알카리 내성 Bacillus속 Promoter의 Cloning (Cloning of Promoters from Alkali-tolerant Bacillus sp.)

  • 토양에서 분리한 알카리 내성 Bacillus sp. YA-14 의 promoter를 Bacillus promoter probe vector인 pPL703을 이용하여 Bacillus subtilis내에 cloning 하였다. 얻어진 형질전환체 중 promoter 활성이 가장 높은 균주의 CAT 비활성은 8.07로 expression vector인 pPL708의 CAT 비활성보다 2.5배 이상 높았으며 대수 증식기가 끝난 이후에 그 활성이 급격하게 증가하였다. 재조합 plasmid내의 삽입 DNA 단편은 그 크기가 2.8kb이고 제한효소 BamHI, Sal I 인식부위가 각각 한군데 존재하였다.

  • In this paper, an efficient feature vector extraction method and MLP neural network are utilized to automatically detect and classify power quality disturbances, where the proposed classification procedure consists of the following three parts: i.e., (i) PQ disturbance detection using discrete wavelet transform. (ii) feature vector extraction from the detected disturbance. using several methods, such as FFT, DWT, Fisher's criterion. etc.. and (iii) classification of the corresponding type of each PQ disturbance by recognizing the pattern of the extracted feature vector. To demonstrate the performance and, applicability of the proposed classification algorithm. some test results obtained by analyzing 10-class PQ disturbances are also provided.

  • By using some existence theorems of maximal elements for a family of set-valued mappings involving a better admissible set-valued mapping under noncompact setting of FC-spaces, we present some non-empty intersection theorems for a family $\{G_i\}_{i{\in}I}$ in product FC-spaces. Then, as applications, some new existence theorems of equilibrium for a system of generalized vector equilibrium problems are proved in product FC-spaces. Our results improve and generalize some recent results.

  • To develop the host-vector system for industrial Coryneform bacteria that seemed to be the most suitable microorganisms for molecular breeding of genes involved in the production of amion acids, nucleotides, and other products of industrial interest, broad host range E. coli plasmid R 1162 DNA was transformed into Brevibacterium ammoniagenes and the plasmids pKU6 isolated from a transformant was physically characterized. All other plasmids from the transformed cells except pKU6 exsisted as multimeric forms in Brevibacterium ammoniagenes. The plasmid DNA was retransformed into Corynebacterium glutamicum with a high frequency ($1.32{\times}10^{-1}$ per cell) and maintained stably both in Brevibacterium ammoniagenes and Corynebacterium glutamicum after 100 generations of cultures with 25-30 copy number per cell. The size of both plasmid pKU6 and plasmid R1162 were the same and restriction maps by EcoR I, Ava I, Pst I, Pvu II and Hinc II were also similar.

  • Let G be a cyclic group of order 2 and let $S^1$ denote the unit circle in $R^2$ with the standard metric. We consider smooth G-vector bundles over $S^1$ when G acts on $S^1$ by reflection.   • Hepatitis B virus (HBV) infection is one of the serious problems in Southeast Asia including Korea because it causes chronic hepatitis, which can easily be transformed In fatal conditions such as cirrhosis and hepatoma. Even though lots of informations on structural characteristics and gene expression mechanisms have been accumulated, the mechanism for HBV-induced hepatocellular injury which is believed to be the consequences of the immunological response is not well understood. In order tn perform immunopathological studies for prevention and treatment of HBV infection, we designed transgenic mice as a disease model which can mimic HBV infection, In this study, a promoter-HBV DNA fragment for the preparation of HBV transgenic mice has been constructed. To add a proper enzyme site on 5' end of HBV gene, total HBV (subtype adr) gene was inserted into BamHI site of pBluescript SK vector and reextracted by PstI-SacI treatment A liver-specific promoter, rat ${\alpha}$ 2u globulin gene promoter, was insrted to pBluescript SK vector and reextracted by BamHI-PstI treatment, Promoter-HBV DNA was constructed by ligation of two fragments using identical PstI sites. For large scale production of promoter-HBV DNA, it was inserted to BamHI-SacI site of pBluescript SK vector.

  • In this paper, I propose an intelligent credit rating system using a bankruptcy prediction model based on support vector machines (SVMs). SVMs are promising methods because they use a risk function consisting of the empirical error and a regularized term which is derived from the structural risk minimization principle. This study examines the feasibility of applying SVM in Predicting corporate bankruptcies by comparing it with other data mining techniques. In addition. this study presents architecture and prototype of intelligeht credit rating systems based on SVM models.

Construction of a Baculovirus Expression System Using Hyphantria cunea Nuclear Polyhedrosis Virus for Eukaryotic Cells

  • Baculovirus transfer and expression vectors with Hyphantria cunea nuclear polyhedrosis virus (HcNPV) were constructed. An initial transfer vector, pHcEV, constructed using HcNPV was previously reported (Park et al. 1993. J. Kor. Soc. Viral. 23: 141-151). Herein, the size of the vector was properly reduced, and a functionally perfect vector was constructed and named pHcEV-IV (6.7 kb). The vector has a 2.2-kb HcNPV DNA sequence in the 5'-flanking region of the vector's polyhedrin gene promoter. The 1.8-kb HcNPV DNA sequence, poly A signal sequence, T3 primer sequence, and 13 multicloning site sequences, in order, were ligated in front of the translation start codon of the polyhedrin gene. The cloning indicating marker lacZ gene was inserted into the pHcEV-IV, named pHcEV-IV-lacZ, and transferred into the wild-type virus. Recombinant expression virus, lacZ-HcNPV, was constructed by replacing the lacZ gene in the pHcEV-IV-lacZ with the polyhedrin gene of the wild-type virus. The recombinant virus was isolated from blue plaques that produce $\beta$-galactosidase without polyhedra. The lacZ gene insertion was confirmed by Southern hybridization analysis. The expression of the lacZ gene in Spodoptera frugiperda cells infected with the lacZ-HcNPV was examined by SDS-PAGE and colorimetric assay. One 116-kDa LacZ protein band appeared on the PAGE. The production rate of the $\beta$-galactosidase was approximately 50 international units (IU) per min per ml between 2 to 5 days postinfection (p.i.). The highest activity occurred at five days p.i. was 170 IU/min/$m\ell$. The enzyme activity first appeared about 20 h p.i. as measured by colorimetric assay.

Investigation on the Effect of Multi-Vector Document Embedding for Interdisciplinary Knowledge Representation

  • Text is the most widely used means of exchanging or expressing knowledge and information in the real world. Recently, researches on structuring unstructured text data for text analysis have been actively performed. One of the most representative document embedding method (i.e. doc2Vec) generates a single vector for each document using the whole corpus included in the document. This causes a limitation that the document vector is affected by not only core words but also other miscellaneous words. Additionally, the traditional document embedding algorithms map each document into only one vector. Therefore, it is not easy to represent a complex document with interdisciplinary subjects into a single vector properly by the traditional approach. In this paper, we introduce a multi-vector document embedding method to overcome these limitations of the traditional document embedding methods. After introducing the previous study on multi-vector document embedding, we visually analyze the effects of the multi-vector document embedding method. Firstly, the new method vectorizes the document using only predefined keywords instead of the entire words. Secondly, the new method decomposes various subjects included in the document and generates multiple vectors for each document. The experiments for about three thousands of academic papers revealed that the single vector-based traditional approach cannot properly map complex documents because of interference among subjects in each vector. With the multi-vector based method, we ascertained that the information and knowledge in complex documents can be represented more accurately by eliminating the interference among subjects.

Inositol(1,4,5)triphosphate 3-Kinase의 유전자 재조합과 CCL39 Hamster Lung Fibroblasts에서 발현

  • IPSKCDNA gene(1.8 kbp) encoding rat brain IP3K enzyme contained Not I restric site in open reading frame. The Not I sequence, GCGGCCGC, was converted to GCAGCCGC by site-directed mutagenesis. The mutated IP3KcDNA was digested with EcoR I and ligated with EcoR I-restricted psp72·Not2 vector. The resulting psp72 · Not2-IP3KCDNA was digested with the Not I restriction enzyme and then subcloned into the Not I -digested PZIP · NeoSV(X) mammalian expression vector. The PZIP · NeoSV(X) -IPSKCDNA was transfected into CCL39 hamster lung fibroblast cells. The efficiency of the expressed IPSKCDNA gene was significantly higher than expected generally, not only a mean 5-fold increase in the amount of enzyme, but also 16-fold increase in enzyme activity from tractsfected CCL39 cells by the method of Western blot using anti-lP3K antibodies. Both distribution of IPSK in various rat tissues and biochemical properties were discussed.
