Effects of Leonuri Herba and Leonuri Semen on Hypercholesterolemia (익모초(益母草)와 충울자(茺蔚子)의 고지혈증(高脂血症) 억제 효능)

  • Objectives:The present study purposed to evaluate the effects of Leonuri Herba and Leonuri Semen on hypercholesterolemia and provide data for the appropriate clinical application of Leonuri Herba and Leonuri Semen. Methods:We applied the water extracts of Leonuri Herba and Leonuri Semen to rats with hypercholesterolemia induced by high-cholesteroldiet, and examined their effects. Results:The levels of the total cholesterol and LDL-cholesterol were reduced by the uptake of Leonuri Herba to rats with hypercholesterolemia, but there was no significant effect on the level of TG(triglyceride), HDLc(HDL-cholesterol), FFA(free fatty acid), and Fc(free cholesterol), while the amount of AST and ALT was inhibited. The application of Leonuri Semen brought about the decrease of the total cholesterol and Fc, the increase of TG, HDLc, and FFA, and the significant decrease of the activation of AST. Leonuri Herba had only a weak effect on hypercholesterolemia, while Leonuri Semen had a strong effect on the reduction of the symptoms of hypercholesterolemia. Conclusion:Based on the fact that Leonuri Herba had only a weak effect on hypercholesterolemia, while Leonuri Semen had a strong effect on the reduction of the symptoms of hypercholesterolemia, it is concluded that Leonuri Semen is a better solution for hypercholesterolemia than Leonuri Herba.

The effects of Hagochosan(HGCS) on hyperlipidemia in rats induced by high cholesterol diet (하고초산(夏枯草散)이 고지혈(高脂血) 유발(誘發) SHR 백서(白鼠)에 미치는 영향(影響))

  • In order to investigate the effect of HGCS on Hypercholesterolemia in rats diagnosed with hypertension, changes of related factors in blood and tissues of animal models were studied. The results are as following: At 250 ${\mu}g/m{\ell}$ or lower doses of HGCS, human fibroblast cells showed a survival rate of more than 90%. HGCS significantly reduced the total cholesterol and LDL cholesterol levels in blood observed in hypercholesterolemia rats. On the other hand, HGCS signicantly increased the blood HDL cholesterol level, compared to that of the control group. HGCS significantly reduced triglycerides caused by hypercholesterolemia. However, glucose level was decreased insignificantly. Significant decrease of hyperoxidative lipid level in tissues due to hypercholesterolemia was observed in HGCS treated group. Significant increase of SOD and catalase activities in tissues due to hypercholesterolemia were observed in HGCS treated group. From the results above, the anti-hypercholesterolemic effect of clinically used HGCS has been experimentally proved. More active prescription of HGCS to hypercholesterolemia and related patients should be available in the future.

  • In order to study the effects of Ohwangsahwatang on Hyperchole-sterolemia, experimental study were performed on Hypercholesterolemia Rats induced by 2% cholesterol diet. Also the level of total cholesterol. triglyceride, LDL-cholesterol. HDL-cholesterol, phospholipid, transami-nase(GOT, GPT) were measured. The results were summerized as follows : In the model of Hypercholesterolemia induced by 2% cholesterol diet in rats, Ohwangsahwatang showed decreasing effects on total cholesterol, triglyceride, LDL-cholesterol, phospholipid level, and transaminase (GOT, GPT) activity in serum significantly, depending on the concentration, and showed increasing effects on HDL-cholesterol level in serum significantly depending on the concentration. According to the above results, Ohwangsahwatang showed significant decreasing effects on Hypercholesterolemia, and it is considered that it is appropriate to apply for Hypercholesterolemia.

The Antithrombotic and Fibrinolytic Effect of Natto in Hypercholesterolemia Rats

  • Antithrombotic and fibrinolytic activity of natto was evaluated on platelet aggregation in vitro and in vivo. Natto showed inhibitory effects on platelet aggregation induced by adenosine 5’diphosphate (ADP) and collagen. Orally administered natto also showed fibrinolytic activity in hypercholesterolemia rats. Normal levels of natto, when administered for four weeks, shortened euglobulin clot lysis time (ECLT) and prolonged partial thromboplastin time (PATT) significantly compared to non-treated group. In addition, the natto treatment decreased total cholesterol in serum. These results showed that intake of normal levels of natto can elicit antithrombotic and fibrinolytic effects, suggesting its consumption may improve blood circulation.

Dietary chitosan enhances hepatic CYP7A1 activity and reduces plasma and liver cholesterol concentrations in diet-induced hypercholesterolemia in rats

  • The present study was performed to elucidate the hypocholesterolemic action of chitosan on the diet-induced hypercholesterolemia in rats. Male Sprague-Dawley rats (n=24) were fed with chitosan-free diet (Control), diets containing 2% or 5% chitosan for 4 weeks. Hypercholesterolemia was induced by adding 1% cholesterol and 0.5% cholic acid to all diets. Body weight gain and food intake of rats did not differ among the groups. The chitosan treated groups showed significant improvement in the plasma concentration of total cholesterol and LDL-cholesterol compared to the control group (p<0.05). Also, the chitosan treated groups decreased the liver concentration of total lipid and total cholesterol compared to the control group (p<0.05). The activity of hepatic cholesterol $7{\alpha}-hydroxylase$ (CYP7A1), the rate-limiting enzyme in the conversion of cholesterol to bile acids, was increased by 123% and 165% for the 2% or 5% chitosan diets, respectively. These findings suggest that enhancement of hepatic CYP7A1 activity may be a mechanism, which can partially account for the hypocholesterolemic effect of dietary chitosan in cholesterol metabolism.

The Inhibiton Effects of Hypercholesterolemia and Platelet in Fermented and Non-Fermented Preparation of Garlic

  • This Dietary cholesterol augments lipid profile and primes production and activation of platelets, leading to development of atherosclerosis which produce several detrimental effects on cardiovascular health. Ethnomedicine and Mediterranean diet are natural sources and cost effective modes against several ailments including cardiovascular diseases while fermented foods have gained interest due to their increased nutrient profile, enhanced bioavailability and efficacy. Garlic has been known to reduce cholesterol and inhibit platelet activation. We examined whether fermented garlic ameliorates effects of hypercholesterolemia and platelet functions in rats. Methodology: Male SD rats were fed with hypercholesterolemia diet and treated with spirulina, fermented and non-fermented preparations of garlic for one month. Platelet aggregation and granule secretion were assessed to evaluate platelet activation. Liver and kidney weights, lipid and enzymatic profile of serum and whole blood analysis was performed. Expressions of SREBP, ACAT-2 and HMG-CoA were assessed using RT-PCR while liver and adipose tissues were analyzed for histological changes. Both fermented and non-fermented garlic inhibited platelet aggregation and granule secretion while fermented garlic showed greater inhibitor tendency. Fermented garlic significantly reduced liver weight and triglycerides concentrations than non-fermented garlic. Similarly, fermented garlic greatly abrogated the detrimental effects of steatosis on liver and adipose tissues. Fermented garlic significantly improved lipid profile and modulated platelet functions, thereby inhibiting atherosclerosis and platelet related cardiovascular disorders.

Effects of dietary taurine supplementation on plasma and liver lipids in OVX rats fed calcium-deficient diet

  • Taurine supplementation has been shown to have an effect on lowering blood lipids in ovariectomized (OVX) rats. It therefore seemed desirable to find out whether the beneficial effect of taurine on OVX rats fed calcium-deficient diet could also be reproduced. Forty female Sprague-Dawley rats were divided into two groups. One group was OVX and the other group received a sham operation (Sham). Each rat group was further divided into the control diet and the taurine supplemented (2.0g/100g diet) diet group. All rats were fed on calcium-deficient diet and deionized water ad libitum for 6 weeks. Plasma and liver lipids were determined by using commercial kits. LDL-cholesterol concentrations were estimated with the equation of Friedewald et al. (1972). There were no significant differences in body weight gain and food intake between the control and taurine group within Sham and OVX groups, but body weight gain, food intake, and food efficiency ratio was higher in the OVX group. Concentrations of plasma total cholesterol, triglyceride, LDL-cholesterol were significantly lower in the taurine fed group of OVX rats fed Ca deficient diet, while HDL-cholesterol concentration was increased in the taurine fed group. Therefore, in this study, we examined whether taurine also prevented hypercholesterolemia induced by ovarian hormone deficiency in ovariectomized rats when they were fed a calcium-deficient diet. These results indicate that taurine may have some beneficial effects on hypercholesterolemia and hypertriglyceridemia in OVX rats fed calcium-deficient diet.

Influences of Korean Haw (Crataegus pinnatifida BUNGE) on Lipid Concentration in Hypercholesterolemia (한국산 산사(Crataegus pinnatifida BUNGE)가 고콜레스테롤혈증 지질농도 변화에 미치는 영향)

  • This study was conducted to observe the influences of Crataegus pinnatifida BUNGE extract on lipid concentration in hypercholesterolemic rats (Sprague Dawley, male) fed the experimental diets for 5 weeks. Body weight gain was remarkably lower in the Crataegus pinnatifida BUNGE extract intake group than in the hypercholesterolemic rats. Total cholesterol concentration was remarkably lower in the extract of Crataegus pinnatifida BUNGE supplement in hypercholesterolemic rats than in the hypercholesterolemic model. Whereas, concentrations of HDL-cholesterol, ratio of HDL-cholesterol concentration to total cholesterol were remarkably higher in the extract of Crataegus pinnatifida BUNGE supplement in hypercholesterolemic rats than in the hypercholesterolemic model. Atherosclerotic index (AI), cardiac risk factor (CRF) were remarkably lower in the extract of Crataegus pinnatifida BUNGE supplement in hypercholesterolemic rats than in the hypercholesterolemic model. From the above research, Crataegus pinnatifida BUNGE extract was effective on the lipid concentrations in sera of dietary hypercholesterolemic rats.

Effects of Portulaca oleracea Powder on the Lipid Levels of Rats Fed a Hypercholesterolemia Inducing Diet

  • This study was initiated to investigate the effects of lyophilized purslane (Portulaca oleracea) powder (5% and 10%) on lipid levels in rats fed a hypercholesterolemia-inducing diet (1% cholesterol). During the four week study, there were no significant differences in either weight change or the food efficiency ratio between the group fed the hypercholesterol diet alone (HC) and the groups fed the purslane powder (HCPO-I and HCPO-II, 5% and 10% purslane, respectively). In serum, the levels of total lipid, total cholesterol and LDL-C decreased significantly for the group fed the 10% purslane powder (HCPO-II) in comparison with the group fed the 5% purslane powder (HCPO-I). The atherogenic index (AI) was reduced by about 51% for the group fed the 10% purslane powder (1.47) in comparison with the HC group (3.03). The activities of GOT, GPT, ALP and LDH decreased significantly for the groups fed the purslane powder in comparison with the HC group. Regarding liver tissue, the levels of total lipid, total cholesterol and triglyceride decreased significantly for the purslane powder-fed rats compared to the HC group. The fecal lipid profiles increased significantly as the amount of purslane powder was increased. Compared to the HC group, the fecal total cholesterol and triglyceride levels were higher in the group fed the 10% purslane powder by about 2.8 times and 2.3 times, respectively. For the serum and liver tissue, the content of lipid peroxide decreased significantly in the groups fed purslane powder compared to the HC group. The data from this experiment show an increase in the lipid levels discharged in feces, suggesting that the supplementation of purslane powder to a hypercholesterolemia-inducing diet reduces lipid levels.

Effects of Cinnamomi Cortex on Hypercholesterolemia Induced by Estradiol Valerate in Rats (육계(肉桂)가 Estradiol valerate에 의하여 유발된 고콜레스테롤 혈증에 미치는 영향)

  • This study was designed to investigate the effects of Cinnamomi Cortex extract (CC) on hypercholesterolemia induced by Estradiol valerate (EV) in female rats. We investigated the effects of CC on Changes in body weights for 5 weeks. In addition, we examined the effects on fasting blood glucose (FBS) and tested oral glucose tolerance test after oral administration of CC for 5 weeks. We also investigated the effects on levels of total cholesterol, HDL-cholesterol and LDL-cholesterol in serum. Treatment with CC restored weight gain rates suppressed by EV significantly. CC did not affect serum glucose level and glucose tolerance. Levels of total cholesterol and triglyceride were lowered by administration of CC compared to EV treated control respectively. These result suggest that CC can lower serum cholesterol level then prevent weight loss induced by EV. We also suggest the possibility as anti-hyperlipidemic agent of CC.
