천연물로부터 $5{\alpha}-Reductase$ 활성억제 및 면포용해효과의 검색 (Screening of $5{\alpha}-Reductase$ Inhibition and Comedolytic Effects from Natural Products)

  • The antibacterial activity against Propionibacterium acnes (P. acnes), $5{\alpha}-reductase$ inhibition and comedolytic effects are the important pharmacological target sites of antiacne drughs. We previously reported on the antibacterial activities against P. acnes by natural products. In the present study the screening of $5{\alpha}-reductase$ inhibition and comedolytic effects from natural products were performed. Seven natural products such as Angelica koreana, Sophora flavescens, Prunus persica, Bombyx mori, Areca catechu, Galla rhois and Gleditschia koraiensis perfectly inhibited the activity of $5{\alpha}-reductase$ at the concentration of 0.01% (w/v). Sixteen natural products which were shown to have the potent antibacterial activities against P.acnes or $5{\alpha}-reductase$ inhibition activities were assayed for the comedolytic test. In the results of comedolytic effects on experimentally-induced comedones (EIC), Sophora flavescens showed the strongest comedolytic effect on EIC, and Polygonum cuspidatum and Angelica koreana showed stronger comedolytic effects on EIC than azelaic acid used for a positive control at the concentration of 3% (w/v). These results suggest that several natural products including Sophora flavescens can be developed as noble antiacne agents.

The pathogenesis of acne involves various factors; excessive androgen, excessive sebum production, abnormal alteration of follicular epithelium, proliferation of Propionibacterium acnes, and inflammation. We investigated acne therapy using oriental herbs described in the Korean traditional medical book(Dong-ui-bo-gam). Oriental herbs(Angelica daurica, Arctium lappa, Coptidis rhizoma, and Glycyrrhiza glabra) were chosen based on their respective property of sebum control, anti-inflammatory activity, and anti-bacterial activity. We examined the effect of acne treatment, in terms of chemotactic inhibition, lipogenesis inhibition, and anti-bacterial activity for P. acnes. 1. Neutrophil chemotaxis assay; P. acnes secrete chemotactic factors and other pro-inflammatory extracellular products. Neutrophil chemotactic activity of P. acnes was measured by 48-well chemotaxis method. Angelica daurica clearly suppressed chemotactic activity of P. acnes. 2. Using sebaceous gland of hamster ear lipogenesis assay; Sebaceous lipogenesis was measured using ear biopsies by incubation or $C^{14}$-acetate in culture media. The $C^{14}$-labeled lipids were extracted and determined by liquid scintilation counting. Coptidis rhizoma markedly inhibited sebum production. 3. Anti-bacterial assay for P. acnes(MIC test); Glycyrrhiza glabra showed anti-bacterial activity. P. acnes did not develop resistance against Glycyrrhiza glabra. Retinoids are effectively to inhibit sebum production and regulate follicular keratinization process, with little anti-inflammatory activity. Angelica daurica suppressed neutrophil chemotaxis, Coptidis rhizoma inhibited sebum production, and Glycyrrhiza glabra showed anti-bacterial activity against P. acnes. A combined formulation of Angelica daurica, Coptidis rhizoma. and Glycyrrhiza glabra is expected to provide effective acne treatment.

현호색(玄胡索)과 초초현호색(醋炒玄胡索)의 여드름균에 대한 항균효과 비교 (Comparison of Antimicrobial effects of Corydalis tuber and processed Corydalis tuber against Propionibacterium acnes)

  • Objectives : Corydalis tuber has been used for promoting blood circulation and for relieving pain in Oriental medicine. In the present study, we focused on the antimicrobial activity of Corydalis tuber and compared its antimicrobial activity with the processed Corydalis tuber. Methods : Processing of Corydalis tuber was accomplished by immersing in 5% of acetic acid for 12 h and then by roasting at $250^{\circ}C$ for indicated time periods(0-30 min). Minimum inhibitory concentration(MIC) and the zone of growth inhibition were determined against Propionibacterium acnes(P. acnes). Results : The methanolic extracts of Corydalis tuber showed potent antimicrobial effect(MIC 62.5 ${\mu}g/ml$). Its alkaloidal component, dehydrocorydaline, also exhibited antibacterial activity(MIC 25.0 ${\mu}g/ml$). After processing of Corydalis tuber, its inhibitory effect on the growth of P. acnes was significantly enhanced compared with that of unprocessed Corydalis tuber. Furthermore, elevated content of dehydrocorydaline was found in the processed than the unprocessed Corydalis tuber. However, the different roasting minutes effected on antimicrobial activity. The best roasting time of Corydalis tuber was 10 min, while roasting for the time above 15 min resulted in diminishing antimicrobial activity. Thus, it was concluded that the standardized processing condition of Corydalis tuber should be established to obtain enhanced antimicrobial(P. acnes) activity. Conclusion : For antimicrobial effect against P. acnes, the best processing condition of Corydalis tuber is immersing in 5% of acetic acid for 12 h and by roasting at $250^{\circ}C$ for 10 min.

목향(木香) 에탄올추출물의 항균효과(抗菌效果) (Antibacterial Activity of Ethanol Extract of Saussurea lappa)

  • Objective : The purpose of this study is to investigate the antibacterial activity of Saussurea lappa for the applications of herb-based extracts to both cosmetic and medicinal industries. Methods : Antibacterial activity of ethanol extract of Saussutes lappa (ESL) was assessed using agar diffusion and broth dilution methods, and determined by whether clear zone was formed around paper disc and in terms of the size (mm) of clear zone, Results : ESL provided activity against Staphylococcus aureus KCTC 1927 in concentration of 1 mg/ml with an clear zone of 16 mm, and showed an activity against Staphylococcus epidermids KCTC 1917 in concentration of 1 mg/ml with an clear zone of 18mm. Moreover, the minimum inhibitory concentration (MIC) of ESL against S. sureus and S. epidermidls were 1 mg/ml for both bacterial species. However, ESL showed no growth inhibition against Pseudomonas aeruginosa KCTC 12513, Enterobactet aerogenes KCTC 2190, Escherichia coli KCTC 2571, Salmonella typhimurium KCTC 1925 and Propionibacterium acnes KCCM 41747. Conclusions : Antibacterial activity of ESL against Staphylococcus sureus and Staphylococcus epidermids causing eye and skin diseases was proved. The result suggest that ESL may be useful as a natural preservative on behalf of synthetic preservatives.

  • The effects of four extracts of medicinal herbs, Rubus coreanum, Acorus calamus, Morus alba and Rehmannia glutinosa on hair growth activity and acnes control were investigated. In the course of screening natural extracts for hair growth, we found that the extract of dried root of Rubus coreanum has the hair growth promoting effect. After topical application of these extracts to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. The growth of dermal papilla cells and mouse vibrissae hair follicle cultured in vitro, however, was not affected by treatment of these extracts. Furthermore these extracts do not possesspotent inhibitory effect on $5{\alpha}-reductase$ I and II activity and anti-bacterial effect on Escherichia coli , Propionibacterium acnes, Pityrosporum ovale, Staphylococcus aureus, Staphylococcus epidemidis, and Candida albicans. RT-PCR analysis showed that these extracts did notinduce mRNA levels of growth factors such as insulin-like growth factor-I, keratinocyte growth factor, hepatocyte growth factor and vascular endothelial growth factor in dermal papilla cells. These results suggest that Rubus coreanum has hair growth promoting effect. However, the effects of these materials on the hair growth promotion are not mediated through inhibition of $5{\alpha}-reductase$ I and II activity, stimulation of hair follicle cells and expression of growth factors in the dermal papilla cells.

苦蔘抽出物이 毛髮成長 促進 및 面疱 抑制에 미치는 영향 (Studies on the effect of Sophora flavescens extract on the hair growth stimulation and acne inhibition)

After topical application of Sophora flavescens extract to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. In addition, the Sophora flavescens extract revealed to possess potent inhibitory effect on $5{\alpha}$-reductase Ⅰ and Ⅱ activity. Furthermore, Sophora flavescens extract also showed anti-bacterial effect on Propionibacterium acnes. These results suggest that Sophora flavescens can be used as a potent treatment agent for helping hair growth stimulation and acne inhibition.

적하수오 및 백하수오 추출물의 항산화 및 항균 활성 (Antioxidant and antimicrobial activities of Jeok Hasuo (Polygonum multiflorum Thunb.) and Baek Hasuo (Cynanchi wilfordii Radix) root extracts)

  • 본 논문은 국내 적하수오 및 백하수오 추출물의 항산화능 및 항균활성을 구명하기 위한 목적으로 진행하였다. 각 하수오의 열수, 에탄올, 부탄올 추출물에 대하여, 일반성분을 분석한 후, 항산화능에 대해서는 총 페놀함량 및 DPPH 라디칼 소거능을 측정하였으며, 항균활성에 대해서는 Staphylococcus aureus 및 Propionibacterium acne에 대한 최소생육저해농도(minimum inhibitory concentration, MIC) 및 생육저해환을 통하여 평가하였다. 적하수오 및 백하수오 부탄올 추출물의 총 페놀함량은 각각 1,212.6 및 1,454.5 mg/g GAE로 다른 유기용매 추출물에 비해 유의적으로 높은 함량을 보였다. 이에 비해 DPPH 라디칼 소거능은 열수추출물에 비해 에탄올 추출물(89.0%), 부탄올 추출물(88.9%)이 유의적으로 높은 값을 나타내었다. MIC 측정 결과, 적 백하수오 에탄올 추출물만이 0.8 mg/mL로 S. aureus와 P. acne에 대해서 항균효과를 나타냈다. 디스크 확산법 측정 결과, 모든 유기용매 추출물이 5 mg/disc 농도부터 생육저해환을 형성하였으며, 농도가 증가함에 따라 생육저해환의 크기도 증가하였다. 적하수오 및 백하수오 에탄올 추출물의 조사균주에 대한 생육저해완의 직경은 10 mg/mL 농도에서 각각 8.9 및 9.2 mm를 나타내, 추출물 중 가장 우수한 항균활성을 나타냈다. 본 연구는 하수오 추출물이 항산화 효과 뿐만 아니라 피부 건강과 관련된 세균에 대해 항균활성을 보여줌으로써, 하수오 추출물이 피부 건강을 위한 천연 유래의 기능성 화장품 소재로도 활용될 수 있는 잠재적 가능성을 제시하였다.

백두옹(白頭翁) 및 선퇴(蟬退) 추출 혼합물의 분자 개선 효과 (Acne-remedy Effects of Extract Mixture of Pulsatillae Radix and Cicadidae Periostracum)

  • Pulsatillae Radix and Cicadidae Periostracum have long been used for treatment of injuries, inflammations or itchiness in Korea. This study was carried out to examine the acne-remedy effects of the mixture made with equal parts of Pulsatillae Radix extract and Cicadidae Periostracum extract (PRCP) compared with 2%-benzoyl peroxide (Standard) used commercially as anti-acne. The results showed the mixture (PRCP) was more effective than each crude drug extract. The 10%-PRCP showed significant antimicrobial activity against Propionibacterium acnes, the major cause of acne, and also showed significant inhibition from type 1 $5{\alpha}$-reductase ($5{\alpha}R$), another cause of acne, And the cream made with PRCP demonstrated anti-sebum and acne-improvement effects on examinee's facial skin.

토사자(菟絲子)의 추출용매에 따른 항염, 항산화 및 항균 효과에 대한 비교 연구 (The Comparative Study of Anti-inflammatory, Antioxidant and Antibacterial Effects with Regard to the Extraction Solvents of Cuscutae Semen)

  • Objective : This study was performed to compare anti-inflammation, anti-oxidation and anti-bacterial effects of Cuscutae Semen(CS) extracted with two kinds of solvents, ethanol and distilled water. Methods : Two kinds of CS extractions were prepared 20, 50, 100 ${\mu}l/mg$. The cytotoxicity was measured by MTT assay in Raw 264.7 cell. The anti-inflammation effect was measured by inhibitory efficacy of NO Production in Raw 264.7 cell. The anti-oxidation effect was measured by DPPH Radical scavenging ability in HaCaT cell. The anti-bacterial effect was measured by inhibition zone diameter on Propionibacterium acnes. Results : 1. Two kinds(100 ${\mu}l/mg$) of CS extraction groups had 50% cytotoxicity in Raw 264.7 cell. 2. All of CS extraction groups were not showed significantly inhibitory effect on NO production. 3. All of CS extracted with ethanol only showed dose-dependently significantly scavenging effect of DPPH radicals. 4. Two kinds of CS extractions did not have a inhibitory effect on Propionibactrium acnes. Conclusion : Two kinds(100 ${\mu}l/mg$) of CS extraction groups have 50% cytotoxicity. Two kinds of CS extractions have not the inhibitory effect on NO production and Propionibactrium acnes. CS groups extracted with ethanol only have a significantly scavenging ability of DPPH radicals. This study suggests that CS extracted with ethanol was effective in anti-oxidation.
