Effects of Vitamin C Supplementation on Immune Status in Smoking and Nonsmoking Male College Students (흡연자에 있어 비타민 C 보충이 면역능력에 미치는 영향)

  • This study investigated the effects of vitamin C supplementation on immune status in smoking and nonsmoking male college students. Blood samples were obtained from 15 healthy smoking and from 15 healthy nonsmoking volunteers before and 4 weeks after vitamin C supplementation (1000mg/day). Daily nutrient intakes was also calculated. Plasma vitamin C, E, and A concentrations, white blood cells and subpopulations, lymphocytes and subpopulations, NK cell percentage,plasma immunoglobulin A, G, M and complement C3, plasma interleukin-2 , and prostagladin E2 were measured . Daily vitamin C consumption was lower in smokers than in nonsmokers. Vitamin C supplementation significantly increased plasma vitamin C in smokers, and increased NK cell percentage in smokers and nonsmokers. Plasma IL-2 concentrations were significantly increased by vitamin C supplementation in nonsmokers, and decreased helpe $r^pressor T cell ratio were decreased by vitamin C supplementatiion , but the differences were not significant. White blood cells and subpopulation percentage and plasma prostaglandin E2 concentration were not affected by smoking and vitamin C supplementation. These findings as nonsmokers.rs.

  • Bickerstaff's brainstem encephalitis (BBE) is an autoimmune central nervous system disorder. It can occur in more limited forms and may overlap with Guillain-Barr$\acute{e}$ syndrome (GBS). A 49-year-old female presented with rapidly progressive paralytic ileus, urinary retention, deep drowsiness, ophthalmoplegia, dysarthria, ataxia, quadriparesis and hyporeflexia after viral meningitis. She was diagnosed as BBE with GBS and treated with immunoglobulin. She was completely recovered after 1 month. It is a rare case of BBE overlapping with GBS presenting with severe paralytic ileus.

Inhibitory Effect of Isodon japonicus Hara on Mast Cell-Mediated Immediate-Type Allergic Reactions (비만세포 매개 즉시형 알레르기 반응에 대한 연명초의 억제 효과)

  • The effect of aqueous extract of Isodon japonicus Hara (Labiatae) (IJAE) on mast cell-mediated immediate-type allergic reactions was investigated. IJAE inhibited compound 48/80-induced systemic anaphylaxis and immunoglobulin E (IgE)-mediated local anaphylaxis. When IJAE was pretreated at the same concentration with systemic anaphylaxis, serum histamine levels were reduced in a dose-dependent manner. IJAE dose-dependently inhibited histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80. The level of cAMP in human mast cell line (HMC-1) cells, when IJAE was added, significantly was increased, compared with that of normal control. These results indicate that IJAE will beneficial in the treatment of immediate-type allergic reaction.

Immunomodulatory Effects by Bifidobacterium longum KACC 91563 in Mouse Splenocytes and Macrophages

  • The present study evaluates the immunomodulatory effect of Bifidobacterium longum KACC 91563 in murine primary splenocytes and macrophages. B. longum KACC 91563 regulated T- and B-cell proliferation and inhibited the Th1 (IL-2, IFN-γ)/Th2 (IL-4, IL-10) cytokine imbalance and immune cytokine production. Moreover, immunoglobulin E (IgE) levels were significantly lower after treatment with B. longum KACC 91563. These findings suggest that B. longum KACC 91563 could modulate the systemic immune system toward both IgE production and regulation of the Th1/Th2 balance.

Guillain-barré Syndrome after Multiple Bee Stings (다발성 벌 자상에 의한 길랑 바레 증후군 1례)

  • Severe systemic responses including neurologic complications such as myasthenia gravis, myeloradiculopathy, optic neuropathy, parkinsonism, stroke and Guillain-$barr{\acute{e}}$ syndrome can occur after bee stings. This case describes a 78-year-old female who presented with symptoms of acute progressive bilateral symmetrical weakness in both lower legs after multiple bee stings. Nerve conduction study findings were consistent with acute sensorimotor axonal neuropathy and recovered by treatment with intravenous immunoglobulin. This case highlights that bee stings can result in acute onset Guillain-$barr{\acute{e}}$ syndrome, although the pathophysiologies of bee venoms need to be investigated accurately.

The Effects of Oldenlandiae Diffusae Herba Extract on Eosinophil, IgE and IL-4 in Experimental Asthma induced by Ovalbumin (백화사설초(白花蛇舌草)가 ovalbumin으로 유도된 천식동물모델에서 Eosinophil의 수, IgE 및 IL-4에 미치는 영향)

  • Objective : This study was performed to investigate the effect of oral administration of Oldenlandiae Diffusae Herba (ODH) on eosinophil, immunoglobulin-E and interleukin-4 in the experimental asthma induced by ovalbumin. Methods : Asthma was induced to Balb/c mouse by i.p. injection and aerosol immunization with ovalbumin. It was observed the change of the eosinophil number in the BALF. And, it was observed the change of the concentrations of IL-4 in BALF and splenocyte, IgE in serum by ELISA. Results : 1. The number of eosinophil in BALF was significantly decreased in ODH group copared with control group. 2. Concentration of IL-4 in serum, BALF and splenocyte was significantly decreased in ODH group compared with control group, respectively. 3. Level of IgE in serum was significantly decreased in ODH group compared with control group. Conclusion : We found that the effect of ODH extract in asthma was implicated in reductions of IL-4 released from Th2 cell, and decreases of IgE from plasma cell. These findings suggest that ODH extract can produce anti-asthmatic effect, which may playa role in allergen-induced asthma therapy.

Effects of Olaquindox and Cyadox on Immunity of Piglets Orally Inoculated with Escherichia coli

  • A 2${\times}$3 factorial arrangement of treatments was used to determine the effects of olaquindox and cyadox on immune response of Landrace${\times}$Large-White geld piglets that had been orally given 10$^{10}$ CFU of Escherichia coli (E. coli, O$_{139}$:K$_{88}$). Factors included (1) E. coli inoculation or control, and (2) no antimicrobials, 100 mg/kg olaquindox and 100 mg/kg cyadox in the basal diet respectively. E. coli inoculums were orally administered 7 days after the diets were supplemented with olaquindox and cyadox. The effects of the two antimicrobials were assessed in terms of: (1) average daily gain (ADG), (2) systemic immune response (the number of white blood cells and lymphocytes, leukocyte bactericidal capacity, lymphocyte proliferation response to PHA, immunoglobulin concentrations, and total serous hemolytic complement activity), and (3) intestinal mucosal immunity including the number of intraepithelial lymphocytes (IELs) and immunoglobulin A secreting cells (ASCs) in the intestinal lamina propria. E. coli inoculation reduced ADG (p<0.05) during the period of d 0 to d 14 after the challenge while the antimicrobial supplementations improved ADG (p<0.01) during the experiment. ADG in cyadox-supplemented pigs was higher (p<0.05) than that in olaquindox-supplemented pigs. The antimicrobials decreased IEL and ASC counts in the jejunum and ileum (p<0.01) while E. coli inoculation caused them to increase (p<0.01). Jejunal ASCs in the cyadox-supplemented pigs were lower (p<0.05) than those in the olaquindox-supplemented. E. coli elicited increase (p<0.05) in white blood cell counts, leukocyte bactericidal capacity, lymphocyte proliferation rate, serous IgA concentrations, and serous hemolytic complement activity. The antimicrobials decreased the measured systemic immune parameters, but not significantly (p>0.05). The data suggest that olaquindox and cyadox suppress E. coli-induced immune activation, especially intestinal mucosal immune activation, which may be involved in the observed growth promotion.

Identification of the Chicken Meat Allergens (닭고기 중 알레르기 유발성분의 동정)

  • The chicken meat has been reported as one of the food causing allergic reactions predominantly to Korean. At present, several in vitro tests for immunoglobulinG (IgG)-mediated as well as IgE-mediated food allergy are available. 13 clinically chicken meat-allergic patients were investigated together with 4control subjects for identification of chicken meat-specific reactivity by ELISA. Also, protein profile and IgE, IgGtotal and IgG4-reacting allergens were detected by means of sodium dodecyl sulfate-polyacrylamide gel electro-phoresis (SDS-PAGE)and immunoblotting. Chicken meat extracts were prepared as raw, heated, heat and simulated gastric fluid (SGF) treated samples to characterize the stability of allergen to physicochemical treatment. SDS-PAGE revealed 9~200 kDa bands. And in immunoblotting 7 sera were identified most major bands between 10 and 78 kDa. In case of IgE, six proteins (17, 26, 35, 40, 78 kDa) were predominant in heat-treated extract, and the one (35 kDa) was present in SGF-treated preparations. In case of IgG$_{total}$ and IgG4, most of them showed a patters simmilar to IgE. There were significant differences (P<0.05) in IgE, IgG$_{total}$ , IgG4 Abs to chicken meat between the allergic and control subjects in ELISA. In addition, the concentration of IgG4Abs in the challenge-positive subjects was significantly higher than that of control subjects. It is considered that the specific IgE response to chicken meat was rarely prevalent to Koreans. However, the specific IgG4 response play an important role in the development of allergic symptoms.

External Application of Apo-9'-fucoxanthinone, Isolated from Sargassum muticum, Suppresses Inflammatory Responses in a Mouse Model of Atopic Dermatitis

  • Allergic skin inflammation such as atopic dermatitis is characterized by skin barrier dysfunction, edema, and infiltration with various inflammatory cells. The anti-inflammatory effects of Apo-9'-fucoxanthinone, isolated from Sargassum muticum, have been described in many diseases, but the mechanism by which it modulates the immune system is poorly understood. In this study, the ability of Apo-9'-fucoxanthinone to suppress allergic reactions was investigated using a mouse model of atopic dermatitis. The Apo-9'-fucoxanthinone-treated group showed significantly decreased immunoglobulin E in serum. Also, Apo-9'-fucoxanthinone treatment resulted in a smaller lymph node size with reduced the thickness and length compared to the induction group. In addition, Apo-9'-fucoxanthinone inhibited the expression of interleukin-4, interferon-gamma and tumor necrosis factor-alpha by phorbol 12-myristate 13-acetate and ionomycin-stimulated lymphocytes. These results suggest that Apo-9'-fucoxanthinone may be a useful therapeutic strategy for treating chronic inflammatory diseases.

Topical Application of Herbal-mixed Extract on back skin suppresses inflammation in Ovalbumin-induced Atopic Dermatitis (아토피 피부염 동물모델에서 피부에 도포한 한약 복합추출물의 염증 억제 효과)

  • Objective : This study is conducted to evaluate the efficiency of ethanol extracts of 4 mixed herbs(CP001 or CP002) on mouse model of atopic dermatitis induced by ovalbumin. Methods : Female BALB/c mice were internally sensitized with ovalbumin($20{\mu}g$) plus aluminum hydroxide hydrate(4mg) once per week. After 3 weeks, they were dermally challenged with patches containing ovalbumin ($100{\mu}g$) plus aluminum hydroxide hydrate(20mg) every other day for 3 weeks. After induction of atopic dermatitis, mice back skin were gently rubbed with CP001 or CP002(200mg/$m{\ell}$, $100{\mu}{\ell}$) for two weeks(every 2 days). Results : In CP001 or CP002 treated group, there was a remarkable reduction in infiltration of eosinophils on the skin areas and diminution of mast cells and total T cells in blood samples as compared with control group. Cutaneous expressions of interleukin-13, 17 were also decreased by CP001 or CP002. Moreover, blood immunoglobulin E level was decreased by drug administration while there was no decrease in OVA sensitization group. Conclusion : In summary, our result shows that herbal extracts(CP001 and CP002) could be potential candidates for the treatment of chronic atopic dermatitis.
