Plant Regeneration from Embryogenic Suspension Culture of Orchardgrass (Dactylis glomerata L.) (오차드그래스의 현탁배양으로부터 부정배 형성과 식물체 재분화)

  • This study was carried out to improve the ability of embryo formation and the efficiency of plant regeneration from suspension cultured cells of seed derived calli of orchardgrass (Dactylis glomerata L.). The frequency of formation of round cell and cell colonies was highest at 50 days after suspension culture in $N_6$ medium supplemented with $4\;g/{\ell}$ casein hydrolysate (CH), $20\;g/{\ell}$ sucrose and $30\;g/{\ell}$ sorbitol. The highest frequency of plant regeneration and somatic embryo formation was obtained from suspension cultured cells of 60 days. Addition of CH ($4\;g/{\ell}$) in suspension culture medium gave the highest frequency of embryo formation (39.6%) and plant regeneration (73.0%).

Plastein formation from sunflower seed protein (해바라기씨 단백질에서 plastein의 합성)

  • Optimum conditions for hydrolysis of sunflower seed by pepsin and for plastein formation by pepsin were determined. The optimum conditions for hydrolysis of sunflower seed were pH 1.5, $45^{\circ}C$, enzyme concentration 2%, substrate concentration 2%, and hydrolysis time 24hr. The optimum conditions for sunflower seed-plastein formation were 50% substrate, pH 4.5, $50^{\circ}C$, 0.25% pepsin and 18hrs reaction time. To verify plastein fromation from concentrated prptic hydrolysate of sunflower seed, thin layer chromatography was performed. The TLC pattern of concentrated peptic hydrolysate of sunflower seed was different from that of its plastein. The TLC pattern of concentrated peptic bydrolysate of sunflower seed and at of its plastein indicated that plastein was different material from the hydrolysate.

Enzymatic Hydrolysis of Egg White Protein Exerts a Hypotensive Effect in Spontaneously Hypertensive Rats

  • This study was conducted to investigate the hypotensive effect of egg white protein (EWP) hydrolysate (EWH) in spontaneously hypertensive rats (SHRs). The hydrolysis of EWP was effectively performed with a combination of 0.5% bromelain and 1% papain at 50℃ for 60 min. The resulting hydrolysate did not elicit an allergic reaction as confirmed by human mast cell activation test. The systolic and diastolic blood pressures of the SHRs fed the EWH diet were observed to be significantly or numerically lower than those of the other groups during the experimental period of 28 d. EWH treatment significantly (p<0.05) upregulated the nitric oxide levels in hCMEC/D3 cells and the plasma of the SHRs compared to those in the control. Moreover, EWH ingestion significantly (p<0.01) reduced the plasma angiotensin II level of the SHRs compared with that in the control. In conclusion, beyond its basic nutritional value, EWH prevents and manages hypertension, and thus can be an invaluable resource for functional food development.

Structural Features of Enzymatic Hydrolysate of Porphyran Isolated from Porphyra yezoensis (방사무늬김(Porphyra yezoensis)에서 추출한 Porphyran 효소 분해물의 화학적 결합 특성)

  • Enzymatic hydrolysate of porphyran from Porphyra yezoensis was prepared by treatment with ${\beta}$-agarase. The hydrolysate was fractioned into molecular sizes of <3, 3-30, and 30-300 kDa using an ultrafiltration membrane. The membrane fractions were further separated into neutral and anionic fractions using Dowex $1{\times}8$ ion exchange chromatography. After hydrolysis of porphyran with ${\beta}$-agarase, 23.2% of the starting porphyran was recovered as a neutral fraction of low-molecular weight (<3 kDa), and 28.9% remained as an enzyme-resistant anionic fraction of high molecular weight (>300 kDa). Desulfation of porphyran and $^{13}C$-NMR analysis of the anionic fraction of low molecular weight (<3 kDa) showed that the anionic fraction has a backbone consisting of 3-linked ${\beta}$-D-galactose units alternating with either 4-linked a-L-galactose 6-sulfate or 3, 6-anhydro-a-L-galactose units. These results indicate that porphryan is a copolymer of two moieties, about 25% of which are composed of neoagarose moieties and 75% as anionic moieties.

  • An enzymatic process was developed to produce protein hydrolysater form defatted soya protein. Various unit operations were tried, and the effects of pre- and post-treatments on the product characteristics such as degree of hydroylsis (DH), free amino acid content (%FAA) and average molecular weight (MW) were investigated. The use of acid washes showed no difference in %DH. Increasing pH during pre-cooking gave lower %DH. Alkaline cooking made too much insoluble protein, thus the protein yield was too small. A better hydrolysis with more acceptable taste was obtained when the combination of Neutrase/Alcalase/Flavourzyme was used in place of Alcalase/Flavourzyme combination; Untoasted defatted soya was more effective on the proteolysis than toasted one. The MW of the evaporated and spray dried product was higher than that of undried product, due to precipitation of low-solubility components. When ultrafiltration and the product concentration carried out the product separation by reverse osmosis, the solubility and the taste of the product were improved. The difference between enzyme hydrolysate and acid hydrolysate was significant in free amino acid composition, especially in tyrosine, phenylalanine, glutamine and asparagine.

Effects of Hydro-thermal Reaction Temperature on Anaerobic Biodegradability of Piggery Manure Hydrolysate

  • In order to enhance a biogas production by the hydro-thermal pre-treatment of piggery manure, the effects of hydro-thermal reaction temperature at thermal hydrolysis of piggery manure on the methane potential and anaerobic biodegradability of thermal hydrolysate were analyzed. The increase of hydro-thermal reaction temperature from $170^{\circ}C$ to $220^{\circ}C$ caused the enhancement of hydrolysis efficiency, and most of organic matters were present in soluble forms. However, the methane potentials ($B_u-TCOD$) of hydrolysate were decreased from 0.239 to $0.188Nm^3kg^{-1}-TCOD_{added}$ by increasing hydro-thermal reaction temperature from $170^{\circ}C$ to $220^{\circ}C$, and also the anaerobic biodegradability (DTCOD) decreased from 74.6% to 58.6% with increase of hydro-thermal reaction temperature. The increase of hydro-thermal reaction temperature from $170^{\circ}C$ to $220^{\circ}C$ resulted in the decrease of easily biodegradable organic matter content, while persistent organic matter contents increased.

Antioxidant and Antihypertension Effects of Enzyme Hydrolysate from Hippocampus abdominalis (식용 빅벨리 해마(Hippocampus abdominalis) 유래 단백질 가수분해물의 항산화와 항고혈압 효능)

  • Seahorses have long been used as ornamental and medicinal products. The sea horse Hippocampus abdominalis has a beautiful color and unique shape and is also used for ornamental purposes and as a traditional medicine in China. This study examined the value of H. abdominalis as a health functional food or food additive. H. abdominalis was hydrolyzed using seven proteases: flavourzyme, neutrase, alcalase, trypsin, kojizyme, pepsin and protamex. The yields of all of the enzyme hydrolysates were higher than that of the aqueous extract. Of the enzymatic hydrolysates, seahorse Protamex hydrolysate (SHP) gave the highest yield and had excellent antioxidant and angiotensin-I converting enzyme inhibitory activities. It protected Vero cells against oxidative by 2,2-azobis-(2-amidinopropane) dihydrochloride (AAPH) and antihypertension in Spontaneously Hypertensive Rats. This study attempted to demonstrate H. abdominalis as a health functional food or food additive in the future.

Effect of supplementation with brewer's yeast hydrolysate on growth performance, nutrients digestibility, blood profiles and meat quality in growing to finishing pigs

  • Objective: This study was aimed to investigate the effects of brewer's yeast hydrolysate (YH) on growth performance, nutrients digestibility, blood profiles and meat quality of growing pigs. Methods: A total of 200 growing pigs ($[Landrace{\times}Yorkshire]{\times}Duroc$) (initial body weight, $25.31{\pm}1.29kg$) were allotted to 5 treatments as follow: CON, basic diet; and YH treatment, CON+0.05%, 0.1%, 0.5%, and 1.0% of YH, respectively. Results: On wk 11, 16 and overall phase, pigs fed YH diet showed a linear improvement in average daily body gain and gain/feed (p<0.05). The pigs that received YH linearly increased the digestibility of dry matter, nitrogen, and energy on wk 11 and 16. The concentration of serum urea nitrogen was linearly increased in YH treatments on wk 16. However, the carcass weight, back fat and lean muscle percentage of pigs receiving YH had no significant change. Besides, no difference was observed in creatinine and total protein in the blood among treatments. Conclusion: The pigs fed a graded YH diet had improved growth performance and nutrient digestibility, meanwhile, the YH increased the serum urea nitrogen in the growing pigs.

Quantitative Analysis of Chemical Components of Hydrolysate from Silkworm Fed with Cudrania tricuspidata Leaves

  • The purpose of this study was to develop hydrolysate from silkworm (Cudrania Silkworm Fibroin Peptide; CSFP), a food containing components for improving blood vessel health. In general, Cudrania tricuspidata leaves contain about 5 times more rutin and 25 times more dihydroquercetin than mulberry leaves. 1-Deoxynojirimycine (1-DNJ), dihydroquercetin and rutin inhibit the activity of carbohydrate-digesting enzymes, inhibit blood lipid peroxidation, and regulate insulin secretion, which helps blood vessels to be healthy. When the diet-controlled silkworm was enzymatically hydrolyzed, it was confirmed that rutin content was about 8 times higher than that of the in general silkworm as a control. In the silkworm hydrolysate, CSFP, developed as a final food material, the active ingredients were 65 mg/kg for rutin, 3,328 mg/kg for DNJ, 0.43 mg/kg for dihydroquercetin, and 82,624 mg/kg for total polyphenol, which was confirmed through LC-MS/MS analysis. In conclusion, it was found that silkworms fed with C. tricuspidata leaves as a diet had more active components that can help control blood sugar and improve blood vessel health than silkworms fed with mulberry leaves.

Effects of hydrothermal pretreatment on methane potential of anaerobic digestion sludge cake of cattle manure containing sawdust as bedding materials

  • Objective: The purpose of this study was to analyze the effect of the hydrothermal pretreatment of anaerobic digestion sludge cake (ADSC) of cattle manure on the solubilization of organic matter and the methane yield to improve the anaerobic digestion efficiency of cattle manure collected from the sawdust pens of cattle. Methods: Anaerobic digestion sludge cake of cattle manure was thermally pretreated at 160℃, 180℃, 200℃, and 220℃ by a hydrothermal pressure reactor, and the biochemical methane potential of ADSC hydrolysate was analyzed. Methane yield recovered by the hydrothermal pretreatment of ADCS was estimated based on mass balance. Results: The chemical oxygen demand solubilization degree (CODs) of the hydrothermal hydrolysate increased to 63.56%, 67.13%, 70.07%, and 66.14% at the hydrothermal reaction temperatures of 160℃, 180℃, 200℃, and 220℃, respectively. Considering the volatile solids content obtained after the hydrothermal pretreatment, the methane of 10.2 Nm3/ton-ADSC was recovered from ADSC of 1.0 ton, and methane yields of ADSC hydrolysate increased to 15.6, 18.0, 17.4, and 17.2 Nm3/ton-ADSC. Conclusion: Therefore, the optimal hydrothermal reaction temperature that yielded the maximum methane yield was 180℃ based on mass balance, and the methane yield from cattle manure containing sawdust was improved by the hydrothermal pretreatment of ADSC.
