Effect of Dietary Fat and Genistein on Lipid Metabolism and Antioxidant Activity in Hyperlipidemic Male Rats induced High Fat Diet (고지방식이로 유도된 고지혈증 모델 흰쥐에서 지방과 제니스테인 섭취가 지질대사 및 항산화능에 미치는 영향)

  • This study was conducted to investigate whether dietary factors, normal fat and genistein leads to beneficial improvement of lipid metabolism and oxidative stress in adult hyperlipidemic male rats. Seven wk-old male SD rats were fed high fat diet (15% fat, 1% cholesterol) for 4 wks for induction of hyperlipidemic model rat. Weight-matched rats were then assigned to four groups according to dietary fat level (7% or 15% fat) and genistein contents (0 or 320 mg/kg diet). Food intake was significantly decreased by both high fat intake and genistein supplementation compared with normal fat intake and genistein no supplementaion. But weight gain was significantly decreased by genistein supplementation in normal fat intake compared with the other groups. Total lipid, total cholesterol and triglyceride in serum and liver were significantly decreased by normal fat intake compared with high fat intake. But total cholesterol in liver was significantly increased by genistein supplementation in both high fat and normal fat intake. TBARS in serum and liver was less produced by normal fat intake compared with high fat intake but TBARS in liver was significantly increased by genistein supplementation compared with genistein no supplementation in normal fat intake. Glutathione reductase activity in erythrocytes was significantly reduced by genistein supplementation in normal fat intake compared with the other groups. Glutathione peroxidase and glutathione reductase activities in liver were significantly inhibited by normal fat intake compared with high fat intake. Catalase activity in liver was significantly increased by genistein supplementation compared with genistein no supplementation in high fat intake. Nitrite was significantly decreased by normal fat intake compared with high fat intake. These results suggest that normal fat intake has the treatment effect against risk factors related with cardiovascular disease by reducing lipid profiles, lipid peroxidation. And genistein shows action as a antioxidant replacing antioxidant enzymes but also may act as prooxidant causing the production of TBARS.

Studies on Diet Regimens for the Developement of Hyperlipidemic Animal Model (고지혈증 동물모델 설정을 위한 식이처방의 실험적 연구)

  • Development of hyperlipidemic animal model is prerequisite for the screening of drugs lowering the blood lipoprotein level. In this study, two kinds of diet regimens were tried for the hyperlipidemic effect. The model will be used for the screening of current prescriptions to a variety of diseases related to hyperlipidemia by practioner of traditional herbal medicine. Fifteen male Wister rats of 200g were divided into three groups, and given normal, or either one of the two high cholesterol diets for up to 2 weeks. Regimen HC-1 and regimen HC-2 contained I% and 2% cholesterol in diet, respectively. Total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL) and lower density lipoprotein cholesterol (LDL) levels in the serum were measured before the start of the diet, at 1 week after the start, and at 2 week point. Both regimens resulted in hyperlipidemia with the typical characteristics of increase in TC and LDL, and reduction in HDL, and showed no observable side effects such as diarrhea. In conclusion, both of HC-1 and HC-2 regimens appeared appropriate as diet regimens for the hyperlipidemic rat model.

Effects of Kyeongok-go and Its Two Added Precriptions on Hyperlipidemic Rats Induced by High-fat Diet (경옥고(瓊玉膏) 및 그 가미방(加味方)이 고지방 식이로 유발된 흰쥐의 고지혈증 병태에 미치는 영향)

  • This study was designed to investigate the antihyperlipidemic activity of aqueous extracts of Kyeongokgo (KOG), Kyeongokgo Dansambang (KDB), and Kyeongokgo Gamibang (KGB) on the hyperlipidemic rats induced by high-fat diet. The animals were divided into 4 groups of the hyperlipidemic control group, the KOG-fed group, the KDB-fed group and the KGB-fed group, and received oral administration of each prescription with diet for a period of six weeks. The changes in the body weight, the amount of food intake, the liver weight, the epididymal fat weight, and the plasma levels of total cholesterol, triglyceride, HDL-cholesterol and LDL-cholesterol were measured. In all experimental groups, the weight of liver and epididymal fat decreased significantly. There was a significant decrease in the amount of food intake, the weight of liver and epididymal fat, the plasma level of total cholesterol, and HDL-cholesterol in the KDB-fed and the KGB-fed groups. In addition, KGB-fed group represented a significant decrease in the triglyceride. These results suggest that the aqueous extracts of Kyeongokgo Dansambang and Kyeongokgo Gamibang have a good antihyperlipidemic activity and a potential to treat hyperlipidemia.

Effect of the MeOH Extract of Adventious Root Culture of Panax ginseng on Hyperlipidemic Rat Induced by High Fat-rich Diet (조직배양산삼 부정근 메탄올 추출물이 식이성 고지혈증에 미치는 영향)

  • We have investigated the effect of MeOH extract of the adventious root culture of Panax ginseng on hyperlipidemic rats induced by fat-rich diet. We also analyzed and compared the content ginsenoisdes of the cultured ginsengs by HPLC. After oral administration of the MeOH extract to hyperlipidemic rats for four weeks, the variables including body weight, cholesterol, HDL, LDL, and triglyceride levels in serum were measured. One of the cultured ginseng roots (CBN3) decreased cholesterol and LDL-cholesterol and increased HDL-cholesterol levels in serum. HPLC analysis demonstrated that CBN3 contains more $Rb_1$ and Rd ginsenosides than any other cultured ginsengs and cultivated ginseng. These results suggest that CBN3 with high contents of $Rb_1$ and Rd may be useful in lowering blood cholesterol.

The Effect of Achyranthis Radix Herbal-acupuncture on Hyperlipidemia in Rats (양릉천(陽陵泉) 우슬(牛膝) 약침이 흰쥐의 고지혈증에 미치는 영향)

  • Objective & Methods : The purpose of this study is to observe the effects of herbal- acupuncture with Achyranthis Radix(AR-HA) at GB34(Yangleungchean) on hyperlipidemia in rats. The author performed several experimental items to analyze the levels of various components and enzymes in serum and liver, as well as the histological changes of liver and aorta. Results : 1. Achyranthis Radix herbal acupuncture solution increased DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. AR-HA at GB34 decreased the level of serum total cholesterol, and increased the ratios of HDL-cholesterol to total cholesterol, phospholipid to total cholesterol in hyperlipidemic rat. 3. AR-HA at GB34 decreased the hepatic HMG-CoA reductase activity in hyperlipidemic rat. 4. AR-HA at GB34 increased the hepatic GSH level in hyperlipidemic rat. Conclusion : From the above results, it is suggested that AR-HA at GB34 has a therapeutic effect on hyperlipidemia.

Lipid lowering activity of Ponciri Fructus and Aurantii Fructus Immaturus on hyperlipemia rats induced by Triton WR-1339. (국산과 중국산 지실(枳實)의 Triton WR-1339 유발 고지혈증(高脂血症) 흰쥐에 대한 효능 연구)

  • Objective : In order to elucidate hyperlipidemic effects of the fruits of Poncirus trifoliata and Citrus aurantium, our experimental study was performed on hyperlipidemia rats induced by Triton WR-1339. Method : The hyperlipidemia rats induced by intraperitonial injection of triton WR-1339 were treated with the fruits of P. trifoliata (rare and preparata) and C. aurantium 50 and 200mg/kg. The amount of triglyceride. total cholesterol. HDL-cholesterol, LDL-cholesterol, AST. and ALT levels were measured. Results: The levels of triglyceride. total cholesterol and LDL-cholesterol were significantly reduced in fruits of P. trifoliata (rare and preparata) and C. aurantiu. Simvastatine treated group compared with those of control but the level of HDL-cholesterol was incresed. The level of AST and ALT were not significantly reduced by the treatment to the fruits of P. trifoliata (rare and preparata) and C. aurantium. Conclusion : In the hyperlipidemia rats induced by triton WR-1339. the fruits of P. trifoliata (rare and preparata) and C. aurantium showed a hyperlipidemic effect.

Effects of the Water Extract from the Stem Bark of Acanthopanax senticosus on Hyperlipidemia in Rats (가시오갈피 줄기의 물 추출물이 지질대사에 미치는 영향)

  • The effects of the water extract from the stem bark of Acanthopanax senticosus were evaluated on hyperlipidemic rats induced by lipid rich diet or poloxamer-407. The water extracts, when administered orally for 3 consecutive days in hyperlipidemic rats induced by poloxamer-407 (1 ml of 30%), was found to cause a significant decrease in plasma cholesterol and triglyceride concentrations. The water extracts, when treated orally for 5 consecutive days also showed a significant inhibition of serum total cholesterol and triglyceride level in rats treated with lipid rich diet (15% cholesterol, 1% sodium cholate and 84% com oil). HDL-cholesterol, however, was increased significantly.

Beneficial effects of natural Jeju groundwaters on lipid metabolism in high-fat diet-induced hyperlipidemic rats

  • BACKGROUND: Groundwater is believed to possess many beneficial effects due to its natural source of various minerals. In this study, we examined the effects of natural Jeju groundwater S1 (Samdasoo$^{TM}$), S2 and S3 pumped up from different locations of Jeju Island, Korea, along with local tap water, on body weight gain, serum lipids and lipoproteins, and liver histopathology in high-fat diet-induced hyperlipidemic rats. MATERIALS/METHODS: Rats were randomly and equally divided into 6 groups. Different water samples were supplied to the hyperlipidemic rats as their daily drinking water and the widely-used anti-hyperlipidemic drug simvastatin was used as a positive control. Body weight, serum lipids and lipoproteins were measured weekly. Liver weight, liver index and liver histopathology were examined after the execution of the rats. RESULTS: After drinking Jeju groundwaters for two months, S2 but not S3 significantly reduced weight growth and serum triglycerides levels and increased high density lipoprotein-C (HDL-C) without affecting total cholesterol or LDL-C. S1 and particularly S2 significantly reduced the severity of liver hypertrophy and steatosis. All Groundwaters had much higher contents of vanadium (S3>S2>S1>>tap water) whereas S1 and S2 but not S3 markedly blocked autoxidation of ferrous ions. CONCLUSION: Jeju Groundwater S1 and particularly S2 exhibit protective effects against hyperlipidemia and fatty liver and hypothesize that the beneficial effect of Jeju Groundwaters may be contributed from blockade of autoxidation of ferrous ions rather than their high contents of vanadium.

The therapeutic effects of Bangkeehwangkee-tang and Bangkeebokryeong-tang on the hyperlipidemia in rats (방기황기탕(防己黃?湯) 및 방기복령양(防己茯?陽)이 고지혈증(高脂血症) 흰쥐에 미치는 효과(效果))

  • Objectives : Each 2 types of Bangkeehwangkeetang(BHT) and Bangkeebok-ryeongtang(BBT) was prescribed to examined the therapeutic effects on hyperlipidemia. BHT-1 and BBT-1 were composed of Bunbangkee(粉防己, Stephaniae Radix) for Bangkee with other drugs, but BHT-2 and BBT-2 were composed of Cheongpungdeung(淸風藤, Sinomenii acuti Lignum). Methods : Four decoctions prepared from the prescriptions were respectively administrated to animal models in rats such as hyperlipidemic model induced by high cholesterol diet. Results : On the hyperlipidemic model, BHT-2 and BBT-2 would decrease the levels of total cholesterol(Tc) and triglyceride(TG) in blood, on the other hand, BHT-1 and BBT-1, despite little change of Tc, decrease TG but also HDL-cholesterol(HDLc). Conclusion : BBT, especially composed of Cheongpungdeung(Sinomenii acuti Lignum) for Bangkee, could be used more effectively than the others(Stephaniae Radix) on hyperlipidemia.
