Inhibition of Pacemaker Activity of Interstitial Cells of Cajal by Hydrogen Peroxide via Activating ATP-sensitive $K^+$ Channels

  • To investigate whether hydrogen peroxide($H_2O_2$) affects intestinal motility, pacemaker currents and membrane potential were recorded in cultured interstitial cells of Cajal(ICC) from murine small intestine by using a whole-cell patch clamp. In whole cell patch technique at $30^{\circ}C$, ICC generated spontaneous pacemaker potential under current clamp mode(I=0) and inward currents(pacemaker currents) under voltage clamp mode at a holding potential of -70 mV. When ICC were treated with $H_2O_2$ in ICC, $H_2O_2$ hyperpolarized the membrane potential under currents clamp mode and decreased both the frequency and amplitude of pacemaker currents and increased the resting currents in outward direction under voltage clamp mode. Also, $H_2O_2$ inhibited the pacemaker currents in a dose-dependent manner. Because the properties of $H_2O_2$ action on pacemaker currents were same as the effects of pinacidil(ATP-sensitive $K^+$ channels opener), we tested the effects of glibenclamide(ATP-sensitive $K^+$ channels blocker) on $H_2O_2$ action in ICC, and found that the effects of $H_2O_2$ on pacemaker currents were blocked by co- or pre- treatment of glibenclamide. These results suggest that $H_2O_2$ inhibits pacemaker currents of ICC by activating ATP-sensitive $K^+$ channels.

  • Hydrogen peroxide ($H_2O_2$) biosensor is one of the most developing sensors because this kind of sensors is highly selective and responds quickly to the specific substrate. Hemoglobin (Hb) has been used as ideal biomolecules to construct hydrogen peroxide biosensors because of their high selectivity to $H_2O_2$. The direct electron transfer of Hb has widely investigated for application in the determination of $H_2O_2$ because of its simplicity, high selectivity and intrinsic sensitivity. An electrochemical detection for hydrogen peroxide was investigated based on immobilization of hemoglobin on DNA/Fe(pyterpy)$^{2+}$ modified gold electrode. The pyterpy monolayers were firstly an electron deposition onto the gold electrode surface of the quartz crystal microbalance (QCM). It is offered a template to attach negatively charged DNA. The fabrication process of the electrode was verified by quartz crystal analyzer (QCA). The experimental parameters such as pH, applied potential and amperometric response were evaluated and optimized. Under the optimized conditions, this sensor shows the linear response within the range between $3.0{\times}10^{-6}$ to $9.0{|times}10^{-4}$ M concentrations of $H_2O_2$. The detection limit was determined to be $9{\times}10^{-7}$ M (based on the S/N=3).

  • The purpose of this study was to investigate the characteristics of slurry including phosphoric acid for chemical-mechanical planarization of copper and tantalum nitride. In general, the slurry for copper CMP consists of alumina or colloidal silica as an abrasive, organic acid as a complexing agent, an oxidizing agent, a film forming agent, a pH control agent and additives. Hydrogen peroxide (H$_2$O$_2$) is the material that is used as an oxidizing agent in copper CMP. But, the hydrogen peroxide needs some stabilizers to prevent decomposition. We evaluated phosphoric acid (H$_3$PO$_4$) as a stabilizer of the hydrogen peroxide as well as an accelerator of the tantalum nitride CMP process. We also estimated dispersion stability and zeta potential of the abrasive with the contents of phosphoric acid. An acceleration of the tantalum nitride CMP was verified through the electrochemical test. This approach may be useful for the development of the 2$\^$nd/ step copper CMP slurry and hydrogen peroxide stability.

Electrocatalytic Reduction of Hydrogen Peroxide at Nanoporous Gold Surfaces

  • We report on the electrocatalytic reduction of hydrogen peroxide at nanoporous gold (NPG) surfaces. Various NPG surfaces with different surface structure were prepared by changing the conditions of electrodeposition for Ag-Au layers such as the concentration ratios of $KAu(CN)_2$ over $KAg(CN)_2$ and deposition charges. The effects of different electrochemical conditions on the electrocatalysis of $H_2O_2$ reduction were investigated. The NPG surfaces exhibited sensitive amperometric responses for $H_2O_2$ reduction, from which calibration plots with higher sensitivity than a bare Au surface were obtained.

Effects of cell viability and antioxidant enzyme activity of Phellinus linteus extract on Mouse melanoma cells(B16F10)

  • The effects of oxidative stress on the alterations of different antioxidant enzyme activity on mouse melanoma cells(B16F10) was investigated. Oxidative stress was induced by the exposeure to hydrogen peroxide(H2O2). B 16F 10 cells were exposed Phellinus linteus Ex. in combination with H2O2 and measured the time course of changes in cell viability and antioxidant enzyme activity. CAT activity peaked at 12 hr. On the contrary, SOD and GPX activity was maximum at 6 hr. The cell viability of Pheltinus linteus extracts in combination with hydrogen peroxide was higher than hydrogen peroxide alone.

A Comparative Study of the Degradation of the Erionyl Navy R by Different Oxidation Processes: Chemical, Fenton and Fenton-like

  • The oxidative degradation performance of the Erionyl Navy R dye was studied in this article. The investigation mainly focused on a comparative study between chemical oxidations by sodium hypochlorite (NaClO) and hydrogen peroxide (H2O2), and catalytic oxidations including the Fenton (Fe2+-H2O2) and Fenton-Like (Fe2+/ Fe3+/Co2+/ Mn2+-H2O2) or modified Fenton-like (Fe2+/ Fe3+ -NaClO) reactions. A discoloration and degradation of the Erionyl Navy R occurred after 30 minutes, which varies according to the oxidation system involved; 31%, 54%, <20%, 95%, and >96% losses were observed for Co2+-H2O2, Mn2+-H2O2, Fe2+-NaClO, Fe3+-NaClO), and Fe2+-H2O2 and Fe3+-H2O2, respectively.

Effect of Herba Epimedii on hydrogen peroxide induced neurotoxicity in cultured rat dorsal root ganglion neurons

  • Effects of hydrogen peroxide $(H_2O_2)-induced$ neurotoxicity were investigated in cultured newborn rat spinal dorsal root ganglion (DRG) neurons after DRG neurons were treated in the media containning various concentrations of $H_2O_2$. In addition, the protective effect of Herba Epimedii (HE) extract against $H_2O_2-induced$ neurotoxicity was examined. Cytotoxic values were determined by the cell viability of living cells using 3-(4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide (MTT) assay. In the present study, exposure of neurons to $H_2O_2$ resulted in a significant cell death in a dose- and time-dependent manners in cultured DRG neurons. The decrement of cell viability by $H_2O_2$ was blocked by HE. These results suggest that the neuroprotective effect of HE against $H_2O_2-induced$ cytotoxicity may result from the prevention of injury induced by $H_2O_2$.

  • The aim of this study was to evaluate the chemical quenching system for residual ozone and to determine the operating condition for the quenching system. Hydrogen peroxide ($H_2O_2$) and sodium thiosulfate ($Na_2S_2O_3$) were investigated as quenching reagents for ozone removal, and the tendency of each chemical was notably different. In the case of $H_2O_2$, the degradation rate of ozone was increased as the concentration of $H_2O_2$ increase, and temperature and pH value have a significant effect on the degradation rate of ozone. On the other hand, the degradation rate of ozone was not affected by the concentration of $Na_2S_2O_3$, temperature and pH value, due to the high reactivity between the ${S_2O_3}^{2-}$ and ozone. This study evaluates the decomposition mechanism of ozone by $H_2O_2$ and $Na_2S_2O_3$ with consideration for the water quality and reaction time. Furthermore, the removal test for the quenching reagents, which can be remained after reaction with ozone, was conducted by GAC process.

Hydrogen Peroxide Prompted Lignification Affects Pathogenicity of Hemi-biotrophic Pathogen Bipolaris sorokiniana to Wheat

  • Spot blotch caused by Bipolaris sorokiniana has spread to more than 9 million ha of wheat in the warm, humid areas of the Eastern Gangetic Plains (EGP) of South Asia and is a disease of major concern in other similar wheat growing regions worldwide. Differential lignin content in resistant and susceptible genotypes and its association with free radicals such as hydrogen peroxide ($H_2O_2$), superoxide ($O_2{^-}$) and hydroxyl radical ($OH^-$) were studied after inoculation under field conditions for two consecutive years. $H_2O_2$ significantly influenced lignin content in flag leaves, whereas there was a negative correlation among lignin and $H_2O_2$ to the Area Under Disease Progress Curve (AUDPC). The production of $H_2O_2$ was higher in the resistant genotypes than susceptible ones. The $O_2{^-}$ and $OH^-$ positively correlated with AUDPC but negatively with lignin content. This study illustrates that $H_2O_2$ has a vital role in prompting lignification and thereby resistance to spot blotch in wheat. We used cluster analysis to separate the resistant and susceptible genotypes by phenotypic and biochemical traits. $H_2O_2$ associated lignin production significantly reduced the number of appressoria and penetration pegs. We visualized the effect of lignin in disease resistance using differential histochemical staining of tissue from resistant and susceptible genotypes, which shows the variable accumulation of hydrogen peroxide and lignin around penetration sites.
