자궁내막증이 있는 불임환자에서 복강액내의 Cytokine에 관한 연구 (A study of Cytokine in Peritoneal Fluid of Infertile Patients with Endometriosis)

  • Objective: The presence of the various cytokines in human peritoneal fluid has been evaluated incompletely. Changes in cytokine levels may be related to activation of peritoneal macrophage and T-lymphocyte, development of endometriosis, and infertility. This study assesses peritoneal fluid levels of interleukin-6 (IL-6), interleukin-8 (IL-8), interleukin-10 (IL-10) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in infertile women with endometriosis and normal women without endometriosis. Design: Prospective and case-control study in university hospital. Materials and Methods: Cytokine levels in peritoneal fluid obtained during laparotomy or laparoscopy from 21 patients in infertile patients with endometriosis and 24 controls undergoing laparotomy or laparoscopy with no evidence of pelvic endometriosis were determined by enzyme-linked immunosorbent assay. Results: The mean levels of interleukin-6 in infertile patients with endometriosis and controls were $72.7{\pm}23.7$ pg/ml and $18.5{\pm}9.7$ pg/ml respectively (p=0.02). Similarly, the mean levels of interleukin-8 in infertile patients with endometriosis was significantly higher than that of controls ($445.0{\pm}89.6$, vs $45.1{\pm}48.4$, p=0.04). The mean concentration of interleukin-10 in infertile patients with endometriosis was significantly lower than that of controls ($1.09{\pm}0.04$ vs $2.19{\pm}0.03$, p=0.03). The level of tumor necrosis factor-${\alpha}$ was not significantly different between the two study groups. Conclusions: Increased IL-6 and IL-8 and decreased IL-10 levels in the peritoneal fluid may be related to pathogenesis in the endometriosis and infertility, suggesting that partially contribute to the disturbed immune regulation observed in infertili women with endometriosis.

($Interferon{\gamma}$)로 자극된 치은섬유아세포와 치주인대섬유아세 포에서 항원제시기능과 관련된 세포 표면분자의 발현 (Expression of Antigen Presenting Function-Associated Surface Molecules on $Interferon{\gamma}$-Treated Gingival Fibroblasts and Periodontal Ligament Fibroblasts)

  • It is becoming increasingly clear that human gingival fibroblasts(HGF) may play a role in regulating immune responsiveness in inflammatory periodontal lesions. Stimulation of HGF with locally-secreted T cell cytokine $IFN_{\gamma}$ induces human leukocyte antigen class II(HLA II) expression on HGF, which is one of the characteristic feature of professional antigen presenting cells(pAPC). However, $IFN_{\gamma}$-treated HGF and other nonprofessional antigen presenting cells(npAPC) are known to be ineffective or less effective antigen presenter to resting T cells. This study, therefore, was undertaken in an effort to elucidate the differences in expression of cell surface molecules between npAPC in periodontal tissues, such as HGF and periodontal ligament fibroblasts(PDLF), and pAPC such as monocytes/macrophages. Using flow cytometry, the levels of cell surface expression of HLA-D, ICAM-1, LFA-3, and B7-1, which are involved in antigen presentation, were determined in HGF, PDLF and human myelomonocytic cell line THP-1. $IFN_{\gamma}$ clearly induced HLA-D expression on both of fibroblasts and monocytes dose dependently. However, expression level on monocytes were 4 to 5 times higher than that on fibroblasts, and induction rate was faster in monocytes than in fibroblasts. The levels of ICAM-1 expression on fibroblasts and monocytes were enhanced by $IFN_{\gamma}$ in a dose dependent manner. On the other hand, the expression of LFA-3 molecule, which could be detected in fibroblasts and monocytes without cytokine stimulation, was no more enhanced by addition of $IFN_{\gamma}$. B7-1, important costimulatory molecule in T cell activation and proliferation, was not detected on both of fibroblasts and monocytes even when stimulated with $IFN_{\gamma}$, except on monocytes fully differentiated by pretreatment of PMA and treated by $IFN_{\gamma}$. These results suggest that delayed expression of HLA-D and absence of B7-1 on $IFN_{\gamma}$ - treated fibroblasts may at least in part be involved in the ineffectiveness of fibroblasts as primary APC. And it is postulated that although periodontal fibroblasts may not serve as primary APC in normal periodontium, sustained expression of HLA II on ubiquitous fibroblasts in inflammatory lesions may perpetuate immune responses and produce chronic inflammation and tissue injury.

구멍갈파래의 면역활성 증진을 위한 추출방법 비교 (The Comparison of Extraction Process for Enhancement of Immunomodulating Activities of Ulva pertusa kjellman)

  • 본 연구는 제주도의 녹조현상의 원인인 구멍갈파래의 활용을 위해 고압추출 공정을 통한 추출물의 면역활성 확인 하였다. 150 MPa, $80^{\circ}C$ 조건에서 Superoxide Radical 소거효과에서는 90% 로 활성도가 제일 높았으며, 면역세포 생육도면에서도 B&T cell, 각 각 $14.2{\times}10^4$ cells/mL $14.5{\times}10^4$ cells/mL 나타나 제일 높았다. 반면, NK cell과 NO 면역능 실험에서는 100MPa, $30^{\circ}C$ 조건에서 각각 $11.8{\times}10^2$ cells/mL, 30.0 ${\mu}M$을 보여 주었다. 결과를 통해 구멍갈파래는 면역활성을 가지며 고압 공정을 통한 추출물의 활성이 일반추출에 비해 높은 것을 알 수 있다. 따라서 해조류 및 다른 유용 자원에서도 고압 공정이 기존의 추출방법보다 높은 활성을 나타낼 것으로 판단되며 기능성 식품이나 기능성 소재로서 사용가능성이 있는 것으로 사료된다.

Paired Ig-Like Type 2 Receptor-Derived Agonist Ligands Ameliorate Inflammatory Reactions by Downregulating β1 Integrin Activity

  • The paired immunoglobulin-like type 2 receptor (PILR) family consists of two functionally opposite members, inhibitory $PILR{\alpha}$ and activating $PILR{\beta}$ receptors. PILRs are widely expressed in various immune cells and interact with their ligands, especially CD99 expressed on activated T cells, to participate in immune responses. Here we investigated whether PILR-derived agonists inhibit ${\beta}1$ integrin activity as ligands for CD99. PILR-derived peptides as well as PILR-Fc fusion proteins prevented cell adhesion to fibronectin through the regulation of ${\beta}1$ integrin activity. Especially, PILRpep3, a representative 3-mer peptide covering the conserved motifs of the PILR extracellular domain, prevented the clustering and activation of ${\beta}1$ integrin by dephosphorylating FAK and vinculin, which are major components of focal adhesion. In addition, PILRpep3 inhibited transendothelial migration of monocytes as well as endothelial cell tube formation. Furthermore, upon intraperitoneal injection of PILRpep3 into mice with collagen-induced arthritis, the inflammatory response of rheumatoid arthritis was strongly suppressed. Taken together, these results suggest that PILR-derived agonist ligands may prevent the inflammatory reactions of rheumatoid arthritis by activating CD99.

가시오가피 물 추출물에 의한 마우스 비장세포 및 대식세포 활성의 항진효과 (Enhancing Effect of Acanthopanax senticosus Extracts on Mouse Spleen and Macrophage Cells Activation)

  • In vitro 실험을 통한 가시오가피 물 추출물 첨가가 마우스의 면역세포 증식에 미치는 영향에 대한 연구 결과, 음의 대조군에 비해 가시오가피 물 추출물을 첨가한 모든 농도에서 비장세포 증식능이 증가하였으며, 특히 고농도인 $500{\sim}1,000{\mu}g/mL$ 농도에서 유의적으로 증가하였다. 반면, 복강 대식세포로부터 유도된 사이토카인 생성의 경우, IL-2, IFN-${\gamma}$, TNF-${\alpha}$ 사이토카인 생성량을 측정한 결과, TNF-${\alpha}$ 사이토카인은 가시오가피 물 추출물 $50{\mu}g/mL$ 농도와 $250{\sim}1,000{\mu}g/mL$ 농도에서 대조군보다 유의적으로 높은 분비량을 보인 반면, IFN-${\gamma}$에서는 변화를 보이지 않았다. IL-2의 경우, $100{\sim}500{\mu}g/mL$에서 유의적으로 증가하는 경향을 보여주었다. 이상의 결과에 의하면 가시오가피 물 추출물은 마우스 비장 세포를 증식시키고, 사이토카인 분비량에도 영향을 줄 것으로 보이며, 면역 기관의 주요 기능을 증진시킬 가능성이 있을 것으로 사료된다. 따라서 가시오가피 물 추출물이 면역 증진 기능성식품 개발의 소재로 활용될 가능성이 있을 것으로 기대된다.

Zika Virus Proteins NS2A and NS4A Are Major Antagonists that Reduce IFN-β Promoter Activity Induced by the MDA5/RIG-I Signaling Pathway

  • Zika virus (ZIKV) is a mosquito-transmitted, emerging Flavivirus that causes Guillain-$Barr{\acute{e}}$ syndrome and microcephaly in adults and fetuses, respectively. Since ZIKV was first isolated in 1947, severe outbreaks have occurred at various places worldwide, including Yap Island in 2007, French Polynesia in 2013, and Brazil in 2015. Although incidences of ZIKV infection and dissemination have drastically increased, the mechanisms underlying the pathogenesis of ZIKV have not been sufficiently studied. In addition, despite extensive research, the exact roles of individual ZIKV genes in the viral evasion of the host innate immune responses remain elusive. Besides, it is still possible that more than one ZIKV-encoded protein may negatively affect type I interferon (IFN) induction. Hence, in this study, we aimed to determine the modulations of the IFN promoter activity, induced by the MDA5/RIG-I signaling pathway, by over-expressing individual ZIKV genes. Our results show that two nonstructural proteins, NS2A and NS4A, significantly down-regulated the promoter activity of IFN-${\beta}$ by inhibiting multiple signaling molecules involved in the activation of IFN-${\beta}$. Interestingly, while NS2A suppressed both full-length and constitutively active RIG-I, NS4A had inhibitory activity only on full-length RIG-I. In addition, while NS2A inhibited all forms of IRF3 (full-length, regulatory domain-deficient, and constitutively active), NS4A could not inhibit constitutively active IRF3-5D. Taken together, our results showed that NS2A and NS4A play major roles as antagonists of MDA5/RIG-I-mediated IFN-${\beta}$ induction and more importantly, these two viral proteins seem to inhibit induction of the type I IFN responses in differential mechanisms. We believe this study expands our understanding regarding the mechanisms via which ZIKV controls the innate immune responses in cells and may pave the way to development of ZIKV-specific therapeutics.

돼지에서 분리된 Lactobacillus Strains의 균체분해산물에 의한 RAW 264.7 Macrophage 활성화 (Activation of RAW 264.7 Macrophage by Digested Bacterial Cell of Pig-derived Lactobacillus Strains)

  • 경구로 섭취된 생균제가 소화관 효소의 작용을 받은 경우 장관면역계의 macrophage에 미치는 영향을 조사하기 위하여 돼지의 장내에서 분리된 lactobacilli를 pepsin과 pancreatin과 같은 소화관 효소로 분해한 균체분해산물에 의해 RAW 264.7 murine macrophage에서 유도되는 NO, TNF-$\alpha$ 및 IL-6의 생성을 측정하였다. Macrophage에서 유도되는 NO, TNF-$\alpha$ 및 IL-6는 균종과 균체분해산물의 양에 따라 달랐다. NO의 생성수준은 10~150 ug/ml의 분해물에서 관찰 되었으나 TNF-$\alpha$와 IL-6는 50~300 ug/ml의 고농도처리에서 나타났다. 6개의 Lactobacillus strains 중에서 3149와 3156 균주는 다른 균주에 비해 TNF-$\alpha$와 IL-6 유도능이 높았다. 또한 소화관 효소에 의해 분해된 lactobacilli의 균체분해산물의 성분이 macrophage 활성 증가와 관련이 있으며 생체내에서 숙주면역계를 조절할 것으로 사료된다. 본 시험에서 사용한 방법은 소화관 면역계에 미치는 유산균의 효과를 규명하는데 유익할 것이다.

Proinflammatory Cytokine and Nitric Oxide Production by Human Macrophages Stimulated with Trichomonas vaginalis

  • Trichomonas vaginalis commonly causes vaginitis and perhaps cervicitis in women and urethritis in men and women. Macrophages are important immune cells in response to T. vaginalis infection. In this study, we investigated whether human macrophages could be involved in inflammation induced by T. vaginalis. Human monocyte-derived macrophages (HMDM) were co-cultured with T. vaginalis. Live, opsonized-live trichomonads, and T. vaginalis Iysates increased proinflammatory cytokines, such as TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 by HMDM. The involvement of nuclear factor (NF)-${\kappa}B$ signaling pathway in cytokine production induced by T. vaginalis was confirmed by phosphorylation and nuclear translocation of p65 NF-${\kappa}B$. In addition, stimulation with live T. vaginalis induced marked augmentation of nitric oxide (NO) production and expression of inducible NO synthase (iNOS) levels in HMDM. However, trichomonad-induced NF-${\kappa}B$ activation and TNF-${\alpha}$ production in macrophages were significantly inhibited by inhibition of iNOS levels with L-NMMA (NO synthase inhibitor). Moreover, pretreatment with NF-${\kappa}B$ inhibitors (PDTC or Bay11-7082) caused human macrophages to produce less TNF-${\alpha}$. These results suggest that T. vaginalis stimulates human macrophages to produce proinflammatory cytokines, such as IL-1, IL-6, and TNF-${\alpha}$, and NO. In particular, we showed that T. vaginalis induced TNF-${\alpha}$ production in macrophages through NO-dependent activation of NF-${\kappa}B$, which might be closely involved in inflammation caused by T. vaginalis.

꿀벌 꽃가루 열수 추출물의 큰포식세포 면역활성 효과 (Immunomodulatory effect of bee pollen extract in macrophage cells)

  • 본 연구는 꿀벌 꽃가루 열수 추출물(BPW)의 면역 활성에 관하여 알아보기 위하여, 선천면역과 적응면역에서 중심 역할을 수행하는 큰포식세포에 BPW를 처리하여 세포 생존율, NO 분비능, 사이토카인($TNF-{\alpha}$, IL-6, $IL-1{\beta}$) 분비능과 메커니즘 분석을 통한 신호전달에 관하여 관찰하였다. BPW를 큰포식세포에 처리하여 면역활성에 관하여 알아봤을 때, 큰포식세포 면역활성의 대표적인 바이오마커인 NO, 사이토카인의 분비능과 iNOS의 세포내 발현이 BPW 처리구에서 유의적으로 증가되는 것으로 관찰되었다. 또한 활성화된 탐식 세포의 세포 표면에서 발현되는 CD80과 CD86의 발현과 탐식세포의 항원제시에 밀접한 관련이 있는 주조직적합성 복합체(MHC class I과 II)의 발현이 BPW 처리구에서 유의적으로 증가되는 것으로 관찰되었다. 기전분석 결과, BPW의 처리는 MAPKs의 인산화와 $NF-{\kappa}B$의 핵 내 이동성을 증가시켜 면역활성을 증가시키는 것으로 관찰되었다. 따라서 BPW의 처리는 큰포식세포의 활성을 유도시켜 면역활성을 조절하는 것으로 관찰되었다.

Enterotoxigenic Bacteroides fragilis에 의한 질환과 검출 (Enterotoxigenic Bacteroides fragilis-Associated Diseases and Detection)

  • 정상인에서 장내세균은 숙주의 면역이나 영양 흡수를 돕지만, 때로는 기회감염균으로서 그들을 위협하기도 한다. 그 중 절대 혐기성 세균인 Bacteroides fragilis는 분비되는 장독소(enterotoxin)인 Bacteroides fragilis toxin (BFT)의 유무에 따라 non-enterotoxigenic B. fragilis (NTBF)와 enterotoxigenic B. fragilis (ETBF)로 나뉜다. ETBF는 가축 및 사람에서 설사 질환 및 대장 질환을 유발한다 그러나 때때로 ETBF를 가지고 있으나 증상이 없는 사람도 존재한다. ETBF는 염증성 설사 질환, 여행자 설사 환자의 대변에서 검출되어 주목 받고 있다. 또한, 몇몇 연구를 통해 inflammatory bowel disease (IBD)나 대장염 및 대장암 환자에서 ETBF가 증가한다는 것이 밝혀졌다. 일반 C57BL/6 마우스 및 germ-free 마우스, multiple intestinal neoplasia (Min) 마우스, 토끼, Mongolian gerbil 등 여러 동물 모델에서 ETBF가 IBD나 대장염, 대장암을 유발 또는 촉진한다는 것이 발표되었다. ETBF의 유일한 병원성 인자인 BFT는 E-cadherin의 분절을 유도하여 장상피 세포의 투과성을 높인다. 이어서 ${\beta}$-catenin 신호전달계가 활성화하여 장상피세포의 증식이 증가한다. 또한 ETBF의 감염은 일반 마우스에서 급성이나 만성의 대장염을 일으키고 Min 마우스에서 종양 형성을 촉진한다. 이는 Stat3에 의존한 $T_H17$ 면역반응의 활성화를 통해 일어난다. 현재 ETBF의 검출 방법에는 크게 BFT toxin assay와 몇 가지 PCR 방법이 있다. 최근 real-time PCR과 같은 분자진단학적 기법의 발달로 일반적인 PCR보다 더 정확한 ETBF의 검출이 가능하게 되었다. 이것을 이용하여 앞으로 실제 임상에서 ETBF와 대장염 및 대장암의 발달 관계에 대한 심도 깊은 연구가 이뤄질 것으로 본다.
