  • To develope the novel baculovirus transfer vector, the p10 gene was cloned from the Bombyx mori nuclear polygedrosis virus (BmNPV) vB2 strain isolated from the B. mori larvae of sericultural farms. The novel transfer vector was constructed by using the p10 gene of BmNPV vB2 strain was 210 bp. The TAAG sequence at the -71 bp of upstream from translation initiator ATG and two polyadenylation signal site at the downstream from terminator TAA were also detected in the p10 gene. The 5' and 3' flanking region of the p10 gene amplified by PCR was cloned into pBluescriptII SK(+) and then transfer vector pBm10 was construceted. The 7.9 kb pBm10 was analysed by restriction enzymes and the map was confirmed. In order to determine the expression of foreign gene of pBm10, $\beta$-galactosidase gene was inserted in the SmaI site of foreign gene cloning site of pBm10. The pBm10 containing $\beta$-galactosidase gene was cotranfected wth genomic DNA of BmNPV vB2 into BmN-4 cells. The recombinant baculovirus expressing $\beta$-galactosidase was also produced polygedra in the infected cells. The results indicated that pBm10 is functional, suggesting that in the baculovirus expression vector system, the recombinant virus produced by pBm10 was effective by oral infection for the producing recombinant proteins in in vivo expression.

  • The similarity weight, the pearson's correlation coefficient, which is used in the recommender system has a weak point that it cannot predict all of the prediction value. The similarity weight, the vector similarity, has a weak point of the high MAE although the prediction coverage using the vector similarity is higher than that using the pearson's correlation coefficient. The purpose of this study is to suggest how to raise the prediction coverage. Also, the MAE using the suggested method in this study was compared both with the MAE using the pearson's correlation coefficient and with the MAE using the vector similarity, so was the prediction coverage. As a result, it was found that the low of the MAE in the case of using the suggested method was higher than that using the pearson's correlation coefficient. However, it was also shown that it was lower than that using the vector similarity In terms of the prediction coverage, when the suggested method was compared with two similarity weights as I mentioned above, it was found that its prediction coverage was higher than that pearson's correlation coefficient as well as vector similarity.

  • Let g be a continuous function on an interval I which is not constant on any subinterval of I, and let ${\mu}$ be a Borel measure on I. In this paper we give a necessary and sufficient conditions guaranteeing, for the strongly measurable function f on I with values in a Banach space X, the existence of a continuous primitive function F on I with respect to g.

  • We present a very simple algorithm for reducing the encoding (codebook search) complexity of vector quantization (VQ), exploiting some features of a vector currently being encoded. A proposed VQ of 16 (=$4{\times}4$) vector dimension and 256 codewords shows a slight performance degradation of about 0.1-0.9 dB, however, with only 16 or 32 among 256 codeword searches, i.e., with just 1/16 or 1/8 search complexity compared to a full-search VQ. And the proposed VQ scheme is also compared to and shown to be a bit superior to tree-search VQ with regard to their SNR performance and memory requirement.

  • In this article, using the example of C. Camci([7]) we reconfirm necessary sufficient condition for a slant curve. Next, we find some necessary and sufficient conditions for a slant curve in a Sasakian 3-manifold to have: (i) a $C$-parallel mean curvature vector field; (ii) a $C$-proper mean curvature vector field (in the normal bundle).

  • The authors aim to establish the theory necessary for developing free gyro positioning system and focus on measuring the nadir angle by using the motion rate of a free gyro. The azimuth of a gyro vector from the North can be given by using the property of the free gyro. The motion rate of the spin axis in the gyro frame is transformed into the platform frame and again into the NED (north-east-down) navigation frame. The nadir angle of a gyro vector is obtained by using the North components of the motion rate of the spin axis in the NED frame. The component has to be transformed into the horizontal component of the gyro by using the azimuth of the gyro vector and then has to be integrated over the sampling interval.

  • The interpretations of degree expressions require the postulation of new entities to represent degrees. Diverse entities such as degrees, intervals, and vectors are adopted for degree expressions. Positive degree sentences and questions are properly construed with the introduction of these entities, but their negative counterparts need more consideration. Negative degree sentences show dual patterns of entailments depending on contexts, and negative degree questions are unacceptable, making weak islands. To explicate the distinct nature of negative degree sentences and questions, Fox & Hackl (2006) provide an analysis based on degrees while Abrusan & Spector (2010) suggest a proposal in interval readings of degree expressions. I have pointed out the theoretical problems of these analyses and proposed an alternative in the framework of the vector space semantics, following Winter (2005). Bi-directional scales in vector space fit well with the dual patterns of negative degree sentences, and the notion of a reference vector is useful to accommodate the contextual influence in negative degree sentences and to deal with the unacceptability of negative degree questions.

  • In the present article, a novel fusion expression vector for Escherichia coli was developed based on the pTORG plasmid, a derivative of pET32a. This vector, named pT7MT(GenBank Accession No DQ504436), carries a T7 promoter and it drives the downstream gene encoding Metallothionein 2A(MT2A). There are in-framed multiple cloning sites(MCS) downstream of the MT2A gene. A target gene can be cloned into the MCS and fused to the C-terminal of the MT2A gene in a compatible open reading frame(ORF) to achieve fusion expression. The metal-binding capability of MT2A allows the purification of fusion proteins by metal chelating affinity chromatography, known as $Ni^{2+}$-affinity chromatography. Using this expression vector, we successfully got the stable and high-yield expression of MT2A-GST and MT2A-Troponin I fusion proteins. These two proteins were easily purified from the supernatant of cell lysates by one-step $Ni^{2+}$-affinity chromatography. The final yields of MT2A-GST and MT2A-Troponin I were 30mg/l and 28mg/l in LB culture, respectively. Taken together, our data suggest that pT7MT can be applied as a useful expression vector for stable and high-yield production of fusion proteins.

  • Recombinant baculoviruses are widely used to express heterologous genes in cultured insect cells. Recombinant baculoviruses can serve as gene-transfer vectors for expression of recombinant proteins in a wide range of mammalian cell types. Baculovirus system has significant benefits in view of safety, large-scale, and high level of gene expression. In this study, baculoviral vectors which were reconstructed from pOPINEneo-3C-GFP vector, were recombined with cytomegalovirus (CMV) promoter, green fluorescent protein (GFP), and p53 with NcoI and XhoI. These recombinant vectors were infected with various cells and cell lines. The baculovirus vector thus developed was analyzed by comparing the metastasis and expression of the recombinant genes with conventional vectors. These results suggest that the baculovirus vector has higher efficiency in metastasis and expression than the control vector.
