Bifidobacteria has been suggested to exert health promoting effects on the host by maintaining microbial flora and modulating immune functions in the human intestine. We assessed modulatory effects of the different cell fractions of Bifidobacterium sp. BGN4 on macrophage cells and other immune cells from the spleen and Peyer's patches (PP) of mouse. Cell free extracts (CFE) of the BGN4 fractions induced well-developed morphological changes in the macrophages and increased the phagocytic activity more effectively than other fractions in the mouse peritoneal cells. Nitric oxide (NO) production was significantly reduced by both the cell walls (CW) and CFE in the cultured cells from the spleen and PP. The production of interleukin-6 (IL-6) and interleukin-10 (IL-10) was eminent in the spleen cells treated with experimental BGN4 cell fractions. However, in the PP cells, IL-6 was slightly decreased by the treatment with the whole cell (WC) and CW, whereas IL-10 was significantly increased by the treatment with the CW and CFE. These results suggest that different types of bifidobacterial cell fractions may have differential immunomodulatory activities depending on their location within the host immune system.
Enrofloxacin, a fluoroquinolone, is a broad-spectrum antibiotic widely used in veterinary medicine that inhibits the action of bacterial DNA gyrase, resulting in anti-bacterial effects. This study was performed to examine whether enrofloxacin has modulatory and anti-inflammatory activity on immune cells. A few studies have reported the anti-inflammatory effects of enrofloxacin. In this study, we used mouse spleen cells treated with lipopolysaccharide (LPS) and examined the effects of enrofloxacin. Several assays were performed in LPS-treated spleen cells after the enrofloxacin treatment. Enrofloxacin inhibited the metabolic activity and mitochondrial membrane potential of LPS-treated spleen cells significantly. On the other hand, enrofloxacin did not alter the proportion of the subsets in spleen cells, and did not induce cell death. The production of tumor necrosis factor-alpha in LPS-treated spleen cells was inhibited by enrofloxacin. Overall, enrofloxacin had modulatory activity in spleen cells treated with LPS. These data may broaden the use of enrofloxacin as an antibiotic with anti-inflammatory activity in veterinary clinics.
Objectives: Much evidence exists that herbs have effective immunomodulatory activities. Chrysanthemi Flos (CF) is effective in clearing heat, reducing inflammation, dropping blood pressure and treating headache and is used as a pharmaceutical raw material for an immune enhancer. The purpose of this study was to investigate the modulatory effect of Chrysanthemi Flos pharmacopuncture on nitric-oxide (NO) production in activating macrophages. Methods: After a murine macrophage cell line, RAW 264.7, was cultured in the presence of lipopolysaccharide (LPS), immune-modulating abilities of CF were evaluated by using NO, interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-${\alpha}$) production and phagocytic activity of macrophages. Results: CF enhanced the activities of macrophages by increasing the phagocytic activity and decreasing NO production. Especially, both LPS and CF, 200 ${\mu}g/ml$, treatment could significantly reduce the NO production, but did not change the production of IL-6 and TNF-${\alpha}$. Conclusion: The results of this study indicate that CF may be of immunomodulatory value, especially for adverse diseases due to increased NO production. It may have potential for use as immunoenhancing pharmacopuncture.
The purpose of this study was to characterize and investigate the immune activity of CpG oligodeoxynucleotides (ODNs) from Bifidobacterium longum. Bacterial CpG motifs have attracted considerable interests because of their immunomodulatory activities. Genomic DNA from B. longum was prepared and amplified for 4 different 180-188-mer double-stranded ODNs (BLODN1-BLODN4). When immune cells (RAW 264.7 murine macrophages and JAWS II dendritic cells) with these ODNs were treated, BLODN4 induced the highest immune activity. To assess the effectiveness of the CpG sequences within BLODN4, single-stranded 40-mer ODNs containing CpG sequences (sBLODN4-1, sBLODN4-2) were synthesized. sBLODN4-1 induced higher level of cytokines such as interleukin (IL)-12p40 and tumor necrosis factor (TNF)-$\alpha$ by macrophage and IL-6 and TNF-$\alpha$ by dendritic cells than did sBLODN4-2. The results suggest that CpG ODNs-enriched components of B. longum might be useful as an immunomodulatory functional food ingredient.
In order to evaluate the efficacy of SHT against atopic dermatitis (AD), various immune related cytokines as well as histological comparison were performed in animal models, and the results are described. Clinical skin index of the SHT treated group decreased significantly in weeks 11 and 13, compared to the control group. Also, CD4+ immune cell ratio in the dorsal skin was significantly decreased to 69%, and both epidermal and dermal skin thickness was decreased. Serum IL-4, IL-5, IL-6, IL-13, and TNF-${\alpha}$, which are all important markers of inflammation, were decreased to 64%, 44%, 87%, 48%, and 45%, respectively. The expression of histamine, a chemical transmitter increasingly released during the progression of inflammation, was significantly decreased to 47%. The production of IgE immunoglobulin was significantly decreased to 16% compared to the control group. In conclusion, SHT pacifies the activation of T cells, leading to suppression of both Th2 cytokine overexpression and infiltration of immune cells into skin. As a result, relative thinning of both epidermis and dermis were observed. With the results obtained from in vitro studies, the immune modulatory effect of SHT in AD animal models was experimentally demonstrated. This study should provide solid information to construct EBM and for clinical practice.
Immune-potentiation of Chicorium endivia L. were investigated on follows extracts associated with ultrasonification process at 60 kHz and showed the highest promotion of human B and T cell growth, about $10{\sim}20%$ compared to the control. The secretion of TNF-${\alpha}$ and IL-6 was also enhanced by the addition $(0.5mg/m{\ell})$ of the extracts. NK cell activation was Improved up to 1.37 times higher than the control, through adding extracts. It was also found that extracts from C. endivia L. could yield higher nitric oxide production from macrophage than Lipopolysaccaharides (LPS). It can be concluded that, in general, the extracts treated with ultrasonification has higher immune activity than others, possibly by higher yielding immune-modulatory activity than conventional extraction process. The optimum condition for the extraction of C. endivia L. is ethanol extraction at $60{\sim}100^{\circ}C$ associated with ultrasonification.
The purpose of this research was to investigate the effect of Sojagangqi-tang(SJGQT) on the immune cell activity. The addition of SJGQT enhanced the proliferation of cultured-mice splenocytes and thymocytes. Administration of SJGQT(250 mg/kg) accelerated the subpopulation of splenic T lymphocytes especially CD/sup 4+/-TH cells in BALB/c mice. But high concentration(500 mg/kg) of SJGQT decreased the splenic T, B lymphocytes and thymic Tc (CD/sup 8+/) lymphocytes. Oral administration of SJGQT(250 mg/kg) significantly enhanced the production of IFN-γ and IL-4 in mice serum. And also, the addition of SJGQT(100 ㎍/ml) inhibited the proliferation of cultured-Jurkat leukemia cells in vitro. These results suggest that SJGQT have a cellular immuno-modulatory effect and anti-cancer property action
본 연구는 양조과정의 부산물인 주박으로부터 분리한 다당류(JPS)가 초기 면역반응에 중추적인 역할을 수행하는 대식세포에서 활성화를 유도하는지에 관한 여부를 알아보기 위해서 수행되었다. 주박에서 분리한 다당류를 마우스 유래 대식세포인 RAW264.7 cell에 처리하였을 때 대식세포의 활성화의 지표인 NO와 cytokine(IL-6, TNF-${\alpha}$)의 분비가 증가되었다. 또한 이러한 NO와 cytokine의 증가의 원인에 관한 면역기전에 관하여 알아본 결과 JPS의 처리는 MAPKs(ERK, JNK, p-38)의 인산화를 촉진시켜 NF-${\kappa}B$의 활성을 유도하여 면역세포의 활성인자들의 분비를 촉진시킨 것으로 관찰되었다.
The purpose of the present study was to investigate the immune-enhancing activity of selenizing Codonopsis pilosula polysaccharide (sCPPS5) in nonspecific immune response. In in vitro experiment, the results showed that sCPPS5 could promote the phagocytic uptake, NO production, and TNF-α and IL-6 secretion of RAW264.7 cells. sCPPS5 could also strongly increase the IκB-α degradation in the cytosol and the translocation of NF-κB p65 subunit into the nucleus of RAW264.7 cells. In the vivo experiment, sCPPS5 at medium doses could significantly improve the phagocytic index of peritoneal macrophages and induce the secretion of TNF-α and IL-6. Moreover, the effect of sCPPS5 was significantly better than Codonopsis pilosula polysaccharide (CPPS). These results indicated that selenylation modification could significantly enhance the immune-enhancing activity of CPPS in the nonspecific immune response.
본 연구는 전통약재로 주로 사용되는 구절초에서 분리한 다당류(CZPS)가 선천 및 적응면역에서 중추적인 역할을 수행하는 대식세포의 활성화에 미치는 영향에 관하여 관찰하였다. 구절초에서 분리한 다당류를 마우스 유래 대식세포주인 RAW 264.7 cell에 처리하였을 때, iNOS의 발현을 조절하여 NO의 생성이 증가되었으며, 또한 면역활성 물질인 TNF-${\alpha}$, IL-$1{\beta}$ 및 IL-6 등의 cytokine의 분비능이 증가되었으며, 이러한 면역활성 매개자인 NO 및 cytokine의 증가의 원인에 관한 정확한 면역세포내 신호전달에 관하여 알아본 결과, CZPS 처리는 MAPKs(ERK, p38)의 인산화를 촉진시키며, 이로 인한 전사인자인 NF-${\kappa}B$의 인산화를 증가시켜, 대식세포의 활성화를 유도시키는 것으로 관찰되었다.
