Inhibitory effects of Kimchi lactic acid bacteria on harmful enzymes of human intestinal bacteria

  • Lactic acid bacteria have been considered as the most beneficial probiotic organisms contributing to inhibition of harmful and putrefactive intestinal bacteria. Among them, Bifidobacterium spp. has been considered as one of the most beneficial probiotic organism that can improve the health of humans, since it is one of the major bacteria flora in human intestine. However, the harmful enzyme-inhibitory activity of lactic acid bacteria of Kimchi, which is a representative Korean fermented food has not been evaluated. (omitted)

Inhibitory Effects of Bifidobacterium spp. Isolated from a Healthy Korean on Harmful Enzymes of Human Intestinal Microflora

  • Five hundreds of bifidobacteria were isolated from a healthy Korean and the inhibitory effects of these isloated bacteria on harmful enzymes of human intestinal microflora were examined by cocultivation of the isolated bifidobacteria with E. coli or total human intestinal microflora. In comparison with the results of E. coli or intestinal microflora cultivation, Bifidobacterium breve K-110, B. breve K-111 and B. infantis K-525 effectively inhibited harmful enzymes ($\beta$-glucuronidase and tryptophanase) of E. coli and lowered the pH of the culture media. Also they inhibited the harmful enzymes ($\beta$-glucosidase, $\beta$-glucuronidase, tryptophanase and urease) and ammonia production of intestinal microflora, and lowered pH of the culture media by increasing lactic acid bacteria of intestinal microflora. When these isolated bifidobacteria were administered on mice, fecal harmful enzymes were also inhibited. Among tested bifidobacteria, B. breve K-110 had the highest inhibitory effect of fecal harmful enzymes.

고추 및 토마토 친환경 및 관행재배지에서 분리한 인체 유해세균의 항생제 저항성 평가 (Evaluation of Antibiotics Resistance for Human-harmful Bacteria Isolated from Eco-friendly and Practical Cultivation Farms of Hot Pepper and Tomato)

  • This study was conducted to monitor the antibiotics resistance of human-harmful bacteria isolated in the agricultural environment for hot peppers (Capsicum annuum) and tomato (Lycopersicon esculentum). As a result, we isolated 120 bacterial species (34 on fruits, 48 in soil, 21 in water, and 17 in manure), identified them with the 16S rRNA sequence, analyzed minimum inhibitory concentration (MIC) for 26 antibiotics using Sensititre ARIS Hi-Q system and then evaluated whether each bacterial genus acquired resistance for the tested antibiotics or not, according to the CLSI criteria. From difference in MIC between eco-friendly (EFM) and practical (PFM) cultivation farms, Klebsiella spp. isolated from EFM was resistant to ampicillin (AMP) and nalidixic acid (NAL), and that isolated from PFM was resistant to streptomycin (STR) and tetracycline (TET). Enterobacter spp. isolated from EFM was resistant to AMP and azithromycin (AZI), and that isolated from PFM was resistant to AMP, AZI, and STR. Meanwhile, Pseudomonas spp. isolated from EFM and PFM were all resistant to AMP, AZI, cefotaxime (FOT), cefoxitin (FOX), ceftriaxone (AXO), CHL, NAL, and STR. Staphylococcus spp. isolated from EFM and PFM were resistant to gentamycin (GEN), STR, and kanamycin (KAN), and in particular, that from EFM showed resistance for erythromycin (ERY). In conclusion, our study suggested that EFM lead STR antibiotics resistance for human-harmful bacteria to decrease, because only the bacteria isolated from hot pepper and tomato crop with PFM have showed resistance against STR antibiotics, regardless of bacterial genus.

유산균의 장내환경개선효과 (Isolation of bifidobacteria inhibiting harmful enzymes of Korean intestinal bacteria)

  • Five hundreds of bifidobacteria were isolated from an healthy Korean and the inhibitory effects of these isolated bacteria on harmful enzymes of human intestinal microflora were examined by cocultivation of the isolated bifidobacteria with E. coli HGU-3 or total human intestinal microflora. The inhibitory effect of bifidobacteria on Growth of Helicobacter pylori and Rotavirus infection were exammed. Bifidobacterium K-110 and K-111 inhibited effectively them. When these isolated bifidobacteria were administered to mice, the activities of fecal harmful enzymes were inhibited and the AC and ACF formation were suppressed. Among tested bifidobacteria, B. breve K-110 had high inhibitory effect of fecal harmful enzymes and ACF formation.

Effect of Lentinus edodes on the Growth of Intestinal Lactic Acid Bacteria

  • As the growth factor of lactic acid bacteria, LD (trehalose) was isolated from Lentinus edode5 by using silica gel column chromatography. LD induced the growth of Bifidobacteria breve and Lactobacillus brevis, which were isolated from human feces. LD selectively induced the growth of lactic acid bacteria among total microflora. When total intestinal microflora were cultured in the medium containing LD, it stimulated the growth of lactic acid bacteria and inhibited harmful enzymes, ${\beta}$-glucosidase, ${\beta}$-glucuronidase, and tryptophanase, of intestinal bacteria. LM, which was a monosaccharide from L. edooles, induced the growth of lactic acid bacteria but it seems to be invaluable in vivo. LH isolated from L. edodes by Sephadex G-100 column chromatography was not effective for the growth of lactic acid bacteria.

한국인의 장내 균총 조성 및 분포 (Composition and Distribution of Intestinal Microbial Flora in Korean)

  • Intestinal microbial flora comprise one third of the large intestinal contents in human. They play a significant effects through beneficial and harmful action on the human health. This is the first study which examined the composition of the microflora of the general population in Korea. Bacteroides, Bifidobacterium, Eubacterium, Peptostreptococcus, Lactobacillus, Streptococcus, Escherichia coli, Staphylococcus, Clostridium perfringens, total aerobic bacteria and total anaerrobic bacteria were counted using various selective and non-selective media. Among the bacteria studied the number of Bifidobacterium were greatest in breast-fed infants(30-90 days old), whereas Streptocuccus and Bifidobacterium in bottle-fed infants. In 20-40 age group Bacteroides were predominant followed by Bifidobacterium and Eubacterium. In early group(over 65 years old) Bacteroides were predominant followed by Eubacterium and bifidobacterium. The frequency and number of Cl. perfringens were highest in dlderly group. These results confirm that the microfloral pattern in large intestine change during the life cycle of humans.

결장암 예방에 대한 유산균의 기능 (The Functions of Lactic Acid Bacteria in Colon Cancer Prevention)

  • Certain lactic acid bacteria have anti-tumor activity, especially colon cancer. The fermented milk products containing that kind of lactic acid bacteria have to be recommended for human health as excellent health functional foods. This paper have been classified by 5 regions on the functions of lactic acid bacteria related to prevention of colon cancer. 1) Enhancing of host's immune response; Production of cytokines. 2) Binding and degradation of potential carcinogens; Binding and degradation of mutagenicity. 3) The changes of intestinal microflora and production of antitumorigenic or antimutagenic compounds; Production of azoxymethane. 4) Alteration of the metabolic activity of intestinal microflora; Decrease of harmful enzymes in intestinal tract. 5) Alteration of physicochemical conditions in the colon; Decrease of pH and bile acids contents.

미나리발효액이 장내 유해세균 및 유익균의 In Vitro 생육 및 효소활성에 미치는 영향 (Effect of Extract of Fermented Dropwort on Intestinal Bacteria and Enzymes In Vitro)

  • 미나리(Oenanthe stolonifera) 발효액이 장내 병원성 미생물과 유익균의 생육, 그리고 장내 세균효소에 미치는 영향을 in vitro 에서 조사하였다. 고상 배지 (Agar plate) 에서의 clear zone 형성에 의한 생육저해를 측정한 결과, Vibro, Salmonella 등의 유해 미생물에 대해서 강한 생육저해 효과를 보였으며 Bifidobacterium longum 에 대해서는 생육저해 효과가 크지 않았다. 액체배지에서의 최소저해농도(MIC) 측정에서도 고장배지에서와 같은 경향을 보여 상대적으로 B. longum 에 대한 생육저해가 가장 적었다. 발효 기간에 따른 영향을 보면 발효 기간이 길수록 적은 양으로도 유해한 균들의 생육을 잘 저해할 수 있는 것으로 나타났다. 장내 세균 효소인 ${\beta}$-glucuronidase와 tryptophanase의 활성에 대해 미나리 발효액은 발효하지 않은 액에 비해 저해효과가 컸으며 발효기간이 길수록 저해효과도 증가하였다. 이상의 실험 결과로서 미나리 발효액은 유해세균에 대한 생육저해능이 크고 상대적으로 유익균인 B. longum에 대한 저해는 적어, 음용 시 정장 효과가 있을 것으로 추정된다.

Bacillus licheniformis AJ 균주제제의 설사원인 미생물의 성장 억제효과 (Inhibitory Activity of Bacillus licheniformis AJ on the Growth of Diarrheal Pathogens)

  • The injibitory effect of Bacillus licheniformis AJ isolated from genitourinary normal flora as a new probiotics on the growth of diarrheal pathogens was studied. This B. licheniformis AJ inhibited the growth of E.coli O-157. Salmonella typhi and Shigella sonnei as well as the infectivity of rotavirus. However, it did not inhibit the growth of Helicobacter pyloriand human intestinal bacteria although it inhibited the harmful enzyme activity of human intestinal bacteria. B. licheniformis AJ seems to excret heat-lable growth-inhibitory protein, bacteriocin, into the media. These results suggest that B. lichenoformis AJ could be used as a new type of probiotics.

  • In recent years, harmful substances and fine dust in the air are caused by respiratory and cardiovascular diseases through various mechanisms when they are introduced into the human body through respiration, thereby exacerbating human health and causing cancer by prolonged exposure do. In order to prevent such fine dust from being introduced into the room and to improve indoor air quality, improvement of air quality has attracted attention. Among indoor air pollutants, fine dust and CO2 are pollutants that are directly affected by indoor number and activity. The purpose of this study is to evaluate the basic performance of cement matrix using photosynthetic bacteria as a basic study of fine dust and CO2 adsorption type matrix to suppress indoor air pollution and improve air quality.
