Twelve-oxoeicosatetraenoic acid-induced fetal membrane release improves postpartum ovarian function, milk production, and blood plasma biochemical parameters in cows

  • Objective: We aimed to determine the effects of 12-oxoeicosatetraenoic acid (12-KETE)-induced placenta release on the performance of mother cows (milk yield, ovarian function, and blood plasma biochemical properties). Methods: Experimental treatments were as follows: i) natural delivery including natural placental release (control cows); ii) induced calf delivery with placental retention (RP cows); and iii) induced calf delivery and 12-KETE-induced placental release (KE cows). Delivery in pregnant KE cows was induced with dexamethasone and prostaglandin. These cows were injected with 12-KETE after calf discharge, resulting in the release of the fetal placenta. RP cows were not treated with 12-KETE after inducing delivery, resulting in placental retention. Results: The milk yield in RP cows during the first 50 days after delivery was significantly lower than that in control cows (p<0.05), whereas KE cows exhibited a similar milk yield to that of control cows. The postpartum plasma progesterone levels of control cows increased 14 days after delivery on average; however, its increase was delayed by 10 days in RP cows. Meanwhile, the 12-KETE treatment (KE cows) brought the timing of progesterone increase forward to the normal level (control cows). Among the 20 biochemical parameters examined, the total cholesterol levels in blood plasma 14 days after delivery were lower in RP cows than that in the other two treatment groups (control cows and KE cows) (p<0.05). In addition, the plasma level of haptoglobin tended to be low in cows that discharged their placentas shortly after delivery. Conclusion: These findings indicate that 12-KETE treatment can alleviate the disorder caused by placental retention.

Effects of Soshiho-tang on Hydrogen Peroxide-induced Oxidative Damage in Hepatocytes (과산화수소로 유도된 산화성 간세포 손상에 대한 소시호탕(小柴胡湯)의 효과)

  • Objectives : The aim of this study was to investigate the hepatoprotective effect of Soshiho-tang (SSH) in mouse primary liver cells against hydrogen peroxide ($H_2O_2$)-induced oxidative stress. We also elucidated the molecular mechanism of hepatoprotective effect by SSH. Methods : Cell viability, level of ALT, AST and LDH, intracellular ROS level, mRNA expression and activity of antioxidant enzymes were used to evaluate hepatoprotection of SSH against $H_2O_2$. Target gene expressions were analyzed by real-time PCR. Results : Pre-treatment with SSH for 1 hour prevented cytotoxicity against $H_2O_2$. $H_2O_2$-induced ROS level decreased under SSH pre-treatment. mRNA expression of GPx and SOD increased in SSH-treated cells. In addition, HSP72 and HSP40 gene expression were elevated under SSH-treatment. Conclusions : These results indicate that SSH protects mouse primary liver cells from $H_2O_2$-induced oxidative injury. This hepatoprotective activity of SSH is mediated by decreasing intracellular ROS and increasing antioxidant enzyme expression (GPx and SOD) and stress response protein (HSP72 and HSP40).

Cloning And Characterization of Pathogen-Inducible EREBP-Like Transcription Factor(CaNR19) From Hot Pepper (Capsicum annuum L.)

  • An EREBP/AP2-type transcription factor (CaPFl) was isolated by DDRT-PCR following inoculation of soybean pustule pathogen Xanthomonas axonopodis pv. glycines Bra which induces HR on pepper leaves. Genomic Southern blot analysis revealed that the CaPFl gene is present as a single copy within the hot pepper genome. The deduced amino acid sequence of CaPFl has two potential nuclear localization signals, a possible acidic activation domain, and an EREBP/AP2 motif that could bind to a conserved cis- element present in promoter region of many stress-induced genes. The mRNA level of CaPFl was induced by both biotic and abiotic stresses. We observed higher-level transcripts in resistance-induced pepper tissues than diseased tissues. Expression of CaPFl is also induced upon various abiotic stresses including ethephon, MeJA, cold stress, drought stress and salt stress treatments. To study the role of CPFI in plant, transgenic Arabidopsis and tobacco plants which express higher level of pepper CaPFl were generated. Global gene expression analysis of transgenic Arabidopsis by cDNA microarray indicated that expression of CaPFl in transgenic plants affect the expression of quite a few GCC box and DRE/CRT box-containing genes. Furthermore, the transgenic Arabidopsis and tobacco plant, expressing CaPFl showed tolerance against freezing temperature and enhanced resistance to Pseudomonas syrnigae pv. tabaci. Taken together, these results indicated that CaPFl is a novel EREBP/AP2 transcription factor in hot pepper plant and it may has a significant role(s) in regulation of biotic and abiotic stresses in plant.

EP2 Induces p38 Phosphorylation via the Activation of Src in HEK 293 Cells

  • Prostaglandin $E_2$ ($PGE_2$), a major product of cyclooxygenase, binds to four different prostaglandin $E_2$ receptors (EP1, EP2, EP3, and EP4) which are G-protein coupled transmembrane receptors (GPCRs). Although GPCRs including EP receptors have been shown to be associated with their specific G proteins, recent evidences suggest that GPCRs can regulate MAPK signaling via non-G protein coupled pathways including Src. EP2 is differentially expressed in various tissues and the expression of EP2 is induced by extracellular stimuli. We hypothesized that an increased level of EP2 expression may affect MAPK signaling. The overexpression of EP2 in HEK 293 cells resulted in significant increase in intracellular cAMP levels response to treatment with butaprost, a specific EP2 agonist, while overexpression of EP2 alone did not increase intracellular cAMP levels. However, EP2 overexpression in the absence of $PGE_2$ induced an increase in the level of p38 phosphorylation as well as the kinase activity of p38, suggesting that up-regulation of EP2 may promote p38 activation via non-G protein coupled pathway. Inhibition of Src completely blocked EP2-induced p38 phosphorylation and overexpression of Src increased the level of p38 phosphorylation, indicating that Src is upstream kinase for EP2-induced p38 phosphorylation. EP2 overexpression also increased the Src activity and EP2 protein was co-immunoprecipitated with Src. Furthermore, sequential co-immunoprecipitation studies showed that EP2, Src, and ${\beta}$-arrestin can form a complex. Our study found a novel pathway in which EP2 is associated with Src, regulating p38 pathway.

Effects of Jindangwon extract in streptozotocin-induced diabetic rats (진당원(珍糖元)의 고혈당 조절 작용 및 기전에 관한 연구)

  • Objective : Diabetes is a disease in which the body does not produce or properly use insulin. Etiological studies of diabetes and its complications showed that oxidative stress might playa major role. Therefore, many efforts have been made to regulate oxygen free radicals for treating diabetes and its complications. Because Jindangwon has been known to be effective in treatment of diabetes, the methanol extract of Jindangwon was tested for its effectiveness in reducing the oxidative stress induced by Streptozotocin. Methods : Jindangwon was washed, dried in the shade and crushed. The crushed Jindangwon was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 hours. The extract was filtered and evaporated under reduced pressure using a rotary evaporator to yield 30.6 g. Jindangwon extract was oral-administered to the diabetic rats induced by streptozotocin 50 mg per 1 kg of body weight for 15 days. The efficacy of the Jindangwon extract was examined with regard to the enzymatic pathways involved in the oxygen free radical production and the glutathione balance. Results : he effects of the methanol extract of Jindangwon in streptozotocin-induced diabetics rats with regard to body weight, blood glucose level, hepatic lipid peroxide level, hepatic xanthine oxidase activity and type conversion rate, hepatic glutathione level, hepatic glutathione peroxidase activity, hepatic glutathione reductase activity, hepatic aldose reductase activity, and hepatic sorbitol dehydrogenase activity were favorable enough to suggest that it is a cure for diabetes and its complications. Conclusions :These results support Jindangwon as an effective reducing agent for oxidative stress in the tissues and organs by regulating the production of oxygen free radicals. Jindangwon, in particular, shows promising results for its use as a cure, or preventative medicine for diabetes and its complications by reducing oxidative stress in beta-cells of the pancreas.

Effects of Hydroxybrazilin on Glutathione Depletion Induced by $\textrm{BrCCl}_3$ and Menadione in Cultured Rat Hepatocytes

  • In this study we investigated the effect of hydroxybrazilin on glutathione depletion induced by BrCCl$_3$ and menadione in cultured hepatocytes to understand the cellular mechanisms of hepatoprotective effect of hydroxybrazilin. Hydroxybrazilin alone had no effect on total glutathione level and the ratio of reduced glutathione/total glutathione (GSH/(GSSG+GSH)). BrCCl$_3$ dramatically decreased total glutathione level and hydroxybrazilin significantly prevented glutathion depletion by BrCCl$_3$. The ratio of GSH/(GSSG+ GSH) was also decreased by BrCCl$_3$ and recovered by hydroxybrazilin treatment. Menadione decreased total glutathione level and the ratio of GSH/(GSSG+GSH) but hydroxybrazilin showed no significant effects on menadione-induced glutathione depletion. These data suggest that hydroxybrazilin might prevent the hepatotoxicity induced by chemicalderived radicals but not the toxicity linked with oxidative stress.

Effects of Keonbodan (健步丹) on MIA-Induced Osteoarthritis in Rat (건보단(健步丹)이 MIA로 유도된 골관절염 Rat에 미치는 영향)

  • Objectives This study was carried out to know the effects of Keonbodan (hereinafter referred to KBD) in osteoarthritis induced by Monosodium iodoacetate(hereinafter referred to MIA) on Wistar rat. Methods Osteoarthritis was induced by injection of MIA into left knee joint cavities of rat. Osteoarthritis rats were divided into 4 groups (normal (n=6), control (n=6), indomethacin (n=6), KBD (n=6) group). The control group was administered normal saline and indomethacin group was administered indomethacin (2 mg/kg). And the KBD group was administered KBD (142 mg/kg). Each groups were administered by orally for 4 weeks. This experiment were carried out in vivo. In vivo, at the end of the experiment (5 weeks after MIA injection), effects on hepatotoxicity and nephrotoxicity, cytokines in serum, arachidonic acid, osteocalcin, MMP-9, TIMP-1 and cartilage volume were evaluated. And histopathological examinations on the articular structures of knee joints were performed. Results 1. In weight-bearing measurement, level of weight was increased. 2. In order to hepatotoxicity and nephrotoxicity, ALT, AST, BUN and creatinine were tested. And there were no significant changes. 3. In serum, levels of TNF-$\alpha$, IL-$1{\beta}$ were significantly decreased. IL-6 was insignificantly decreased. 4. In serum, level of MMP-9 and TIMP-1 was decreased. 5. In serum, level of $LTB_4$, $PGE_2$ and osteocalcin was decreased. 6. In ${\mu}$CT-arthrography, the cartilage volume was greater than that of the control group. 7. The joint damage induced by osteoarthritis was lesser than the control group in histopathologic observation (H&E, Safranin-O staining). Conclusions These results demonstrated that KBD suppressed the osteoarthritis- inducing effects of MIA in rat. And further studies are required to find out more effective substance and anti-osteoarthritic mechanism in the future.

Effects of the Extract in Streptozotocin-induced Diabetic Rats (고혈당 흰쥐에서 제조의 혈당 조절과 항산화 작용에 관한 연구)

The efficacy of the Holotrichia extract was examined with regard to the enzymatic pathways involved in the oxygen free radical production and the glutathione balance. Results : The Effects of the methanol extract of Holotrichia in streptozotocin-induced diabolic rats with regards to body weight, blood glucose level, hepatic lipid peroxide level, hepatic superoxide anion radical content. hepatic xanthine oxidase activity and type conversion rate, hepatic glutathione level, hepatic aldose reductase activity, and hepatic sorbitol dehydrogenase activity were shown to be good enough to cure and prevent the diabetes and its complications. Conclusions : These results indicated that Holotrichia might reduce the oxidative stress in the tissues and organs by regulating the production of oxygen free radicals. Especially, Holotrichia might prevent and cure the diabetes and its complications by reducing the oxidative stress in the ${\beta}$-cells of pancreas. Some suggestions on biophoton experiments were made.

Effects of Low Level Laser Treatment Applied to KI 1 on the Gait Behavior in the Rat Model of CFA-Induced Arthritis (흰쥐 관절염 모델에서 용천 저출력 레이저 자극이 보행행동에 미치는 영향)

  • Objectives : The aim of the study is to investigate the effects of low level laser treatment (LLLT) on the gait behavior in the rat model of arthritis. Methods : Knee arthritis was induced by the injection of $125{\mu}l$ of Complete Freund's Adjuvant(CFA) into the right hind knee joint cavity. Arthritic rats were divided 3 groups; arthritis group was used for control(CON), 10 min of laser treated group(LSR10), and 30 min of laser treated group(LSR30). LLLT was applied to KI 1 for 11 times under gaseous anesthesia. We performed several analyses under catwalk test including stand and swing time, duty cycle of paw steps, intensity and print area of steps, and stride length. Results : Stand and duty cycle of paw steps were increased significantly at 12 days after arthritis induction in LSR30 group. Swing time was decreased significantly at 12 days after arthritis induction in LSR10 group. In the analysis of intensity, print area and stride length, however, results did not show statistical significance during the time point of experiments. Conclusions : The data suggest that LLLT on the rat model of CFA induced arthritis showed beneficial effects by increase of stand time and duty cycle of paw steps and decrease of swing time. Therefore, LLLT could be useful option to improve gait discomfort in arthritis patients.

Effects of the Fractions of Oryza sativa cv. Heugjinmi on Plasma Glucose and Lipid Levels in Streptozotocin-Induced Diabetic Rats (흑진미 과피 용매분획물 투여시 당뇨 유발 흰쥐의 혈당 및 혈중 지질함량에 미치는 영향)

  • We have investigated the effects of the solvent fractions from the aleurone layer of Oryza sativa cv. Heugjinmi on plasma glucose and lipid levels in streptozotocin-induced diabetic rats. Sprague-Dawley rats were divided into 6 groups, normal, diabetic control, and four experimental groups. Diabetes mellitus was induced by injection of streptozotocin(45 mg/kg body weight) into tail vein, and fraction of Oryza sativa cv. Heugjinmi was administered orally in experimental rats for 14 days. The body weight, glucose, cholesterol, HDL-cholesterol, triglyceride, free fatty acid level, and aminotransferase activities were analyzed. It was identified that acetone and anthocyanin fractions significantly reduced the levels of glucose, triglyceride, and free fatty acid in plasma of diabetic rats. And HDL-cholesterol levels of acetone and anthocyanin groups were higher than those of diabetic control group. From these data, it can be postulated that acetone and anthocyanin fractions might possess rather potent hypoglycemic and hypotriglyceridemic properties in streptozotocin-induced diabetic rats.
