EPD 방법을 이용한 알루미나-실리카 복합 코팅막의 제조와 전기절연 특성 (Preparation of Alumina-Silica Composite Coatings by Electrophoretic Deposition and their Electric Insulation Properties)

  • Alumina-silica composite coating layers were prepared by electrophoretic deposition (EPD) of plate-shaped alumina particles dispersed in a sol-gel binder, which was prepared by hydrolysis and the condensation reaction of methyltrimethoxysilane in the presence of colloidal silica. The microstructure and the electrical and thermal properties of the coatings were compared according to the EPD process parameter: voltage, time and the content of the plate-shaped alumina particles. The electrical insulation property of the coatings was measured by a voltage test. The coatings were prepared by EPD of the sol-gel binder with 5-30 wt% plate alumina particles on parallel electrodes at a distance of 2 cm for 1-10 min under an applied voltage of 10-30 V. The coatings experienced increased breakdown voltage with increasing thickness. However, the higher the thickness was, the smaller the breakdown voltage strength was. A breakdown voltage as high as 4.6 kV was observed with a $400{\mu}m$ thickness, and a breakdown voltage strength as high as 27 kV/mm was achieved for the sample under a $100{\mu}m$ thickness.

Glucose recovery from different corn stover fractions using dilute acid and alkaline pretreatment techniques

  • Background: Limited availability of corn stover due to the competing uses (organic manure, animal feed, bio-materials, and bioenergy) presents a major concern for its future in the bio-economy. Furthermore, biomass research has exhibited different results due to the differences in the supply of enzymes and dissimilar analytical methods. The effect of the two leading pretreatment techniques (dilute acid and alkaline) on glucose yield from three corn stover fractions (cob, stalk, and leaf) sourced from a single harvest in Uganda were studied at temperatures 100, 120, 140, and $160^{\circ}C$ over reaction times of 5, 10, 30, and 60 min. Results: From this study, the highest glucose concentrations obtained from the dilute acid (DA) pretreated cobs, stalks, and leaves were 18.4 g/L (66.8% glucose yield), 16.2 g/L (64.1% glucose yield), and 11.0 g/L (49.5% glucose yield), respectively. The optimal pretreatment settings needed to obtain these yields from the DA pretreated samples were at a temperature of $160^{\circ}C$ over an incubation time of 30 min. The highest glucose concentrations obtained from the alkaline (AL) pretreated cobs, stalks, and leaves were 24.7 g/L (81.73% glucose yield), 21.3 g/L (81.23% glucose yield), and 15.0 g/L (51.92% glucose yield), respectively. To be able to achieve these yields, the optimal pretreatment settings for the cobs and stalks were $140^{\circ}C$ and for a retention time of 30 min, while the leaves require optimal conditions of $140^{\circ}C$ and for a retention time of 60 min. Conclusions: The study recommends that the leaves could be left on the field during harvesting since the recovery of glucose from the pretreated cobs and stalks is higher.

  • This study investigated some enzymatic properties and bitterness improvement of an aminopeptidase retentate fraction (ARF) from common squid Todarodes pacificus hepatopancreas extract (HPE), obtained by ultrafiltration with a 10 kDa molecular weight cut off membrane. Endoprotease and aminopeptidase (AP) activity, and the purity of the ARF (>10 kDa) increased by 6.69-18.11 U/mg and 1.5-2.6 fold, respectively, compared to HPE (2.63-9.37 U/mg). The AP activity toward LeuPNA was stable at 20-55℃ and pH 5-9, but decreased slightly with increasing concentration of NaCl in the reaction mixture. The ARF was the most active MetPNA and preferentially hydrolyzed Glu, Leu and AlaPNA. The bitterness tryptic casein hydrolysates (BTCHs) were treated with ARF, and the bitterness of ARF-BTCHs significantly decreased with increasing amounts of released amino acids Ala, Val, Met, Ile and Leu, which show strong correlations with bitterness. Therefore, the ARF of T. pacificus HPE obtained by ultrafiltration may have a considerable potential for application in protein hydrolysis and appears to be ideally suited to the purpose of lowing bitterness in protein hydrolysates.

$\alpha$-Galactosidase를 생산하는 Bacillus lichennformis YB-42의 분리와 효소 특성 (Characterization of Extracellular $\alpha$-Galactosidase Produced by Bacillus licheniformis YB-42.)

  • 가정에서 제조된 된장으로부터 -galactosidase의 생산균으로 분리된 YB-42는 형태적 특성, 생화학적 성질 및 16S rRNA의 염기서열에 근거하여 Bacillus licheniformis로 동정되었다. B. lichentyormiE YB-42의 -galactosidase 활성은 균체내ㆍ외에서 모두 관찰되었다. 배양상등액으로부터 DEAE-Sepharose와 Q-Sepharose 컬럼 크로마토그래피를 통해 부분 정제한 -galactosidase을 사용하여 para-nitrophenyl--D-galactopyranoside(pNP-Gal)의 가수분해 반응특성을 조사한 결과 와 pH 6.5에 서 최대 활성을 보였다. Melibiose, raffinose와 stachyose는 부분 정제효소액에 의해 완전히 가수분해 되었으며, 분해산물로 galactose가 생성된 것으로 보아 -1,6 결합이 분해된다는 것이 확인되었다. 한편 pNP-Gal과 melibiose의 가수분해 활성은 galactose에 의해 가장 크게 저해되었으며, galactose보다는 낮지만 pNP-Gal의 가수분해 활성이 mannose와 glucose에 의해서도 저해되는 것으로 나타났다.

우레아 첨가량 변화에 따라 수열합성법으로 제조 된 3mol%Y2O3-ZrO2 분말의 합성 및 기계적 특성 평가 (Hydrothermal Synthesis and Mechanical Characterization of 3mol%Y2O3-ZrO2 by Urea Contents)

  • The industrial manufacturing of YSZ products can be summarized as a three step process: a) hydrolysis of zirconyl chloride and mixing of other solutions, b) precipitation, and c) calcination. The addition of ammonia or OH- is essential in the precipitation process. However, a strong agglomeration was observed in the results of an ammonia or OH- addition. Thus, it is necessary to disperse the powders smoothly in order to improve the mechanical strength of YSZ. In this study, YSZ was synthesized using the urea stabilizer and hydrothermal method. YSZ powders were synthesized using a hydrothermal method with Teflon Vessels at $180^{\circ}C$ for 24 h. The mole ratio of urea to Zr was 0, 0.5, 1, and 2. The crystal phase, particle size, and morphologies were analyzed. Rectangular specimens ($33\;mm{\times}8\;mm{\times}1{\pm}0.5\;mm$) for three-point bend tests were used in the mechanical properties evaluation. The crystalline of YSZ powders observed a tetragonal phase in the sample with a ratio of Zr:urea = 1:2 addition and a hydrothermal reaction time of 24 h. The average primary particle size of YSZ was measured to be 9 nm to 11 nm. The agglomerated particle size was measured from 15 nm to 30 nm. The three-point bending strength of the YSZ samples was 142.47 MPa, which is the highest value obtained for the Zr:urea = 1:2 ratio addition YSZ sample.

나노 ZnO:Ni를 이용한 후막 가스센서의 탄화수소계 가스에 대한 감응특성 (Response Characteristics of Thick Film Sensors Using Nano ZnO:Ni for Hydrocarbon Gas)

  • The effects of a Ni coating on the sensing properties of nano ZnO:Ni based gas sensors were studied for $CH_4$ and $CH_3CH_2CH_3$ gases. Nano ZnO sensing materials were prepared by the hydrothermal reaction method. The Ni coatings on the nano ZnO surface were deposited by the hydrolysis of zinc chloride with $NH_4OH$. The weight % of Ni coating on the ZnO surface ranged from 0 to 10 %. The nano ZnO:Ni gas sensors were fabricated by a screen printing method on alumina substrates. The structural and morphological properties of the nano ZnO : Ni sensing materials were investigated by XRD, EDS, and SEM. The XRD patterns showed that nano ZnO : Ni powders with a wurtzite structure were grown with (1 0 0), (0 0 2), and (1 0 1) dominant peaks. The particle size of nano ZnO powders was about 250 nm. The sensitivity of nano ZnO:Ni based sensors for 5 ppm $CH_4$ gas and $CH_3CH_2CH_3$ gas was measured at room temperature by comparing the resistance in air with that in target gases. The highest sensitivity of the ZnO:Ni sensor to $CH_4$ gas and $CH_3CH_2CH_3$ gas was observed at Ni 4 wt%. The response and recovery times of 4 wt% Ni coated ZnO:Ni gas sensors were 14 s and 15 s, respectively.

  • 항균제인 2-페닐페놀을 아크릴로일클로라이드와 반응시켜 2-비페닐릴아크릴레이트(BPA)를 합성하고 이를 메타크릴산(MA)과 라디칼 공중합시켜 2-페닐페놀 moiety를 가지는 친수성 고분자를 합성하였다 핵자기공명 스펙트라로부터 구한 이들 고분자중의 BPA와 MA 단량체의 조성을 사용하여 Kelen-Tudos 플로트로 결정한 단량체 반응성 비는 0.86과 1.21이였다. 이 결과는 제조된 poly(BPA-co-MA)가 이상적인 랜덤 공중합체라는 것을 뜻한다. 자외선 분광 광도계를 이용하여 이들 공중합체의 가수분해로 방출된 2-페닐페놀의 양을 측정하였다. 그 결과 pH가 증가함에 따라 그리고 공중합체중의 친수성의 MA의 양이 증가함에 따라 방출된 2-페닐페놀의 양이 증가하였다. 이들 결과는 제조한 고분자는 2-페닐페놀을 조절방출하는 고분자 항균제라는 것을 의미한다.

Improvement of ${\beta}-glucosidase$ Activity of Olea europaea Fruit Extracts Processed by Membrane Technology

  • The ${\beta}-glucosidase$ from olive fruit is of particular interest compared to the ones from other sources because it has shown to have high specifity to convert the oleuropein into dialdehydes, which have antibacterial activity and are of high interest for their application in the food and pharmaceutical fields. The enzyme is not yet commercially available and advanced clean and safe technologies for its purification able to maintain the functional stability are foreseen. The purification of this protein from fruit extracts has been already tempted by electrophoresis but either enzyme deactivation or high background with unclear profiles occurred. In this work, fruit extracts obtained from the ripening stage that showed the highest enzyme activity have been processed by diafiltration and ultrafiltration. Asymmetric membranes made of polyamide or polysulphone having 50 and 30 kDa molecular weight cut-off, respectively, were tested for the diafiltration process. Ultrafiltration membranes made of polyethersulfone with 4 kDa molecular weight cut-off were used to concentrate the dia-filtered permeate solutions. The efficiency of the separation processes was evaluated byenzyme activity tests using the hydrolysis of p-D-nitrophenyl-${\beta}$-D-glucopyranoside (pNPGlc) as reaction model. Qualitative and quantitative electrophoresis were applied to analyze the composition of protein solution before and after the membrane separation; in addition dot blot and western blot analyses were applied to verify the presence of ${\beta}-glucosidase$ in the processed fractions. The overall results showed that the ${\beta}-glucosidase$ functional stability was preserved during the membrane operations and the removal of 20 kDa proteins allowed to increase the specific activity of the enzyme of about 52% compared to the one present in the initial fruit extract.

재현성 있는 메조포러스 TiO2 박막의 제조에 대한 연구 (Reproducible Synthesis of Periodic Mesoporous TiO2 Thin Film)

  • 현재 넓은 표면적과 메조기공 뿐만 아니라 $TiO_2$의 넓은 band gap과 그 광학 활성 등으로 인하여 크게 각광받고 있는 메조포러스 $TiO_2$ 박막을 합성하기 위해 여러 가지 합성방법이 제시되고 있으나, 그 합성이 습도나 온도 등의 여러조건에 따라 크게 영향을 받아 재현성이 떨어진다는 치명적인 문제점이 제기되어 왔다. 이는 합성 용액 내에서 $TiO_2$ 전구체가 가수분해 및 축합반응을 하면서 구조유도체와의 자기조립에 의한 나노구조물 형성하는 과정에서 $TiO_2$의 전구체가 온도나 습도 등 주변의 영향을 많이 받기 때문이다. 본 연구에서는 2차원 구조의 메조기공을 가진 $TiO_2$ 박막을 재현성 있게 얻을 수 있는 실험 조건을 찾고자 하였다. 이를 위해 촉매인 HCl과 $TiO_2$ 전구체의 몰비 그리고 $TiO_2$ 전구체와 P-123의 몰비 등의 합성 조건뿐 아니라 코팅과정 도중이나 이후의 습도와 온도가 미치는 영향에 대한 실험을 수행하였고 그 특성을 XRD와 TEM 등으로 분석하였다.

Extracellular Secretion of a Maltogenic Amylase from Lactobacillus gasseri ATCC33323 in Lactococcus lactis MG1363 and its Application on the Production of Branched Maltooligosaccharides

  • A maltogenic amylase gene from Lactobacillus gasseri ATCC33323 (LGMA) was expressed in Lactococcus lactis MG1363 using the P170 expression system. The successful production of recombinant LGMA (rLGMA) was confirmed by the catalytic activity of the enzyme in liquid and solid media. The N-terminal amino acid sequencing analysis of the rLGMA showed that it was Met-Gln-Leu-Ala-Ala-Leu-, which was the same as that of genuine protein, meaning the signal peptide was efficiently cleaved during secretion to the extracellular milieu. The optimal reaction temperature and pH of rLGMA ($55^{\circ}C$ and pH 5, respectively) and enzymatic hydrolysis patterns on various substrates (${\beta}$-cyclodextrin, starch, and pullulan) supported that rLGMA was not only efficiently secreted from the Lactococcus lactis MG1363 but was also functionally active. Finally, the branched maltooligosaccharides were effectively produced from liquefied com starch, by using rLGMA secreted from Lactococcus lactis, with a yield of 53.1%.
