Inhibition of Growth and Collagenase Activity of the Extract from Salvia miltiorrhiza against Microorganisms Causing Periodontal Diseases (단삼(Salvia miltiorrhiza) 추출물의 치주질환유발 세균의 생육억제 및 Collagenase 저해 활성)

  • This study was carried out to evaluate the inhibition of growth and collagenase activity of Salvia miltiorrhiza Bunge against microorganisms causing periodontal diseases. The ethanol extract of Salvia miltiorrhiza showed sig-nificant growth inhibition against microorganisms causing periodontal diseases. Ethanol extract was further fractionated with organic solvents in the order of hexane, chloroform and ethyl acetate. Among the fractions tested, the hexane fraction showed the highest cell growth inhibition. The minimal inhibitory concentration (MIC) of the extract against C. curvus, C. rectus, E. corrodens, F nucleatum, P. gingivalis, P. intermedia and W. succinogenes were 200, 50, 50, 250, 150, 250 and 200 ${\mu}g$/ml, respectively. The inhibition of collagenase activity by organic solvent fractions were higher than that of minocycline, and the inhibition ratio of collagenase activity was $88.2{\pm}2.1$ % in the chloroform fraction.

Inhibitory Effect of Mugwort Extracts on Tyrosinase Activity (쑥 추출물의 Tyrosinase 효소활성에 미치는 억제효과)

  • To determine the inhibition of mushroom tyrosinase activity, fresh and dried mugwort, Artemisia princeps was extracted initially with water and ethyl alcohol, and subsequently with hexane, chloroform and ethyl acetate in that order. The highest yield was obtained from the ethyl acetate (15.2%) and hexane (15.5%) fraction of the ethanolic extract of fresh and dried mugwort, respectively. For all fractions tested, the inhibition of tyrosinase activity by fresh mugwort was higher than that of dried mugwort, and the inhibition ratio of tyrosinase activity was 98.9% in the chloroform fraction and 96.7% in the hexane fraction.

Antibiotic, Antioxidant and Whitening effects of Jwa Kum-Whan and Soo Ryeon-Whan (좌금환과 수련환의 항균, 항산화 및 미백 효과)

  • Objectives : The purpose of this study was to research the antibiotic, antioxidant and whitening effects of Jwa Kum-Whan(JKW) and Soo Ryeon-Whan(SRW). Both Jwa Kum-Whan and Soo Ryeon-Whan are composed of Coptidis rhizoma and Evodiae fructus, but the ratios of the two species are different. JKW is composed of Coptidis rhizoma and Evodiae fructus by ratio of 6:1 and SRW's ratio is 1:1. Methods : Antibiotic activities of JKW and SRW's water extracts were studied by paper disc diffusion method. Four kinds of bacteria were applied in paper disc, and each extract was dropped, individually. The diameter of inhibition zone was measured. DPPH assay was used for studying free radical scavenging activity. DPPH is quantitatively decolorized from purple color by antioxidant material. Change of color was measured by spectrophotometer. Whitening activity was analyzed by tyrosinase inhibition assay. Tyrosinase is major enzyme for control process of making eumelanin. Optical Density was measured by spectrophotometer. Results : Candida albicans and Staphylococcus aureus's inhibition zone were made large available by Both prescriptions. Inhibition zone's diameter of JKW was preferable to SRW's. Radical scavenging activity was better at SRW, but JKW's activity was available, too. Tyrosinase inhibition activity was found out available only for JKW, not for SRW. Conclusions : JKW had antibiotic effects for Candida albicans, Staphylococcus aureus. And had antioxidant, whitening effects. SRW had antibiotic, antioxidant activities but not whitening effect.

Cytotoxicity of Hyaluronic Acid Membrane Cross-linked with Lactide (락타이드로 가교시킨 히아루론산 막의 세포독성)

  • The biodegradable hyaluronic acid(HA) membranes cross-linked with lactide using the crosslinking agent, 1-ethyl-3(3-dimethyl aminopropyl) carbodiimide(EDC) were prepared as a potential biocompatible material for tissue engineering. HA membranes having different mechanical properties were synthesised by varying degree of the mole ratio of lactide to HA, EDC concentration, and crosslinking temperature. HA membranes were degradable in water solution and the degradation became slower with the increasing mole ratio of lactide to HA. HA membranes were sterilized using ethylene oxide gas and extracted with cell culture medium for 24 h at $37^{\circ}C$ and 200 rpm. Cytotoxicity of the extract was tested using NIH/3T3 mouse embryo fibroblast as a model cell. Growth inhibition was not observed in the extracts of HA membranes with the mole ratios of lactide to HA, 5 or 10, and 10% EDC concentration, however 11% of growth inhibition was observed in the extract with the mole ratio of 13. Growth inhibition was not observed in the extracts of HA membranes prepared with 5% EDC or 10% EDC and the mole ratio of lactide to HA, 10, however 12% of growth inhibition was observed in the extract with 20% EDC. Cytotoxicity was not observed in the extracts of HA membranes prepared at varying crosslinking temperatures, $15^{\circ}C,\;25^{\circ}C,\;and\;28^{\circ}C$ with the mole ratio of lactide to HA, 10 and 10% EDC.

  • To investigate antitumor component of Lyophyllum decastes, the aqueous extract of its shake-cultured mycelia was subjected to antitumor test against sarcoma 180 cells implanted in ICR mice. The extract showed an inhibition ratio of 65.4% and was found to consist of a polysaccharide moiety and a protein moiety. After purification with DEAE-Sephadex A-50 ion exchange chromatography, Fraction D showed the highest inhibition ratio of 75.7%. The antitumor constituent was examined for immunoaccelerating activity and was found to increase macrophage accumulation in the peritoneal cavity and plaque forming cells of the spleen cells. It was named lyophyllan after the genus name.

Scavenging Effects of Lonicera Japonica Extracts on Paraquat Induced Toxicity(IV) (Paraquat 유도독성에 대한 금은화 엑스의 효과(IV))

  • Scavenging effects on paraquat induced toxicity were investigated by using methanol (MeOH) and ethylacetate (EtoAC) extracts of Lonicera japonica. The results are summerized as follows: 1. To Fe(III)-ADP-NADPH induced microsomal lipid peroixdation, MeOH and EtoAC extracts showed antioxidative activiies and inhibition ratio at 100 $\mu\textrm{g}$/$m\ell$ 44.4% and 73.8% respectively 2. To microsomal NADPH dependent cytochrome p -450 reductase in rat liver, MeOH and EtoAC extracts inhibited the enzyme activiies and inhibition ratio were 26.3% and 44.8% respectively. 3. Administration (30 mg/kg, iv) of paraquat to rats caused the marked elevation of GOT, GPT, LDH, ALP in the serum and lipid peroxides in the microsome as compared to the control group. Serum GTP, LDH, ALP and liver microsomal LPO were reduced significantaly by administration of MeOH extract. (1,000 mg/kg), EtoAC extract (40 mg/kg) and Silymarin (150 mg/kg) as compared to the paraquat group. From the results, MeOH and EtoAc exuacts. of Lonicera japonica showed the useful scavenger and reducer on the paraquat induced hepatotoxicty.

  • The action of acetylcholine on the sodium plus potassium activated ATPase activity in the rabbit red cell membrane has been investigated and the experiments were also designed to determine the mechanism of action of acetylcholine on the ATPase activity. The following results were observed. 1. The activity of the NaK ATPase from red cell membrane is inhibited by acetylcholine. 2. The ratio of inhibition of NaK ATPase by acetylcholine is decreased by raising the potassium concentration, and is increased by raising the sodium concentration. 3. The ATPase activity is increased by small amounts of calcium but inhibited by larger amounts. The ratio of inhibition of the enzyme by acetylcholine is increased by raising the calcium concentration. 4. The inhibitory action of acetylcholine on the NaK ATPase activity was not related to the sulfhydryl group of cysteine, the hydroxyl group of threonine, or the carboxyl group of aspartic acid. 5. The inhibitory action of acetylcholine on the ATPase activity is due to amino group of the enzyme of NaK ATPase.

Antitumor Components of Korean Basidiomycetes

  • To find antitumor components in the shake-cultured mycelia of Volvariella bombycina, the mycelia were extracted with hot water. After the extract was dialyzed and freeze-dried, a protein-polysaccharide fraction was obtained and examined for antitumor activity against the solid form of sarcoma 180 in ICR mice. It showed 60.3% inhibition ratio at a dose of 20mg/kg/day for 10 days. It was found to consist of a polysaccharide moiety and a protein moiety. After gel filtration on Sepharose 4B, Fraction B was obtained and showed the highest inhibition ratio of 71.1%. When the antitumor component was examined for immunopotentiating activity, it was found to increase the macrophage accumulation in the peritoneal cavity as well as the antibody production of the spleen cells of the mice.

  • To find antitumor components in the hot water extract from the basidiocarps of Hypsizigus mamomus. protein-bound polysaccharides were purified and fractionated by DEAE-cellulose ion exchange column chromatography and Sepharose CL-4B gel filtration chromatography. When a dose of 20 mg/kg/day was injected intraperitoneally into ICR mice. fraction IV of the component showed the highest inhibition ratio of 73.8% against the count of hemolytic plaque forming cells in mice to 3.2 times. when IV was about 30 KD and the fraction was composed of 76.1% polysaccharide and 4.9% protein. The hexosamine was detected in all the fractions, showing that the polysaccharide and protein moieties were bound each other.

Anticancer Effect of Extracts from the Marine and Salted Fish Products. (수산물 및 수산 발효식품의 암세포 억제효과)

  • This study was performed to observe the cytotoxic effect of the various salted fish extracts against cancer cell line, human hepatocellular carcinoma (HepG2) using MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide) method. Urechis unicinctus was the strongest cytotoxic effects among any other traditional salted fish products. The growth inhibition ratio of Urechis unicinctus hot-water extracts was 94.5% at the concentration of 1000$\mu\textrm{g}$/$m\ell$. On the other hand, in case of salted fish methanol extracts, salt-fermented shad gizzard was showed the strongest cytotoxic effects. The growth inhibition ratio of salt-fermented shad gizzard methanol extracts was investigated 90% at the concentration of 1000$\mu\textrm{g}$/.$m\ell$.
