Oxygen Supersaturation in Korean Streams as a Stress Factor to Fish (어류 스트레스 요인으로서 우리나라 하천의 산소과포화 실태)

  • Abnormal dissolved oxygen concentration in aquatic habitat, both depletion and supersaturation, can be stress factor to aquatic animals. In this study the nationwide distribution of oxygen supersaturation was analyzed for three categories of streams (43 urban streams, 15 rural streams, and 14 forest streams) by using monitoring network data of the Korean Ministry of Environment. From the distribution analysis 30% of urban streams showed hyperoxic condition of eutrophic level, while no forest stream showed hyperoxic condition. The physiological effect of hyperoxia on fish was examined using two species of fish Zacco koreanus, which resulted in higher concentration of a stress hormone (cortisol) in fish exposed to hyperoxic concentration (196%) of oxygen. This study shows that hyperoxic condition is ubiquitous in urban and rural Korean streams, and it can be a stress factor to aquatic animals.

Lipid Peroxidation in Vivo Monitored as Ethane Exhalation in Hyperoxia (호기중 에탄(ethane)측정을 통한 산소중독시 지질과산화평가에 관한 실험적 연구)

  • In vivo ethane production in rats was used as an index of oxygen toxicity. The rats were allocated to four exposure conditions; hyperbaric oxygenation (HBO=5 ATA, 100% $O_2$), normobaric oxygenation (NBO=1 ATA,100% $O_2$), hyperbaric aeration (HBA=5 ATA, 21% $O_2$) and normobaric aeration (NBA=1 ATA, 21% $O_2$). After 120 minutes of exposure, the rats exposed to high concentration and/or high pressure oxygen exhaled significantly larger amounts of ethane than those exposed to NBA, and the differences in ethane production between any two groups were statistically significant (p<.01). This finding supports the hypothesis that hyperoxia increases oxygen free-radicals and the radicals produce ethane as a result of lipid peroxidation. It is notable that the ethane exhalation level of the HBA group was significantly higher than that of the NBO group. This difference could not be accounted for by the alveolar oxygen partial presure difference between the two groups.

The Effect of 30% Oxygen on the Memory Performance, Hyperoxia and Heart Rate (30% 농도의 산소 공급이 기억력, 혈중 산소 농도, 심박률에 미치는 영향)

  • In this study, changes in memory performance, blood oxygen saturation and heart rate according to 30% concentration oxygen supply were observed. Ten healthy male and female college students (male:$25.8{\pm}0.8$ years, female: $24.2{\pm}1.9$ years) participated in the study. The experiment was performed as Rest (1min.), Control task (1min.), Word presentation (1min.), Reaction time task (1min.), Distractor (2min.), and Word recall (1min.) and the physiological signals such as blood oxygen saturation and heart rate were measured throughout the stages. Subjects who received 30% oxygen recalled more words than those who received 21% oxygen, which shows 30% oxygen supply has influenced positively on memory cognitive performance. When 30% concentration oxygen is supplied, the blood oxygen saturation in the task phases was increased and the heart rate decreased when comparing to 21%. It means that 30% oxygen can stimulate brain activation by increasing actual blood oxygen concentration in the process of cognitive performance, and the heart rate decreases because enough oxygen is supplied to process the cognitive performance.

  • This study aimed to investigate the hypothesis that administration of the air with 30% oxygen compared with normal air (21% oxygen) enhances cognitive functioning through increased activation in the brain. Seventeen college students (right­handed, average age of 24.3) were selected as subjects for this study. An oxygen supply equipment that provides 21% and 30% oxygen at a constant rate of 8L/min was developed. In order to measure the performance level of visuospatial and verbal cognition, two psychological tests were developed. The experiment consisted of two runs, one for cognition task with normal air (21% oxygen) and the other for cognition task with hyperoxic air (30% oxygen). Visuospatial and verbal tasks were presented while brain images were scanned by a 3T fMRI system using the single-shot EPI method. The results showed that there was an improvement in performance and also increased activation in several brain areas in the higher oxygen condition. These results suggest that while performing cognitive tasks, high concentrations of oxygen administration make oxygen administration sufficient, thus making neural network activate more, and the ability to perform cognitive tasks increase.

Neutrophil Chemotactic Activity in Bronchoalveolar Lavage Fluid of the Rats Exposed to Hyperoxia (고농도의 산소에 노출시킨 쥐의 기관지폐포세척액내 호중구 화학주성활성화도)

  • Background : An excessive accumulation of neutrophils in lung tissue has been known to play an important role in mediating the tissue injury among the adult respiratory distress syndrome, idiopathic pulmonary fibrosis and cystic fibrosis by releasing toxic oxygen radicals and proteolytic enzymes. Therefore, it is important to understand a possible mechanism of neutrophil accumulation in lung tissue. In many species, exposure to hyperoxic stimuli can cause changes of lung tissues very similar to human adult respiratory distress syndrome and neutrophils are also functioning as the main effector cells in hyperoxic lung injury. The purpose of the present study was to examine whether neutrophils function as a key effector cell and to study the nature of possible neutrophil chemotactic factors found in bronchoalveolar lavage fluid from the hyperoxia exposed rats. Methods : We exposed the rats to the more than 95% oxygen for 24, 48, 60 arid 72 hours and bronchoalveolar lavage(BAL) was performed. Neutrophil chemotactic activity was measured from the BAT- fluid of each experimental groups. We also evaluated the molecular weight of neutrophil chemotactic tractors using fast performance liquid chromatography and characterized the substances by dialyzer membrane and heat treatment. Results : 1) The neutrophil proportions in bronchoalveolar lavage fluid began to rise from 48 hours after oxygen exposure, and continued to be significantly increased with exposure times. 2) chemotactic index for neutrophils in lung lavages from rats exposed to hyperoxia was significantly higher in 48 hours group than in control group, and was significantly increased with exposure time. 3) No deaths occured until after 48 hours of exposure. However, mortality rates were increased to 33.3 % in 60 hours group and 81.3 % in 72 fours group. 4) Gel filtration using fast performance liquid chromatography disclosed two peaks of neutrophil chemotactic activity in molecular weight of 104,000 and 12,000 daltons. 5) Chemotactic indices of bronchoalveolar lavage fluid were significantly deceased when bronchoalveolar lavage fluid was treated with heat ($56^{\circ}C$ for 30 min or $100^{\circ}C$ for 10 min) or dialyzed (dialyzer membrane molecular weight cut off : 12,000 daltons). Conclusion : These results suggested that the generation of neutrophil chemotactic factor and subsequent neutrophil influx into the lungs are playing an important roles in hyperoxia-induced acute lung injury. Neutrophil chemotactic factor in the lung lavage fluids consisted of several distinct components having different molecular weight and different physical characteristics.

Effects of Three Levels of Flow Rate of Highly Concentrated Oxygen Administration on SpO2 and HR of 60s Male and Female (세가지 유량의 고농도 산소 공급이 60대 남녀의 혈중 산소 포화도와 심박동률에 미치는 영향)

  • This study investigated differences in blood oxygen saturation($SpO_2$) and heart rate(HR) according to flow rate, gender, and phase in males and females in their 60s when the supply of 93% highly concentrated oxygen administration was changed (1L/min, 3L/min, and 5L/min). It recruited totally 20 elderly subjects including 10 males($68.0{\pm}2.6$ years) and 10 females ($65.5{\pm}3.1$ years). The experiment consisted of three phages of Rest 1(5 min), Hyperoxia(10 min), and Rest 2(10 min), and $SpO_2$[%] and HR[bpm] were measured during all phages. $SpO_2$ was higher in Hyperoxia phase supplied with highly concentrated oxygen than in Rest phases. Higher flow rate was associated with more increase in $SpO_2$. HR was reduced in Hyperoxia phase compared to Rest phases. More supply of highly concentrated oxygen was associated with more decrease of HR. However, there were no differences in both $SpO_2$ and HR according to gender.

Hyperoxia-Induced ΔR1: MRI Biomarker of Histological Infarction in Acute Cerebral Stroke

  • Objective: To evaluate whether hyperoxia-induced ΔR1 (hyperO2ΔR1) can accurately identify histological infarction in an acute cerebral stroke model. Materials and Methods: In 18 rats, MRI parameters, including hyperO2ΔR1, apparent diffusion coefficient (ADC), cerebral blood flow and volume, and 18F-fluorodeoxyglucose uptake on PET were measured 2.5, 4.5, and 6.5 hours after a 60-minutes occlusion of the right middle cerebral artery. Histological examination of the brain was performed immediately following the imaging studies. MRI and PET images were co-registered with digitized histological images. The ipsilateral hemisphere was divided into histological infarct (histological cell death), non-infarct ischemic (no cell death but ADC decrease), and nonischemic (no cell death or ADC decrease) areas for comparisons of imaging parameters. The levels of hyperO2ΔR1 and ADC were measured voxel-wise from the infarct core to the non-ischemic region. The correlation between areas of hyperO2ΔR1-derived infarction and histological cell death was evaluated. Results: HyperO2ΔR1 increased only in the infarct area (p ≤ 0.046) compared to the other areas. ADC decreased stepwise from non-ischemic to infarct areas (p = 0.002 at all time points). The other parameters did not show consistent differences among the three areas across the three time points. HyperO2ΔR1 sharply declined from the core to the border of the infarct areas, whereas there was no change within the non-infarct areas. A hyperO2ΔR1 value of 0.04 s-1 was considered the criterion to identify histological infarction. ADC increased gradually from the infarct core to the periphery, without a pronounced difference at the border between the infarct and non-infarct areas. Areas of hyperO2ΔR1 higher than 0.04 s-1 on MRI were strongly positively correlated with histological cell death (r = 0.862; p < 0.001). Conclusion: HyperO2ΔR1 may be used as an accurate and early (2.5 hours after onset) indicator of histological infarction in acute stroke.

  • 본 연구에서는 언어과제 수행 시 일반 공기 중의 산소 농도 (21%) 환경에 비해 외부에서 고 농도 (30%)의 산소 공급이 혈중 산소 포화도(SPO$_2$), 심박동율(Heart Rate), 정답률(Accuracy), 반응속도(Reaction Time)에 어떠한 영향을 미치는지를 검증하고자 한다. 30%와 21%의 산소를 8L/min의 양으로 일정하게 공급할 수 있는 산소 공급 장치를 이용하였고, 10명의 대학생(오른손잡이, 평균나이 23.4세)을 대상으로 실험을 수행하였다. 난이도가 비슷한 두 가지 언어과제를 28문제씩 피험자에게 풀게하여 정답률과 반응속도를 계산하였다.(중략)

  • The adverse effect of diving on the fetus may extend beyond n gestational process and outcome. Primiparous Sprague-Dawley rats were assigned to one of ten exposure schedules during gestatred $PO_2$ level, the following question about the effect of exposing a pregnant female to high partial pressure of inspired oxygen has been raised. 'What effect does an increased maternal $PIO_2$ have on fetal arterial $PO_2$ and therefore on possible fetal oxygen poisoning?' This study was carried out to observe the effects of maternal hyperoxia on gestational process and outcome. Primiparous Sprague-Dawley rats were assigned to one of ten exposure schedules during gestation. The treatment groups were subjected to either the high concentration of oxygen, or the high atmospheric pressure. On day 21 of gestation, laparotomy was performed to examine for number and distribution of implantations and live and resorbing embryos. Fetuses were weighed, and examined for gross malformations. Subsequently, they were fixed, measured in physical parameters, and examined for visceral anomalies. Minor visceral anomalies and anatomical variation was not found. Similarily, there were no significant differences when number of resorptions, mean fetal weights, pregnancy interruption rate were compared by analysis of variance. These results indicate that exposing rats to oxygen at increased atmospheric pressure doese not affect fetal health or survival.

The Optimal Pulse Oxygen Saturation in Very Low Birth Weight or Very Preterm Infants (극소 저체중 출생아에서 경피적 산소포화도의 적정 범위)

  • Purpose: To determine the effect of changing practice guidelines designed to avoid hyperoxia or hypoxia in very low birth weight or very preterm infants. Methods: We analyzed a database of <1,500 g birth weight or <32 weeks of gestation infants who were born and admitted to the neonatal intensive care unit of Chungnam National University Hospital from January 2007 to July 2010. First, we defined the relationship between arterial partial pressure of oxygen ($PaO_2$) and pulse oxygen saturation ($SpO_2$). When we evaluated 96 pairs of $PaO_2$ and $SpO_2$ measurements, oxygen saturation was 90-94% at a $PaO_2$ of 43-79 mmHg on the oxyhemoglobin dissociation curve, according to pulse oximetry. Based on this observation, a change in practice was instituted in August 2008 with the objective of avoiding hypoxia and hyperoxia in preterm infants with targeting a $SpO_2$ 90-94% (period II). Before the change in practice, high alarms for $SpO_2$ were set at 100% and low alarms at 95% (period I). Results: Sixty-eight infants the met enrollment criteria and 38 (56%) were born during period II, after the change in $SpO_2$ targets. Demographic characteristics, except gender, were similar between the infants born in both periods. After correcting for the effect of confounding factors, the rates for mortality, severe retinopathy of prematurity, and IVH attended to be lower than those for infants in period II. No difference in the rate of patent ductus arteriosus needed to treat was observed. Conclusion: A change in the practice guidelines aimed at avoiding low oxygen saturation and hyperoxia did not increase neonatal complication rates and showed promising results, suggesting decreased mortality and improvements in short term morbidity. It is still unclear what range of oxygen saturation is appropriate for very preterm infants but the more careful saturation targeting guideline should be considered to prevent hypoxemic events and hyperoxia.
