Antioxidative and anti-inflammatory activities of Petasites japonicus extract were evaluated. P. japonicus extract showed 70.1% inhibition on peroxidation of linoleic acid. In the experiment using the cell permeable dye, 2',7'- dichlorofluorescein diacetate (DCFDA) as an indicator of reactive oxygen species (ROS) generation, intracellular oxidative stress in UVB-irradiated keratinocytes was shown to be decreased by P. japonicus extract. Also, UVB-induced production of interleukin-$1{\alpha}$ and prostaglandin $E_2$ in human HaCaT keratinocytes was reduced in a dose-dependent manner by treatment with P. japonicus extract. All these results suggest that P. japonicus extract can be effectively used for prevention of UV-induced adverse skin reactions such as radical production and inflammation.

Acne vulgaris is a common chronic skin disorder that affects millions of people. The pathogenesis of acne has been known to be closely associated with the bacterium Propionibacterium acnes. Here we investigated the anti-acne activity of Lactobacillus plantarum K-1 BR by observing the expressions of proinflammatory cytokines, TNF-α, IFN-γ and IL-8, of human keratinocytes. When we applied heat-killed P. acnes to HaCaT cells, the inflammatory cytokines were induced by two- to four-fold compared to the normal control. When the bacteriocin, purified from L. plantarum K-1 BR, was pretreated to the HaCaT cells, the expression levels of TNF-α and IFN-γ stimulated by P. acnes significantly decreased to 25% and 30% of the induced levels, respectively. The IL-8 levels also significantly decreased with the concentration dependent manner of the bacteriocin. These results suggest that the bacteriocin from L. plantarum K-1 BR could reduce the expression levels of inflammatory cytokines and thus may relieve inflammations caused by acne.

Hydrogen peroxide (H2O2) is a type of active oxygen species (ROS) that causes oxidative stress in cells and affects cell growth, proliferation, senescence, and death. The purpose of this study is to find active peptides that attenuate cytotoxicity of H2O2. A positional scanning synthetic tetrapeptide combinatorial library was screened to predict the sequence of potentially active peptides. As a result of comparing the effect of peptide pools on H2O2-induced death of human keratinocytes (HaCaT cells), various active peptide sequences were predicted. Especially, peptides containing cysteine (C) residue were predicted to be active. In follow-up experiments, the cytotoxicity and activity of cysteine-containing peptides of different lengths, such as C-NH2, CC-NH2, CCC-NH2, and CCCC-NH2 were examined. C-NH2 and CC-NH2 showed no significant cytotoxicity up to 1.0 mM, but CCC-NH2, and CCCC-NH2 showed relatively strong cytotoxicity. C-NH2 and CC-NH2 alleviated H2O2-induced cytotoxicity. CC-NH2 was more cytoprotective compared to C-NH2, C, N-acetyl cysteine (NAC), and glutathione (GSH). When intracellular ROS was measured by flow cytometry, H2O2 increased ROS production, and CC-NH2 suppressed ROS production more effectively than C-NH2, and it was as effective as C, NAC, and GSH. This study suggests that CC-NH2 of the cysteine-containing peptides of different lengths has an antioxidant property that safely and effectively alleviates H2O2-induced cytotoxicity and ROS production.
The skin plays a key role in protecting the body from the environment and from water loss. Cornified envelope (CE) and natural moisturizing factor (NMF) are considered as the primary regulators of skin hydration and barrier function. The CE prevents loss of water from the body and is formed by cross-linking of several proteins. Among these proteins, filaggrin is an important protein because NMF is produced by the degradation of filaggrin. Proteases, including matriptase and prostasin, stimulate the generation of filaggrin from profilaggrin and caspase-14 plays a role in the degradation of filaggrin. This study elucidated the effects of an ethanol extract of Boesenbergia pandurata (Roxb.) Schltr., known as fingerroot, and its active compound panduratin A on CE formation and filaggrin processing in HaCaT, human epidermal keratinocytes. B. pandurata extract (BPE) and panduratin A significantly stimulated not only CE formation but also the expression of CE proteins, such as loricrin, involucrin, and transglutaminase, which were associated with $PPAR{\alpha}$ expression. The mRNA and protein levels of filaggrin and filaggrin-related enzymes, such as matriptase, prostasin, and caspase-14 were also up-regulated by BPE and panduratin A treatment. These results suggest that BPE and panduratin A are potential nutraceuticals which can enhance skin hydration and barrier function based on their CE formation and filaggrin processing.

The matrix metalloproteinase (MMP) family is involved in the breakdown of the extracellular matrix during normal physiological processes such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes such as pathological aging, arthritis, and metastasis. Oxidative conditions generate reactive oxygen species (ROS) (e.g., hydrogen peroxide [$H_2O_2$]) in cells, which subsequently induce the synthesis of matrix metalloproteinase-1 (MMP-1). MMP-1, an interstitial collagenase, in turn stimulates an aging phenomenon. In this study, baicalein (5,6,7-trihydroxyfl avone) was investigated for its in vitro activity against $H_2O_2$-induced damage using a human skin keratinocyte model. Baicalein pretreatment signifi cantly inhibited $H_2O_2$-induced up-regulation of MMP-1 mRNA, MMP-1 protein expression and MMP-1 activity in cultured HaCaT keratinocytes. In addition, baicalein decreased the transcriptional activity of activator protein-1 (AP-1) and the expression of c-Fos and c-Jun, both components of the heterodimeric AP-1 transcription factor. Furthermore, baicalein reduced phosphorylation of extracellular signal-regulated kinase (ERK) and c-Jun-N-terminal kinase (JNK), which are upstream of the AP-1 transcription factor. The results of this study suggest that baicalein is involved in the inhibition of oxidative stress-induced expression of MMP-1 via inactivation of the ERK/JNK/AP-1 signaling pathway.
Chronological aging and photoaging affect appearance, causing wrinkles, pigmentation, texture changes, and loss of elasticity in the skin. Phragmites communis is a tall perennial herb used for its high nutritional value and for medicinal purposes, such as relief from fever and vomiting and facilitation of diuresis. In this study, we investigated the effects of ethanol extract of P. communis rhizome (PCE) on skin aging. The total flavonoid and total phenolic content in PCE were 2.92 ± 0.007 ㎍ of quercetin equivalents (QE) and 231.8 ± 0.001 ㎍ of gallic acid equivalents (GAE) per 100 mg of dried extract (n = 3). The half-maximal inhibitory concentration (IC50) values of PCE for 2,2-diphenyl-1-picrylhydrazyl radical (DPPH) and hydrogen peroxide scavenging activities were 0.96 and 0.97 mg/mL, respectively. PCE showed inhibitory effects on tyrosinase when L-tyrosine (IC50 = 1.25 mg/mL) and L-3,4-dihydroxyphenylalanine (IC50 = 0.92 mg/mL) were used as substrates. PCE treatment up to 200 ㎍/mL for 24 h did not cause any significant cytotoxicity in B16F10 melanocytes, human dermal fibroblasts (HDFs), and HaCaT keratinocytes. In B16F10 melanocytes, PCE (25 and 50 ㎍ /mL) inhibited melanin production and cellular tyrosinase activity after challenge with α-melanocyte-stimulating hormone (α-MSH; p < 0.05). In HDFs, PCE suppressed the mRNA expression of matrix metalloproteinase-1 (MMP-1) and reduced the activity of elastase (p < 0.05). In addition, ultraviolet B (UVB)-mediated downregulation of hyaluronic acid synthase-2 gene expression in HaCaT keratinocytes was also effectively suppressed by PCE treatment. Overall, our results showed that PCE has potential anti-skin aging activity associated with the suppression of hyperpigmentation, wrinkle formation, and reduction in dryness. PCE is a promising candidate for the development of an anti-skin aging cosmetic ingredient.

Yuza (Citrus junos Sieb ex TANAKA) is a citrus fruit that is cultivated in northeast Asia. Citron is known for containing abundant antioxidants such as vitamin C, flavonoids, for example hesperidin and hesperetin, and terpenoids such as limononin. When mature citron is processed for tea or other beverage food products in Korea, massive amounts of seeds and pericarp are remained as waste. This study aimed to exploit the processed remnant of Citron for developing functional cosmetic applications. Ethanol extracts of Yuza seed and pericarp did not show significant radical scavenging activities measured by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. But they contained significantly high phenolic compounds. Cultured human dermal fibroblasts and HaCaT keratinocytes were irradiated with 25 mJ UVB and the citron extracts were added to the medium of each culture. Cellular damages caused by UVB irradiation were prevented by the addition of the Yuza extract. In addition, the reduction of the enhanced MMP-1 expression after irradiation of UVB in human dermal fibroblasts was observed. Also the increased level of pro-inflammtory TNF-$\alpha$ in the UVB irradiated HaCaT cells was decreased. The collagen expression was enhanced by the extract. Yuza extract markedly inhibited melanin production from $\alpha$-MSH treated B16F1 melanoma cells. Melanin assay, tyrosinase zymography results indicated that Yuza extract had strong depigmenting activity. In conclusion, Yuza ethanol extracts have good anti-photoaging and strong anti-melanogenic efficacies.
In this study, to develop a new anti-aging agent, we will examine the antioxidant activity and the inhibitory effects of the mosidae for the MMP-1 activities in UVB-irradiated HS68 human foreskin fibroblast cell lines and human keratinocytes (Ha- CaT cell line). And the consomme soup being prepared with mosidae according to different levels (3, 6, 9, 12%) and then quality characteristics will be determined by sensory evaluations and color values. It is also observed that the mosidae has the inhibition of UVB-induced MMP-1 activities. The consomme soup prepared with mosidae is found to reduce the level of $IC_{50}$ in SOD-like activities and hydroxy radical scavenging activities, respectively. And consomme soup prepared with mosidae (CS6) is the best by sensory evaluations. In conclusion, these results suggest that the consomme soup prepared with mosidae can be further developed as new anti-aging food.

Nuclear-kappa B(NF-$\kappa$B) plays a role in the regulation of genes responsible for inflammatory and immune responses as well as growth control of cells. A cell-based assay system for guiding NF-KB activity was developed to determine the influence of activated NF-KB in human keratinocytes. It suggested that this system could be used to determine the quantitative measurement of NF-$\kappa$B activity in the human skin and allow the monitoring of anti-inflammatory agent for dermatological means from various environmental stimuli. Objective : UVB damages skin health by causing skin redness and intense inflammation, sunburn, and skin cancer. Paeoniae Radix Alba has been used to relieve gynecological symptoms, muscle spasms, and skin ailments. This study was conducted to confirm whether it has a protective effect against UVB photodamage. Methods : Ethanol extract of Paeoniae Radix Alba (PRA) was prepared by extracting 100 g Paeoniae Radix Alba in 1 L of ethanol for 48 h. Apoptosis was monitored by the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay and expression levels of apoptosis indicator proteins, and tyrosinase activity was measured with a colorimetric commercial kit. Results : In human keratinocyte HaCaT cells, PRA reduced UVB-induced cell death through apoptosis by inhibiting PARP cleavage and caspase-3 and -9. UVB-induced increase in cellular reactive oxygen species (ROS) was suppressed by PRA pretreatment. PRA also showed dose-dependent ABTS and DPPH radical scavenging activities. Furthermore, the inhibitory effect of tyrosinase activity by PRA was confirmed. Conclusion : These results demonstrated the protective role of PRA in UVB photodamage of human keratinocytes, mainly due to its antioxidant and antiapoptotic properties. We also suggest that PRA can be considered as an effective natural agent to prevent skin photodamage.
