Phenogenetical Relationships of Sex Comb and Genital Arch in Drosophila melanogaster Complex and Their Hybrids (Drosophila melanogaster comple 3종 및 그들간 잡종의 성즐과 생식궁의 표현형적 유연관계)

  • Drosophila melanogaster, D. simulans, and D. sechellia are closely related species which belong to the D. melanogaster complex; the first two cosmopolita and the last one restricted to th Seychelles archipelago. The phenogenetical relationship between this complex and their hybrids were investigated by the comparison of sex-comb tooth number an genital arch of male. In interspecific hybrids of all crosses between three species four hybrid males were produced and completely sterile. Males of D. simulans (${O}_{9}$) have significantly less sex-comb teeth (mean 8.35) than either D. melanogaster (OR, mean 10.73) and D. sechellia (Ja, mean 10.60). From the analysis by the number of sex-comb tooth in interspecific hybrids we could not represent the direction of heredity nature. each species of D. melanogaster complex were characteristic in the shape of the genital arch, which readily allows these species to be distinguished. The common structure of the genital arch in the interspecific hybrids were mosaic-like structure between parental species.

  • This study was conducted to identify inbreds and hybrids by measuring plant height, root and other characters at early growing stage. Five inbreds and four hybrids were used for the study and results obtained are as follows; 1. Shoot and root length were significantly different between inbreds and hybrids even 5 days after germination. Slight differences within inbreds and hybrids were also observed. 2. Significantly different observations between inbreds and hybrids for stem diameter, fresh weight, and dry matter of root and stem began to show 23 days after germination.

  • Morphological characteristics of leaves, trichomes and acorns were investigated in 6-year-old artificial hybrids of Q. aliena ${\times}$ Q. mongolica var. crispula and Q. serrata ${\times}$ Q. mongolica var. crispula. Leaf shapes of Q. aliena ${\times}$ Q. mongolica var. crispula $F_{1}$ were obovate and resembled to that of Q. aliena. But several characters including the size of leaf and petiole and the shape of leaf base resembled to those of Q. mongolica var. crispula. In F1 hybrids, small stellate hairs distributed sparsely on the abaxial surface and their lay length was intermediate between both parents. There were no big differences on characters of nuts and cupules between both parents and $F_{1}$ hybrids. Leaf shapes of Q. serrata ${\times}$ Q. mongolica var. crispula $F_{1}$ were obovate-elliptic, and the leaf shape and leaf base and the length of petiole resembled to those of Q. mongolica var. crispula, but leaf size and serration resembled to those of Q. serrata. The number of serration in a leaf was intermediate between both parents. Small stellate hairs distributed sparsely and large single hairs were mixed on the reverse side of leaves. there were no big differences on the number and size of stellate hairs between $F_{1}$ hybrid and Q. serrata. It is able to distinguish $F_{1}$ hybrids from both parents by the size leaf size and shapes, leaf base and serration, petiole length and trichome type in the leaf.

Enhanced Seed Development in the Progeny from the Interspecific Backcross (Fagopyrum esculentum ${\times}$ F. homotropicum) ${\times}$ F. esculentum

  • To facilitate the introgression of F. esculentum into the traits of F. homotropicum, several accessions of the hybrids between these two species were pollinated with F. esculentum as the recurrent parent. The embryo in vitro rescue was performed to increase the recovery of backcross progenies. The $F_{2}$ generation was more amenable than $F_{1}$ hybrids to produce backcross progenies. The $F_{1}$ hybrids were backcrossed twice with common buckwheat (pin-type F. esculentum) (recurrent backcrossing). Also, alternate backcrosses with common buckwheat and F. homotropicum (congruity backcrossing) were carried out. Pollen tube growth of BC$F_{1}$ ${\times}$ F. esculentum (thrum) and F. homotropicum ${\times}$ BC$F_{1}$ was the disturbed penetration exceeded for all initial interspecific hybrids, and its requirement was proportionally lower when the common buckwheat was used as the recurrent parent and as the last parent of congruity hybrids. Effects of both common buckwheat and F. homotropicum on seed success rate for hybridization were observed. Growth of hybrid embryos before rescue, regeneration of mature hybrids all increased recurrent and congruity backcrosses and inter-crosses between $F_{1}$ plants and selected fertile plants of the second congruity backcrosses.

Effects of Maturing Stage of Corn Hybrids on Silage Yield, Feeding Value for Dairy Cows and Milk Production in a Cold Region of Japan

  • This experiment was conducted to evaluate the effects of differently maturing corn hybrids on silage production and milk production per unit area in the northern part of Japan, where grain development occurs under decreasing ambient temperature. Both hybrids were harvested at the same time. The stages of maturity for the early-maturing hybrids (EH; 80 d relative maturity) and the mid-maturing hybrids (MH; 93 d relative day) were early dent and late dough stage, respectively. The plant yields for MH were higher than those for EH. The dry matter (DM) content of MH was lower than that for EH, and the effluent loss for MH silage was greater than that for EH silage. Therefore, the DM yields of prepared silage per area were similar for both treatments. Twelve multiparous mid-lactation Holstein cows ($58{\pm}13$ days in milk) were fed diets based on EH or MH silage in a crossover design with two 3-week periods. Cows were fed 3 kg of hay crop silage (DM basis) and either EH or MH silage ad libitum, and concentrates were supplied to meet NRC requirement for dairy cows. Silage DM intake for EH was found to be higher (p<0.05) than that for MH (10.0 vs. 9.1 kg/day). Milk production and milk composition for EH were similar to those for MH. Feed efficiency per total feed intake was similar in both treatments, although the feed efficiency per concentrate intake tended to be higher for the EH than that for the MH diet. These results indicate that differences in maturation in corn hybrids affect the effluent production of silage and the silage intake of dairy cows. It may be advantageous to plant early hybrid corn with a reduction in effluent production of silage as well as a reduction in purchased feed costs for dairy cows under the climatic conditions of the northern part of Japan.

  • In order to preparr the hybridoma cells which produce a monoclonal antibody to human fibroblast interferon(Hu IFN-$\beta$), spleen cells from BALB/cmice immunized with the purified Hu IFN-$\beta$ were fused with NS-O cells, a myeloma cell line. Forty hybrids with high titer among 1300 hybrids Isolated by an ELISA screening method were subcloned using the soft agarose cloning and limiting dilution methods, and 11 hybrids were selected. As a result of iso-typing the hybrids using the mouse monoclonal typing kit, two hybridomas were found to produce 1gG 2a type of monoclonal an-tibodies. The ascites fluid from nude mice inoculated intraperitoneally with the above hybridomas was removed and purified using a protein A-Sepharose CL-4B. Monoclonal antibody was proven to have only the heavy and light chains on SDS-polyacrylamide gel electrophoresis.

  • Protein content of waxy corn hybrids ranged from 9.3 to 17.1%, and amylopectin content ranged from 83.9 to 94.5%. Three waxy corn hybrids including CNU043 showed less than 10 percent in protein content, and seven waxy corn hybrids including CNU052 were higher than 94% at the amylopectin content. At the amylogram test the ranges of the pasting temperature, peak viscosity, hot viscosity, cool viscosity, breakdown, setback, and consistency were 71.6~$77.5^{\circ}C$, 58~210 RVU, 36.1~114.2 RVU, 47~145 RVU, 21.4~97.8 RVU, -66.4~-9.6 RVU, and 11.1~36.3 RVU, respectively.

  • Reciprocal interspecific hybrids between two bitterling species Rhodeus uyekii (RU) and R. notatus (RN) were genetically identified based on the partial sequence analysis of recombination activating gene-1 (RAG-1) gene. Out of 863 bp positions analyzed, 13 nucleotide substitutions were detected between the two parental species (RU and RN genotypes). Both the induced hybrids (RU female$\times$RN male; UN genotype) and their reciprocal counterparts (RN female$\times$RU male; NU genotype) displayed the double peaks (or polymorphism) of sequence chromatograms at the 13 diagnostic positions, indicating that those hybrids were actual karyogamy derived from the two parental haploid genomes. However, it was not possible to distinguish between the reciprocal interspecific hybrids.

Studies on the Taxonomical Characters in Populus tomentiglandulosa and P. glandulosa (Populus tomentiglandulosa와 P. glandulosa의 분류학적(分類學的) 연구(硏究))

  • Populus glandulosa and Populus tomentiglandulosa, which were known to be natural hybrids, were examined for morphological, physiological and karyological traits to illucidate its hybridity and taxonomical importance. The results abtained were as follows; 1. Survival rate in rooting of cuttings and grafting was different between the hybrids and their rooting abilities showed incomplete dominance. 2. Their leaf openings showed incomplete dominance. The leaf longevities of P. alba ${\times}$ glandulosa and P. tomentiglandulosa were stronger than the other hybrids. 3. There were differences in resistance to toxicity of $KClO_3$ between the hybrids. 4. Many external leaf characters of the hybrids also showed incomplete dominance. P. tomentiglandulosa was similar in those characters to P. alba ${\times}$ glandulosa while P. glandulosa was similar to hybrids crossed, reciprocally crossed or back-crossed between P. davidiana and P. alba. 5. Their numbers of male flower showed incomplete dominance or hybrid vigor. The numbers of P. tomentiglandulosa were similar to thosa of P. alba ${\times}$ glandulosa while those of P. glandulosa to those of P. alba ${\times}$ davidiana or P. davidiana ${\times}$ alba. 6. Morphology and band color of male catkin bract showed incomplete dominance. Those of P. glandulosa were similar to those of P. alba ${\times}$ davidiana while those of P. tomentiglandulosa to those of. P. alba ${\times}$ glandulosa. 7. There were differences in vascular bundle number and arrangement of petiole between the hybrids. 8. Differences in the anatomical traits of stem did not exist between the hybrids but those in wood fiber size existed. 9. The chromosomes of artificial hybrids, P. glandulosa and P. tomentiglandulosa showed irregular behavior in metaphase I and II. 10. All hybrids including P. glandulosa and P. tomentiglandulosa showed small number of P.M.C. with 19 II but many univalent chromosomes were exhibited in metaphase I. 11. All hybrids including P. glandulosa and P. tomentiglandulosa showed a little abnormal nuclear plates as laggard chromosome and chromosome bridge in anaphase I and II. 12. The frequency of pollen tetrad and fertile pollen was low in most of the hybrids including P. glandulosa and P. tomentiglandulosa.

Nuclear DNA inheritance of intra-specific somatic hybrids by di-mono cross in Pleurotus ostreatus based on URP-PCR analysis (URP-PCR 분석에 의한 느타리 이핵-단핵 계통간 교잡주의 핵 DNA 유전)

  • The primary objective of the present study is the characterization of the somatic hybrids of dikaryon-monokaryon (di-mono) crosses in mushroom breeding. We employed this technique for developing superior strains from Pleurotus ostreatus strains with 48 intraspecific hybrids of 12 combinations between six P. ostreatus strains and one P. florida strain. The results on the experiments of hybridization rate, nuclear DNA patterns, and colors and morphology of fruit-bodies, are presented. In di-mono crosses, somatic hybrids among Pleurotus strains showed 100% of crossability as seen in those between P. ostreatus and P. florida strains indicating that the nuclei of a dikaryon is inferred to be migrated to a recipient. 87.5% of the somatic hybrids among Pleurotus strains were similar to the donor dikaryons, and 12.5% of the somatic hybrids presented DNA patterns of both parents. In 16.6% of di-mono crosses between P. ostreatus and P. florida, the nuclear DNA patterns of all hybrids showed the same or similar patterns compared to the donor dikaryons. 70.9% of the hybrids between P. ostreatus and P. ostreatus were similar to the donor dikaryons and 12.5% of them presented the DNA patterns of both parents. 79.2% of fruiting body morphology of the hybrids among Pleurotus strains were similar to the dikaryons and 20.8% of them were similar to both parents. Interestingly, the morphology of all dikaryons were dissimilar each other. All hybrid strains between dikaryon P. florida and monokaryon P. ostreatus showed the fruiting body of which colors were similar to those of the dikaryon, while the hybrids between dikaryon P. ostreatus and monokaryon P. florida were showed the combined colors of both parents. Therefore, the fruiting body color of P. florida tends to be generally dominant. In conclusion, the present study provides a way to find out and suggest superior hybrid strains using the nuclear DNA patterns of hybrids between Pleurotus strains as well as the characteristics of their fruiting bodies. The advantages of the di-mono crossing are needs to be fully utilized in mushroom breeding because it is an ideal way to develop the superior strains of Pleurotus.
