Screening of Promoters from Metagenomic DNA and Their Use for the Construction of Expression Vectors

  • This study was focused on the screening of valuable genetic resources, such as promoters from metagenome, and describes a promoter trapping system with a bidirectional probe concept, which can select promoters or operons from various biological resources including metagenomic DNA. A pair of reporters, GFP and DsRed, facing the opposite direction without promoters, is an effective system that can function regardless of the direction of inserted promoters. The feasibility of this system was tested for the isolation of constitutively expressed promoters in E. coli from a soil metagenome, resulting in a potential pool of various promoters for practical application. The analyses of structural organization of the trapped genes demonstrated that constitutively expressible promoters in E. coli were broadly distributed within the metagenome, and suggested that some promoters were useful for the construction of expression vectors. Based on these observations, three constitutive promoters were employed in the expression vector system and their potentials for practical application were evaluated in terms of expression level, protein solubility, and effects on host growth.

In-wheel 모터를 이용한 전기자동차 구동시스템의 구현 (Implementation of In-wheel Motor Driving System for Electric Vehicle)

  • In-wheel motor system gets the driving force from direct-driven motor in the wheel of electric vehicle. It is known as good system for vehicles, from an efficiency, packaging, handling and safety. This paper describes motor and inverter technologies, system configuration and control algorithms for in-wheel type electric vehicle. It is necessary to control on an interrelation perspective because this system drives two motors at same time. In system design, IPMSM(Interior Permanent Magnet Synchronous Motor) including a wide operating range and high-speed rpm is used and flux weakening control is performed in constant power range. Under the torque command from the host controller, auto control box, inverter's output torque is calculated with using torque estimation technique and applied to actual vehicle driving system. It is verified that the configuration and the algorithm are suitable for the in-wheel motor system.

An Implementatin of a Multi-Channel Speech Surveillance System Over Telephone Lines

  • This paper presents an implementation of a multi-channel speech surveillance system over telephone lines using TMS320C31 DSP chips. The incoming speech into each telephone line are first compressed simultaneously in real-time by the popular vector-sum excited linear predictive (VSELP) speech coding algorithm at the rate of 8 Kbps. The compressed steech bit streams are then multiplexed with those of other users. The multiplexed speech bit streams are transferred to the system storage equipments with some other required information so that a system operator can later monitor the stored speech data whenever it is necessary. The host program runs under Microsoft Windows95 for an efficient man-machine interface and a future upgrade-ability. We have confirmed that the overall 64-channel system operates satisfactorily in realtime. We also have checked approximately up to 2,880 total hours of recording capability of the system on a playback module and two removable backup drives.

Development of Leuconostoc sp. Host Vector System

  • Leuconostoc citreum CBUE isolated from kimchi proved to harbor a small cryptic plasmid, pNS75. The complete nucleotide sequence of pNS75 was 1,821 bp and had a low G+C content of 39.2%. Computer analysis using DNASIS revealed one open reading frame (ORF), having ATG as putatitive start condon and potentially encoding proteins with molecular mass of 38 kDa. The chimeric plasmid pLeuCM was first constructed wih pNS75, pUC19 and chroamphenicol acetyltransferase (CAT) from Staphylococcus sp.. pLeuCM replicated and expressed chroamphenicol acetyltransferase in Leuconostoc citerum CBNF after transformation. To test the availability of shuttle vector as cloning vehicle of foreign gene, $\alpha$-amylase gene of Streptococcus bovis was cloned and all transformants secreated the $\alpha$-amylase successfully. The result indicates that pLeuCM is a potential shuttle vector for Leuconostoc spp. and lactic acid bacteria.

A Gammaherpesvirus Establishes Persistent Infection in Neuroblastoma Cells

  • Gammaherpesvirus (${\gamma}HV$) infection of the central nervous system (CNS) has been implicated in diverse neurological diseases, and murine ${\gamma}HV$-68 (MHV-68) is known to persist in the brain after cerebral infection. The underlying molecular mechanisms of persistency of virus in the brain are poorly understood. Here, we characterized a unique pattern of MHV-68 persistent infection in neuroblastoma cells. On infection with MHV-68, both murine and human neuroblastoma cells expressed viral lytic proteins and produced virions. However, the infected cells survived productive infection and could be cultured for multiple passages without affecting their cellular growth. Latent infection as well as productive replication was established in these prolonged cultures, and lytic replication was further increased by treatment with lytic inducers. Our results provide a novel system to study persistent infection of ${\gamma}HVs$ in vitro following de novo infection and suggest application of MHV-68 as a potential gene transfer vector to the brain.

파밤나방 세포주에서 Autographa californica 핵다각체병 바이러스의 감염 특성 (Characteristics of Autographa californica Nuclear Polyhedrosis Virus in Spodoptera exigua Cell Line.)

  • AcNPV에 대해 감수성을 보이는 파밤나방 세포주인 Se301 세포주의 AcNPV를 이용한 발현 벡터계에서의 유용성을 알아보기 위하여 Se301세포주에서 AcNPV의 감염 특성을 Sf-21 세포주에서와 비교하였다. 그 결과, 바이러스의 감염상은 두 세포주에서 거의 유사하였으나 감염 7일 후에 세포로부터 방출된 다각체의 비율은 Se301 세포주에서 훨씬 높았다. 또한 전체 다각체 생성율은 Se301 세포주에서 약간 높았지만 그 차이는 매우 근소하여 대체적으로 두 세포주에서 거의 비슷하였으나 생성된 다각체의 크기는 Se301세포주에서 더욱 컸다. 한편, 세포주나 바이러스의 증식율과는 무관하게 AcNPV에 의한 다각체 단백질의 발현량이 Se301 세포주에서 약 2.4배 높게 나타남으로써 Se301 세포주가 AcNPV를 이용한 발현 벡터계에서 숙주 세포로서 매우 유용하게 이용될 수 있음을 보여 주었다.

Mycobacteria에 적용 가능한 genetic tool로서의 새로운 vector system 개발 (Development of New Vector Systems as Genetic Tools Applicable to Mycobacteria)

  • Mycobacterium 속은 Mycobacterium tuberculosis, Mycobacterium leprae, Mycobacterium bovis와 같은 동물과 인체에 병원성을 나타내는 세균 종을 다수 포함하고 있다. 이들의 숙주에서의 생존과 병원성에 관한 유전학적 정보를 확보하는 것은 매우 중요하지만, 효과적인 유전학적 도구가 부족하였기 때문에 이들에 관한 연구가 미비하였다. 따라서 mycobacteria의 연구를 위한 분자생물학적 실험 도구로서 다양한 기능성 vector들이 고안되었고, 이러한 기능성 vector의 개발은 실질적으로 mycobacteria에서의 연구 효과를 증진시켰다. 본 연구에서는 Mycobacterium smegmatis에 적용 가능하고 기존에 제시되었던 mycobacteria 연구에 있어서의 한계점을 극복하기 위한 노력의 일환으로, 기능성 vector인 temperature-sensitive replication origin (TSRO)과 counterselectable marker로 levansucrase를 암호화하는 sacB 유전자를 포함하는 suicide vector pKOTs, chromosomal DNA로 site-specific recombination을 통해 삽입되는 lacZ transcriptional fusion vector pMV306lacZ, 그리고 TSRO를 가지는 minitransposon vector pTnMod-OKmTs를 개발하였다. 이 vector들은 실질적으로 M. smegmatis에서 효과적으로 작동하는 것이 확인되었으며 목적으로 하는 실험 결과 도출 가능성 또한 보여주었다. 따라서 이들 vector는 앞으로의 mycobacteria에 대한 효과적인 연구 기반이 될 것으로 기대된다.

Large-Scale Production of Rotavirus VLP as Vaccine Candidate Using Baculovirus Expression Vector System (BEVS)

  • Rotavirus virus-like particle (VLP) composed of VP2, VP6, and VP7 was expressed in the Baculovirus Expression Vector System (BEVS). Sf9 cell, a host of the baculovirus, was cultured from a 0.5-1 spinner flask to the 50-1 bioreactor system. Sf9 cell was maintained at cell density between 3.0E+05 and 3.0E+06 cells/ml and grew up to 1.12E+07 cells/ml in the bioreactor. Growth kinetics was compared under different culture systems and showed similar growth kinetics with 20.1-25.2 h of doubling time. Early exponentially growing cell culture was infected with three recombinant baculoviruses expressing VP2, VP6, and VP7 protein at 1.0, 2.0, and 0.2 moi, respectively. The expression of rotavirus proteins was confirmed by Western blot analysis and its three-layered virus-like structure was observed under an electron microscope. Rotavirus VLP was semipurified and immunized in ICR mice intramuscularly. Rotavirus-specific serum antibody was detected from 2 weeks after the immunization and lasted at least 21 weeks of the post-immunization, indicating its possible use as a vaccine candidate.

The Study of Mutation Spectrum in Iac / Gene of Transgenic Big Blue$\textregistered$ Cell Line Following Short-Term Exposure to 4-Nitroquinoline N-oxide

  • Transgenic animal and cell line models which are recently developed in toxicology field combined with molecular biological technique, are powerful tools for studying of mutation in vivo and in vitro, respectively. The Big Blue mutagenesis assay system is one of the most widely used transgenic systems. Especially, for the study of direct acting mutagens, Big Blue cell line is very useful and powerful to evaluate mutagenicity because the mutation frequency and mutationspectrlun showed no distinct differences between cell line and animal. The Big Blue cell lines carry stably integrated copies of lambda shuttle vector containing lac I gene as a mutational target. These lambda shuttle vectors are useful for various mutagenesis related studies in eukaryotic system due to their ability to be exposed mutagen and then transfer a suitable target DNA sequence to it convenient organism for analysis. We tried to assess the mutagenic effect of 4-NQO with Big Blue cell line. After the treatment of 4-NQO, genomic DNA was isolated and lambda shuttle vector was packaged by in Vitro packaging and then these were plated on bacterial host in the presence of X-gal to screen mutation in the lac I. We determined MF as a ratio of blue plaques versus colorless plaques and now undergoing the mutation spectrum of 4-NQO in lac J gene sequence.

Adenovirus vs AAV Vectors for Gene Delivery: Their Advantages and Disadvantages

  • Gene therapy is to treat and cure diseases by an introduction of therapeutic genes in defective cells or tissues of human body. Gene delivery system, gene expression system, and therapeutic gene are three core elements for gene therapy. The efficient delivery of therapeutic genes and appropriate gene expression are the crucial issues for therapeutic outcome of gene delivery. Because it can be used in common for the treatment and cure of various diseases, gene delivery system is the most important core element for a successful gene therapy. Viruses are naturally evolved to transfer their genomes into host cells efficiently. This ability has made vectorologists exploit viruses as attractive vehicles for the delivery of therapeutic genes. Viral vectors based on adenovirus (Ad) and adeno-associated virus (AAV) have been often used for gene delivery in laboratory. Ad and AAV vectors derived from human DNA viruses differ greatly in their life cycle, expression level and duration of transgenes, immunogenicity, and vector preparation. Both vectors can be used as effective tools for gene therapy and more recently in functional genomics. Here, the characteristics of Ad and AAV vectors are discussed.
