Structure Formation in Multilayered Films Prepared by the Layer-by-Layer Deposition using PAA and HM-PEO

  • In present study, poly(acrylic acid) (PAA) and hydrophobically modified poly(ethylene oxide) (HM-PEO) multilayers based on the hydrogen bonding between the component polymer pair have been prepared by the LbL deposition method. Dip assembled HM-PEO/PAA multilayers yield unique film morphologies in comparison with PEO/PAA multilayers due to the micellar formation of HM-PEO owing to the hydrophobic attraction between alkyl chains end-capped with the PEO chains. Individual HM-PEO micelles were connected through the bridging PEO chains to form temporary networks on multilayer surface and induced peculiar surface morphology on HM-PEO/PAA multilayers above the critical number of bilayers.

Effect of Hydrophobic Excipients on the Properties of Fast Disintegrating Tablets

  • Highly hydrophobic lubricants including magnesium stearate may hinder water penetration into the tablet core resulting in delayed disintegration of fast disintegrating tablets. Alternative lubricants with equivalent lubricating properties may need to be incorporated into the tablet formulations. Sodium stearyl fumarate, glyceryl behenate and polyethylene glycol were evaluated regarding the tablet ejection energy, mechanical strength and disintegration time using Texture analyzer (TA). Resulting tablets were also compared with different particle sizes of granules and various compression forces. Among the tested lubricants, sodium stearyl fumarate was less sensitive to mixing time and also showed better or competitive tablet properties. During the experiments, TA was found to be very useful tool to investigate the tablet properties.

Mutation Effects on FAS1 Domain 4 Related to Protein Aggregation by Molecular Dynamics Simulations and Solvation Free Energy Analysis

  • Fasciclin 1 (FAS1) is an extracellular protein whose aggregation in cornea leads to visual impairment. While a number of FAS1 mutants have been studied that exhibit enhanced/decreased aggregation propensity, no structural information has been provided so far that is associated with distinct aggregation potential. In this study, we have investigated the structural and thermodynamic characteristics of the wild-type FAS1 and its two mutants, R555Q and R555W, by using molecular dynamics simulations and three-dimensional reference interaction site model (3D-RISM) theory. We find that the hydrophobic solvent accessible surface area increases due to hydrophobic core repacking in the C-terminus caused by the mutation. We also find that the solvation free energy of the mutants increases due to the enhanced non-native H-bonding. These structural and thermodynamic changes upon mutation contribute to understand the aggregation of these mutants.

Urokinase Inhibitor Design Based on Pharmacophore Model Derived from Diverse Classes of Inhibitors

  • A three-dimensional pharmacophore model was developed based on 24 currently available inhibitors, which were rationally selected from 472 compounds with diverse molecular structure and bioactivity, for generating pharmacophore of uPA (Urokinase Plasminogen Activator) inhibitors. The best hypothesis (Hypo1) comprised of five features, namely, one positive ionizable group, one hydrogen-bond acceptor group and three hydrophobic aromatic groups. The correlation coefficient, root mean square deviation and cost difference were 0.973, 0.695, and 94.291 respectively, suggesting that a highly predictive pharmacophore model was successfully obtained. The application of the model showed great success in predicting the activities of 251 known uPA inhibitors (test set) with a correlation coefficient of 0.837, and there was also none of the outcome hypotheses that had similar cost difference and RMS deviation (RMSD) with that of the initial hypothesis generated by Cat-Scramble validation test with 95% confidence level. Accordingly, our model should be reliable in identifying structurally diverse compounds with desired biological activity.

Clonazepam Release from Core-shell Type Nanoparticles In Vitro

  • AB-type amphiphilic copolymers (abbreviated as LE) composed of poly (L-leucine) (PLL) as the A component and poly (ethylene oxide) (PEO) as the B component were synthesized by the ring-opening polymerization of L-leucine N-carboxy-anhydride initiated by methoxy polyoxyethylene amine $(Me-PEO-NH_2)$ and characterized. Core-shell type nanoparticles were prepared by the diafiltration method. Particle size distribution obtained by dynamic light scattering was dependent on PLL composition and the size for LE-1, LE-2 and LE-3 was $369.6{\pm}267$, $523.4{\pm}410$ and $561.2{\pm}364 nm$, respectively. Shapes of the nanoparticies observed by transmission electron microscope (TEM) were almostly spherical. The critical micelle concentration (CMC) of the nanoparticles determined by a fluorescence probe technique was dependent on the composition of hydrophobic PLL, and the CMC for LE-1, LE-2 and LE-3 was $2.0{\times}10^{-6},1.7{\times}10^{-6}$ and $1.5{\times}10^{-6}(mol/l) $, respectively. Clonazepam release from core-shell type nanoparticles in vitro was dependent on PLL composition and drug loading content.

Adenosine Kinase Inhibitor Design Based on Pharmacophore Modeling

  • Adenosine kinase (AK) is a ubiquitous intracellular enzyme, which catalyzes the phosphorylation of adenosine (ADO) to adenosine monophosphate (AMP). AK inhibitors have therapeutic potential as analgesic and antiinflammatory agents. A chemical feature based pharmacophore model has been generated from known AK inhibitors (26 training set compounds) by HypoGen module implemented in CATALYST software. The top ranked hypothesis (Hypo1) contained four features of two hydrogen-bond acceptors (HBA) and two hydrophobic aromatics (Z). Hypo1 was validated by 124 test set molecules with a correlation coefficient of 0.905 between experimental and estimated activity. It was also validated by CatScramble method. Thus, the Hypo1 was exploited for searching new lead compounds over 238,819 chemical compounds in NCI database and then the selected compounds were screened based on restriction estimated activity and Lipinski's rules to evaluate their drug-like properties. Finally we could obtain 72 new lead candidates and the two best compound structures from them were posted.

  • Rod-coil amphiphilic block copolymers, PALG-PEOs, poly(alkyl $\alpha$, L-glutamate-co-ethylene oxide)s, were successfully synthesized in three steps: 1) esterification of L-glutamic acid, 2) synthesis of ${\gamma}$-alkyl L-gultamate N-carboxyanhydride, and 3) polymerization of NCA monomers. These molecules form polymeric micelles with the hydrophobic core and hydrophilic corona in aqueous solution, which were characterized by light scattering and static fluorescence measurement.

  • The metastable native state of proteins plays an important role in regulating biological functions. The native strain of serpins (serine protease inhibitors) are considered to be crucial for the inhibitory function. Several thermostable mutations of human $\alpha$$_1$-antitrypsin, a prototype inhibitory serpin, were identified in a systematic search targeted at the hydrophobic core of the molecule [Nature structural biology, vol. 3, no. 6, 497-500(1996)].
