The goal of this research was (1) to describe the conditions and parameters required for the cell cycle synchronization and the accumulation of large number of metaphase cells in maize and other cereal root tips, (2) to isolate intact metaphase chromosomes from root tips suitable for characterization by flow cytometry, and (3) to construct chromosome-specific libraries from maize. Plant metaphase chromosomes have been successfully synchronized and isolated from many cereal root-tips. DNA synthesis inhibitor (hydroxyurea) was used to synchronize cell cycle, follwed by treatement with trifluralin to accumulate metaphase chromosomes. Maize flow karyotypes show substantial variation among inbred lines. thish variation should be sueful in isolating individual chromosome types. In addition, flow cytometry is a useful method to measure DNA content of individual chromosomes in a genotyps, and to detect chromosomal variations. Individual chromosome peaks have been sorted from the maize hybrid B73/Mol7. Libraries were generated form the DOP-PCR amplification product from each peak. To date, we have analyzed clones from a library constructed from the maize chromosome 1 peak. Hybridization of labeled genomic DNA to clone inserts indicated that $24\%,\;18\%,\;and\;58\%$ of the clones were highly repetitive, medium repetitive, and low copy, respectively. Fifty percent of putative low cpoy clones showed single bands on inbred screening, blots, and the remaining $50\%$ were low copy repeats. Single copy clones showing polymorphism will be mapped using recombinant inbred mapping populations. Repetitive clones are being characterized by Southern blot analysis, and will be screened by in situ hybridization for their potential utility as chromosome specific markers.
Eleven inbred lines of silkworm, Bombyx mori L. were exposed to two main different temperature (23C and 25C) during the larval period in order to elucidate the effect of the temperature differences on their main characters under the, prevailing in Egyptial agro-climactic conditions. The results show that the temperature differences did not affect significantly the pupation ratio and cocoon shell ratio. However, they affected their larval duration to become shorter with the high temperature treatment ($27^{\circ}C$) and their cocoon shell weight and cocoon yield per box of silkworm eggs to be higher in the lower temperature($23^{\circ}C$). It was recommended that some inbred lines should be bred at the 23C throughout the whole larval period, particularly in being reared with high nutritional leaves in order to maximize the hybridization of cocoon yields.
The rice quality related traits including physico-chemical traits were evaluated with one hundred sixty-four MG RILs derived from the cross between Milyang 23, Indica/Japonica hybrid type, and Gihobyeo, Japonica type. The variation distribution of all traits examined approximately fit normal distribution and transgressive segregants over parents were observed for all traits. The occurrence of such transgression could be associated with the interactions of complementary QTL alleles from two parents. However in this study, it could not be concluded that our results of segregation were due to either complementary gene effects or overdominance of a major gene. These factors should be verified by further studies. Correlations between traits were evaluated by regressing phenotypic values of one trait on those of another traits. There are highly significant correlation between grain thickness with grain width, white core and white belly. But between white belly and alkali digestion value showed highly negative significant correlation. Contents of protein showed highly negative correlation with amylose and Mg/K ratio but showed highly correlation with K and Fat contents. Hardness of cooked rice showed highly correlation with adhesiveness, elasticity, gumminess, chewiness.
This study was conducted to identify the differences in proteomic characteristics of 'Agakong', recombinant inbred line, and its parental genotypes 'Eunhakong' (Glycine max) and 'KLG10084' (G. soja). The isoflavone content of 'Agakong' was 3 times higher than that of its parental lines. A combined high-throughput proteomic approach was employed to determine the expression profile and identity of proteins using 2-dimensional gel electrophoresis and matrix-assisted laser desorption ionization time-of-flight (MALDI-TOF) mass spectrometry. The overall distribution patterns of proteins are quite similar, but lots of protein spot intensities varied among the genotypes. A total of 41 proteins, representing significant difference in the quantities of protein among the lines, were successfully identified. Among them, more than 50% of the proteins identified were subunits of glycinin and $\beta$-conglycinin, 2 major storage proteins. This study showed that the proteomic analysis could help to define specific changes in protein level and composition, which can occur in the generation of new soybean varieties.
This study was carried out to find a new corn cultivar with multiple tillers that is suitable for forage. Materials were developed at the Corn Breeding Laboratory of Chungnam National University. A total of 3,650 accessions were collected domestically and from other countries, including China. Among them, 16 CNU inbreds, tillering and non-tillering, were compared to each other in terms of stem heigh, number of tillers, fresh weight, and dry weight per plant. Of these surveyed traits, the stem heigh at tasseling stage was 140~190 cm and number of tillers per plant ranged from two to four. Fresh weight and dry weight of these lines were higher in the tillered than in the non-tillered sample.
Protoplasts were isolated from hypocotyls, cotyledons, and young leaves of Chinese cabbage grown under in vitro environmental condition. An enzyme mixture of 1% Cellulysin and 0.5% Macerozyme in combination with 0.4 M mannitol was most effective condition for protoplast isolation. The highest yield of protoplasts, 7.6$\times$10$^{5}$ protoplast/g of fresh weight, was obtained from the treatment of leaves for 12~16 hours at 27~28$^{\circ}C$ with shaking at 30 rpm. The most suitable medium for an initial cell division was K8p basal medium supplemented with 5 mg/L 2,4-D and 2 mg/L kinetin. Within 7~10 days, protoplasts derived from hypocotyl and cotyledon tissues formed cell colonies. When the cells were grown at the size of 8~10 cells, they were embedded into semi-solid medium containing 0.2% agarose. Calli derived from protoplast culture were transferred to the 100 different types of plant regeneration media, but no completely regenerated plants from inbred lines of Chinese cabbage used for this study wore obtained, though frequent rooting took place in several media tested.

Soybean oil is an important source of vegetable oil for human food and nonfood applications and accounts for approximately 22% of the world's total edible oil production. Improvement of the quality and quantity of soybean seed oil constituents is one of the most important objectives in soybean breeding. The objective of this study was to identify quantitative trait loci (QTLs) that control oleic, linoleic, and linolenic acid contents in soybean. The 117 $F_{2:10}$ recombinant inbred lines (RIL) developed from a cross of 'Keunolkong' and 'Shinpaldalkong' were used. Narrow-sense heritability estimates based on a plot mean on seed weight, protein and oil content were 0.85, 0.82 and 0.81, respectively. Eight independent QTLs for oleic acid content were identified from linkage group (LG) A2, C1, D2, F, G, L, and O. Seven QTLs for linoleic acid content were located on LG D1b, E, H, I and L. Oil content was related with five QTLs located on LG C1, H, J, K, and L. Oleic, linoleic, and linolenic acid have two common QTLs on LG C1 and L. Thus, we identified major loci improving soybean oil quality.
Phylogenetic relationships in Korean watermelons were evaluated by genetic similarity coefficients using 15 SSR(simple sequence repeat), 14 SCAR(sequence characterized amplified region) and 14 CAPS(sequence characterized amplified region) markers. The SSR markers were selected from previously reported melon and watermelon SSRs through testing polymorphisms within a set of commercial $F_1$ varieties. The SCAR and CAPS markers were developed from polymorphic AFLP(amplified fragment length polymorphism) markers between inbred lines 'BN4001' and 'BN4002'. From the AFLP analysis, 105 polymorphic fragments were identified between the inbred lines using 1,440 primer combinations of EcoRI+CNNN and XbaI+ANNN. Based on the sequencing data of these polymorphic fragments, we synthesized sequence specific primer pairs and detected clear and reliable polymorphisms in 27 primer pairs by indels(insertion/deletion) or RFLP(restriction fragment length polymorphism). A total of 43 sequence-based PCR markers were obtained and polymorphic information content(PIC) was analyzed to measure the informativeness of each marker in watermelon varieties. The average PIC value of SCAR markers was 0.41, which was similar to that of SSR markers. Genetic diversity was also estimated by using these markers to assess the phylogenetic relationships among commercial varieties of watermelon. These markers differentiated 26 Korean watermelon varieties into two major phylogenetic groups, but this grouping was not significantly correlated with their morphological and physiological characteristics. The mean genetic similarity was 66% within the complete set of 26 commercial varieties. In addition, these sequence-based PCR markers were reliable and useful to identify cultivars and genotypes of watermelon.
Anthers of hybrids and inbred lines of Chinese cabbage (Brassica campestris ssp. pekinensis) were inoculated on the modified solid or liquid B$_{5}$ medium after several pre-treatments. Anthers were then maintained at 25 $^{\circ}C$ after being subjected to various post-treatment. Somatic embryos began to appear 9 days after inoculation and ended at 13th days. Low temperature pre-treatment did not increase embryoid production whereas high post-treatment temperature of 4$0^{\circ}C$ or 45$^{\circ}C$ enhance the production. There were considerable levels differences in embryogenesis between lines used, but not between the culture methods. Somatic embryo yield was also increased by subjecting anthers to one day at 35$^{\circ}C$ and then another one day at 3$0^{\circ}C$ after plating. Histological observations showed several stages in haploid development ranging from a few celled to large multiple-celled embryoids.s.
Fusarium and Rhizoctonia genera are important pathogens of many field crops worldwide. They are constantly evolving and expanding their host range. Selecting resistant cultivars is an effective strategy to break their infection cycles. To this end, we screened a collection of Medicago truncatula accessions against Fusarium oxysporum, Fusarium solani, and Rhizoctonia solani strains isolated from different plant species. Despite the small collection, a biodiversity in the disease response of M. truncatula accessions ranging from resistant phenotypes to highly susceptible ones was observed. A17 showed relative resistance to all fungal strains with the lowest disease incidence and ratings while TN1.11 was among the susceptible accessions. As an initiation of the characterization of resistance mechanisms, the antioxidant enzymes' activities, at the early stages of infections, were compared between these contrasting accessions. Our results showed an increment of the antioxidant activities within A17 plants in leaves and roots. We also analyzed the responses of a population of recombinant inbred lines derived from the crossing of A17 and TN1.11 to the infection with the same fungal strains. The broad-sense heritability of measured traits ranged from 0.87 to 0.95, from 0.72 to 0.96, and from 0.14 to 0.85 under control, F. oxysporum, and R. solani conditions, respectively. This high estimated heritability underlines the importance of further molecular analysis of the observed resistance to identify selection markers that could be incorporated into a breeding program and thus improving soil-borne pathogens resistance in crops.
