Regulation of Estrogen Receptor Under Hypoxia in Breast Cancer Cells

  • Previously, we have shown that hypoxia, through HIF-1, induces ligand-independent $ER{\alpha}$ activation and the physical interaction of HIF-1 and $ER{\alpha}$. However, the effect of hypoxia on the transactivation of $ER{\beta}$ is not yet known. In the present study, we found that hypoxia activated the $ER{\beta}$-mediated transcriptional response in the HEK 293 cell line, as determined by the transient expression of$ER{\beta}$ and ER-responsive reporter plasmids. The hypoxia-induced estrogen response element-mediated transcriptional response was dependent on $ER{\beta}$ expression and was inhibited by the ER antagonist ICI 182,780. Transactivation of $ER{\beta}$ was induced by the expression of HIF-$1{\alpha}$ under normoxic conditions, as determined by the expression of oxygen-independent stable GFP-HIF-$1{\alpha}$. HIF-$1{\alpha}$-induced $ER{\beta}$ transactivation was abolished by the inhibition of HIF-$1{\alpha}$ activation. This was determined by using chemical inhibitors for the MAPK pathway. In addition, HIF-$1{\alpha}$ interacted with $ER{\beta}$ in a mammalian-two hybrid assay. We conclude that hypoxia activates $ER{\beta}$ in a ligand-independent manner, possibly through the interaction of HIF-$1{\alpha}$ and $ER{\beta}$.

  • Sweat-based glucose sensors are being widely investigated and researched as they facilitate painless and continuous measurement. However, because the concentration of sweat glucose is almost a hundred times lower than that of blood glucose, it is important to develop electrochemical sensing electrode materials that are highly sensitive to glucose molecules for the detection of low concentrations of glucose. The preparation of a flexible and ultra-sensitive sensor for detection of sweat glucose is presented in this study. Oxygen and nitrogen are removed from the surface of a polyimide film by exposure to a CO2 laser; hence, laser-induced graphene (LIG) is formed. The fabricated LIG electrode showed favorable properties of high roughness and good stability, flexibility, and conductivity. After the laser scanning, Pt nanoparticles (PtNP) with good catalytic behavior were electrodeposited and the glucose sensor thus developed, with a LIG/PtNP hybrid electrode, exhibited a high order of sensitivity and detection limit for sweat glucose.

혼재망에서 IPTV 서비스를 위한 계층부호화 방식 결정 방법에 대한 연구 (A study on decision on scalable coding method for IPTV service over heterogeneous network)

  • 혼재 네트워크 (heterogeneous networks) 환경에서는 IPTV 서비스를 위해 계층부호화 방식 (SVC, Scalable Video Coding)이 사용될 것이다. 본 논문은 공간적 (spatial), 품질적 (quality), 시간적 (temporal) 세 가지의 계층화 방식을 (scalable coding) 혼합한 혼합 계층화 방식 (hybrid scalable coding)에서 최종 디스플레이 되는 레벨에 따라 계층간 참조 (inter-layer reference)하는 계층을 결정하는 방식에 대하여 분석하였다. 공간적 계층화에서 위신호(僞信號) (aliasing) 방지를 위해 제거되는 고주파 성분과 품질적 계층화에서 양자화에 의해서 생기는 잡음간의 관계에 따라 공간적 하부 계층에서 요구되는 품질은 어느 정도가 적합한지를 결정한다. 본 논문은 공간적 계층화와 품질적 계층화의 레벨 선택은 비디오 시퀀스의 특성에 따라 결정되어야함을 보인다.

Tobamovirus Coat Protein CPCg Induces an HR-like Response in Sensitive Tobacco Plants

  • When inoculated into sensitive tobacco Xanthi-nn plants, the crucifer and garlic-infecting Tobacco mosaic virus (TMV-Cg) induces local necrotic lesions that resemble those seen in the hypersensitive response (HR) of resistant tobacco plants. However, unlike these, tobacco Xanthi-nn plants do not become resistant to infection and the virus spreads systemically causing a severe disease characterized by necrotic lesions throughout the plant. To identify the viral protein that elicits this necrotic response, we used a set of hybrid viruses constructed by combination of TMV-Cg and the tobacco mosaic virus strain U1 (TMV-U1). In this study we present evidence that the coat protein of TMV-Cg (CPCg) is the elicitor of the necrotic response in tobacco Xanthi-nn plants. Local and systemic necrotic lesions induced by TMV-Cg and by the hybrid U1-CPCg -that carries CPCg in a TMV-U1 context- are characterized by cell death and by the presence of autoflorescent phenolic compounds and $H_2O_2$, just like the HR lesions. In addition, defense-related genes and detoxifying genes are induced in tobacco Xanthi-nn plants after TMV-Cg and U1-CPCg inoculation. We postulate that in our system, CPCg is recognized by sensitive tobacco plants that mount an incomplete defense response. We call this an HR-like since it is not enough to induce plant resistance.

유도된 넙치와 범가자미간 잡종 자어의 생존 및 성장에 관한 연구 (Survival and Growth in Larval Stage of Induced Hybrid between Olive Flounder (Paralichthys olivaceus) and Spotted Flounder (Verasper variegatus))

  • 넙치(Paralichthys olivaceus) 암컷과 범가자미(Verasper variegatus) 수컷간의 잡종을 유도하여 넙치, 범가자미 및 잡종을 대상으로 변태기 전인 부화후 25일째까지 자어의 생존율 및 성장을 비교 조사하였다. 실험 종료시 잡종의 생존율과 성장은 양친으로 사용된 두 종 중 넙치와 유사하였다(P>0.05). 부화자어의 형태에서 잡종은 난황의 길이 대 폭의 비에서 넙치와 좀 더 많이 닮았고, 수온 $17\pm1^{\circ}C$에서 유구가 완전히 소실되는 기간은 잡종이 18일, 넙치가 14일이었으며 범가자미에서는 유구가 관찰되지 않았다. 조사 결과 변태기전까지의 잡종은 그의 생존율 및 성장에서 범가자미 보다 넙치와 더 유사한 경향을 나타내었다(P>0.05).

Control of wind-induced motion in high-rise buildings with hybrid TM/MR dampers

  • In recent years, high-rise buildings received a renewed interest as a means by which technical and economic advantages can be achieved, especially in areas of high population density. Taller and taller buildings are being built worldwide. These types of buildings present an asset and typically are built not to fail under wind loadings. The increase in a building's height results in increased flexibility, which can lead to significant vibrations, especially at top floors. Such oscillations can magnify the overall loads and can be annoying to the top floors' occupants. This paper shows that increased stiffness in high-rise buildings may not be a feasible solution and may not be used for the design for comfort and serviceability. High-rise buildings are unique, and a vibration control system for a certain building may not be suitable for another. Even for the same building, its behavior in the two lateral directions can be different. For this reason, the current study addresses the application of hybrid tuned mass and magneto-rheological (TM/MR) dampers that can work for such types of buildings. The proposed control scheme shows its effectiveness in reducing floors' accelerations for both comfort and serviceability concerns. Also, a dissipative analysis carried out shows that the MR dampers are working within the possible range of optimum performance. In addition, the design loads are dramatically reduced, creating more resilient and sustainable buildings. The purpose of this paper is to stimulate, shape, and communicate ideas for emerging control technologies that are essential for solving wind related problems in high-rise buildings, with the objective to build the more resilient and sustainable infrastructure and to optimally retrofit existing structures.

Thrust Characteristics of a Laser-Assisted Pulsed Plasma Thruster

  • An assessment of a novel laser-electric hybrid propulsion system was conducted, in which a laser-induced plasma was induced through laser beam irradiation onto a solid target and accelerated by electrical means instead of direct acceleration only by using a laser beam. A fundamental study of newly developed rectangular laser-assisted pulsed-plasma thruster (PPT) was conducted. On discharge characteristics and thrust performances with increased peak current compared to our previous study to increase effects of electromagnetic forces on plasma acceleration. Maximum peak current increased for our early study by increasing electromagnetic effects in a laser assisted PPT. At 8.65 J discharge energy, the maximum current reached about 8000 A. Plasma behaviors emitted from a thruster in various cases were observed with an ICCD camera. It was shown that the plasma behaviors were almost identical between low and high voltage cases in initial several hundred nanoseconds, however, plasma emission with longer duration was observed in higher voltage cases. Canted current sheet structures were also observed in the higher voltage cases using a larger capacitor. With a newly developed torsion-balance type thrust stand, thrust performances of laser assisted PPT could be estimated. The impulse bit and specific impulse linearly increased. On the other hand, coupling coefficient and the thrust efficiency did not increase linearly. The coupling coefficient decreased with energy showing maximum value (20.8 ?Nsec/J) at 0 J, or in a pure laser ablation cases. Thrust efficiency first decreased with energy from 0 to 1.4 J and then increased linearly with energy from 1.4 J to 8.6 J. At 8.65 J operation, impulse bit of 38.1 ?Nsec, specific impulse of 3791 sec, thrust efficiency of 8 %, and coupling coefficient of 4.3 ?Nsec/J were obtained.

붉바리(Epinephelus akaara)와 Brown-Marbled Grouper (E. fuscoguttatus) 종간 잡종 수정란의 부화율 (The Hatchability of Fertilized Eggs of Interspecific Hybrid between Red Spotted Grouper (Epinephelus akaara) and Brown-Marbled Grouper (E. fuscoguttatus))

  • 서식 환경이 다른 바리과(family Serranidae) 어류 두 종, 붉바리 (Epinephelus akaara)와 brown-marbled grouper (E. fuscoguttatus)의 종간 잡종 수정란의 부화율을 조사하였다. 잡종(붉바리 ♀ ${\times}$ brown marbled grouper ♂)의 수정 후 각 발생단계까지 소요시간은 붉바리 순종과 차이가 없었으며, 부화 소요시간은 $25^{\circ}C$에서 25.6시간이었다. 잡종은 붉바리 순종과는 달리 비균등 난할이 분명하게 관찰되었다. 잡종의 부화율은 네 수온(22, 25, 28, $31^{\circ}C$)에서 대체적으로 낮았으며, $25^{\circ}C$에서 9.8%로 가장 높았다. 이상의 연구에서 서식환경이 서로 다른 바리과 두 종간 잡종이 생산 가능한 것으로 조사되었으며, 향후 산업적 활용성을 검증하기 위하여 발생 양상, 기형률 및 초기 생존율 등에 관한 면밀한 연구가 필요할 것으로 판단된다.

지지격자가 있는 봉다발과 축방향으로 평행한 유동의 압력손실에 관한 실험적 연구 (Experimental Study on Pressure Loss of Flow Parallel to Rod Bundle with Spacer Grid)

  • 지지격자가 있는 봉다발과 축방향으로 평행한 유동에서, 봉다발 마찰계수와 지지격자 손실계수를 평가하였다. 시험부는 외경 9.5 mm, 길이 2000 mm 인 봉 25 개를 $5{\times}5$ 정사각 구조로 배열하여 제작하였으며, 봉 중심간 거리와 봉 외경의 비는 1.35 였다. 지지격자로는 plain 지지격자, split-vane 지지격자, hybrid-vane 지지격자를 이용하였다. 지지격자가 없는 봉다발의 마찰계수는 기존 상관식과 비교적 잘 일치하였다. 지지격자가 있는 봉다발 실험의 경우, hybrid-vane 지지격자에서 봉다발 마찰계수 및 지지격자 손실계수가 가장 크게 측정되었으며, 이는 지지격자의 유동단면 막음비 증가와 혼합날개 형상에 의한 유동 교란이 증가되기 때문인 것으로 판단된다. Re=$5{\times}10^5$ 조건에서 plain 지지격자, split-vane 지지격자, hybrid-vane 지지격자의 손실계수는 약 0.79, 0.80, 0.88 로 예측되었다.

석회석 광산 내 광주의 안정성 분석을 위한 미소진동 계측기술의 현장적용 (Case study of microseismic techniques for stability analysis of pillars in a limestone mine)

  • 본 연구는 미소진동 계측기술을 국내 광산의 안정성 분석에 적용한 사례연구로서, 계측자료의 분석을 통해 미소진동 기법의 광산 적용성과 한계성을 알아보았다. 적용 광산은 채수율 향상을 위해 주방식하이브리드 채광법이 적용된 석회석광산으로, 수평 단면 $50m{\times}50m$의 시험영역에 대해 각각의 수직 광주에 미소진동 센서를 설치하였다. 측정된 미소진동 신호는 발파와 천공작업으로 인한 신호, 손상에 의한 신호, 전기 잡음에 의한 신호로 구분되었으며, 손상에 의한 신호를 중심으로 안정성 분석을 실시하였다. 시험영역에 근접한 채굴부의 발파작업 후 광주의 손상이 증가하였으며, 주변에서 발생한 낙반을 미소진동 신호로부터 추정할 수 있었다. 또한 일일 미소진동 발생량의 변화로부터 광주와 채굴주변 암반의 안정성을 평가할 수 있었으며, 누적된 계측정보를 토대로 본 광산의 시험영역에 대한 안전관리 기준안을 제시하였다. 그러나 국부적인 센서 배열에 따라 3차원 음원위치를 산정하는 데 어려움이 존재하고, 실시간 계측을 위한 현실적인 대안의 필요성이 제기되었다. 향후 광산적용에서 제기된 문제점을 보완하고, 광산 현장작업과의 유기적인 비교, 분석을 통해 보다 좋은 안전감시의 지시자로서 미소진동 계측기술이 활용될 수 있을 것으로 사료된다.
