동충하초(Cordyceps Militaris)가 고콜레스테롤혈증 흰쥐의 지질대사에 미치는 영향 (Effect of Cordyceps Militaris on Lipid Metabolism in Rats Fed Cholesterol Diet)

  • The effects of Cordyceps militaris on the weight gain, the food intake, the food efficient ratio, and the lipid concentrations of serum, liver, pancreas, heart and aorta in male rats fed cholesterol diet were studied. Male Sprague-Dawley rats were received 4 types of diets for 4 weeks, respectively : a normal diet(without cholesterol), a control diet(hypercholesterolemic diet), a hypercholesterolemic diet supplemented with 3% fruiting body of Cordyceps militaris(CF), a hypercholesterolemic diet supplemented with 3% mycelium of Cordyceps militaris(CM). The body weight gain of rats fed the control diet were significantly increased compared to those of the rats fed diets with CF or CM. But, the hepatic and pancreatic weights of rats fed diets with CF or CM were heavier than those of rats fed the normal diet. In serum of rats fed the CM diet, the concentrations of total cholesterol, triglyceride, LDL-cholesterol and the atherogenic index were significantly lower than those of rats fed the control diet. Also, serum HDL-cholesterol concentration and HDL-cholesterol/total cholesterol ratio were significantly lower in rats fed the control, CF or CM diets than those in rats fed the normal diet. Whereas, the hepatic concentrations of the total lipid, the total cholesterol and the triglyceride were significantly higher in rats fed 3 hypercholesterolemic diets than those in the rats fed normal diet. The heart and pancreatic concentrations of total cholesterol were the highest in the rats fed diet with the CR among rats fed experimental diets. In conclusion, the CM feeding decreased the concentrations of the cholesterol, triglyceride, and LDL-cholesterol and the atherogenic index in the serum of the rats. But the CF or CM feeding could not decrease the hepatic concentrations of the total lipid, cholesterol, triglyceride in the rats fed hypercholesterolemic diets. Hypolipidemic Activities of Dietary Pleurotus ostreatus in Hypercholesterolemic Rats

  • This work was conducted to investigate dietary supplementation of oyster mushroom fruiting bodies on biochemical and histological changes in hyper and normocholesterolemic rats. Six-week old female Sprague-Dawley albino rats were divided into three groups of 10 rats each. Feeding a diet containing a 5% powder of Pleurotus ostreatus fruiting bodies to hypercholesterolemic rats reduced plasma total cholesterol, triglyceride, low-density lipoprotein (LDL), total lipid, phospholipids, and LDL/high-density lipoprotein ratio by 30.18, 52.75, 59.62, 34.15, 23.89, and 50%, respectively. Feeding oyster mushrooms also significantly reduced body weight in hypercholesterolemic rats. However, it had no adverse effects on plasma albumin, total bilirubin, direct bilirubin, creatinin, blood urea nitrogen, uric acid, glucose, total protein, calcium, sodium, potassium, chloride, inorganic phosphate, magnesium, or enzyme profiles. Feeding mushroom increased total lipid and cholesterol excretion in feces. The plasma lipoprotein fraction, separated by agarose gel electrophoresis, indicated that P. ostreatus significantly reduced plasma ${\beta}$ and pre-${\beta}$-lipoprotein but increased ${\alpha}$-lipoprotein. A histological study of hepatic cells by conventional hematoxylin-eosin and oil red O staining revealed normal findings for mushroom-fed hypercholesterolemic rats. These results suggest that a 5% P. ostreatus diet supplement provided health benefits by acting on the atherogenic lipid profile in hypercholesterolemic rats.

고콜레스테롤혈증을 유도한 쥐를 대상으로 천년초 추출물의 콜레스테롤 저하 효과 (Hypocholesterol Effect of Opuntia humifusa Extract on High Cholesterol Diet-induced Hypercholesterolemic Rats)

  • 본 연구는 흰쥐를 이용하여 고콜레스테롤 식이로 고콜레스테롤혈증을 유도하고 이에 천년초 추출물을 2, 4% 식이에 포함시켜 경구로 섭취 시의 천년초 추출물의 콜레스레롤 개선 효과를 알아보고자 하였다. 이에 혈중 및 간 조직 내 지질성분, 혈중 간 손상 지표, 분변 내 지질 성분 및 담즙산을 분석하였다. 천년초 추출물 섭취는 고콜레스테롤 식이로 상승된 혈청 총 콜레스테롤과 저밀도 지단백 콜레스테롤을 정상 수준으로 감소시켰으며 고밀도 콜레스테롤은 증가시켜 혈중 콜레스테롤 개선 효과를 나타내었다. 고콜레스테롤 식이 유도로 인해 상승된 혈중 ALT와 AST는 천년초 추출물 처치로 감소되는 경향을 보였으며 고콜레스테롤 식이로 인해 상승된 간 조직 내의 총 콜레스테롤과 중성 지방 역시 천년초 추출물 처치로 인해 용량 의존적으로 감소되었다. 분변 내 중성 지방과 담즙산은 천년초 추출물 처치로 유의적으로 상승되어 천년물 추출물 섭취로 중성 지방과 담즙산 배설이 유도가 이루어짐을 알 수 있었다. 본 연구 결과 천년초 추출물은 고콜레스테롤 식이로 고콜레스테롤이 유도된 쥐에 있어 콜레스테롤 개선 효과의 가능성 있는 식품 소재로 판단되며, 이를 인체에 적용시키기 위해서는 이에 대한 기전연구와 활성 물질 동정에 대한 계속적인 연구가 이루어져야 할 것으로 생각되어진다.

Antihyperlipidemic Effect of Dietary Lentinus edodes on Plasma, Feces and Hepatic Tissues in Hypercholesterolemic Rats

  • We investigated diet supplementation with shiitake mushroom fruiting bodies on biochemical and histological changes in hypercholesterolemic rats. Six-wk old female Sprague-Dawley albino rats were divided into three groups of 10 rats each. A diet containing 5% Lentinus edodes fruiting bodies given to hypercholesterolemic rats reduced plasma total cholesterol, triglyceride, low-density lipoprotein (LDL), total lipid, phospholipids, and the LDL/high-density lipoprotein ratio by 34.33, 53.21, 75.00, 34.66, 25.73, and 71.43%, respectively. Feeding mushroom also significantly reduced body weight in hypercholesterolemic rats. However, it had no detrimental effects on plasma albumin, total bilirubin, direct bilirubin, creatinine, blood urea nitrogen, uric acid, glucose, total protein, calcium, sodium, potassium, chloride, inorganic phosphate, magnesium, or enzyme profiles. Feeding mushroom increased total lipid and cholesterol excretion in feces. The plasma lipoprotein fraction, separated by agarose gel electrophoresis, indicated that L. edodes significantly reduced plasma ${\beta}$ and pre-${\beta}$-lipoprotein but increased ${\alpha}$-lipoprotein. Porphyran 음료 투여가 고콜레스테롤 유발 흰쥐의 혈청과 간중의 콜레스테롤함량에 미치는 영향 (Effect of Porphyran Drink on Serum and Liver Cholesterol Contents in Hypercholesterolemic Rat)

  • 본 연구는 김에서 추출한 porphyran을 음료로 이용하여 Sprague-Dawley계 흰쥐를 5군으로 나누어 정상식이에 물, 고콜레스테롤 식이에 물, 고콜레스테롤 식이에 1, 2, $3\%$ porphyran 음료를 4주간 급여하여 혈액과 간중의 지방과 콜레스테롤 함량을 측정한 결과는 다음과 같다. 혈청중 중성지방과 총콜레스테롤 함량은 정상군에 비해 대조군에서 높게 나타났으며, porphyran 음료군은 대조군보다 유의하게 낮았으며, 이는 섭취수준이 높을수록 더 낮아지는 경향을 나타냈다. 혈청중 HDL-콜레스테롤 함량은 대조군에서 가장 낮게 나타났으나 porphyran 음료 섭취수준이 높아질수록 높아지는 경향을 보였다. 간중의 중성지방과 총콜레스테롤 함량 역시 대조군에 비해 porphyran음료 섭취수준이 높을수록 유의하게 낮은 경향을 나타냈다. 이들 결과로 볼 때 고 콜레스테롤 식이를 섭취시킨 흰쥐에서 김에서 추출한 porphyran 을 음료의 형태로 섭취 시 체내 중성지방과 콜레스테롤을 낮추어 지방개선의 효과가 있음을 알 수 있었다.

  • This study was designed to observe the effects of the feeding Callophyllis japonica extracts on the improvement of the blood glucose, lipid in the serum of dietary hypercholesterolemic rats(S.D. strain, ?) fed the experimental diets for 4 weeks. Concentrations of total cholesterol and LDL-cholesterol in serum were significantly lower in the Callophylis japonica extracts group than in the choloesterol supplemented diet group. Concentrations of HDL-cholesterol in serum were higher in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. Concentrations of free cholesterol, triglyceride, phospholipid and blood glucose in serum were significantly lower in the Callophyllis japonica extracts group than in the cholesterol supplemented diet group. The activities of aspatate aminotransferae(AST), alanine aminotransferase(ALT), lactate dehydrogenase(LDH) and alkaline phosphatase(ALP) in serum were lower in the CAllophyllis japonica extracts supplemented group than in the cholesterol diet group. From the above research, the physiological activity substances in Callophylis japonica extracts were effective on the improvement of the blood glucose, lipid compositions in serum of dietary hypercholesterolemic rats.

Appraisal of Antihyperlipidemic Activities of Lentinus lepideus in Hypercholesterolemic Rats

  • The wild edible mushroom, Lentinus lepideus has recently been cultivated for commercial use in Korea. While the mushroom has been widely used for nutritional and medicinal purposes, the possible anti-hyperlipidemic action is unclear. The effects of dietary L. lepideus on plasma and feces biochemical and on the liver histological status were investigated in hypercholesterolemic rats. Six-wk-old female Sprague-Dawley albino rats were divided into three groups of 10 rats each. Biochemical and histological examinations were performed. A diet containing 5% L. lepideus fruiting bodies reduced plasma total cholesterol, triglyceride, low-density lipoprotein, total lipid, phospholipids, and the ratio of low-density to high-density lipoprotein. Body weight was reduced. The diet did not adversely affect plasma biochemical and enzyme profiles. L. lepideus reduced significantly plasma ${\beta}$- and pre-${\beta}$-lipoprotein, while ${\alpha}$-lipoprotein content was increased. A histological study of hepatic cells by conventional hematoxylin-eosin and oil red O staining revealed normal findings for mushroom-fed hypercholesterolemic rats. The present study suggests that a diet supplemented with L. lepideus can provide health benefits by acting on the atherogenic lipid profile in hypercholesterolemic rats.

Effects of the Cynanchum wilfordii Ethanol Extract on the Serum Lipid Profile in Hypercholesterolemic Rats

  • The purpose of this study was to investigate the effects of the ethanol extract of Cynanchum wilfordii (ECW) on the blood lipid profile of hypercholesterolemic rats. Thirty 7-week-old male Sprague-Dawley rats were allowed free access to either a normal diet (AIN-93 diet), or 1% high-cholesterol diet with or without 0.5% or 1% ECW for 5 weeks. After sacrifice, the rat serum lipid profile was analyzed. The diets containing ECW decreased body weight gains compared to the normal diet. Serum HDL-cholesterol levels of ECW-fed groups were significantly increased in the hypercholesterolemic groups and normal groups (P<0.05). When 1% ECW was fed to the normal group, total cholesterol level was increased. Moreover, treatment of ECW in hypercholesterolemic groups yielded a dose-dependent and highly significant decrease in the atherogenic index as compared to the control. These results suggest that intake of Cynanchum wilfordii may help reduce the risks of hypercholesterolemia by increasing blood HDL-cholesterol and lowering the atherogenic index.

동충하초 엑스의 고콜레스테롤혈증 대사효소 활성 변동에 관한 효과 (Studies of the Cordyceps militaris Extract Administration on the Metabolic Enzyme Activities in Hypercholesterolemia)

  • The objective of this study was to investigate the effects of the feeding Cordyceps militaris extract on the improvement of the free fatty acid, lipid peroxide, creatinine and enzyme (creatine phosphokinase; CPK, lactate dehydrogenase; LDH, alkaline phosphatase; ALP, lecithin cholesterol acyltransferase; LCAT) activities in the sera of dietary hypercholesterolemic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of free fatty acid, lipid peroxide and CPK, LDH, ALP activities in sera were fairly reduced in the Cordyceps militaris extract administration group (CHE) than in the hypercholesterolemic diet group (CHD). However, no significance was found in the effect of an creatinine concentration among the groups. The LCAT activity in serum was increased in the Cordyceps militaris extract administration (CHE) than in the hypercholesterolemic diet group (CHD). From these results, Cordyceps militaris extracts were effective on the improvement of the lipid components and metabolic enzyme activities in sera of dietary hypercholesterolemic rats.

Antioxidant Effects of Cranberry Powder in Lipopolysaccharide Treated Hypercholesterolemic Rats

  • This study was conducted to investigate the effects of cranberry power on antioxidant defense system in rats fed an atherogenic diet and injected with lipopolysaccharide (LPS). Sprague-Dawley rats were divided into the following 5 groups: normal diet+saline (NS), atherogenic diet+saline (AS), atherogenic diet+LPS (AL), atherogenic diet with 5% cranberry powder+LPS (AL-C5), and atherogenic diet with 10% cranberry powder+LPS (AL-C10). Total antioxidant status measured by ferric reducing ability of plasma (FRAP) was significantly reduced by LPS injection (24%) and was restored by the cranberry powder treatment (P<0.05). In addition, the mean level of plasma total phenolics was significantly decreased by LPS injection (P<0.05) and tended to be increased when cranberry powder was incorporated in to the diet. Activity of serum superoxide dismutase (SOD) tended to be lowered by LPS injection and declined further in cranberry powder fortified groups. Overall results indicate that dietary cranberry powder may provide appropriate antioxidants to counter the diminished antioxidant status induced by exposing hypercholesterolemic rats to LPS.
