bacillus subtilis (ATCC 19659, 1 mm inhibition zone at $150{\mu}g/disc$)와 피부곰팡이균인 Trichophyton mentagrophytes (ATCC 28185, 6 mm inhibition zone at $150{\mu}g/disc$)에 대한 억제효과가 관찰되었으며, 이들 추출물은 P388 murine leukaemia cells ($IC_{50}$ $23.96{\mu}g/mL$)에서도 세포독성이 나타났다. B. monroi의 에탄올추출물의 항바이러스효과는 T. hatcheri의 에탄올추출물보다 강한 활성이 관찰되었으며, T. hatcheri의 에탄올추출물의 항균효과는 B. monroi의 에탄올추출물보다 강한 활성을 나타났다. 그렇지만 P388 murine leukaemia cells ($IC_{50}>12,500{\mu}g/mL$)에 대한 세포독성은 아주 미미한 것으로 나타났다.

상황미를 이용한 항로타바이러스 효과와 항균성 측정에 관한 연구 (Studies on the Anti-rotaviral and Anti-bacterial Effects of Phellinus linteus Mushroom Rice)

  • 본 연구에서는 상황미 추출물을 이용하여 병원성 식중독 균(S aureus 305, L. monocytogenes ATCC 19114, E. coli 0157:H7 ATCC 42894, E. coli O55)에 대한 항균효과와 human rotavirus(KU, S2, YO, K-21)에 대한 억제효과를 알아보기 위해서 수행되었다. 그 결과는 다음과 같다. 항균성 테스트에서 S. aureus 305는 18mm, L. monocytogenes ATCC 19114는 20 mm, E. coli O55는 13mm의 inhibition zone이 생성되었으나, E. coli O157:H7 ATCC 42894는 inhibition zone이 생성되지 않았다. MTT assay 방법에 따라 실시한 l/3% 상황미 추출물의 human rotavirus에 대한 억제효과는 K-21이 $97.56{\pm}12.50%$, S2가 $94.74{\pm}8.68%$ YO가 $59.77{\pm}8.68%$, KU가 $90.52{\pm}18.42%$로 각각 나타났다. 이러한 결과로 보아 상황미 추출물은 병원성 식중독균과 다양한 유아 rotavirus 모두에게 억제 효과가 있는 것으로 확인되었다.

잔디 근권에서 분리된 Actinomycetes균주의 생리학적 특성 (Physiological Characteristics of Actinomycetes Isolated from Turfgrass Rhizosphere)

  • 방선균을 대상으로 생물적 방제 균주를 선발한 결과 총 248개의 균주가 분리되었다. 분리된 균주의 셀룰로스 분해능력은 S11 균주와 S4 균주의 투명환의 크기는 약 1.2mm로 셀룰로스 분해 능력이 가장 좋은 것으로 나타났다. 키틴분해 능력을 검정한 결과 S12 균주의 투명환의 크기가 2.1mm로 가장 키틴분해 능력이 우수한 것으로 나타났다. Skim milk agar 배지를 이용하여 15개 균주의 단백질 분해 능력을 검정한 결과 S2 균주가 접종된 배지의 투명환의 크기가 7.5mm로 단백질 분해 능력 가장 우수한 것으로 나타났다. Chrome azurol S agar 배지를 이용하여 병원균에 대한 중요한 항균 기작으로 알려져 있는 siderophore 생성정도를 조사한 결과 S1 균주의 투명환의 크기가 0.6mm으로 가장 높게 나타났다. 선발된 4개 균주가 분비하는 휘발성 물질의 달라스팟과 라지패취 병원균에 대한 항균활성 능력 검정한 결과 달라스팟의 경우 S8 균주가 94.8% 억제하는 것으로 나타났다. Rhizoctonia solani AG2-2균사생장에 미치는 영향은 S8 균주가 72.9%로 가장 높게 억제하였으며 다음으로 S2 균주가 62.1%로 S5 균주가 37.8%로 S12 균주가 27% 억제하는 것으로 나타났다.

수종 근관 충전재의 항균 효과에 관한 연구 (ANTIMICROBIAL EFFECT OF ROOT CANAL CEMENTS ON MICROORGANISMS FROM INFECTED ROOT CANALS)

  • The purpose of this study was to evaluate the antimicrobial effect of root canal cements such as AH-26, Sealapex, Canals and Apatite root sealers(Type I, II, III) and to determine the efficacy of their activities. S.mutans(AHT), S.sanguis(NCTC 9811) and B.gingivalis(381) were streaked on blood agar and the PVC tybes filled with root canal cements were applied on. Then the microorganisms were cultured for 48 hours, anaerobically. B.cereus(KCTC 1012) was streaked on nutrient agar, PVC tubes were applied on and were cultured for 48 hours, aerobically. The inhibition zones of root canal cements were measured with vernier caliper. The data statistically analyzed, and the results were as followed. 1. Apatite root sealers(Type I, II, III) showed no inhibition zones. 2. AH-26, Sealapex and Canals had inhibition zones with varying degrees. The inhibition zone of AH-26 was greatest and followed by Canals and Sealapex(P<0.01). 3. As time goes by after mixing the root canal cements, AH-26, Canals and Sealapex showed significantly reducing inhibition zones(P<0.01). 4. There were the least inhibition zones of all the root canal cements on S.mutans and followed in such order as; B.gingivalis, S.sanguis and B.cereus(P<0.01).

상온에서 보관한 반코마이신 함유 항생제 시멘트의 메티실린 내성 포도상구균에 대한 항균력 조사 (In Vitro Study Evaluating the Antimicrobial Activity of Vancomycin-Impregnated Cement Stored at Room Temperature in Methicillin-Resistant Staphylococcus aureus)

  • Purpose: Treatment of diabetic foot infection due to methicillin-resistant Staphylococcus aureus (MRSA) remains challenging. Applying vancomycin-impregnated cement is one of the best methods of treatment. Vancomycin-impregnated cement has been used worldwide; however, to date, there is a limited number of studies regarding its use. We evaluated the duration of antimicrobial activity of vancomycin-impregnated cement stored at room temperature after manufacturing. Materials and Methods: The vancomycin-impregnated cement was manufactured by mixing 1 g of vancomycin with 40 g of polymer and adding 17.90 g of liquid monomer. The cement dough was shaped into flat cylinders with diameter and height of 6 mm and 2 mm, respectively. Another cement of the same shape without mixing vancomycin was prepared as the negative control. All manufactured cements were sterilized with ethylene oxide gas and stored at room temperature. Each cement was placed on Mueller Hinton agar plate lawned with standard MRSA strain. Standard vancomycin disk and gentamicin disk were placed together. After 24 hours, the diameter of inhibition zone was measured, and if the diameter was less than 15 mm, vancomycin-impregnated cement was regarded as a loss of antimicrobial activity. The study was repeated every 2 weeks until vancomycin-impregnated cements lost their antimicrobial activity. Results: Vancomycin-impregnated cement stored for a duration of 16 weeks created a 14 mm inhibition zone, while vancomycin disk created a 15 mm inhibition zone. Vancomycin-impregnated cement stored for a duration of 17 weeks created 7 mm and 9 mm inhibition zones, while vancomycin disk created 16 mm and 15 mm inhibition zones, respectively. Conclusion: We found a decrease of antimicrobial activity in vancomycin-impregnated cements after 16 weeks. After 17 weeks, they showed definite loss of antimicrobial activity. Therefore, we recommend not using vancomycin-impregnated cement spacers that has been stored for more than 16 weeks at room temperature.

Effect of Kimchi and Its Ingredients on the Growth of Helicobacter pylori

  • Effects of kimchi and its ingredients, vitamin C and $\beta$-sitosterol on the growth of Helicobacter pylori were investigated. Three kimchi variations were studied: a standard recipe (kimchi I) and two functional variations for cancer prevention and treatment made with organically grown ingredients (kimch II and III). Methanol extracts and juices from kimchi I and III did not inhibit the growth of H. pylori. However, 10 mm and 12 mm inhibition zones were formed by methanol extract and juice from kimchi II, which had higher concentrations of red pepper powder (RPP) than those of kimchi I and III. Among the major kimchi ingredients, methanol extracts of RPP, garlic and ginger substantially inhibited the growth of H. pylori. The maximal inhibition zone (30 mm) was attained with garlic treatment. Inhibitory effects of the RPP, garlic and the sub-ingredient mixture (prepared with radish, garlic, RPP, ginger, green onion, sugar and fermented anchovy juice) on H. pylori were decreased by lactic acid bacteria fermentation. Neither the fermented garlic nor the fermented sub-ingredient mixture inhibited the growth of H. pylori. But, the inhibition Bone of fermented RPP was 12 mm, which was less than the 16 mm inhibition zone formed by the non-fermented RPP. Vitamin C and $\beta$-sitosterol which are known to be functional active compounds of kimchi also showed no inhibitory effect on the growth of H. pylori after 3 days of incubation. Further study is needed to determine why the inhibitory effect is removed or decreased by lactic acid fermentation, and to determine if fresh kimchi and lactic acid bacteria of kimchi can inhibit the growth of H. pylori.

마늘 추출물의 항균, 항고혈압 및 항암활성 (Antimicrobial, Antihypertensive and Anticancer Activities of Garlic Extracts)

  • 국내산 마늘과 중국산 마늘을 methanol, ethyl acetate, chloroform. hexane으로 추출하여 항균, 항 곰팡이, 항암, 항고혈압 활성을 조사하였다. 그 결과 추출수율은 methanol추출물이 7.9-8.8 brix의 농도로 가장 높은 추출수율을 보였다. 마늘 용매추출물을 E. coli, B. subtilis, S. aureus, P. aeruginosa 및 S. typhimurium균주에 대해 항균활성을 조사한 결과, ethyl acetate와 chloroform추출물이 B. subtilis, P. aeruginosa에 대해 8-16mm의 clear zone으로 강한 항균활성을 나타내었으며, A. niger, M. miehei, P. rugullosum, A. oryzae, T. reesei균주의 항 곰팡이 활성은 ethyl acetate, chloroform추출물이 A. niger M. miehei, T. reesei에 대해 8-30mm의 clear zone으로 좋은 활성을 나타내었다. ACE 저해 활성은 완도 65.6%, 단양 60.4%, 서산 55.3% 및 중국산 70.1%으로 methanol추출물이 다른 추출물에 비해 높은 활성을 나타내었다. 마늘 용매추출물의 암세포 성장 억제효과는 methanol 추출물이 위암세포(SNU-1)에 대해 완도 42.3%, 단양 53.8%. 중국산 50.4% 및 서산 54.5%의 효과를 나타내었다. 또한 자궁암세포(HeLa)에 대해서는 hexane추출물에서 각각 완도 11.3%,단양 20.2%, 중국산 36.6% 및 서산 30.5%의 활성을 나타내었다.
