  • The Effects of Hoichunyanggyuksan on the Anti-allergic Effect, Analgesic Action, Anti-inflammatory Action and Antipyretic Action. Experimental studies were done to research the clinical effects of Hoichunyanggyuksan on the Anti-allergic effect, Analgesic action, Anti-inflammatory action and Antipyretic action. The results obtained as follows; 1. On vascular permeability responses to intradermal histamine, Hoichnyanggyuksan showed significant effect. 2. In the homologous PCA provoked by the IgE-like antibody against white egg albumin, Hoichunyanggyuksan showed the decreasing tendency, but was none significant effect. 3. In the delayed type hypersensitivity responses to Picryl chloride, Hoichunyanggyuksan was proved significant effect. 4. In the delayed type hypersensitivity resposes to SRBC, Hoichunyanggyuksan revealed significant effect. 5. In Anti-pyretic action by yeast method, Hoichunyanggyuksan showed significant effect. 6. In Anti-inflammatory action by carrageenin method, Hoichunyanggyuksan showed significant effect. 7. In analgesic action by acetic acid method, Hoichunyanggyuksan was recognized significantly.

Inhibitory Effect of Carbamylated Staphylococcal Enterotoxins B on Inflammatory Response in HL-60 Cells

  • Staphylococcal enterotoxin B (SEB) is bacterial toxin that induces the activation of immune cells. Because the inhibition of pro-inflammatory effect of SEB can resolve the inflammation, I determined the influence of functional or structural change of SEB on immune cells. The post translational modification of protein occurs through carbamylation. Carbamylation can change the structure of proteins and can modify the biological activity of protein. In the present study, I investigated the effect of carbamylated SEB (CSEB) on the inflammatory response mediated by LPS in HL-60 cells. To determine the anti-inflammatory effect of CSEB, I produced carbamylated SEB using potassium cyanate (KCN) and then examined whether CSEB involved in cytokine releases and apoptosis of LPS-stimulated HL-60 cells. Although CSEB had not any effect on the LPS-stimulated HL-60 cells, the protein levels of IL-8, TNF-${\alpha}$ and IL-$1{\beta}$ were significantly decreased by CSEB without cytotoxicity. CSEB also blocked Akt and NF-${\kappa}B$ activation. These results indicate that the suppressive effect of CSEB in LPS-stimulated cytokine releases is occurred by inhibition of Akt and NF-${\kappa}B$ activity. Through further studies, CSEB may be used as anti-inflammatory molecule that makes the immune system more efficient.

Anti-Inflammatory Effects of Rice Bran Ethanol Extract in Murine Macrophage RAW 264.7 Cells (미강에탄올추출물의 RAW264.7 세포에서 항염증효과)

  • The aim of the present study is to investigate the anti-inflammatory effect of a Rice Bran Ethanol Extract (RBE). Inflammation, such as a bacterial infection in vivo metabolites, such as external stimuli or internal stimuli to the defense mechanisms of the biological tissue a variety of intracellular regulatory factors deulin inflammatory TNF-${\alpha}$, IL-$1{\beta}$, IL-6, IL-8, such as proinflammatory cytokines, prostagrandin, lysosomal enzyme, free radicals are involved in a variety of mediators. The present study was designed to determine the effect of the RBE on pro-inflammatory factors such as NO, iNOS expression and TNF-${\alpha}$, IL-$1{\beta}$, IL-6 in lipopolysaccharide (LPS) - stimulated RAW264.7 macrophages cells. The cell toxicity was determined by MTS assay. To evaluate of anti-inflammatory effect of RBE, amount of NO was measured using the NO detection kit and the iNOS expression was measured by reverse transcriptase polymerase chain reaction (RT-PCR). And proinflammatory cytokines were measured by ELISA kit. As a result, the RBE reduced NO, iNOS expression and TNF-${\alpha}$, IL-$1{\beta}$, IL-6 production without cytotoxicity. Our results suggest that the RBE may have an anti-inflammatory property through suppressing inflammatory mediator productions and appears to be useful as an anti-inflammatory material.

The anti-inflammatory effect of Portulaca oleracea 70% EtOH Extracts on lipopolysaccharide-induced inflammatory response in RAW 264.7 cells (LPS로 유도한 RAW 264.7 세포의 염증반응에서 마치현(馬齒莧) 70% 에탄올 추출물의 항염증 효과)

  • Objectives : Portulaca oleracea (PO) have been used as a traditional medicine to treat inflammatory diseases in Korea. However, the anti-inflammatory effect of PO ethanol extract on lipopolysaccharide (LPS)-induced inflammation is not well-known. Therefore, this study was performed to identify the anti-inflammatory effect of PO on LPS induced inflammatory.Methods : Identification of PO was conducted by comparison with purified standards by HPLC. To measure out the cytotoxicity of PO, author performed the MTT assay. To evaluate the anti-inflammatory effects of PO, author examined the inflammatory mediators such as nitric oxide (NO) and pro-inflammatory cytokines (tumor necrosis factor (TNF)-α, interleukin, (IL)-1β and IL-6) on RAW 264.7 cells. Author also examined molecular mechanisms such as mitogen-activated protein kinases (MAPKs) and nuclear factor-B (NF-κB) activation by western blot.Results : Three major components (peaks 1, 2, 3) were detected in both varieties and peak 1 was characterized as caffeic acid, peak 2 as p-coumaric acid, and peak 3 as ferulic acid by comparison of chromatographic properties with authentic standards. Extract from PO itself did not have any cytotoxic effect in RAW 264.7 cells. PO inhibited LPS-induced productions of inflammatory mediators such as NO and pro-inflammatory cytokines in RAW 264.7cells. In addition, PO inhibited the phosphorylation of extracellular signal-regulated kinase1/2 (ERK1/2), c-Jun NH2-terminal kinase (JNK) and NF-κB activation in RAW 264.7 cells.Conclusions : Above experiment data can be an important indicator for the identification of PO and this study suggest that treatment of PO could reduce the LPS-induced inflammation. Thereby, PO could be used as a protective agent against inflammation.

Evaluation of anti-inflammatory effect of traditional medicinal plants, Gynura segetum

  • Gynura segetum, family Asteraceae is a cultivated species and can be found growing in the tropical regions of Indonesia and Malaysia. The plant is known for its use for the treatment of cancer, inflammation, diabetes, hypertension and skin afflictions. In the current study, anti-inflammatory effect of Gynura segetum leaf has been investigated. The present study investigated the in vivo anti-inflammatory effect of Gynura segetum leaf by using hen's egg test-chorioallantoic membrane (HET-CAM) assay. Different solvent extract of Gynura segetum leaf were tested, the most active methanol extract was further fractionated and tested. Among the extracts tested, the methanol extract showed a significant good anti-inflammatory effect (76.8% inhibition at 50 ${\mu}g/disc$) on the HET-CAM assay as compared with the anti-inflammatory drug indomethacin (82.1% inhibition). However, the fractionated sample exhibited a significantly lower activity in comparison to crude methanol extract. The results demonstrated that Gynura segetum leaf displayed remarkable anti-inflammatory effects, which support the folkloric uses of this plant for treatment of inflammation.

Anti-Inflammatory Effect of Extracts from Ligustrum obtusifolium S. fruits in RAW 264.7 Macrophages (RAW 264.7 대식세포 내에서 남정목 열매 추출물의 항염증 효과)

  • Objectives This study was designed to investigate the anti-inflammatory effect of extracts from Ligustrum obtusifolium S. fruits(LOF) in RAW 264.7 Macrophages stimulated with lipopolysaccharide(LPS). Methods We examined productions of nitric oxide(NO), reactive oxygen species(ROS), inducible isoforms of NO synthase(iNOS), cyclooxygenase-2(COX-2) to investigate the anti-inflammatory effect of LOF extracts. In addition, we measured generation of pro-inflammatory cytokines(TNF-${\alpha}$, IL-6). Results Cell viability showed that LOF extracts had no cytotoxicity in Raw 264.7 cells. The treatment with LOF extracts significantly decreased the generation of NO and pro-inflammatory cytokines(TNF-${\alpha}$, IL-6) in LPS-stimulated macrophage cells. Furthermore LOF extracts inhibited intracellular ROS generation dose dependently and reduced the expression of iNOS, COX-2 proteins. Conclusions These results showed that the LOF extracts had an anti-inflammatory effect on LPS-stimulated Raw 264.7 cells. These findings provide scientific support for the use of this Ligustrum obtusifolium S. for inflammatory-related diseases.

Anti-inflammatory Effects of Earthing Mattress in Mouse (Balb/c 생쥐에 대한 어싱 매트리스에 의한 항염 효과)

  • Earthing, caused by direct skin contact with the Earth's surface, is used to reduce the symptoms of inflammation (fever, fever, swelling and pain). However, there is little evidence to support the anti-inflammatory effects of earthing mattresses. Therefore, this study was conducted to investigate whether anti-inflammatory effect of earthing mattress using an in vivo animal model. The anti - inflammatory effect was evaluated by measuring ear thickness and foot volume in 12-O-tetradecanoylphorbol-13 acetate (TPA) - induced ear edema and carrageenan - induced paw edema model, respectively. Balb/c mouse in carrageenan paw edema model showed significant anti - inflammatory effect in the group treated with earthing mattress for 4 hours or 24 hours for 3 days. For females, the anti-inflammatory effect was greater when the earthing mattress was added to the mattress than the mattress alone treatment. From the above results, it was found that the female responds more to the effect of the earthing as well as the mattress effect. In addition, when the male and female Balb/c mice were exposed to mattresses and earthing mattresses for 24 h for 3 days, respectively, the mattress and earthing mattresses showed significant inhibition of IL (Interleukin)-1β levels compared to the control. In the TPA ear edema model, Balb/c mouse showed significant anti - inflammatory effect in the group treated with the earthing mattress for 4 hours or 24 hours for 3 days. Both males and females showed more anti-inflammatory effects when they were exposed to earthing mattresses with mattresses added to the mattresses. From the above results, it was found that both male and female respond to the effect of earthing as well as the mattress effect in the TPA ear edema model. In conclusion, in this study, we have verified that earthing mattress shows inhibitory effects on TPA and carrageenan-induced inflammation. From these results, it is suggested that the anti-inflammatory effect can be expected by applying the earthing mattress to patients suffering from inflammatory diseases. However, there is a need to pinpoint exactly how the earthing mattress relieves inflammation, and further research is needed to investigate the mechanism.

An Experimental Study on the Anti-inflammatory Effects of Cheonggisan Extract in Allergic Late Inflammation (알레르기 후기 반응 염증 억제효과에 관한 청기산(淸肌散)의 실험적 연구)

  • Objectives : Allergic diseases have a various symptoms of hyperresponsiveness and recently hyperresponsive reaction in the chronic phase is reported as the important mechanisms. Cheonggisan(CGS) is used in oriental clinics for curing various skin diseases due to effect of controlling of pruritus. There have been studies on the anti-allergic effect and anti-inflammatory effect of CGS, but there had no study of anti-allergic effects in allergic late inflammation of CGS, so we aimed to find out the effects of CGS in allergic late inflammation in our study.Methods : To investigate the anti-allergy effect and anti-inflammatory effect of CGS, RAW 264.7 macrophage cells and CSG water-extracts were used. Cytotoxic effect of CSG was examined by MTT assay, an oxidative product of NO was measured in the culture medium by the Griess reagent assay. The level of prostaglandin E2(PGE2) was measured by competitive enzyme-linked immunoassay. Cytokine(PGE2, IL-1β, IL-6, TNF-α) was measured by Bio-Plex suspension assay system and quantitative multiplexed cytokine/chemokine assay.Results : We investigated that there was no cytotoxic effect of CGS water-extract at any levels of concentration on RAW 264.7 macrophage cells by MTT assay. CGS water-extracts significantly suppressed the levels of the inflammatory mediators such as NO and PGE2, cytokine of IL-1β, TNF-α at the level of 400 ㎍/㎖ CGS concentration. But there was no significant effect on IL-6 production suppression.Conclusions : These results suggest that CSG water-extract has and anti-inflammatory effects in allergic reaction. These properties may contribute to the allergic diseases and inflammatory related disease care.

Anti-oxidant and anti-inflammatory effect of Allium Hookeri water extracts in RAW 264.7 cells (삼채(三菜) 물추출물이 RAW 264.7 세포의 항산화 및 염증반응에 미치는 영향)

  • Objectives : Allium hookeri is a well-known traditional herbal remedy and its root used for treatment of inflammation and tumor. However, the mechanism of anti-inflammatory effect of Allium hookeri is still unknown. This study aims to examine the mechanism of anti-inflammatory effect of Allium hookeri on mouse macrophage cell line, RAW 264.7 cells. Methods : Anti-oxidant effect of water extract of Allium hookeri (WEAH) was measured by 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay. 3- (4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was performed to determine the effect of WEAH on cell viability in RAW 264.7 cells. In addition, anti-inflammatory effect of WEAH was investigated in RAW 264.7 cells. Inflammation of RAW 264.7 cells induced by lipopolysarccharide (LPS) treatment and expression levels of inflammatory cytokine interleukin 1 β (IL-1β) and interleukin 6 (IL-6) gene were analyzed using quantitative reverse transcription PCR (qRT-PCR) analysis. Furthermore, the phosphorylation of inhibitor of nuclear factor kappa B (IκBα) after LPS treatment with WEAH-treated RAW 264.7 cells was confirmed by immunoblot analysis. Results : WEAH showed a strong anti-oxidant effect and no cytotoxicity to RAW 264.7 cells up to 2 mg/㎖ concentration. The LPS-induced mRNA expression levels of IL-1β and IL-6 were decreased by WEAH treatment. Furthermore, the LPS-induced phosphorylation of IκBα is attenuated by WEAH treatment. Conclusions : Through experimental demonstration of anti-oxidant and anti-inflammatory effects of WEAH, we suggest that Allium hookeri is a valuable material for prevention and treatment of various inflammatory diseases.

  • Experimental studies were done to research the clinical effect of Bojeosodokeum on the Analgesic, Antipyretic, Anti-inflammatory, Antimicrobial and Anti-convulsive effects in mice and rats. The results obtained as follows : 1. The solid extract powders of Bojeosodokeum was revealed significant action on analgesic effect. 2. the solid extract powders of Bojeosodokeum was revealed significant action on Anti-inflammatory effect. 3. The solid extract powders of Bojeosodokeum was revealed significant action on antipyretic effect. 4. Antimicrobial effects of solid extract powders of Bojeosodokeum against Escherichia coli and staphylococcus aureus was observed. 5. The solid extract powders of Bojeosodokeum was revealed significant action on anticonvulsive effect. According to the above results, it is confirmed that analgestic, anti-inflammatory, antipyretic, antimicrobial and anticonvulsive effect were recognized in solid extract powders of Bojeosodokeum.
