As a first step for improvement of breeding and productivity of Epimedii Herba, the patterns of components and the difference in icariin, a main component of this plant, in compliance with seasons and places of collection were evaluated by HPLC. As a result, the patterns of components in samples collected in July-September were shown to be markedly increased compared to those collected in May. Corresponding to the alternations of the pattern, the amount of icariin, which was estimated to be 0.017% in May was found to be increased markedly up to 0.82% in September. The samples collected at Chul-won district showed the highest icariin contents (0.94%), however, those collected in China was only 50% level (0.4%) of domestic ones. These results strongly suggest that the quality of Epimedii Herba might be quite different with respect to the seasons and places of the collection.

Epimedium koreanum Nakai is a wild herb commonly consumed in South Korea due to its beneficial health effects. In the present study, the antimutagenic and immunoactivities of extracts from E. koreanum Nakai containing different icariin quantities were investigated for food use. In the Ames test, both the water and ethanol extracts were found not to have a mutagenic effect on Salmonella typhimurium TA98 and TA100, respectively. The E. koreanum Nakai extracts showed over 80 and 90% antimutagenic effects on benzo(${\alpha}$)pyrene (B(a)P) in S. typhimurium TA98 and TA100, respectively. Moreover, all the extracts showed over 70% antimutagenicity on S. typhimurium TA98 and TA100 against 4-nitroquinoline-1-oxide (4NQO). The E. koreanum Nakai extract with ethanol showed strong antimutagenic activity, higher than that of the water extract and the sequenced KE9412, KE9408, and KE9405. In the immunomodulating activity test, the effect of E. koreanum Nakai on the B (Rhamos) and T (Jukat) cells were investigated. The immunoactivity results showed that the growth and viability of the B and T cells increased and were activated more in KE9405 (1.8 times), KE9408 (1.6 times), and KE9412 (1.32 times) in the water extracts, and least in KE9412 (1.74 times), KE9408 (1.52 times), and KE9405 (1.4 times) in the ethanol extracts. In the case of both the water and ethanol extracts ($500{\mu}g/mL$) from E. koreanum Nakai, the highest cell number of the human B (Rhamos) and T (Jukat) cells was observed on day 4 in KE9405 and KE9412, and on day 5 in KE9408. Based on the obtained results, the development of E. koreanum Nakai as a food material is recommended.
Three flavonoids were isolated from Epimedium koreanum and identified as quercetin(1), $anhydroicaritin-3-O-{\alpha}-rhamnoside(2)$ and icariin(3) by spectroscopic methods. The former two compounds are the first isolation from this plant.

Near infrared reflectance spectroscopy (NIRS) has become widely accepted for rapid quantitative analysis of components in some crops. Our object was to determine icariin contents in whole plant of Epimedium koreanum by using an NIRS system. Total 150 plant samples previously analyzed by HPLC were scanned by NIRS and 68 samples were selected for calibration and validation equation. A calibration equation calculated by MPLS(modified partial least squares) regression technique was developed and a coefficient of determination in calibration and validation sets were 0.95 and 0.82, respectively. A comparison between NIRS estimation and HPLC value was performed with the remaining samples not included in the calibration and validation sets. Most of samples also showed a positive correlation like a validation set. Our results demonstrate that this developed NIRS equation can be practically used as a mass screening method for rapid quantification of icarin contents in Epimedium koreanum N.
Epimedii Herba is a traditional medicinal herb used in Korea and China and exerts estrogenic activity. In this study, we investigated the effect of peripubertal administration of Epimedii Herba on pubertal development in female rats using a modified protocol of the rodent 20-day pubertal female assay. Female Sprague-Dawley rats (21 days old after weaning, 10 rats per group) were divided into five groups: saline (Con), ethinyl estradiol (E2), Epimedii Herba ext (Ext), icariin (ICI), and icaritin (ICT), which were administered by oral gavage (E2 by subcutaneous injection) from postnatal day (PND) 21 through PND40. The time to vaginal opening (VO) was shorter for the Epimedii groups, particularly for the ICT group (p<0.05). Treatment with ICI and ICT significantly increased the duration of the estrus cycle (ICI, 2.78 days; ICT, 4.0 days; control, 1.78 days). Ovary weight was reduced by E2 treatment and increased by the Ext, ICI, and ICT treatments while the weight of the uterus and pituitary glands increased significantly only in the E2 and ICT groups. Although Epimedii Herba displayed relatively weak estrogenic activity, its repeated administration could affect pubertal development in female rats.
The seasonal variation of two flavonol glycosides, icariin and epimedoside A, in the aerial parts and underground parts of Epimedium koreanum from June through September was investigated. The icariin concentration was decreased with time in both parts. Epimedoside A concentration was fluctuated, being highest in June and lowest in July in the underground parts. In the aerial parts, however, it was almost same in concentration. Determinations were made of the occurrence of two new flavonol glycosides, 2‘-O-rbamnosyl ikarisoside A and 2’-O-rhamnosyl icarisid II in the aerial parts and their changes in concentration were similar to those in the underground parts, being highest in July in both parts.

In order to obtain basic data on the shelf life of Epimedii Herba (EH), a physicochemical characteristics including weight loss rate, loss on drying, ash, acid-insoluble ash, and quantitative determination were evaluated to compare the data by packaging methods including general (G), general + silica gel (GS), alumium (AL), and alumium + silica gel (ALS) packaging and storage periods (0-66 months). All physicochemical analyzes were conducted using the method of "The Korean Pharmacopoeia (KP) 9th edition". As a result, weight loss rate (%), loss on drying (%), ash (%), acid-insoluble ash (%) of EH according to the packaging methods and storage periods were -0.27-4.73%, 3.06-9.35%, 5.48-7.37%, and 0.45-0.99, respectively. In the quantitative analysis of icariin, the marker compound of EH using high-performance liquid chromatography, the initial content was 0.44% at 0 month in all packaging methods. After that, the result of analyzing the content of icariin every 6 months was detected 0.44-1.18%. All of the above results satisfied the acceptance criteria for loss on drying, ash, acid-insoluble ash, and quantification contained in the KP. This study can be used as a basic data for establishing the packaging method and shelf life of EH.
The air dried of Epimedium koreanum Nakai was extracted with MeOH and its extractives were concentrated with a vacuum evaporator. The extractives were fractionated with a series of n-hexane, chloroform (${CHCl}_3$), butanol (BuOH), ethyl acetate (EtOAc) and water on a separately funnel. Each fraction was freeze dried to give some dark brown powder. The EtOAc and BuOH soluble fractions were chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-n-hexane mixture as eluents. The isolated compounds were tested with a cellulose TLC developed with TBA and 6% acetic acid and then visualized on UV lamp or sprayed with vanillin-HCl-EtOH. The purified compounds were flavonoids and their glycosides, and organic acid as follows : (+)-catechin, icariin, hyperoside, Ikarisoside A and caffeic acid. The structures of each compounds were confirmed by $^1H-NMR,\;^{13}C-NMR$ and Mass spectra. Also, executed qualitative analysis as use GC/MS(Libraries search) about ${CHCl}_3$ soluble compounds of each part.

Three kinds of prenylated flavonols, icariside I, icariside II, and icaritin, were isolated from an icariin hydrolysate and their effects on melanogenesis evaluated based on mushroom tyrosinase inhibition and quantifying the melanin contents in melanocytes. Although none of the compounds had an effect on tyrosinase activity, icariside II and icaritin both effectively inhibited the melanin contents with an $IC_{50}$ of 10.53 and $11.13{\mu}M$, respectively. Whereas icariside II was obtained from a reaction with ${\beta}$-glucosidase and cellulase, the icariin was not completely converted into icariside II. Thus, for the high-purity production of icariside II, the reaction was optimized using the response surface methodology, where an enzyme concentration of 5.0mg/ml, pH 7, $37.5^{\circ}C$, and 8 h reaction time were selected as the central conditions for the central composite design (CCD) for the enzymatic hydrolysis of icariin into icariside II using cellulase. Empirical models were developed to describe the relationships between the operating factors and the response (icariside II yield). A statistical analysis indicated that all four factors had a significant effect (p<0.01) on the icariside II production. The coefficient of determination $(R^2)$ was good for the model (0.9853), and the optimum production conditions for icariside II was an enzyme concentration of 7.5mg/ml, pH 5, $50^{\circ}C$, and 12 h reaction time. A good agreement between the predicted and experimental data under the designed optimal conditions confirmed the usefulness of the model. A laboratory pilot scale was also successful.
