시상은 신경 기능 조절에 중요한 역할을 하는 것으로 알려져 있다. 뇌의 중앙에 위치한 시상은 수면, 각성, 감정 조절에 관여하며, 다발성 경화증, 본태성 떨림, 파킨슨병과 같은 신경퇴행성 질환과 관련이 있는 것으로 보고되고 있다. 또한, 시상의 철 침전물이 나이가 들면서 우울 증상을 유발할 수 있다는 보고가 있다. 연구 간에 차이가 있지만, 시상과 감정 조절, 처리 등의 신경 기능이 밀접한 관계가 있어 시상 영역이 신경 장애에 분명한 영향을 미친다고 추론할 수 있다. Tractography 분석을 통해 각 피질하 영역의 세부 영역 간의 연결성을 매트릭스 형태로 조사하여 강한 연결성과 약한 반구간 연결성을 보였다. 60세 이상 그룹에서 시상의 WM 연결성이 두 그룹보다 약한 것으로 나타났다. 두 그룹을 비교한 결과 젊은 그룹(10-39세 및 40-59세)가 60세 이상 그룹보다 연결 강도가 높았고 각 반구에서 3개의 연결 경로에서 통계적으로 유의한 차이가 발견되었다. 노화로 인한 시상 관련 연결 강도의 감소는 불안 및 우울증과 같은 정서적 및 신경학적 장애에 영향을 미칠 수 있으며 네트워크 측정은 임상 조건 전반에 걸쳐 인지 장애를 평가하는 데 도움이 될 수 있음을 보여주었다.
자기공명영상(MRI)은 뇌의 구조적 및 기능적 연구에서 핵심 기술로 필요성이 증가하고 있다. Tractography 분석을 이용하는 뇌지도(Connectome)는 MRI를 통해 뇌의 구조적 연결성을 확인하고 연결성의 변동성을 이용해 질병 병리학에 대한 이해를 높이는 방법으로 인간을 대상으로 활발한 연구가 진행되고 있다. 하지만 마우스 같은 작은 동물의 경우 분석 방법의 표준화가 부족하고 영상에 대한 정확한 전처리 전략 및 아틀라스 기반 신경 정보학에 대한 과학적 합의가 없다. 또한, 인간의 뇌에 비해 마우스의 뇌는 매우 작기 때문에 높은 해상도를 갖는 영상을 획득하는 것에도 어려움이 있다. 연구에서는 구조적 영상과 확산 텐서 영상을 이용해 구조 영역 세분화를 포함한 구조적 연결성 분석을 가능하게 하고 마우스 뇌 데이터를 처리하는 Allen Mouse Brain Atlas 기반 영상 데이터 분석 파이프라인을 제시한다. 각 분석 방법은 마우스 뇌 영상 데이터의 분석을 가능하게 하고 이미 인간 영상데이터로 검증된 소프트웨어를 이용해 신뢰성을 가질 수 있게 하였다. 또한, 연구에서 제시되는 파이프라인은 복잡한 분석 과정과 다양한 기능들 중 마우스 Tractography에 필요한 기능들을 정리하여 사용자가 효율적으로 데이터 처리를 하는데 최적화되었다.
목적 : 개인의 성장과 사회적 활동에 영향을 미치는 국내 가족기능 요소를 개발하기 위해, 가족기능 평가도구인 McMaster 모델을 기반으로 국내 문화와 적합한 내용을 도출하여 가족기능에 영향을 주는 요소들을 델파이 기법을 통해 확인하고자 하였다. 연구방법 : 가족기능과 관련이 깊은 분야의 전문가 패널 12명을 대상으로 델파이 기법을 실시하였다. 기간은 2020년 5월부터 6월까지 총 7주간 진행되었으며, 델파이 조사는 2회 실시하였다. 1차 조사에서는 McMaster 모델의 국내 문화와 적합한 내용을 선정 및 한글화를 진행한 후, 구성요소에 관한 폐쇄형 항목 및 개방형 항목을 사용하여 전문가들의 의견을 수집하였다. 2차 조사에서는 구성요소의 적합도 및 중요도를 조사하였다. 결과 : 1차 델파이 조사 결과, 제시된 53개의 항목에서 18개의 항목이 삭제되었고, 11개의 항목이 추가되었으며, 의미가 중복되는 항목을 제외하여 40개의 항목이 선정되었다. 이 후 이해가 어려운 문장을 친숙한 어휘로 수정하고 2차 조사지를 구성하였다. 2차 델파이 결과에서는 총 33개의 항목이 선정되었으며, 최종 선정된 구성요소에 대한 내용타당도 비율은 0.76, 안정도는 0.28로 도출되었다. 결론 : 본 연구를 통해 도출된 국내 가족기능 평가를 위한 영향요소들은 가족기능을 포함하고 임상현장 또는 관련연구에서 가족기능 평가 및 중재를 시행하는데 활용되기를 기대한다.
Background: The recent COVID-19 pandemic is one of the worst disease outbreaks of the 21th century. Due to a lack of reliable antiviral therapeutics, wearing face masks is recommended to prevent airborne infection originating from virus-contaminated bioaerosols. Objectives: The aim of this study was to evaluate the filtration efficiencies of face masks that are commercially available in South Korea for a biological aerosol of Staphylococcus aureus (S. aureus) and murine coronavirus, a well-known surrogate for human coronaviruses. Methods: We collected six different kinds of commercial masks: two Korea Filter (KF)94 (KF94-1, KF94-2) masks, one surgical (Surgical-1) mask, one anti-droplet (KF-AD-1) mask, and two dust (Dust-1, Dust-2) face masks. S. aureus (ATCC 6538), a well-performing test bacteria and murine coronavirus (ATCC VR-764) were prepared under a suitable culture condition. Then, a mask biological filtration tester was used to examine the microbial filtration efficiencies of masks. Test microorganisms were quantitatively measured via cultivation methods and microbial filtration efficiencies were calculated appropriately. Results: All face masks showed over 99.6% filtration efficiency for S. aureus or murine coronavirus. There were no significant differences among the bacterial filtration efficiencies of the face masks. KF94-1 (99.97±0.08%) and Dust-1 mask (99.97±0.07%) showed the highest (over 99.9%) filtration efficiency for murine coronavirus. KF94-1 or Dust-1 masks showed a significant virus filtration efficiency compared to Surgical-1 mask (p<0.05; Mann-Whitney U test). Conclusions: All the commercially available face masks used in this study can filter S. aureus or murine coronavirus in bioaerosols efficiently, regardless of the mask type. Therefore, our results suggest that wearing a certified face mask is a reliable means to prevent the transmission of infectious airborne diseases via biological aerosols.
Background: The coronavirus disease (COVID-19) has caused changes in human activity, and these changes may possibly increase or decrease exposure to fine dust (PM2.5). Therefore, it is necessary to evaluate the exposure to PM2.5 in relation to the outbreak of COVID-19. Objectives: The purpose of this study was to compare and evaluate the exposure to PM2.5 concentrations by the variation of dynamic populations before and after the outbreak of COVID-19. Methods: This study evaluated exposure to PM2.5 concentrations by changes in the dynamic population distribution in Guro-gu, Seoul, before and after the outbreak of COVID-19 between Jan and Feb, 2020. Gurogu was divided into 2,204 scale standard grids of 100 m×100 m. Hourly PM2.5 concentrations were modeled by the inverse distance weight method using 24 sensor-based air monitoring instruments. Hourly dynamic population distribution was evaluated according to gender and age using mobile phone network data and time-activity patterns. Results: Compared to before, the population exposure to PM2.5 decreased after the outbreak of COVID-19. The concentration of PM2.5 after the outbreak of COVID-19 decreased by about 41% on average. The variation of dynamic population before and after the outbreak of COVID-19 decreased by about 18% on average. Conclusions: Comparing before and after the outbreak of COVID-19, the population exposures to PM2.5 decreased by about 40%. This can be explained to suggest that changes in people's activity patterns due to the outbreak of COVID-19 resulted in a decrease in exposure to PM2.5.
This study aimed to investigate the effects and potential mechanisms of Chikusetsusaponin V (CsV) on endothelial nitric oxide synthase (eNOS) and vascular endothelial cell functions. Different concentrations of CsV were added to animal models, bovine aorta endothelial cells (BAECs) and human umbilical vein endothelial cells (HUVECs) cultured in vitro. qPCR, Western blotting (WB), and B ultrasound were performed to explore the effects of CsV on mouse endothelial cell functions, vascular stiffness and cellular eNOS mRNA, protein expression and NO release. Bioinformatics analysis, network pharmacology, molecular docking and protein mass spectrometry analysis were conducted to jointly predict the upstream transcription factors of eNOS. Furthermore, pulldown and ChIP and dual luciferase assays were employed for subsequent verification. At the presence or absence of CsV stimulation, either overexpression or knockdown of purine rich element binding protein A (PURA) was conducted, and PCR assay was employed to detect PURA and eNOS mRNA expressions, Western blot was used to detect PURA and eNOS protein expressions, cell NO release and serum NO levels. Tube formation experiment was conducted to detect the tube forming capability of HUVECs cells. The animal vasodilation function test detected the vasodilation functions. Ultrasonic detection was performed to determine the mouse aortic arch pulse wave velocity to identify aortic stiffness. CsV stimulus on bovine aortic cells revealed that CsV could upregulate eNOS protein levels in vascular endothelial cells in a concentration and time dependent manner. The expression levels of eNOS mRNA and phosphorylation sites Ser1177, Ser633 and Thr495 increased significantly after CsV stimulation. Meanwhile, CsV could also enhance the tube forming capability of HUVECs cells. Following the mice were gavaged using CsV, the eNOS protein level of mouse aortic endothelial cells was upregulated in a concentration- and time-dependent manner, and serum NO release and vasodilation ability were simultaneously elevated whereas arterial stiffness was alleviated. The pulldown, ChIP and dual luciferase assays demonstrated that PURA could bind to the eNOS promoter and facilitate the transcription of eNOS. Under the conditions of presence or absence of CsV stimulation, overexpression or knockdown of PURA indicated that the effect of CsV on vascular endothelial function and eNOS was weakened following PURA gene silence, whereas overexpression of PURA gene could enhance the effect of CsV upregulating eNOS expression. CsV could promote NO release from endothelial cells by upregulating the expression of PURA/eNOS pathway, improve endothelial cell functions, enhance vasodilation capability, and alleviate vessel stiffness. The present study plays a role in offering a theoretical basis for the development and application of CsV in vascular function improvement, and it also provides a more comprehensive understanding of the pharmacodynamics of CsV.
지하수의 비소(As) 오염은 방글라데시의 주요 문제 중 하나이다. 독성은 부종, 피부암, 방광암, 폐암, 각화 과다증, 조산, 흑사병과 같은 심각한 건강 문제를 야기한다. 지하수의 오염은 주로 인위적 활동의 영향을 받는 지역의 지질학적 특성에서 발생된다. 방글라데시 국민의 대부분이 지하수에 식수를 의존하고 있기에 비소 오염 현황을 조사하고 물이 부족한 지역에 안전한 식수를 제공하기 위하여 비소 처리를 위한 수처리 기술이 필요하다. 따라서 본 연구는 92개의 논문을 기반으로 방글라데시의 지하수 As 오염 관련 최근 현황과 다양한 저비용 정화 기술에 대한 트렌드를 조사하였다. 방글라데시에서 지하수 비소 오염이 가장 높은 지역은 Brahamanbariya, Tangail, Barisal, Pabna, Patuakhali 및 Kurigram등 총 4곳이며, Magura 및 Faridpur 등의 지역에서는 비소 농도가 0.05mg/L를 초과한다. WHO 표준 가이드라인 값(<0.01 mg/L)을 만족하는 지역은 Kushtia, Khagrachari, Jessore, Dinajpur, Meherpur 및 Munshiganj 등 총 6곳으로 나타났다. 수중에서 As를 처리하는 기술이 다양하다. 시간-비용효율적인 처리 방법은 Mg-Fe계 Hydrotalcite와 유사 화합물, electro-chemical As remediation (ECAR) reactor, aerated electrocoagulation 등이 적용되고 있다. 전반적으로 예산, 운영 및 유지 관리 비용, 재료의 가용성 및 전문 지식 요구 사항 등을 고려하여 수질 내 비소를 처리해야 한다.
본 논문에서는 자체 제작한 움직임 팬텀을 이용하여 다양한 호흡모사를 하고, PET/CT 장비 내의 데이터기 기반 보정프로그램을 적용하여 영상을 획득하고, 영상의 SUVmax 및 병변 부피를 분석하여 호흡에 따른 장기 또는 종양의 움직임으로 인한 인공물의 감소효과를 확인하였다. 이동거리 3 cm에서 데이터 기반 게이팅을 적용한 영상이 데이터 기반 게이팅을 적용하지 않은 영상보다 SUVmax가 팬텀 구의 지름이 10 mm, 13 mm, 17 mm, 22 mm, 28 mm, 37 mm에서 각각 2.37, 2.02, 1.44, 1.20, 0.42, 0.52 향상시키는 것으로 나타났고, 향상률로 표시하면 각각 72.5%, 73.3%, 51.3%, 25.8%, 8.6%, 7.2%이었다. 크기가 작은 병소의 SUVmax의 개선효과가 크게 나타났다. 데이터 기반 게이팅을 적용하지 않았을 때에 비하여 적용하였을 때에 1 cm, 2 cm, 3 cm에서 각각 5%, 12%, 18%의 영상의 면적이 감소하는 개선효과가 있었고, 그리고 움직임에 의한 인공물이 클 때 감소 효과도 커졌다. 데이터 기반 게이팅을 적용하면 검사절차가 간소화되고, 사술자의 피폭선량 감소효과와 더불어 호흡으로 인한 인공물을 감소시켜 영상의 질을 개선할 수 있음을 확인하였다.
일반 X-선 검사는 간단하고 많은 정보를 얻을 수 있어 가장 기본이 되는 검사방법이며 획득된 영상에서는 인체 해부학적 구조와 시간 경과에 따른 질병의 변화 정보를 쉽게 획득할 수 있다. 그럼에도 불구하고 영상이 확대되는 단점으로 인하여 병변의 크기와 형태가 왜곡되어 나타나기 때문에 현재 X-선 영상의 깊이 있는 관찰은 이루어지지 않고 있다. 본 연구에서는 후전촬영(PA)과 측방향촬영(LAT) 영상을 획득하고 각각의 영상에서 암 시료가 위치하는 거리를 측정하여 암 시료의 확대율을 계산하고 측정한 암 시료 길이에 보정한다. 암 시료의 길이와 두께에 따른 확대율 보정 값을 전산화단층촬영장치(Computed Tomography, CT)로 획득된 영상 및 실제 제작한 암 시료 크기와 각각 비교하였다. 기존의 확대율은 검출기에서 암의 거리를 측정하여 계산할 수 있었으나 본 연구에서는 획득된 PA와 LAT 영상을 이용하여 확대율을 계산하였다. 6 mm 암 시료를 PA와 LAT 영상을 획득하여 확대율을 구한 후 보정한 결과 길이는 5.9 mm, 두께는 6.1mm로 실제와 비슷한 값이 측정되었으며 영상을 이용한 확대율 계산이 가능하다는 것을 알 수 있었다. X-선 영상만으로도 병변의 확대율을 손쉽게 보정하여 정확한 길이 측정이 가능하고 이는 영상 판독 및 정확한 진단에 유용한 정보를 제공할 것이다.
Pulmonary arterial hypertension (PAH) is a progressive and devastating disease whose pathogenesis is associated with a phenotypic switch of pulmonary arterial vascular smooth muscle cells (PASMCs). Bone morphogenetic protein (BMP) signaling and potassium two pore domain channel subfamily K member 3 (KCNK3) play crucial roles in PAH pathogenesis. However, the relationship between BMP signaling and KCNK3 expression in the PASMC phenotypic switching process has not been studied. In this study, we explored the effect of BMPs on KCNK3 expression and the role of KCNK3 in the BMP-mediated PASMC phenotypic switch. Expression levels of BMP receptor 2 (BMPR2) and KCNK3 were downregulated in PASMCs of rats with PAH compared to those in normal controls, implying a possible association between BMP/BMPR2 signaling and KCNK3 expression in the pulmonary vasculature. Treatment with BMP2, BMP4, and BMP7 significantly increased KCNK3 expression in primary human PASMCs (HPASMCs). BMPR2 knockdown and treatment with Smad1/5 signaling inhibitor substantially abrogated the BMP-induced increase in KCNK3 expression, suggesting that KCNK3 expression in HPASMCs is regulated by the canonical BMP-BMPR2-Smad1/5 signaling pathway. Furthermore, KCNK3 knockdown and treatment with a KCNK3 channel blocker completely blocked BMP-mediated anti-proliferation and expression of contractile marker genes in HPAMSCs, suggesting that the expression and functional activity of KCNK3 are required for BMP-mediated acquisition of the quiescent PASMC phenotype. Overall, our findings show a crosstalk between BMP signaling and KCNK3 in regulating the PASMC phenotype, wherein BMPs upregulate KCNK3 expression and KCNK3 then mediates BMP-induced phenotypic switching of PASMCs. Our results indicate that the dysfunction and/or downregulation of BMPR2 and KCNK3 observed in PAH work together to induce aberrant changes in the PASMC phenotype, providing insights into the complex molecular pathogenesis of PAH.

제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
