Immune-triggering effect of the foodborne parasite Kudoa septempunctata through the C-type lectin Mincle in HT29 cells

  • Kudoa septempunctata is a myxozoan parasite that causes food poisoning in individuals consuming olive flounder. The present study aimed to investigate the currently insufficiently elucidated early molecular mechanisms of inflammatory responses in the intestine owing to parasite ingestion. After Kudoa spores were isolated from olive flounder, HT29 cells were exposed to spores identified to be alive using SYTO-9 and propidium iodide staining or to antigens of Kudoa spores (KsAg). IL-1β, IL-8, TNF-α and NFKB1 expression and NF-κB activation were assessed using real-time PCR, cytokine array and western blotting. The immunofluorescence of FITC-conjugated lectins, results of ligand binding assays using Mincle-Fc and IgG-Fc, CLEC4E expressions in response to KsAg stimulation, and Mincle-dependent NF-κB activation were assessed to clarify the early immune-triggering mechanism. Inflammatory cytokines (IL-1β, GM-CSF and TNF-α), chemokines (IL-8, CCL2, CCL5 and CXCL1) and NF-κB activation (pNF-κB/NF-κB) in HT29 cells increased following stimulation by KsAg. The immunofluorescence results of spores and lectins (concanavalin A and wheat germ agglutinin) suggested the importance of Mincle in molecular recognition between Kudoa spores and intestinal cells. Practically, data for Mincle-Fc and KsAg binding affinity, CLEC4E mRNA expression, Mincle immunofluorescence staining and hMincle-dependent NF-κB activation demonstrated the involvement of Mincle in the early immune-triggering mechanism. The present study newly elucidated that the molecular recognition and immune-triggering mechanism of K. septempunctata are associated with Mincle on human intestinal epithelial cells.

비만 억제를 위한 췌장 리파아제 도메인에 대한 특이 난황항체의 개발 (Development of Egg Yolk Antibody Specific to the Pancreatic Lipase Domain for Anti-Obesity)

  • 사람 췌장 리파아제는 췌장에서 합성되어 소장으로 분비된 후, 음식물속의 지방을 가수분해하는 소화효소이다. 췌장 리파아제 단백질은 촉매활성도메인과 코리파아제 결합도메인의 2개의 도메인으로 구성되어 있다 본 연구에서는 리파아제 전체단백질과 촉매활성도메인 및 코리파아제 결함도메인 유전자를 PCR 방법으로 증폭하여 발현벡터에 넣은 후, 대장균에서 발현시켜 얻은 3가지 재조합 단백질을 산란계에 3차례 주사하여 면역반응을 유도하였다. 난황단백질을 분리하여 항체가를 측정한 결과, 코리파아제 결합도메인의 항원성이 가장 높은 것으로 밝혀졌다. 또한, 돼지 췌장 리파아제의 활성저해 실험에서도 코리파아제 결합도메인에 의해 만들어진 난황항체가 지방분해활성 저해작용이 가장 큰 것으로 밝혀졌다. 코리파아제 결합도메인 난황항체가 첨가된 지방을 실험동물에게 급여하고 혈중 주요 성분을 분석한 결과, 코리파아제 결합도메인에 의한 난황항체를 섭취한 경우에 혈중 중성지방의 수치가 대조군에 비해서 유의적으로 감소했음을 화인하였다. 결론적으로 코리파아제 결합도메인을 항원단백질로 사용하여 얻은 난황항체가 생체 내에서 췌장 리파아제의 활성을 억제할 수 있으며 비만억제제로의 개발 가능성이 있음을 보여주었다.

한국산(韓國産) 사독(蛇毒)에 관(關)한 실험적(實驗的) 연구(硏究) (Experimental Studies on Korean Snake Venoms)

  • Agkistrodon halys (Crotalidae) is the only species of poisonous snakes in Korea, and is divided into three subspecies; Agkistrodon bromhoffii brevicaudus, Agkistrodon calaginosus and Agkistrodon saxatilis. With the three venoms, the pharmacological actions on the cardiovascular system and intestine as well as some toxicological characteristics were studied. In addition, the precipitin test in an agar gel medium was employed for immunological comparison of the venoms and the sera of envenomed patients. The results obtained were as follows: Lyophilized venoms contained solids of $211{\sim}273mg/ml$, and LD50 to mice were 1.73 and 0.86 mg/kg in venoms of Agkistrodon bromhoffii brevicaudus obtained on July and October respectively, and 0.40 and 0.32 mg/kg in Agkistrodon calaginosus and the venoms of Agkistrodon saxatilis obtained on October was 2.29 mg/kg. Isoelectric focusing of lyophilized snake venoms showed 19 to 22 protein fractions and 2 to 3 isoamylase fractions. Acute irreversible hypotension was caused by the intravenous injection of large doses of venoms in rabbits and cats, but at the small doses, acute hypotension followed by slow recovery. Little changes of cardiac movements by the venom injection despite of marked hypotension were showed except bradycardia and arrhythmia prior the death. Also no changes on the isolated rabbit atria by the snake venoms were noted. The hypotensive effect of the snake venoms was prevented by the bilateral vagotomy or atropine pretreatment (1 mg/kg), but they did not affect when already the hypotension has undergone. In the isolated rabbit duodenum, small doses of venom increased the phasic movement, while large doses decreased after spastic contraction. With the injection of venoms in dog, strong contraction of gall-bladder was caused and it was not blocked by the pretreatment with phenoxybenzamine (10 mg/kg) or atropine (1.4 mg/kg). In the venoms of Agkistrodon bromhoffii brevicaudus and Agkistrodon calaginosus, at least 5 antigenic components were detected, and four of them were shared in common with each other. Polyvalent antivenin (Wyeth Lab. USA) had three common precipitating antibodies with the venom of Agkistrodon bromhoffii brevicaudus and Akistrodon calaginosus. In the serum of envenomed patients, no precipitating antibodies were seen to the venoms and little changes in serum protein, GOT and GPT were observed. In conclusion, the snake venoms obtained in Korea were highly toxic and caused chiefly the vascular collapse leading to death. This vascular collapse was resulted largely by cholinergic effects, and not cardiotoxin of venoms. In human, it is likely that precipitating antibodies to venom were not produced by an envenomed incidence to poisonous snakes.

표고버섯 추출액이 loperamide로 유도된 변비에 미치는 영향 (Effects of Lentinus edodes Extract on the Loperamide-induced Constipation in Rats)

  • 표고버섯 추출액을 10, 20 및 40%로 30일간 음용수에 섞어 실험동물에 투여하고 실험 28-30일에 loperamide(4 mg/kg/day, s.c.)로 변비를 유도한 뒤 표고버섯 추출액의 변비 예방효과를 측정하였다. Loperamide를 단독 투여한 군은 정상 대조군과 비교하여 변의 개수 및 중량이 유의적으로 감소하였으며 원위 결장 내 변잔류의 증가 및 cecocolonic segment의 무게가 증가하였다. 표고버섯 추출액 및 loperamide를 투여한 군은 loperamide를 단독 투여한 군과 비교하여 변의 개수 및 중량이 유의적으로 증가하였으며 원위 결장 내 잔류 변 및 cecocolonic segment의 무게도 감소하였다. 이러한 결과는 표고버섯 추출액이 in vivo에서 변비 개선효과가 있음을 보여준다. 변의 수분함량은 실험군 간의 유의한 차이가 없었지만 loperamide에 의한 변비 유발 시 감소하는 경향이 있었으며, 표고버섯 추출액 투여군에서는 변의 수분함량 감소가 다소 완화되었다. 한편, 혈 중 WBC, RBC, HGB, HCT, PLT, MCV, MCH, MCHC치 및 혈청 ALT, AST, ALP, BUN 및 creatinine 수치는 정상 대조군, loperamide 처리군 및 표고버섯 추출액 처리군 사이에서 차이가 없었다. In vitro 실험결과, 회장 적출 절편에 표고버섯 추출액 동결건조물(0.5 mg/mL)을 전 처리 시 loperamide에 의한 장력과 진폭 억제가 부분적으로 차단되었으며, atropine에 의해 유도된 진폭의 억제도 효과적으로 차단되었다. 이러한 결과는 표고버섯 추출물의 변비 개선효과가 장의 운동성 촉진과 관련이 있음을 시사한다.

흰쥐에서 Divalent Metal Transporter 1의 조직내 분포와 Iron에 의한 조절 (Tissue Distribution of Divalent Metal Transporter 1 and Regulation by Dietary Iron in Rats)

  • Iron (Fe) is an essential metal in biological processes, which maintains a homeostasis in the human body. Divalent metal transporter 1 (DMT1) has been known as an iron transporting membrane protein, which is involved in the uptake Fe at the apical portion of intestinal epithelium, and may transport Fe across the membrane of acidified endosome in peripheral tissues. In this study, we studied the tissue distribution of DMT1 in the Fe supplemented (FeS) diet fed rats, and the regulation of DMT1 expression by depleting body Fe. Sprague-Dawley rats were divided into two groups, and fed FeS (120 mg Fe/kg) diet or Fe deficient (FeD, 2∼6 mg Fe/kg) diet for 4 weeks. The evaluation of body Fe status was monitored by measuring sFe, UIBC and tissue Fe concentration. Additionally, DMT1 mRNA levels were analyzed in the peripheral tissues by using the quantitative real time RT-PCR method. In the FeS diet fed rats, the tissue Fe was maintained at a relatively high level, and DMT1 was eventually expressed in all tissues studied. DMT1 was highly expressed in the testis, kidney and spleen, while a moderate levels of DMT1 expression was detected in the brain, liver and heart. In the digestive system, the highest level of DMT1 was found in the duodenum. Feeding the FeD diet caused a reduced body weight gain and depletion of body Fe with finding of decreased sFe, increased UIBC and decreased tissue Fe concentration. The depletion of body Fe upregulated DMT1 expression in the peripheral tissue. The expression of DMT1 was very sensitive to the body Fe depletion in the small intestine, especially in the duodenum, showing dramatically higher levels in the FeD rats than those of the FeS group. In the FeD diet fed animals, the expression of DMT1 was low significantly in other tissues compared with the duodenum. The expression of DMT1, however, was 60∼120% higher in the testis, kidney and spleen, and 30∼50% higher in the lung, liver and heart, compared to the FeS diet fed rats. In summary, DMT1 expression was ubiquitous in mammalian tissue, and the level of expression was the organ-dependent. The expression of DMT1 in peripheral tissues was upregulated by depletion of body Fe. Duodenum was the most sensitive tissue among organs studied during Fe depletion, and expressed the greatest level of DMT1, while other tissues were less higher than in duodenum. This study supports that DMT1 plays a role in maintaining the body Fe level through intestinal uptake as well as homeostasis of Fe in the peripheral tissue.

『영추ㆍ경맥편』과 『상한론』의 양명병에 대한 상관성 연구 (Study of Relationship between the Chapter of Channels in Miraculous Pivot of Emperor′s Classic of Internal Medicine and Yangmyung disease in Sanghanron)

  • Chang Chung-ching(張仲景) in the Later Han(Eastern Han) Dynasty of Chinese history wrote the treatise on Diseases Caused by Cold Factors(傷寒論; Shang Han Lun) on the basis of the fundamental theory of Emperor's Classic of Internal Medicine(黃帝內經; ECIM) after collecting medical treatment experiences until the Han Dynasty. It had great significance that Shang Han Lun was the origin of treating six-channels(六經) and there showed the peculiar guidelines on the diagnosis and treatment of oriental medicine to divide diseases into six-channels. The oriental medical doctors who had studied Shang Han Run thought highly of meridians and until now it was generally known that the chapter of heat in the Plain Questions of ECIM(黃帝內經, 素問ㆍ熱論) was the basis of Shang Han Run. The chapter of heat in the Plain Questions of ECIM was the first text in which the basic theory on six-channels according to the types of illness was introduced. In my point of view, the theory of treating six-channels had close relation to the Chapter of Channels in Miraculous Pivot of ECIM(黃帝內經, 靈樞ㆍ經脈篇) as well as the chapter of heat in the Plain Questions of ECIM. Therefore I took a look at the origin of treating six-channels in Shang Han Lun and illuminated again the meaning to compare the parts of in Shang Han Lun with the Chapter of Channels in Miraculous Pivot of ECIM. Conclusion: The, symptoms divided into six-channels in the chapter of channels in ECIM gave the fundamental basis of diagnosis and treatment basesd on overall analysis of signs and symptoms(辨證論治) an illness in the Zangfu(臟腑) in respect of meridians. Viewed in the light of diagnosis and treatment basesd on overall analysis of signs and symptoms(辨證論治), the symptoms of YangMing-channel(陽明經) in the Chapter of Channels in Miraculous Pivot of ECIM were, for the most part, accord with those of YangMing-disease in Shang Han Lun. Furthermore, the symptoms in Shang Han Lun were explained definitely and in detail. Therefore the theory of Shang Han Lun has been developed on the basis of ECIM with the changes of the times. YangMing-disease in Shang Han Lun implied medical cases in stomach meridian(胃經) and large intestine meridian(大腸經). Therefore Shang Han Lun was the foundation of treatment basesd on overall analysis of signs and symptoms(辨證論治) in respect of meridian as well as the text in which the steps of infectious diseases(外感病) of human bodies were explained.

수막뇌염을 동반한 진국인 유극악구충증 1예 (Description of a Male Gnathostoma spinigerum Recovered from a Thai Woman with Meningoencephalitis)

  • 서울에 있는 미군 병원에서 시행한 신경외과 수술에서 검출 의뢰된 충체를 유극악구충으로 동정하여, 국내에서 충체로 화인된 첫 증례로 보고한다. 환자는 젊은 태국인 여성으로 태국에 체재할 때부터 수막뇌염 증세를 앓았고, 뇌척수액에서는 수많은 호산구를 검출하였다. 등글게 말려서 고정된 충체는 길이가 12.3mm, 폭이 0.9mm이고 앞쪽 끝에 근 head bulb가 있었다. 이 기관의 표피에는 고양이 발톱모양의 피극이 8줄로 배열되어 있었다. 충체 전반부의 표피에는 수많은 피극이 가로로 열지어 있는데 head bulb 가까이 에서는 등근 기저부와 4분지의 끝이 있는 굵은 피극이 분포하나 점차 뒤쪽으로 3분지되고 2분지되면서 가늘고 걸어지는 피극으로 이행되었다. 중간부에서는 짧고 작은 가시모양의 피극이 관찰되고 그 수도 크게 감소하였고 후반부에서는 피극이 관찰되지 않았다. 길이 12.4mm의 식도와 두 개의 식도선(cervical sacs)이 있고, 장이 식도에서 항문까지 곧게 이어져 있었다. 장은 검고 불투명하며, 심하게 굴곡하는 고환과 겁쳐져서 관찰되었다. 뒤 끝에서는 작은 피극이 다시 나타나서 가로로 배열하였고 항문 주위에 Y형으로 피극이 없는 부분이 있었다. 끝의 복면·양쪽에 네 쌍의 큰 감각유두((pedunculated papillae)가 배열하였다. 이상의 소견은 enathostoma spinigerum 성충 수컷의 형태와 일치하였다. 따라서 이 증례는 태국에서 감염되어 국내로 유입된 수막뇌염이 유발된 유극악구충 증례로서, 국내에서는 처음으로 기록되는 것이라 하겠다.

흰쥐소장 점막의 허혈재관류손상에서 프로스타글란딘 E1이 세포자멸사에 미치는 영향 (The Effect of Prostaglandin E1 on Apoptosis Induced by Ischemia Reperfusion Injury in Rat Intestinal Mucosa)

  • Apoptosis is a physiologic or programmed cell death process which is controlled by genes. It is essential for the function and the appropriate development of multicellular organism. It is also thought to be one of the main mechanisms of cell death in ischemic tissues. The effect of prostaglandin $E_1$($PGE_1$) is proven to be useful in the recovery of ischemic changes by inducing vasodilation of peripheral vessels and platelet disaggregation. $PGE_1$ is also known to suppress apoptosis in human liver sinusoidal endothelial cell from ischemia-reperfusion injury. The purpose of this study is to evaluate the effects of $PGE_1$ on the apoptosis in the ischemia reperfusion injury of rat intestine. Thirty Sprague-Dawley rats were used. In control group(N=15), superior mesenteric artery was occluded for 60 minutes and after removing the vessel clamp, it was reperfused for 60 minutes and harvested. In experimental group(N=15), a jejunal flap was also made as in the control group except for the intraarterial administration of the $PGE_1$ right after clamping the artery and removing the clamp. H&E, TUNEL and immunohistochemical stains for p53, bax, and bcl-2 were performed. There were ischemic changes in gross and microscopic findings in both groups. The apoptotic index was significantly lower in the experimental group($1.29{\pm}0.82$(p=0.003)) than in the control group ($2.33{\pm}0.95$). The rat intestinal ischemia apoptosis by ischemia-reperfusion was partly related to the modulating of bcl-2, bax, and p53 expression. Our results indicate that $PGE_1$ suppresses the apoptosis in the ischemic jejunal flap and this effect is probably the result of a increase in expression of bcl-2.

"황제내경영추(黃帝內經靈樞)"에 기재된 삼초(三焦)에 관한 소고(小考) (A Study on San Jiao(三焦) stated on "Ling Shu(靈樞)")

  • According to the result about 'San Jiao(三焦)'recorded on "Yellow Empero's Canon Internal Medicine Ling Shu(黃帝內經 靈樞)", we achieved following results. 1. As we consider the concept of 'San Jiao' recorded on "Ling Shu", in early time, it was related to bladder(膀胱) and there was no divided concept into 'Shang Jiao(上焦)', 'Zhong Jiao(中焦)' and 'Xia Jiao(下焦)'. Later, there was appearance of body metabolism concept in anatomic way and started to emphasize that stomach(胃) is the very beginning of digestion. This point then adjusted into the concept which the core theory of digestion and water metabolism begins with 'Zhong Jiao' and processes to 'Shang Jiao' and 'Xia Jiao' then it established the 'San Jiao' theory adjusted to the functional change than property change. Later as there is set theoretical structure of 'three Yin and three Yang(三陰三陽)', it included the concept of 'San Jiao' onto meridian system theory to complete as a theory. Finally, it completed the theoretical structure that 'San Jiao' runs water metabolism of circulation, body fluid and urine for body to produce blood and Qi to protect and provide nutrition to the human body. 2. From the point of each part, 'San Jiao' means all body composition factor related to the relation to the digestion and water metabolism to produce Qi and blood. Also, the details of entire function of 'San Jiao' tells that 'Zhong Jiao' intakes food and divides the clarity and turbidity of digested substances. The origin of this digestion and water metabolism lies at Xia Jiao. The clean substance including the mood and taste climbs via Shang Jiao. The vapor like substance climbed to Shang Jiao becomes 'defensive Qi(衛氣)' and controls body temperature and sweat by supporting and spreading the Qi by Shang Jiao. The liquid substance climbed to Shang Jiao becomes blood. The blood has stronger character as substance than defensive Qi so Zhong Jiao becomes the base and the way for the blood. The turbid Qi separated at Zhong Jiao passes large intestine and the solid substance is excreted and the liquid is absorbed into bladder. The Xia Jiao that controls this process controls the liquid state of water matabolism so control the urine with bladder. Therefore, 'San Jiao' can be understood as a general concept that controls entire water metabolism as a way of food, Qi and blood.

임신기간 중 철 섭취가 모체와 제대 혈청의 미량 무기질 농도에 미치는 영향 (Iron Intake During Pregnancy on Serum Concentrations of Trace Minerals in Mothers and Neonates)

  • It is a common clinical practice to recommend taking iron supplements for pregnant women during gestation. Although it is required to ensure adequate iron stores during pregnancy, there has been some debate over the interference effects of excessive iron load, because it is possible to compete in the transport in the intestine and placenta and in binding to serum proteins of other trace minerals. In this study, maternal and neonatal serum concentrations of Fe, Zn, Cu, Se, Cr, Mn, and Co were assessed along with maternal Fe intakes. A total of 124 pregnant women and their term neonates participated voluntarily in this research. The women were divided into one of the three groups {high Fe intake (HFI), median Fe intake (MFI), and low Fe intake (LFI)} by their total Fe intakes and one of the two groups (Anemic and Normal) by their Fe nutritional status. All the data were compared among the three groups and between the two groups also. Total Fe intakes of HFI, MFI, and LFI groups were 140.8 ${\pm}$ 76.1, 68.0 ${\pm}$ 11.2, and 30.2 ${\pm}$ 8.6 mg/day, respectively. Those of Anemic and Normal groups were 90.1 ${\pm}$ 74.8 and 86.6 ${\pm}$ 46.8 mg/day, respectively. Maternal Hb concentration and Hct were not significantly different among HFI, MFI, and LFI groups but those were significantly different between Anemic and Normal groups. However, neonatal Hb concentration was not significantly different among HFI, MFI, and LFI groups and between Anemic and Normal groups either. Maternal serum Fe concentrations of the three groups, HFI, MFI, and LFI, were similar but that of Anemic group was significantly lower compared to Normal group. However, there was no significant difference in neonatal serum Fe concentrations among the three groups and between the two groups either. Serum concentrations of the other trace minerals in both mothers and neonates were not significantly different among HFI, MFI, and LFI groups and between Anemic and Normal groups. In addition, in the maternal serum, Fe concentration was positively correlated to Zn and Se concentration, respectively. As for the neonatal serum, Fe concentration showed a positive correlation to Zn, Cu, Mn, Se, and Co concentration, respectively. No trace mineral concentration was found to correlate negatively to Fe concentration in both maternal and neonatal serum, The results in this study indicate that Fe intakes of pregnant women, even if it is considerably above the level of estimated average requirement (EAR), may not affect serum Fe concentration in both mothers and neonates. In addition it might not influence adversely on the availability of other trace minerals including Zn and Cu in both mothers and neonates.
