Characteristic Variations of H2O2 Concentrations Observed in Seoul (서울시 대기 중 과산화수소 농도 변화 특성)

  • During January $2002{\sim}April\;2004$, hydrogen peroxide ($H_{2}O_{2}$) measurements were performed at the campus of Korea University, which is located in the northeastern part of Seoul. Gas phase hydroperoxide was collected in aqueous solution and separated by HPLC. Concentrations were determined by fluorescence using postcolumn enzyme derivatization. This measurement system was improved to be run automatically from sample collection at every 10 minutes through chemical analysis for data collection. Detection limits of $H_{2}O_{2}$ is $10{\sim}17\;pptv$, and the overall uncertainty of the measurements is better than 8%. Two-year measurements of $H_{2}O_{2}$ show typical seasonal variations. Concentrations of $H_{2}O_{2}$ were higher during $June{\sim}October$ and lower during $January{\sim}February$. Maximum concentration of 1-hour averaged $H_{2}O_{2}$ was 6.5 ppbv, which was observed in August and September. In general $H_{2}O_{2}$ concentrations were well correlated with $O_{3}$ concentrations and largely affected by meteorological factors such as temperature and wind direction.

Physiological and Biochemical Responses of Sedum kamtschaticum and Hosta longipes to Ozone Stress (기린초와 비비추의 오존에 대한 생리·생화학적 반응)

  • In this study, the resistance to ozone and characteristics of ozone-induced damage were investigated on the perennial ground cover plant species. Sedum kamtschaticum and Hosta longipes were exposed to $200{\mu}g{\cdot}kg^{-1}$ ozone for 8 hours per day (from 08:00 to 16:00) in the naturally irradiated phytotron. The extent of ozone-induced damage was measured through the analysis of physiological parameters, such as water use efficiency (WUE), chlorophyll content (Chl. a, Chl. b, Chl. a + b, and Chl. a/b ratio), carotenoid contents, and the induction of reactive oxygen species (ROS). Ozone exposure significantly reduced the daytime WUE in both species. The contents of chlorophyll and carotenoid were also decreased and ROS, such as hydrogen peroxide ($H_2O_2$) and superoxide radical ($O_2{^-}$) were accumulated after ozone exposure. The above results of this study suggested that S. kamtschaticum is more resistant to atmospheric ozone than Hosta longipes. Considering its fast responses to ozone, it was also assumed that Hosta longipes can be used as an indicator plant of an increase in atmospheric ozone concentration.

Effect of Sodium Hydroxide plus Hydrogen Peroxide Treated Mustard (Brassica campestris) Straw Based Diets on Rumen Degradation Kinetics (In sacco), Fermentation Pattern and Nutrient Utilization in Sheep

  • Two experiments were conducted to determine the effect of alkaline hydrogen peroxide (AHP) treatment (1% NaOH+1.5% $H_2O_2$; 1 AHPMS, 2% NaOH+1.5% $H_2O_2$; 2AHPMS) on rate and extent of degradation of mustard straw (MS) in sacco in sheep, and its in vivo digestion and ruminal fermentation characteristics when fed to sheep with concentrate (200 g per sheep daily). The treatment of straw with 1 and 2% AHP increased its sodium content by 148 and 296% to that of untreated straw (UMS). There was significant decrease in NDF and hemicellulose contents of AHP treated straw and increase in cellulose and lignin contents. Phenolic acids like ferrulic, $\rho$-coumaric and o-coumaric significantly (p<0.001) reduced by AHP treatment of mustard straw. In first experiment the in sacco degradation of DM, OM and NDF was significantly (p<0.01) greater for 2 AHPMS than for UMS at all incubation periods. The disappearance of nutrient from 1 AHPMS and 2 AHPMS treated straws continue to increase up to 96 h whereas in UMS the peak disappearance was found at 48 h. By using the equation {(y=a+b) ($1-e^{-ct}$)} the degradation rates (c) for DM, OM, and NDF were significantly higher for UMS than AHP treated straws. Level of alkali (1 and 2%) had significant effect on degradation characteristics (a, b, c and $P_{0.05}$) of DM and NDF fraction of MS. However, the effect was not pronounced on OM fraction of MS. In feeding experiment, the intake of nutrients for DM, OM, cell wall constituents and energy was higher on 2 AHPMS, whereas no effect on the digestibility of these nutrients was observed. The apparent nitrogen retention was higher (p<0.05) both in 1 and 2 AHPMS groups. Water intake by animals was significantly increased due to AHP treated mustard straw feeding. Rumen liquor pH was higher in 2 AHPMS fed animals. The $NH_3-N$ of rumen liquor was not affected by feeding of AHP treated MS based diets. Total VFA concentration was significantly (p<0.01) higher in UMS fed group. The fractional out flow rate of DM was higher (p<0.05) in animals fed on 2 AHPMS diets compared to UMS and lAHPMS fed groups. The population of large holotrichs was higher (p<0.05) on AHP treated MS fed diets compared to UMS. The study indicated that treatment of mustard straw with AHP changed its chemical composition towards a better feed. The nutritive value of 2% AHP treated mustard straw was better in terms of dry matter intake and apparent nitrogen retention. The higher in sacco DM, OM and NDF disappearance however, was not confirmed by in vivo data in this study.

EFFICACY OF HAND REAMER AND ENGINE REAMER TO PREPARE ROOT CANAL (수동(手動)리머와 전동(電動)리머의 근관형성효과(根管形成效果))

  • This experimental study was made to evaluate the efficacy of root canal preparation of engine reamer versus hand reamer. Eighty extracted human teeth were prepared with the following treatments and devided into 4 groups; Group 1: Canal preparation with hand reamer, and irrigation with normal saline solution. Group 2: Canal preparation with engine reamer, and irrigation with normal saline solution. Group 3: Canal preparation with hand reamer, and irrigation with 3% hydrogen peroxide and 3.5% sodium hypochlorite solution. Group 4: Canal preparation with engine reamer, and irrigation with 3% hydrogen peroxide and irrigation with 3% hydrogen peroxide and 3.5% sodium hypochlorite solution. After decalcification, 5p. sections at levels 1-, 3-, and 5-mm from the apex were evaluated microscopically. The results were as follows; The effectiveness of hand reamer and engine reamer were equal in preparing the root canal at all levels from the apex, and were equal in normal saline solution groups, and 3% $H_2O_2$ and 3.5% NaOCl solution groups. Both in hand reaming groups and in engine reaming groups, some canal walls had still untouched portion, and canal irregularities and debris were still remained.

The Study on the Reactivity of Dioxygen Bridged Palladium Complexes Having ${\pi}$-Allyl Ligands (${\pi}$-알릴리간드를 갖는 산소가교 팔라듐착화합물의 반응성에 관한 연구)

  • This study is related to reactivity of dioxygen bridged palladium complexes having ${\pi}$-allyl ligands. In this case, new dioxygen bridged palladium complexes were prepared using superoxide ion$(O_2^-)$ as an oxygen source. Reactions of the dioxygen palladium complexes prepared in the study were examined in order to clarify the nature of the coordinated dioxygen. Treatments of a solution of the dioxygen bridged palladium complexes in benzene by water, methanol and acetic acid gave hydrogen peroxide $(H_2O_2)$ as hydroxy-, methoxy-, and acetoxybridged palladium complexes, respectively. The dioxygen bridged palladium complexes reacted also with substitution phenols of salicylaldehyde, 8-hydroxyquinoline and active methylenes of acetylacetone, dimethyl malonate to afford mononuclear complexes of palladium and hydrogen peroxide. The results suggest that dioxygen is coordinated as peroxo $(O_2^{2-})$ in the complexes and behaves as a strong base.

Lipid Peroxidation Induced by the Reaction of Cytochrome c with Hydrogen Peroxide

  • Lipid peroxidation induced by the reaction of cytochrome c with $H_2O_2$ was investigated. When linoleic acid micelles or phosphatidyl choline liposomes were incubated with cytochrome c and $H_2O_2$, lipid peroxidation was increased in cytochrome c and $H_2O_2$ concentrations-dependent manner. Radical scavengers, azide, formate and ethanol prevented lipid peroxidation induced by the cytochrome c/$H_2O_2$ system. Iron specific chelator, desferoxamine also prevented the cytochrome c/$H_2O_2$ system-mediated lipid peroxidation. These results suggest that lipid peroxidation may be induced by the cytochrome c/$H_2O_2$ system via the generation of free radicals. Carnosine, homocarnosine and anserine are present in the muscle and brain of many animals and human. Previous studies show that these compounds have an antioxidant function. In the present study, carnosine, homocarnosine and anserine significantly prevented the cytochrome c/$H_2O_2$ system-mediated lipid peroxidation. Carnosine and related compounds also inhibited the free radical-generating activity of cytochrome c. The results suggest that carnosine, homocarnosine and anserine may prevent lipid peroxidation induced by the cytochrome c/$H_2O_2$ system through a free radical scavenging.

Antioxidant Effect of Citri Reticulatae Pericarpium Extract on Oxidative Stress-Mediated Cytotoxicity In Cultures

  • In order to examine oxidative stress of reactive oxygen species and the antioxidant effect of Citri Reticulatae Pericarpium (CRP) extract, human skin melanoma cells were treated with various concentrations of hydrogen peroxide ($H_2O_2$). Antioxidant effect of CRP extract on $H_2O_2$-induced cytotoxicity, cell viability, DPPH-radical scavenging activity and superoxide dismutase (SOD)-like activity. In this study, $H_2O_2$ decreased cell viability of cultured human skin melanoma cells in dose- and time-dependent manners, and then, midcytotoxicity value (MCV) was determined at $60\;{\mu}M$ after human skin melanoma cells were cultured for 5 hours in the media containing $20{\sim}60\;{\mu}M$ of $H_2O_2$, respectively. The $H_2O_2$ was on cultured human skin melanoma cells because MCV of $H_2O_2$ was lower than $100\;{\mu}M$. In the antioxidant effect of CRP extract, CRP extract increased cell viability DPPH-radical scavenging activity and SOD-like activity. From these results, it is suggested that $H_2O_2$ was very toxic on cultured human skin melanoma cells. And also, CRP extract has the antioxidant effect on $H_2O_2$-induced cytotoxicity.

Reaction of ferritin with hydrogen peroxide induces lipid peroxidation

  • Lipid peroxidation is known to be an important factor in the pathologies of many diseases associated with oxidative stress. We assessed the lipid peroxidation induced by the reaction of ferritin with $H_2O_2$. When linoleic acid micelles or phosphatidyl choline liposomes were incubated with ferritin and $H_2O_2$, lipid peroxidation increased in the presence of ferritin and $H_2O_2$ in a concentration-dependent manner. The hydroxyl radical scavengers, azide and thiourea, prevented lipid peroxidation induced by the ferritin/$H_2O_2$ system. The iron specific chelator desferoxamine also prevented ferritin/$H_2O_2$ systemmediated lipid peroxidation. These results demonstrate the possible role of iron in ferritin/$H_2O_2$ system-mediated lipid peroxidation. Carnosine is involved in many cellular defense processes, including free radical detoxification. In this study, carnosine, homocarnosine, and anserine were shown to significantly prevent ferritin/$H_2O_2$ system-mediated lipid peroxidation and also inhibited the free radical-generation activity of ferritin. These results indicated that carnosine and related compounds may prevent ferritin/$H_2O_2$ system-mediated lipid peroxidation via free radical scavenging.

Determination of Optimum pH of Enzyme by Electrochemical Method (전기화학적 방법에 의한 효소의 최적 pH 결정)

  • A biosensor for the determination of hydrogen peroxide was constructed by immobilizing of porcine small instestinal tissue in a plain carbon paste, and the effect of varying the $H_2O_2$ concentration and pH on the rate of catalytic reaction was evaluated. For the mathematical simplicity, no mediator was added. Electrochemical properties and the maximal rate could be derived from the quantitative analysis of the observed phenomena related to the electrode reaction. Also, pH dependence of the Michaelis constant enabled to calculate various thermodynamic parameters and subsequently to design a electrochemical method to determine the optimum pH of enzyme.

Chemical Oxidation of Cu - and CN - contained Wastewater (Cu - CN 함유 폐수의 화학적 산화)

  • In the treatment of Cu - and CN - contained wastewater by using Fenton oxidation-flocculation-precipitation, the optimal removal efficiencies of the cyanide and copper were investigated according to pH, reaction time, the molar ratio of cyanide and hydrogen peroxide and the mass ratio of ferrous sulfate and hydrogen peroxide for Fenton oxidation, and pH for hydroxide precipitation, respectively. As a result, the $CN^-$ removal efficiency in our experimental wastewater by the Fenton oxidation was 81.2%~99% at its optimal conditions of pH ranging from 3 to 5 and reaction time of 30 minutes. And the optimal dosage of hydrogen peroxide and ferrous sulfate was 214, $428mg/{\ell}$, 107, $161mg/{\ell}$, 214, $214mg/{\ell}$ and 520, $500mg/{\ell}$, respectively when the molar ratio of $Cu^{2+}$:CN is 2:1, 1:1, 1:2, 1:10, and was 900, $1050mg/{\ell}$ when the molar ratio of $Cu^+$: CN is 1:10. When the copper was precipitated by sodium hydroxide after Fenton oxidation, the copper removal efficiency in the wastewater at pH 7 was 98.92%, 98.52%, 92.46%, 90.6% and 95%, respectively.
