김(Pyropia yezoensis)에서 분리한 포피란 효소가수분해물의 화학적 및 유동 특성 (Chemical Composition and Rheological Properties of Enzymatic Hydrolysate of Porphyran Isolated from Pyropia yezoensis)

  • The chemical and rheological properties of natural and enzymatically hydrolyzed porphyran isolated from Pyropia yezoensis were investigated. The enzymatic hydrolysate was prepared by hydrolysis of porphyran using ${\beta}$-agarase followed by fractionation based on molecular weight (>300 kDa (Fr-1), 100-300 kDa (Fr-2), 10-100 kDa (Fr-3) and 1-10 kDa (Fr-4) using an ultrafiltration membrane. Each hydrolysate fraction consisted mainly of galactose (42.7-57.5%), 3,6-anhydro galactose (6.5-15.1%) and ester sulfate (8.6-14.1%). The sulfate content of the enzymatically hydrolyzed fractions decreased with an increase in molecular weight, whereas the 3,6-anhydro galactose content increased significantly. The rheological behavior of porphyran and enzymatically hydrolyzed porphyran solutions demonstrated a pseudoplastic behavior, which agrees with the Herschel-Bulkley model. The effect of temperature on the viscosity of the porphyrans and hydolysate fractions were measured and modeled using the Arrhenius equation. The activation energy of the porphyrans and enzymatically hydrolyzed porphyran (Fr-1) increased from 12.30 to 20.29 kJ/mol and 9.06 to 23.84 kJ/mol, respectively with increasing concentrations from 3% to 7%. These data indicate that the extent of the apparent viscosity of porphyran and enzymatically hydrolyzed porphyran are influenced by both temperature and concentration.

Development of a Practical and Cost-Effective Medium for Bioethanol Production from the Seaweed Hydrolysate in Surface-Aerated Fermentor by Repeated-Batch Operation

  • To develop a practical and cost-effective medium for bioethanol production from the hydrolysate of seaweed Sargassum sagamianum, we investigated the feasibility and performance of bioethanol production in CSL (corn-steep liquor)-containing medium, where yeast Pichia stipitis was used and the repeated batch was carried out in a surface-aerated fermentor. The optimal medium replacement time during the repeated operation was determined to be 36 h, and the surface aeration rates were 30 and 100 ml/min. Under these conditions, the repeated-batch operation was successfully carried out for 6 runs (216 h), in which the maximum bioethanol concentrations reached about 11-12 g/l at each batch operation. These results demonstrated that bioethanol production could be carried out repeatedly and steadily for 216 h. In these experiments, the total cumulative bioethanol production was 57.9 g and 58.0 g when the surface aeration rates were 30 ml/min and 100 ml/min, respectively. In addition, the bioethanol yields were 0.43 (about 84% of theoretical value) and 0.44 (about 86% of theoretical value) when the surface aeration rates were 30 ml/min and 100 ml/min, respectively. CSL was successfully used as a medium ingredient for the bioethanol production from the hydrolysate of seaweed Sargassum sagamianum, indicating that this medium may be practical and cost-effective for bioethanol production.

민태(Johnius belengeri) frame 단백질 가수분해물의 인산화에 의한 기능성 개선 (Improvement of the Functionalities by Phosphorylation of Hoki(Johnius belengeri) Frame Protein Hydrolysates)

  • 참치내장 중 유문수 조직에 내재되어 있는 단백질 분해효소를 부분정제하여 추출한 후 민태 frame 단백질의 효소적 가수분해에 이용하였으며, 제조된 가수분해물을 한외여과막을 이용하여 분자량 크기에 따라 분획한 후 기능성을 개선시킬 목적으로 cyclic sodium trimetaphosphate를 이용하여 인산화 민태 frame 단백질 가수분해 물을 제조하고, 용해도, 유화성 및 포말성 등과 같은 기능성을 검토하였다. 1 가수분해물의 기능성 개선을 목적으로 가수분해물에 STMP를 처리하여 인산화된 가수분해물을 제조하였으며, IR 스펙트럼으로 인산화를 확인한 결과, $1300\;cm^{-1}$에서 P=O의 흡수띠를, $1000{\sim}1100\;^{-1}$에서 P-O-C와 alkyl group에 결합된 P-O-C의 흡수띠를 확인하였다. 2. 민태 frame 단백질 가수분해물을 인산화시킴으로써 용해도는 물론 유화성 및 유화 안정성 그리고 포말 안정성 등 기능성의 개선에 크게 효과를 나타내었다. 특히 분자량이 가장 큰 획분인 P-30K 가수분해물은 모든 기능성 부분에서 좋은 결과를 나타내어 미이용자원으로부터 기능성이 부여된 새로운 가치의 물질로서 활용이 기대된다.

Enzyme hydrolysate of silk protein suppresses tert-butyl hydroperoxide-induced hepatotoxicity by enhancing antioxidant activity in rats

  • The purpose of current study is to investigate the beneficial effect of enzyme (Alcalase) hydrolysates of silk protein in rat. Alcalase-treated silk protein hydrolysate (ATSH) itself did not show any cytotoxicity on the hepatic tissues and blood biochemistry, similar to the normal condition. ATSH played a protective role in tert-butyl hydroperoxide (t-BHP)-induced hepatotoxicity and liver damage. The values of AST (aspartate aminotransferase) and ALT (alanine aminotransferase), which are the indicators of the liver function, were effectively alleviated with the ATSH treatment in a dose dependent manner. The level of Lactate dehydrogenase (LDH) and Malondialdehyde (MDA), which were increased with t-BHP treatment, were significantly reduced by ATSH. High dose of ATSH (2 g/kg) reduced the t-BHP-induced LDH release by 48%. Antioxidant and antioxidant enzymes in liver cells were significantly increased by ATSH treatment in their level and activities. ATSH (2 g/kg) increased glutathione (GSH), an intracelluar antioxidant, by 2.5-fold compared with the t-BHP treated group. The activities of glutathione-s-transferase (GST), superoxide dismutase (SOD), and catalase were also elevated by 38%, 60%, and 45%, respectively, with ATSH (2 g/kg) treatment. The antioxidative effect of ATSH was recapitulated to the protection from t-BHP induced liver damages in hematoxylin and eosin (H&E) staining. Thus, ATSH might be used as a hepatoprotective agent.

새조개 처리동결 가공부산물을 이용한 페이스트 젓갈 제조 (Processing of Squeezed-Type Cockle Shell By-Product Paste)

  • 새조개 여체부의 긴발을 제외한 나머지인 가공 부산물을 식량자원으로서 효율적 이용을 위한 한 방안으로 효소분해 후 가수분해물의 물성을 개선하여 다소의 조직감을 갖는 반고형 상태의 페이스트 젓갈의 제조와 품질개선에 관하여 검토하였다. 새조개 가공부산물에 절반량의 물과 4% Protease N. P.를 첨가하여 가수분해 후 여과하여 그 여액에 4% glucose를 첨가하여 10$0^{\circ}C$에서1시간 열처리하여 풍미개선, 효소의 불활성화 및 멸균을 시켰다. 물성개량제 즉 0.5% alginic acid, 1% pectin, 0.2% agar를 병용 첨가한 제품이 대조구에 비하여 조직감 및 물성개선의 효과를 나타내었고, 10,000 rpm에서 60분간 원심분리 후에도 조직감이 파괴되지 않아 그 때의 혼합안정성은 98.1%였다. 페이스트 젓갈의 성분은 수분 함량 57.7%, 총당 함량 20.6%, 총질소함향 1,458% 및 아미노질소량 1,187mg%이었으며, 총질소중 아미노질소가 차지하는 비율은 81.4%였다.

Antioxidant Effect and Functional Properties of Hydrolysates Derived from Egg-White Protein

  • This study utilized commercially available proteolytic enzymes to prepare egg-white protein hydrolysates (EPHs) with different degrees of hydrolysis. The antioxidant effect and functionalities of the resultant products were then investigated. Treatment with Neutrase yielded the most ${\alpha}$-amino groups (6.52 mg/mL). Alcalase, Flavourzyme, Protamex, and Ficin showed similar degrees of ${\alpha}$-amino group liberation (3.19-3.62 mg/mL). Neutrase treatment also resulted in the highest degree of hydrolysis (23.4%). Alcalase and Ficin treatment resulted in similar degrees of hydrolysis. All hydrolysates, except for the Flavourzyme hydrolysate, had greater radical scavenging activity than the control. The Neutrase hydrolysate showed the highest 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity ($IC_{50}=3.6mg/mL$). Therefore, Neutrase was identified as the optimal enzyme for hydrolyzing egg-white protein to yield antioxidant peptides. During Neutrase hydrolysis, the reaction rate was rapid over the first 4 h, and then subsequently declined. The $IC_{50}$ value was lowest after the first hour (2.99 mg/mL). The emulsifying activity index (EAI) of EPH treated with Neutrase decreased, as the pH decreased. The EPH foaming capacity was maximal at pH 3.6, and decreased at an alkaline pH. Digestion resulted in significantly higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ABTS radical scavenging activity. The active peptides released from egg-white protein showed antioxidative activities on ABTS and DHHP radical. Thus, this approach may be useful for the preparation of potent antioxidant products.

Enzyme Hydrolysates of Ginseng Marc Polysaccharides Promote the Phagocytic Activity of Macrophages Via Activation of TLR2 and Mer Tyrosine Kinase

  • Although ginseng marc is a by-product obtained during manufacturing of various commercial ginseng products and has been routinely discarded as a waste, it still contains considerable amounts of potential bioactive compounds, including saponins and polysaccharides. Previously, we reported that ginseng oligosaccharides derived from ginseng marc polysaccharides by enzymatic hydrolysis exert immunostimulatory activities in macrophages and these activated macrophages are in turn able to inhibit the growth of skin melanoma cells by inducing apoptosis. In the present study, a more detailed investigation of the immunostimulatory activity and underlying action mechanisms of an enzymatic hydrolysate (GEH) containing these oligosaccharides derived from ginseng marc polysaccharides was performed. The levels of proinflammatory cytokines and anti-inflammatory cytokines were measured in GEH-stimulated RAW264.7 macrophages using RT-PCR analysis and ELISA. The expression levels of Toll-like receptor 2 (TLR2) and TLR4, Dectin-1, and MerTK were measured by RT-PCR analysis or western blot analysis, and the phagocytic activities of GEH-challenged bone marrow-derived macrophages toward apoptotic Jurkat cells were assayed using fluorescence microscopy. GEH induced the production of both proinflammatory cytokines $TNF-{\alpha}$ and IL-6, and anti-inflammatory cytokine IL-10 in RAW 264.7 cells. The expression of the TLR2 and MerTK mRNAs was increased upon GEH treatment. Phagocytosis of apoptotic Jurkat cells was enhanced in GEH-treated macrophages. Based on the results, this enzymatic hydrolysate (GEH) containing oligosaccharides exerts immunostimulatory effects by maintaining the balance between M1 and M2 cytokines, facilitating macrophage activation and contributing to the efficient phagocytosis of apoptotic cells. Therefore, the GEH could be developed as value-added, health-beneficial food materials with immunostimulatory effects.

Isolation and Charaterization of Bioactive Peptides from Hwangtae (yellowish dried Alaska pollack) Protein Hydrolysate

  • Hwangtae, dried Alaska pollack, is a major storage product in the fish processing industry. Hwangtae is prepared by removing the internal organs and drying outdoors during the cold witner months by allowing it to thaw during the daytime and re-freeze at night under sub-zero ($-10^{\circ}C$) conditions and gradually dry from December until the next April for around 5 months from Myungtae. In this study, ground Hwangtae was hydrolyzed using two proteolytic enzymes (pepsin and alcalase) which produced five soluble active peptides from Hwangtae (yellowish dried Pollack, Theragra chalcogramma) protein. Two different peptides with strong antioxidative activity were isolated from the hydrolysate using consecutive chromatographic methods of Sephadex G-25 gel, ion-exchange chromatography on a Sepharose-Sephadex C-25 gel, and high-performance liquid chromatography. The isolated peptides, APO1 and APO2, were composed of 16 and 13 amino acid residues, respectively. Both peptides contained a Gly residue at the C-terminus and the repeating motif Gly-Pro-Hyp. The peptide with a molecular weight less than 1,000 Daltons (APACE) obtained from enzymatic hydrolysates of Hwangtae exhibited the highest ACE inhibitory activity. The APACE peptides was composed of 4 amino acid residues (Gly-Leu-Leu-Pro). These results suggest that Hwangtae hydrolysates could be a good source of peptides with ACE inhibitory activity. Biochemical analysis indicated that two 70 kDa peptides (APG1 and APG2) isolated from the hydrolysate had gelatinoytic activity, which was shown to be a calcium dependent protease type as showed by gelatin SDS PAGE.

미역의 아미노酸 및 비타민에 對한 營養學的 硏究 (Studies on the Nutrition of Amino Acids and Vitamins in Undaria pinnatifida (Harvey) Suringar)

  • Leuconostoc mesenteroides P-60, Lactobacillus arabinosus 17-5, Streptococcus faecalis R have been successfully used for the quantitative determination of sixteen amino acids in Undaria pinnatifida (Harvey) Suringar hydrolysate by alkaline and hydrolysis for succesive two hours from two to twelve hours, by means of microbiological assay. And thiamine and riboflavin were fluorometrically determinated by thiochrome and lumiflavin in powder (80mesh) of Undaria pinnatifida (Harvey) Suringar. The results were as follows: 1) Arginine contents was the highest in hydrolysate for two hours, but longer the hydrolysis, the more content Undaria pinnatifida was decreased. 2) The adequate contents of other amino acids were obtained by hydrolysis for six hours. 3) Growth check and improve of Lactobacillus were not identified in determination by microbiological assay for Undaria pinnatifida. 4) The following values were obtained in Undaria pinnatifida hydrolysate six hours: asparatic acid 466, arginine 230, lysine 317, histidine 74, isoleucine 242, methionine 202, phenylalanine 256, proline 231, threonine 231, tyrosine 161, valine 415, glycine 302, leucine 414, glutamic acid 625, cystine (5 hrs.) 53 and tryptophan (8 hrs.) 90mg per nitrogen one gram. 5) Protein score was 81 (limiting factor was isoleucine) and essential amino acids pattern was of satisfactory results. And methionine contained was higher than FAO value or milk value. 6) Sulphur contained amino acids (methionine plus cystine) contained in Undaria pinnatifida were 225mg/N-g. That was satisfactory results. 7) Absorption spectrum of wave length were not different 1% HAc from buffer-sol. (pH 6.8) in dilution for determination of riboflavin. Both methods might be suitable. 8) Thiamine and riboflavin contained in Undaria pinnatifida were ($B_1,\;82.51{\pm}1.1){\gamma}/N-g\;and\;(B_2,\;115.29{\pm}1.5){\gamma}/N-g.$.

유기인산계 추출제를 이용한 에멀젼형 액막법에 의해 푸란유도체를 함유하는 모사 바이오매스 가수분해액으로부터 초산의 분리 (Separation of Acetic Acid from Simulated Biomass Hydrolysates Containing Furans by Emulsion Liquid Membranes with an Organophosphorous Extractant)

  • 바이오매스 가수분해액으로부터 당을 정제하는 동안 발효 저해물질들의 선택적인 제거 및 회수는 목질계 바이오매스로부터 바이오 알코올을 생성하는 전체 공정의 경제적 효율성을 높일 수 있다. 이 연구에서는 에멀젼형 액막계에서 페놀화합물이 없는 모사 바이오매스 가수분해액에 있는 푸란유도체가 초산 추출에 미치는 영향이 조사되었다. 특별한 조작 조건하에서 5분내에 99%이상의 초산을 추출할 수 있었으며, 이 때 푸르푸랄과 5-hydroxymethylfurfural의 추출율은 각각 10%와 4% 내외의 값을 가졌다. 또한, 원료상에 있는 푸란유도체들의 초기 농도가 높을수록 초산의 추출속도는 낮았는데, 푸르푸랄이 5-hydroxymethylfurfural 보다 그 영향이 더 컸다. 따라서, 초산 추출 전에 가수분해액으로부터 푸르푸랄을 우선적으로 제거한다면 에멀젼형 액막법이 보다 경제성이 있는 초산 제거법이 될 것으로 보인다.
