접촉성 피부염 마우스 모델에서 단핵구의 유출 및 염증 반응에 대한 CD99-유래 펩타이드 CD99CRIII3의 억제 효과 (Inhibitory Effects of CD99-derived Peptide CD99CRIII3 on the Extravasation of Monocytes and Inflammatory Reactions in Contact Dermatitis Mouse Model)

  • 접촉피부염은 피부가 이물질과 접촉함으로써 일어나는 염증반응으로 백혈구 혈관외유출이 매우 중요한 역할을 한다는 것은 잘 알려진 사실이다. 선행연구 결과 CD99가 ${\beta}1$ 인테그린 의존적 메커니즘을 통하여 단핵구의 혈관외유출을 조절하며, 따라서 염증 질환에 대한 치료제 개발의 신규 표적 분자일 가능성이 제시되었다. 본 연구에서는 CD99 유래 펩타이드인 CD99CRIII3가 단핵구의 혈관외유출과 접촉피부염 생쥐 모델에서 염증 반응을 억제하는지를 조사하였다. 인간 단핵구 세포주인 U937를 CD99CRIII로 처리할 경우 농도의존적으로 ${\beta}1$ integrin의 활성도가 감소되었으며, 이 세포주의 사람제대정맥내피세포에의 부착과 혈관외유출도 억제되었다. 나아가 CD99CRIII3는 포르볼 미리스테이트 아세테이트 처리에 의해 유발된 생쥐 접촉피부염 모델에서 Evans Blue의 혈관투과와 귀조직 무게를 농도 의존적으로 억제하였다. 이러한 결과들은 CD99CRIII3가 단핵구의 혈관외유출과 접촉피부염 동물 모델에서 일어나는 염증반응을 억제함을 보여준다. 이와 같이, 본 연구는 CD99 유래 펩타이드가 피부 염증질환에 대한 치료제로 개발될 가능성이 있음을 제시한다.

Radiographic patterns of periosteal bone reactions associated with endodontic lesions

  • Objectives: The formation of new bone by periosteum due to an insult is called periosteal bone reaction (PBR). This study assessed the cone beam computed tomography (CBCT) patterns of periosteal bone reactions associated with periapical inflammatory lesion (apical periodontitis/periapical rarefying osteitis). Materials and Methods: Twenty-two small field of view CBCT images of patients with PBR were selected from a database of a private practice limited to endodontics. The volume of the periapical inflammatory lesion, the presence of cortical fenestration, the distance of the root apices to the affected cortex, and the location, pattern, and longest diameter of the periosteal reaction were recorded. Statistical analysis was performed using Wilcoxon Ranksum, Fischer's exact, Spearman Correlation Coefficient, and paired t-test. Results: In all cases, periosteal bone reaction manifested as either parallel (90.9%) or irregular (9.1%). No correlation was found between periapical inflammatory lesion volume and the periosteal reaction's longest diameter (p > 0.05). Cortical fenestration was noted in 72.7% of the cases. In addition, the findings showed that periosteal reactions were located mostly on the buccal and were present 53.8% and 100% of the time in the mandible and maxilla, respectively. Conclusions: The periosteal reactions of endodontic origin had a nonaggressive form (i.e., parallel or irregular), and none of the lesions resulted in a periosteal reaction with an ominous Codman's triangle or spicule pattern.

수종치아수복재료의 조직반응에 관한 연구

  • The purpose of this study was to investigate biological tissue reactions to various restorative dental materials. An experimental pellects was implanted into subdermal tissue in dog subjects observing 1 week, 3 weeks, and 8 weeks respectively. The obtained histo-pathological findings were as follows: 1. Experimental pellets such as gold, ticonium and amalgam alloy are considered biologically acceptable. 2. Experimental results in 1 week dog showed acute inflammatory changes. 3. Experimental results in 8 week dog showed fibrotic and chronic inflammatory changes. 4. Active irritans such as silicate cements revealed acute inflammatory changes in all observed period. 5. Biological tissue to irritants observed.

Formocresol, Glutaraldehyde가 유견 계승치 치배 및 주위조직에 미치는 영향에 관한 병리조직학적 연구 (THE EFFECTS OF FORMOCRESOL AND GLUTARALDEHYDE ON THE PERFORATED INTERRADICULAR TISSUES AND TOOTH GERMS OF PRIMARY TEETH IN DOGS)

  • The purpose of this study is to observe the effect of formocresol and glutaraldehyde to tooth germs and periapical tissues after perforation of interradicular portion of pulpal floor and application of physiological saline solution in control groups, formocresol and glutaraldehyde in experimental groups. The following results were obtained 1. In control groups, normal healing processes were seen, and, on the sixteenth day, the epithelization of injured areas was completed. Inflammatory reactions were limited to the injured surface, and the underlying alveolar bone were normal and successive tooth germs were normal. 2. In both formocresol groups and glutaraldehyde groups, tissue reactions were identical. Inflammatory reactions were slightly compared with control groups, but the surface epithelizations were delayed compared with control group. 3. In both formocresol and glutaraldehyde groups, necrosis was seen in superficial tissue of bone marrow, and, at 24th day, center area of bone marrow on the successive tooth germs were losed and replaced with connective tissue, and superficial soft tissue of the injured area was connected with soft tissue on the successive tooth germ. In remaining alveolar bone, osteoclastic reaction was remarkable. 4. In both formocresol and glutaraldehyde groups, there is no injury to the successive tooth germs. 5. In both formocresol and glutaraldehyde groups, periodontal membrane was normal, but the partial resorption of cementum and dentin near the injured area were seen.

자소엽의 항알레르기와 항염증효과 (Antiallergic and Anti-inflammatory Effects of Perilla frutescens var. acuta)

  • In the present study, we investigated the effect of the water extract of Perilla frutescens var. acuta (Labiatae; WEPF) on the mast cell-mediated allergic reactions. WEPF was anally administered to mice for high and fast absorption. WEPF inhibited compound 48/80-induced systemic allergic reaction. WEPF attenuated immunoglobulin E (IgE)-mediated local allergic reaction. In addition, WEPF decreased the gene expression of pro-inflammatory cytokines in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated HMC-1 cells. These results indicate that WEPF inhibits mast cell-mediated allergic reactions in vivo and in vitro.

사물탕가미방이 염증치료 및 예방에 미치는 영향에 대한 연구 (Effects of SaMulTangGamibang(SMTG) on the Inflammatory Reactions)

  • This study was carried out to investigate the effects SMTG on the skin inflammatory reactions. Among the herbal ingredients of SMTG, ethanol extracts of Paeoniae obovata Radix and Acanthopanacis Cortex showed potent radical scavenging activity, more than 90$\%$ at a concnentration of 0.01$\%$, tested by DPPH(1,l-diphenyl-2-picryl-hyrazyl) method. And ethanol extract of Saururi Herba Seu Rhizoma inhibited the lipopolysaccharide-induced release of nitric oxide(NO), 94$\%$ at 50 ㎍/㎖, by the macrophage RAW 246.7 cells. In addition, the ethanol extract of Acanthopanacis Cortex showed antibacterial activities on Staphylococcus aureus that was reported to cause impetigo and atopic disease. Taken together, I expect that SMTG may be used as a drug for treatment on skin inflammation.

Among the herbal ingredients of herbal extracts, ethanol extract of Glycyrrhizae Radix showed potent radical scavenging activity, more than 86$\%$ at a concentration of 0.01$\%$, tested by DPPH(I,1-diphenyl-2-picryl-hyrazyl) method. And ethanol extract of Aurantii Immaturus Fructus inhibited the lipopolysaccharide-induced release of nitric oxide(NO), 99$\%$ at 50㎍/㎖, by the macrophage RAW 246.7 cells. In addition, the ethanol extracts of Sophorae Radix, Glycyrrhizae Radix showed antibacterial activities on Propionibacterium acnes that causes acne, Sophorae Radix, Glycyrrhizae Radix and Aurantii Immaturus Fructus on Corynebacterium xerosis that cause osmidrosis axillae, Glycyrrhizae Radix and Aurantii Immaturus Fructus on Staphylococcus aureus that was reported to cause impetigo and atopic disease. Taken together, I expect that herbal extracts may be used as a drug for treatment on skin inflammation.

And ethanol extract of Saussureae Radix inhibited the lipopolysaccharide-induced release of nitric oxide(NO), 86$\%$ at 50㎍/㎖, by the macrophage RAW 246.7 cells. In addition, the ethanol extracts of Cinnamoni Ramulus and Polygoni Avicularis Herba showed antibacterial activities on Staphylococcus aureus that was reported to cause impetigo and atopic disease, Cinnamoni Ramulus, Saussureae Radix and Plantaginis Semen on Propionibacterium acnes that causes acne, and Cinnamoni Ramulus on Candida albicans that causes cadidiasis, Saussureae Radix on Corynebacterium xerosis that cause osmidrosis axillae. Taken together, I expect that YSTG may be used as an effective drug for treatment on skin inflammation.

Eosinophils and childhood asthma

  • Eosinophils are a type of granulocyte with eosinophilic granules in the cytoplasm that play an important role in allergic and parasitic diseases. Eosinophils are important in the pathogenesis of asthma, and many studies have examined the relationship between them. In allergic eosinophilic asthma, eosinophils act not only as important effector cells but also as antigen-presenting cells in allergic inflammatory reactions. In nonallergic eosinophilic asthma, type 2 innate lymphoid cells in the airways play an important role in eosinophil activation. Direct methods, including bronchial biopsy, bronchoalveolar lavage, and the induced sputum test, are used to evaluate eosinophilic inflammatory reactions in patients with asthma, however, because of difficulty with their implementation, they are sometimes replaced by measurements of blood eosinophils, fraction of exhaled nitric oxide, and serum periostin level. However, these tests are less accurate than direct methods. For the treatment of patients with severe eosinophilic asthma, anti-interleukin-5 preparations such as mepolizumab, reslizumab, and benralizumab have recently been introduced and broadened the scope of asthma treatment. Although eosinophils are already known to play an important role in asthma, we expect that further studies will reveal more details of their action.

소건중탕이 알레르기에 미치는 영향 (Inhibitory Effects on the Type I hypersensitivity and Inflammatory Reaction of Sogunjung-tang)

  • Sogunjung-tang(SGJT) has been used for the treatment of general weakness, digestive organ disease and so on. This study was carried out for the purpose of knowing the inhibitory effect on the Type I hypersensitivity and Inflammatory reactions of SGJT. The reasults were obtained as follows: SGJT(0.1, 0.5, 1, 2mg/g) concentration of dependently inhibited compound48/80 induced anaphylaxis reaction in mice. SGJT(2mg/g) also inhibited permeability of evans blue into peritoneal cavity in mice. SGJT reduced IgE, CRP, WBC and Platelets on egg albumin induced hypersensitivity. but serum NO was grown. According to above results, SGJT may be beneficial in the type I hypersensitivity and Inflammatory reactions by inhibition of histamine release from mast cells.
