Screening of the Physiological Activity of Solvent Extracts of Paulownia coreana Bark and Antioxidative Effect of the Extracts on an Edible Oil

  • Paulownia coreana is a medicinal, edible and industrial plant with the largest leaf, and is native to Korea. We evaluated the reducing power activities, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities (RSAs), nitrite scavenging activities (NSAs) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation RSAs by solvent extraction of P. coreana bark by using $50^{\circ}C$ hot water and $25^{\circ}C$ methanol. The antioxidative effect of P. coreana bark extract on corn seed oil was evaluated using the Rancimat test. The extraction yields on dry weight basis with 15 folds of hot water and methanol were 23.88% and 5.30%, respectively; further the flavonoid content in the hot water extract was over 2.5 times more than that in the methanol extract. The DPPH RSA of the methanol extract was substantially higher than that of the water extract, whereas the NSA of the water extract was higher than that of the methanol extract at pH 1.2. The ABTS RSAs of the two extracts were almost the same as that of ascorbic acid and 2,6-ditertiarybutyl hydroxytoluene. The two extracts of P. coreana bark in this study were found to slightly improve the oxidation stability of corn seed oil.

추출방법별 식용 및 약용버섯류의 총 당함량 비교분석 (Total sugar contents of edible and medicinal mushrooms comparative analysis by the extraction method)

  • 이번 연구에서는 식용 및 약용버섯 자실체를 가지고 버섯의 주요 기능성 성분인 총 당의 함량을 추출방법을 달리하여 비교분석하였다. 열수추출, 산추출, 알칼리 추출을 통해 총 당 측정법으로 미량의 시료에 적합한 페놀-황산법으로 측정하였다. 표준 곡선은 glucose solution을 사용하여 작성하고 측정한 흡광도를 표준곡선에 적용하여 시료의 농도를 얻었다. 열수추출에서는 동충하초가 658.4(${\mu}g/g$)로 가장 높았고, 산추출과 알칼리 추출에서는 운지버섯이 각각 649.7(${\mu}g/g$)와 679.0(${\mu}g/g$)로 가장 높음을 알 수 있었다. 추출방법별로 열수추출과 마이크로웨이브 추출로 총 당 함량을 정량한 결과 열수 추출에서는 동충하초, ASI 7025가 높았고, 1 hr 마이크로웨이브 추출 및 2 hr 마이크로웨이브 추출에서는 운지버섯, 꽃송이가 높게 나타났다. 이러한 결과를 통해 각기 다른 추출용매와 추출방법에 따라서 추출되는 총 당 함량이 다름을 알 수 있었다.

In Vitro Evaluation of Antioxidant Activity of Lycium barbarum Hot Water Extract and Optimization of Production Using Response Surface Methodology

  • This study is concerned with the optimization of the manufacturing process of a hot water extract containing antioxidant activity from Lycium barbarum, traditionally known to have various physiological activities. For the establishment of the optimization process, the central composite design of response surface methodology(RSM) was used. Thirteen extraction processes were performed by encoding the independent variables, extraction temperature (65.9℃-94.1℃) and extraction time (2.59 hr-5.41 hr). As a result of the experiment, the optimal manufacturing conditions for the extract were 340.0 mg/100 g of GAE at an extraction temperature of 94.1℃ and an extraction time of 5 hr. The maximum yield of flavonoids was 22.44 mg/100 g of HES at an extraction temperature of 94.1℃ and an extraction time of 4 hr. The conditions for producing the extract with the maximum antioxidant capacity (DPPH 92.12%) were 90℃ and 4.5 hr extraction time. Therefore, the optimal manufacturing process conditions for extracts containing total phenol content, flavonoid content, and DPPH radical scavenging activity, which are dependent variables, were extraction temperature of 90-95℃ and extraction time of 4 hr, which were not significantly different from the actual values. Therefore, Lycium barbarum extract rich in total phenol and flavonoid content related to antioxidant function is expected to be used as a functional food and cosmetic material.

오존수로 세척한 상어연골로부터 Crude Chondroitin Sulfate의 열수추출 (Hot Water Extraction of Crude Chondroitin Sulfate from Shark Cartilage Treated with Ozone Water)

  • 상어연골에서 발생하는 갈변과 냄새를 제거하기 위한 오존수 세척효과와 crude chondroitin sulfate의 추출특성을 조사하기 위하여 알칼리 및 열수추출 효과를 조사하였다. SC 분말을 용존 오존농도 0.3 ppm의 오존수로 세척한 결과 일반성분의 함량에는 영향을 미치지 않으면서 $L^*$값은 증가하고 $a^*$, $b^*$값은 감소되어 탈색효과가 뚜렷하였다. 또한 ammonia 유사취가 크게 감소되었으며 종합적 기호도가 크게 향상되었다. NaOH에 의한 CS의 최적 추출농도는 1 N이었으며 최적 추출시간은 $100^{\circ}C$에서 120분이었고 이때의 수율은 10.02%이었다. $100^{\circ}C$의 열수로 7회 반복추출 할 경우 1 N NaOH 추출수율과 대등하였다.

  • 본 연구에서는 전통적인 열수추출법과 초음파 추출법을 이용하여 민들레에서 항산화성분을 추출하였다. 추출 후 추출수율과 항산화성분인 플라보노이드 함량을 최적화하기 위해 반응표면분석법 중 Box-Behnken 설계모델을 이용하여 추출하였다. 열수추출법의 경우 추출온도, 주정/초순수 부피비, 추출시간을 변수로 설정하였으며, 초음파 추출법의 경우 초음파 조사세기, 주정/초순수 부피비, 초음파 조사시간을 변수로 설정하였다. 열수추출법의 최적 추출조건은 추출온도($45.76^{\circ}C$), 주정/초순수 부피비(41.92 vol.%), 추출시간(1.75 h)이었다. 또한 초음파 추출법의 최적조건은 초음파 조사세기(512.63 W), 주정/초순수 부피비(56.97 vol.%), 추출시간(20.79 min)이었다. 이 조건에서 예상되는 반응치는 수율(22.09 wt.%), 플라보노이드 함량(28.98 mg QE/mL dw)으로 산출되었다. 또한 실제실험 결과 오차는 3% 이하로 나타나 최적화의 타당성을 입증하였다.

반응표면분석법에 의한 동충하초 열수추출공정의 최적화 (Optimization for Hot Water Extraction Process of Cordyceps militaris using Response Surface Methodology)

  • 동충하초를 음료로 개발하기 위한 전단계로 열수추출공정을 반응표면분석법으로 최적화하였다. 추출시간에 따라서는 큰 변화가 없어 추출온도와 농도를 요인변수로 하여 실험을 수행하였다. 열수추출액의 당도(brix)와 고형분 함량은 농도에 따라서 큰 영향을 받는 것으로 나타났다. 추출액의 탁도와 총당, 환원당 및 단백질 함량은 낮은 농도에서 추출수율이 좋은 것으로 나타났다. 각 반응변수 모두 농도에 대하여는 유의성이 있는 것으로 나타났으며 수립된 이차 회귀식에 대한 적합성 결여분석 결과 수립된 반응표면 모형이 통계적으로 유의하였다. 동충하초 열수추출에 있어 고형분함량은 2.0~2.4%, brix 1.5~l.8, 총당 l14~120 $\mu\textrm{g}$/g, 환원당 10.5~11.0 $\mu\textrm{g}$/g 그리고 단백질함량 110~l15 $\mu\textrm{g}$/g인 영역을 만족하는 조건은 추출온도 95~10$0^{\circ}C$, 농도 4.0~4.1%로 결정할 수 있었다.

아가리쿠스 버섯에서 생리활성물질의 추출 및 정제 (Extraction and Purification of Bioactive Materials from Agaricus blazei Fruiting Bodies)

  • ${\beta}$-Glucan a kind of polysaccharide which is particularly abundant in Agaricus blazei is known as the bioactive materials especially anticancer agents. The process development of the isolation and the purification process of water soluble ${\beta}$-glucans from A. blazei was achieved. and the process operation variables were optimized. Crude polysaccharides (CR.PS) were obtained from A. blazei by hot water extraction filtration solvent precipitation dialysis and freeze drying. Neutral and acidic fraction of polysaccharides were separated from crude polysaccharides by ion chromatography and then high molecular weight and low molecular weight fraction were separated from neutral fraction by gel chromatography. Quantitative and qualitative analysis of each compounds were performed with FT-IR NMR spectroscopy. Based on these analysis the optimal conditions of temperatures operating time organic solvent volume for precipitation and dialysis time were determined.

목질(木質)의 열수추출(熱水抽出) 및 CaCl2 첨가(添加)가 목질(木質)-세멘트 보드의 휨강도(强度) 및 팽윤율(膨潤率)에 미치는 영향(影響) (The Effects of Hot Water Extraction of Wood Meal and the Addition of CaCl2 on Bending Strength and Swelling Ratio of Wood-Cement Board)

  • The effects of pre-treatments, the hot water extraction of wood meal and the addition of chemical ($CaCl_2$) to wood-cement water system on the properties of wood-cement composite such as modulus of rupture (MOR), modulus of elasticity (MOE), water sorption ratio and swelling ratio of resulting boards were studied in this experiment. The wood meals through 0.83mm(20 mesh) and retained on 0.42mm(35 mesh) screen were prepared from Pinus densiflora S. at Z. and Larix leptolepsis G. For hot water extraction, 500 grams of wood meal for each species were heated to boiling with 1,500ml of distilled water in 2-liter beaker for 6 hours. Every 2 hours, the wood meals were washed with boiling distil1ed water and reheated to boiling again. After 6 hours boiling, the boiled wood particles were collected by pouring this particles on 200 mesh screen. The collected particles then washed twice with hot distilled water and dried for 24 hours in an oven at $109{\pm}20^{\circ}C$. A mixture of 663.4 grams of cement with 331.7 grams of wood meal based on oven-dry weight were dry-mixed in a plastic vessel. The mixture was kneaded with 497.6ml of distilled water in the ratio of 1.5ml of water to a gram of wood meal. To add calcium chloride to the mixture as an accelerator, $CaCl_2$ 4% solution by weight per volume, was added to pine-or larch-cement board in the ratio of 3% to cement weight. To set wood-cement board, this mixture was clamped at 30cm ${\times}$ 30cm, in thickness of 1.5cm for 3 days at room temperature, declamped and then placed at open condition for 17 days. The target density was 1.0. The four specimens sized to 5cm in width and 28cm in length were used for MOR and MOE test for each treatment. After MOR test, the tested specimens were cut to the size of 5cm ${\times}$ 5cm for water sorption and swelling test. The twenty specimens used to measure the water sorption ratio (soaking 24 hours) and ten of these were used for swelling ratio measurement The results obtained were as follows: 1) Larch was not suitable for wood-cement boards because larch-cement board developed no strength, but pine showed 97.9kg/$cm^2$ by hot water extraction. 2) To increase MOR, hot water extraction was more effective than the addition of $CaCl_2$ in pine and larch because the $CaCl_2$ addition was seemed to speed up the ratio of cement hydration without reacting with the wood substances. 3) The water sorption ratio was lowered by the addition of $CaCl_2$ to wood-cement system because the chemical additive accelerated the rate of cement hydration. 4) In pine-cement board, the swelling ratio from 0.37 to 0.42 percent was observed in length and the swelling ratio from 0.88 to 2.0 percent in thickness. As a rule, the swelling ratio of wood-cement board was very low and the swelling ratio in thickness was higher than in length.

Taguchi 법에 의한 흑목이버섯의 항산화활성 증진을 위한 열수추출 최적화 (Optimization of Hot Water Extraction for Enhancing Antioxidative Activity from Auricularia auricula Using Taguchi Approach)

  • Optimization study of the hot water extraction for enhancing antioxidative activity from Auricularia auricula was performed by Taguchi approach using orthogonal matrix $L_9(3^4)$ method. The correlation between DPPH radical scavenging activity and the components of samples extracted from different extraction conditions were also analyzed. The correlation coefficient between DPPH radical scavenging activity and melanin content of A. auricula were 0.93, indicating 'good correlation'. The optimum extraction conditions was obtained at the extraction time of 1 hr. temperature of $85^{\circ}C$, solid: water ratio of 1: 40(w/v) and frequency of 2 times. Under these conditions, values of maximum DPPH free radical scavenging activity and melanin contents of A. auricula were $67.21{\pm}2.17$ and $52.94{\pm}2.10 mg/g$, respectively. Melanin content of 1.6 times and DPPH free radical scavenging effect of 130% were enhanced by optimization.

추출조건에 따른 블루베리 열수추출물의 항산화 활성 비교 (Antioxidant activities of blueberry hot water extracts with different extraction condition)

  • 본 연구에서는 추출방법을 달리하여 블루베리 열수추출물들간의 항산화 활성 차이를 비교하였다. Autoclave를 이용한 가압가열추출(AE), 오븐을 이용한 추출(OE), 끓인 증류수에 혼합 후 sonication한 추출(HWSE), 끓인 증류수에 산을 첨가한 후 sonication한 추출(HWASE) 및 15분간 끓여 추출(BE) 등의 방법으로 추출물을 제조하고, 총 폴리페놀, 플라보노이드 및 안토시아닌 함량을 확인하고 ABTS, alkyl 및 DPPH radical 소거능 실험과 $Fe^{2+}$ ion chelating 활성을 확인하였다. 총 폴리페놀 및 플라보노이드 함량 모두 AE에서 각각 $3.47{\pm}0.16mg\;GAE/g$$1.59{\pm}0.19mg\;RE/g$으로 가장 높은 값을 나타냈다. 한편 이와는 반대로 총 안토시아닌 함량은 AE가 가장 낮은 함량을 나타냈으며, HWSE(5.29 mg/g) 에서 가장 높은 함량을 나타냈다. ABTS와 alkyl radical 소거능 측정 결과, AE가 가장 우수한 소거능을 나타냈으며, DPPH radical 소거능 실험에서는 AE와 더불어 안토시아닌 함량이 가장 높은 HWSE이 우수한 활성을 나타냈다. $Fe^{2+}$ ion chelating 활성 측정 실험에서는 모든 추출물이 유의적 차이 없이 높은 활성을 나타냈다. 또한 SOD activity 측정 실험에서 AE가 가장 우수한 활성을 나타냈으며, $H_2O_2$에 의해 유도된 세포독성 또한 AE가 가장 효과적으로 억제시켰다. 이상의 결과를 토대로 추출 조건에 따른 항산화 활성 차이를 수율 대비로 비교하였을 때, 가장 낮은 활성을 나타냈던 HWASE에 대비하여 AE는 ABTS radical scavenging activity 1.90배, alkyl radical scavenging activity 2.32배, DPPH radical scavenging activity 1.72배, SOD assay 1.65배, $H_2O_2$에 의해 유도된 세포독성에 대한 보호효능 1.84배의 우수한 항산화 활성을 나타냈다.
