Dopaminergic Inhibition of Dorsal Horn Cell Activity in the Cat

  • Dopamine has been generally known to exert antinociceptive action in behavioral pain test, such as tail flick and hot plate test, but there appears to be a great variance in the reports on the antinociceptive effect of dopamine depending on the dosage and route of drug administration and type of animal preparation. In the present study, the effects of dopamine on the responses of wide dynamic range (WDR) cells to mechanical, thermal and graded electrical stimuli were investigated, and the dopamine-induced changes in WDR cell responses were compared between animals with an intact spinal cord and the spinal animals. Spinal application of dopamine (1.3 & 2.6 mM) produced a dose-dependent inhibiton of WDR cell responses to afferent inputs, the pinch-induced or the C-fiber evoked responses being more strongly depressed than the brush-induced or the A-fiber evoked responses. The dopamine-induced inhibition was more pronounced in the spinal cat than in the cat with intact spinal cord. The responses of WDR cell to thermal stimulation were also strongly inhibited. Dopamine $D_2$ receptor antagonist, sulpiride, but not $D_1$ receptor antagonist, significantly blocked the inhibitory action of dopamine on the C-fiber and thermal responses of dorsal horn cells. These findings suggest that dopamine strongly suppresses the responses of WDR cells to afferent signals mainly through spinal dopamine $D_2$ receptors and that spinal dopaminergic processes are under the tonic inhibitory action of the descending supraspinal pathways.

새로운 캅사이신유도체 DA-5018의 피하주사 및 국소도포시 진통효과 (Analgesic Effects of DA-5018, a New Capsaicin Derivative, after Subcutaneous Injection and Topical Application)

  • The analgesic effects of DA-5018, a new caosaucin derivative, were evaluated in various experimental pain models. Drugs were administered subcutaneously or topically. When drugs were administered subcutaneously, 1) the $ED_{50}$ of DA-5018, morphine . HCI, capsaicin and acetaminophen were 0.091-2.0, 0.3-4.3, 1.4-26.5 and 45.4-643 mg/kg, respectively in various pain or inflammatory models including acetic acid writhing, formalin, tail flick, Randall-Selitto, hot plate and crouton oil-induced ear edema test, 2) the AD2 values (the dose for doubling of pain threshold of vehicle control) of DA-5018, capsaicin and ketoprpgin were 1.07 $\pm$ 0. 18, 23.47$\pm$4.46 and 2.97$\pm$0.43 mg/kg in Freund's complete adjuvant (FCA)-induced arthritic pain model. And by topical application, 1) neither DA-5018 0.3% cream nor Zostrix-HP (capsaicin 0.075%) were effective in formalin test, 2) although DA-5018 0.3% cream significantly inhibited the croton oil-induced ear edema being better than Zostrix-HP and Kenofen (ketoprofen 3%). 3) In FCA model, DA-5018 0.3% cream reversed the decreased pain threshold of arthritic rat from 136.4 g (day 0) to 289.0 g (day 5) and 250.1 g (day 10), which was similar to Zostrix-HP. These results suggest that DA-5018 administered subcutaneously has a potent and broad analgesic spectrum than nonsteroidal antiinflammatory drugs against acute and chronic pain, and by topical application it exerts comparable analgesic and antiinglammaatory effects to capsaicin cream.

신통축어탕(身痛逐瘀湯)의 항염(抗炎), 진통(鎭痛) 및 항열전효과(抗血栓效果)에 관(關)한 연구(硏究) (Study on the Anti-inflammatory, Analgesic and Anti-thrombotic Effects of Shintongchugeotang in the Experimental Animals)

  • This study was designed to elucidate the anti-inflammatory, cardiovascular, anti-thrombotic and analgesic effects of Shintongchugeotang. The anti-inflammatory effect was measured by the method of carragenin induced edema, protein leakage test using CMC-pouch, and the analgesic effect was measured by the acetic acid method and hot plate method, and the effect of Shintongchugeotang on the cardiovascular system was observed by the change of flow rate of Ringer solution in the vascular system in the ear of rabbit, and the contraction and dilatation of rat tail artery. Death rate, platelet aggregation, plasma coagulation activity was observed for the measurement of the anti-coagurative effect of Shintongchugeotang. The result was as follows : 1. After the administration of Shintongchugeotang extract, Carragenin induced edema and CMC-pouch protein leakage were significantly decreased. 2. The slight analgesic effect of Shintongchugeotang extract was confirmed by the observation of writhing syndrome, paw licking time, and escape time. 3. The drug increased the auricular blood flow in rabbit. 4. The drug relaxed the artery contraction by pretreated norepinephrine in rat. 5. The drug inhibited the death rate of mouse which was led to thromboembolism by serotonin and collagen. 6. The drug inhibited the platelet aggregation in rat. 7. The drug prolonged the prothrombin time and activated partial thromboplastin time on the test of plasma coagulation factor activity in rat, but was not valuable.

중심압축을 받는 고강도강 중간주의 좌굴강도 평가 (Buckling Strength of Concentrically Loaded High-Strength Steel Columns with Intermediate Slenderness)

  • 본 연구에서는 공칭인장강도 800MPa급 고강도강재(HSA800)로 제작된 중간주의 중심압축실험을 통해 좌굴강도를 평가하여 현행 구조설계기준(KBC2009, 2010 AISC-LRFD) 기둥곡선의 비탄성영역 적용성 여부를 검토하였다. 강도로 무차원화한 판폭두께비와 판단부 지지조건 및 기둥 세장비를 변수로 하였으며, 고강도강재와 일반강재의 좌굴거동 차이 여부를 확인하기 위해 일반강재(SM490)로 제작된 비교실험체를 포함시켰다. 실험결과 중심압축을 받는 모든 HSA800 중간주 실험체는 현행 강구조기준의 설계좌굴강도를 충분히 발휘하였으나 일부 SM490 실험체는 설계강도를 하회하였다. 이는 고강도강재의 잔류응력에 의한 강도저하가 일반강재에 비해 작기 때문으로 판단된다.

학교급식 HACCP 검증 체크리스트 개선 - 오징어채소무침을 중심으로 - (Improvement of HACCP Verification Checklist in School Foodservices - A Case Study on Cooked Squid with Seasoned Fresh Vegetable -)

  • The purpose of this study was the improvement and modification of the HACCP verification checklist in school foodservices. For this, the HACCP verification checklist was modified on the basis of an existing school foodservice format. The modified checklist was composed of 28 items, including CCPs (critical control points), microbial test, and other components of the HACCP system than CCPs. To confirm the suitability of the modified checklist, comparisons were made based on the microbiological quality of cooked foods, utensils, and number of aerial microbes in the working area. In this study, the applicability of the modified checklist was determined by focusing on cooked squid with seasoned fresh vegetables (Ojingeochaesomoochim). The following results were obtained from 14 schools in Changwon. The checklist scores for maintaining hot foods over $60^{\circ}C$ or serving within 2 hours, microbial tests of drinking water, food contact surfaces and cooking utensils, monitoring tools, and usage of suitable sanitizers were 2 points each (The possible highest score is 2 points). On the contrary, the checklist score for microbial test of cooked foods was the lowest of all the items. The correlation coefficient (r) between the improved checklist and microbiological quality of cooked foods was 0.699 (P<0.01), whereas that between the improved checklist and microbiological quality of cooking utensils was 0.612 (P<0.05). The correlation coefficient between the improved checklist and aerial plate count in the working area was -0.556 (P<0.05). Our results indicate the potential possibility of using the HACCP verification checklist in school foodservices.

Anti-nociceptive and Anti-inflammatory Properties of Ilex latifolia and its Active Component, 3,5-Di-caffeoyl Quinic Acid Methyl Ester

  • The present study was conducted to investigate anti-nociceptive and anti-inflammatory effects of the leaves of Ilex latifolia Thunb (I. latifolia) in in vivo and in vitro. Writhing responses induced by acetic acid and formalin- and thermal stimuli (tail flick and hot plate tests)-induced pain responses for nociception were evaluated in mice. I. latifolia (50 - 200 mg/kg, p.o.) and ibuprofen (100 mg/kg, p.o.), a positive non-steroidal anti-inflammatory drug (NSAID), inhibited the acetic acid-induced writhing response and the second phase response (peripheral inflammatory response) in the formalin test, but did not protect against thermal nociception and the first phase response (central response) in the formalin test. These results show that I. latifolia has a significant anti-nociceptive effect that appears to be peripheral, but not central. Additionally, I. latifolia (50 and $100{\mu}g/mL$) and 3,5-di-caffeoyl quinic acid methyl ester ($5{\mu}M$) isolated from I. latifolia as an active compound significantly inhibited LPS-induced NO production and mRNA expression of the pro-inflammatory mediators, iNOS and COX-2, and the pro-inflammatory cytokines, IL-6 and $IL-1{\beta}$, in RAW 264.7 macrophages. These results suggest that I. latifolia can produce antinociceptive effects peripherally, but not centrally, via anti-inflammatory activity and supports a possible use of I. latifolia to treat pain and inflammation.

Cisternography 검사 시 사용되는 방사성의약품의 안정성 확보를 위한 검사도입 및 유용성 평가 (Evaluation of Usefulness and Procedures for Safety of Radiopharmaceuticals in Cisternography)

  • 뇌조 조영술에 이용되는 방사성의약품들 중 최근에는 Tc-99m DTPA를 이용한 뇌 척수액 영상이 대부분이다. 하지만 Tc-99m DTPA는 사용 시 무균수막염, 근육강직, 경련 등 여러 부작용이 발생할 우려가 있다. 따라서 Tc-99m DTPA의 이러한 부작용의 발생을 사전에 예방하기 위하여 안정성검사를 시행하였으며 그 유용성을 알아보고자 한다. 방사성의약품 Vial검사는 2008년 12월 16일 - 2009년 12월 30일에 뇌조조영술을 시행한 환자(n=12)를 대상으로 시행하였으며, 시술시 사용한 vial내의 DTPA 성분 (Mallinckrodt사의 Techscan-DTPA사용), NaPertechnate radiation dose 및 volume, 생리 식염수량, 환자주사용 용량 및 activity 등을 측정하여 순수 DTPA의 양을 계산하였다. 방사성의약품의 내독소평가(Bacterial endotoxin)는 2008년 12월 16일-2009년 12월 30일에 뇌조 조영술을 시행한 환자(n=12)를 대상으로 하였고, 이 때 사용한 DTPA vial의 Pyrogen test (LAL test)를 시행하였다. 시약이 함유된 Positive/Negative control vial(표준액)에 normal saline 0.2 mL을 주입하고, 동일한 Test control vial(실험액)에 normal saline 0.1mL 와 Tc-99m DTPA 0.1 mL로 주입하였으며 Digital hot plate에서 $37.5^{\circ}C$로 1시간 반응시킨 후 응고여부를 표준액과 대조하여 관찰하였다. 안정적인 제조절차 준수는 보존제(방부제)가 없는 미 개봉한 CaNa3 DTPA kit를 사용하였으며 Tc-99m DTPA는 한 Vial당 한명의 환자선량으로 뽑아 투여한다. 이때 적정 용량을 맞추기 위해 0.9% NaCl 멸균 생리 식염수를 사용하여 희석하였다. 방사성의약품 Vial검사는 측정된 성분값들로 순수 DTPA의 양을 얻어내었으며 이는 2가지의 계산식을 이용하여 선량 대비 DTPA의 용량(평균 0.88 mg), 용량 대비 DTPA의 용량(평균 0.74 mg)을 도출해내었다. 방사성의약품의 내독소 평가에서는 동일한 조건으로 시험(n=12)을 시행한 결과 모두 Bacterial endotoxin이 기준치인 단위 선량 당 14 endotoxin units (EU)미만의 반응인 Negative(-)로 나왔다. 안정적인 제조절차 준수에서는 검사 관련 sheet를 만들어 작성 및 제조절차 준수 여부를 확인하였으며 결과 모두 준수하여 제조과정 내에서 방사성의약품에 의하여 발생할 수 있는 부작용이 없었다. Tc-99m DTPA를 이용한 Cisternography 검사 시 항상 방사성의약품의 안정성문제가 제기되어 왔다. 수막강 내 주입용 방사성의약품으로서 제조 및 사용 시 불안정한 취급으로 부작용이 발생할 우려가 있으며, 이에 따른 사용지침이 확립되어야 한다고 생각되었다. 수막강 내 주입용 방사선의약품으로써 free acid 혹은 sodium을 포함하는 DTPA는 적절하지 않으며, 반면 Calcium이 포함된 DTPA는 적절한 것으로 알려져 있다. 위와 같이 Techscan-DTPA(Mallinckrodt): CaNa3으로 간단하며 편리하게 안정성검사를 시행할 수 있었으며 각 검사결과(n=12) 안정성검사를 모두 통과하여 검사 시 동반될 수 있는 부작용은 나타나지 않았다. 앞으로 위와 같은 SOP (Standard Operating Procedures)를 적용하여 보다 쉽고, 편리하게 안정성검사를 시행할 수 있으리라 사료된다.

음나무 잎 및 수피의 진통소염효과 및 아주반트로 유발된 산화적 스트레스에 대한 효과 (In vivo Antinociceptive Antiinflamatory and Antioxidative Effects of the Leaf and Stem Bark of Kalopanax pictus in Rats)

  • The leaves (KPL) of Kalopanax pictus (KP) are used as a vegetable or a functional food in Korean society. The stem bark (Kalopanacis Cortex, KPS) has been traditionally used to treat neurotic pain, rheumatoid arthritis and diabetic disease. This research was undertaken to demonstrate that the leaf extract of KP (KPL) has also the antinociceptive and antiinflammatory effects like the extract (KPS) of Kalopanacis Cortex and to compare the activity levels of several extracts obtained from KP. Antinociceptive and antiinflammatory effects were measured against the extracts described as followings; KPL-1 (the MeOH extract obtained from the leaf shoot of KP collected on May), KPL-2 (the MeOH extract from KP collected on June), KPL-3 (the MeOH ectract from KP with no thorns), KPS-1 (MeOH extract from KPS of a Korean habitat), KPS-2 (MeOH extract from KPS of a Chinese habitat). The antimociceptive test undertaken by acetic acid-induced writhing, hot plate-, and tail-flick methods using mice. The anti-inflammatory test was also undertaken by measuring the edema in the carrageenan-induced test. The order of activity potency in the antinociceptive and antiinflammatory assays was commonly shown as followings: KPL-3>KPS>1>KPS-2>KPL-1>KPL-2. This order was also observed in acetic acid-induced vascular permeability test. The antiinflammatory activity in carrageenan-induced assay was also observed as the following order: KPL-3>KPS- 1>PS-2>KPL-1>KPL-2. In addition, adjuvant-induced rats were used for a model to assess the oxidative stress. Treatment of the rat with the extracts reduced serum thiobarbituric acid-reactive substances (TBARS), hydroxy radical(OH) and superoxide dismutase(SOD) activity caused by FCA together together with the inhibition of hepatic TBARS level and lipofuscin content. The above finding suggests that the leaf extract has the antinociceptive and antinflammatory activity. It is also suggested that KPL-3 with more potent activity than other tested extracts could be developed for a new available biomaterial.

Antibacterial, Anti-Diarrhoeal, Analgesic, Cytotoxic Activities, and GC-MS Profiling of Sonneratia apetala (Buch.-Ham.) Seed

  • Fruits of Sonneratia apetala (Buch.-Ham.), (English: mangrove apple, Bengali: keora) both seeds and pericarps, are largely consumed as food besides their enormous medicinal application. The fruit seeds have high content of nutrients and bioactive components. The seeds powder of S. apetala was successively fractionated using n-hexane, diethyl ether, chloroform, ethyl acetate, and methanol. The fractions were used to evaluate antibacterial, anti-diarrhoeal, analgesic, and cytotoxic activities. Methanol fraction of seeds (MeS) stronly inhibited Escherichia coli strains, Salmonella Paratyphi A, Salmonella Typhi, Shigella dysenteriae, and Staphylococcus aureus except Vibrio cholerae at $500{\mu}g/disc$. All the fractions strongly inhibited castor oil induced diarrhoeal episodes and onset time in mice at 500 mg extract/kg body weight (P<0.001). At the same concentration, MeS had the strongest inhibitory activity on diarrhoeal episodes, whereas the n-hexane fraction (HS) significantly (P<0.05) prolonged diarrhoeal onset time as compared to positive control. Similarly, HS (P<0.005) inhibited acetic acid induced writhing in mice at 500 mg extract/kg, more than any other fraction. HS and diethyl ether fractions of seed strongly increased reaction time of mice in hot plate test at 500 mg extract/kg. All the fractions showed strong cytotoxic effects in brine shrimp lethality tests. Gas chromatography-mass spectrometry analysis of HS led to the identification of 23 compounds. Linoleic acid (29.9%), palmitic acid (23.2%), ascorbyl palmitate (21.2%), and stearic acid (10.5%) were the major compounds in HS. These results suggest that seeds of S. apetala could be of great use as nutraceuticals.
