장기간 플루세틴 처리에 의한 흰쥐 해마에서의 NCAM140 유전자 발현의 증가 (Chronic Treatment of Fluoxetine Increases Expression of NCAM140 in the Rat Hippocampus)

  • Objectives : Most of the mechanisms reported for antidepressant drugs are the enhancement of neurite outgrowth and neuronal survival in the rat hippocampus. Neural cell adhesion molecule 140(NCAM140) has been implicated as having a role in cell-cell adhesion, neurite outgrowth, and synaptic plasticity. In this report, we have performed to elucidate a correlation among chronic antidepressant treatments, NCAM140 expression, and activation of phosphorylated cyclicAMP responsive element binding protein(pCREB) which is a downstream molecule of NCAM140-mediated intracellular signaling pathway in the rat hippocampus. Methods : Fluoxetine(10mg/kg) was injected acutely(daily injection for 5days) or chronically(daily injection for 14days) in adult rats. RNA and protein were extracted from the rat hippocampus, respectively. Real-time RT-PCR was performed to analyze the expression pattern of NCAM140 gene and western blot analyses for the activation of the phosphorylation ratio of CREB. Results : Chronic fluoxetine treatments increased NCAM140 expression 1.3 times higher than control in rat hippocampus. pCREB immunoreactivity in the rat hippocampus with chronic fluoxetine treatment was increased 4.0 times higher than that of control. Conclusion : Chronic fluoxetine treatment increased NCAM140 expression and pCREB activity in the rat hippocampus. Our data suggest that NCAM140 and pCREB may play a role in the clinical efficacy of antidepressants promoting the neurite outgrowth and neuronal survival.

레벨 셋 방법을 이용한 뇌 MR 영상에서 해마영역 분할 (A Hippocampus Segmentation in Brain MR Images using Level-Set Method)

  • 영상분할은 의료 임상연구에서 가장 중요한 과정 중의 하나이다. 특히 뇌 MRI영상에서 해마의 위축은 알츠하이머병 진행과정의 초기 특정 표지자로서 해마의 볼륨은 초기 알츠하이머병의 임상적 진단에 도움이 된다. 정확한 볼륨 측정에 있어서 해마 영역의 분할은 중요한 역할을 한다. 하지만 MRI 영상에서 해마영역은 낮은 대조도, 낮은 신호 대 잡음 비율, 불연속성 경계의 특징을 보이며, 이러한 특징들은 MRI 영상에서 해마의 정확한 분할을 어렵게 만든다. 이 문제를 해결하기 위해 전처리 과정으로 실험영상에서 관심영역을 선택한 후 반전영상과 원본영상과의 차영상 대조도를 향상시킨 후 비등방성 확산(Anisotropic diffusion) 필터링, 가우시안(Gaussian) 필터링을 수행하였다. 마지막으로 두 개의 레벨 셋(Level Set)기반의 동적 윤곽선(Active Contour) 모델을 결합하여 해마를 분할하는 방법을 제안하였다. 제안된 해마분할방법의 유효성을 다양한 방법으로 평가한 결과 제안된 해마분할방법은 분할 속도와 정확도 면에서 뚜렷하게 개선이 되었음을 확인하였다. 결론적으로 제안된 방법이 해마와 같은 특징을 가진 영역을 분할하는데 적합하다고 할 수 있다. 향후 다른 연구 기법들과 결합할 경우 더욱 잠재성이 증대될 수 있을 것이다.

IBASPM을 이용한 해마체적 측정에서 뇌 Atlases에 대한 고찰 (A Study of brain Atlases in Hippocampus Volume Measurement Using IBASPM)

  • IBASPM(Individual Brain Atlases using Statistical Parametric Mapping Software)를 이용하여 해마의 체적 측정시, Atlas의 종류(Atlas69, Atlas71, Atlas84, Atlas116)에 따른 체적의 변화를 평가하기 위해 20대 정상 성인 10명(남5/여5, $23.3{\pm}2.66$)으로부터 뇌 영상을 획득하였다. 1.5T MRI system(Siemens, Avanto, Erlangen, Germany)의 머리 격자코일(Head matrix coil)을 사용하여 3차원 경사에코 펄스 열(3-D gradient echo pulse sequence)인 MPRAGE(Magnetization Prepared Rapid Acquisition Gradient Echo)영상을 획득하였다. Atlas의 종류에 따라 획득된 해마의 체적을 이용하여 대응표본 t 검정(Paired t-test)을 수행한 결과, 좌측 해마옆이랑(parahippocampal gyrus - left) Atlas69-Altas84, Atlas69-Atlas116(p=0.729, 0.729), 우측 해마형성체(hippocampal formation - right) Atlas69-Atlas84, Atlas69-Atlas116(p=0.219, 0.219)는 유의한 차이가 없었으며, 이를 제외한 부분에서 유의한 차이가 있었다(p=0.000). 그러고 Atlas84와 Atlas116을 이용한 해마의 체적은 모두 동일한 값을 나타내어 유의한 차이가 없었다(p=0.000). Atlas영상과 원본 영상의 overlay를 이용한 영상분석에서는 Atlas71에서만 해마의 부위가 잘 못 정합되는 것을 발견할 수 있었다. 본 연구에 사용된 Atlas의 경우에는 서양인을 바탕으로 개발되었기 때문에 동양인의 뇌와는 차이가 있으며, 정확성 높은 체적의 측정을 위해서는 상황에 맞는 Atlas의 개발이 필요할 것으로 사료된다.

청뇌명신환(淸腦明神丸)이 뇌혈류저하 흰쥐의 학습 및 기억 장애 개선에 미치는 영향 (Ameliorating Effects of Cheongnoemyeongsin-hwan on Learning and Memory Impairment Induced by Cerebral Hypoperfusion in Rats)

  • Objectives : Cheongnoemyeongsin-hwan (CNMSH) is a herb medicine to treat cognitive impairment. This study was investigated the effects of CNMSH on learning and memory impairment induced by cerebral hypoperfusion. Cerebral hypoperfusion was produced chronically by permanent bilateral common carotid artery occlusion (BCCAO) in rats. Methods : CNMSH was administered orally once a day (250 mg/kg) for 28 days starting at 4th week after the BCCAO. The acquisition of learning and the retention of memory were tested on 9th week after the BCCAO using the Morris water maze. In addition, effect of CNMSH on neuronal apoptosis and ${\beta}-amyloid$ accumulation in the hippocmapus was evaluated with immunohistochemistry and Western blotting. Results : 1. CNMSH and ChAL significantly shortened the escape latencies on the 2nd day of acquisition training trials. 2. ChAL significantly prolonged the swimming time spent in the target and peri-target zones and CNMSH also significantly prolonged the swimming time spent in the peri-target zone. 3. CNMSH and ChAL significantly increased the number of target heading in the retention test. 4. ChAL significantly shortened the time of the 1st target heading in the retention test, but CNMSH insignificantly shortened the time of that. 5. CNMSH and ChAL significantly increased the memory score in the retention test. 6. CNMSH and ChAL significantly attenuated the reduction of CA1 neurons, but insignificantly attenuated the reduction of CA1 thickness. 7. CNMSH and ChAL significantly attenuated the up-regulation of Bax expression in the CA1 of hippocampus. 8. CNMSH and ChAL significantly attenuated the up-regulation of cascapse-3 expression in the CA1 of hippocampus. 9. CNMSH and ChAL significantly attenuated the ${\beta}-amyloid$ accumulation in the CA1 of hippocampus. 10. CNMSH and ChAL significantly attenuated the up-regulation of APP expression in the CA1 of hippocampus. 11. CNMSH and ChAL significantly attenuated the up-regulation of BACE-1 expression in the CA1 of hippocampus. Conclusions : The results show that CNMSH attenuates neuronal apoptosis and ${\beta}-amyloid$ accumulation in the hippocampus and alleviates the impairment of learning and memory produced by chronic cerebral hypoperfusion. These results suggest that CNMSH may be a beneficial medicinal herb to treat cognitive impairment associated with neurodegenerative diseases.

Expression of Kir2.1 Channels in Astrocytes Under Pathophysiological Conditions

  • Astrocyte ion channels participate in ionic homeostasis in the brain. Inward rectifying potassium channels (Kir channels) in astrocytes have been particularly implicated in $K^+$ homeostasis because of their high open probability at resting potential and their increased conductance at high concentrations of extracellular $K^+$. We examined the expression of the Kir2.1 subunit, one of the Kir channel subunits, in the mouse brain by immunohistochemistry. Kir2.1 channels were widely distributed throughout the brain, with high expression in the olfactory bulb and the cerebellum. Interestingly, they were abundantly expressed in astrocytes of the olfactory bulb, while astrocytes in other brain regions including the hippocampus did not show any detectable expression. However, Kir2.1 channel-expressing cells were dramatically increased in the hippocampus by kainic acid-induced seizure and the cells were glial fibrillary acidic protein (GFAP)-positive, which confirms that astrocytes in the hippocampus express Kir2.1 channels under pathological conditions. Our results imply that Kir2.1 channels in astrocyte may be involved in buffering $K^+$ against accumulated extracellular $K^+$ caused by neuronal hyperexcitability under phathophysiological conditions.

혈부축어탕(血府逐瘀湯)이 흰쥐의 전뇌허혈에 미치는 영향 (Neuroprotective Effects of Hyulbuchookau-tang(血府逐瘀湯) on Global Cerebral Ischemia of the Rats)

  • Objectives : This study examined the neuroprotective effect of Hyulbuchookau-tang (血府逐瘀湯, HBCAT) against neural damage following global cerebral infarction. Methods : Sprague-Dawley rats were induced with global cerebral infarction by occlusion of the bilateral common carotid artery with hypotension (CCAO). The rats were divided into 3 groups. We treated extract of HBCAT to one group after operation (sample group), one group wasn't induced with ischemic damage after operation (sham group), and one group was induced with ischemic damage after operation (control group) but not treated. We observed neurological scores and cresyl violet-stained hippocampus CAl area, TUNEL-positive neurons, and Bax-positive neurons in brain regions. Results : HBCAT treatment after CCAO increased pyramidal neurons in CAl hippocampus induced by CCAO. HBCAT treatment after CCAO reduced Bax-positive neurons in CAl hippocampus of brain regions induced by CCAO. HBCAT treatment after CCAO wasn't effective for HSP70-positive neurons in CAl hippocampus induced by CCAO. Conclusions : These results suggest that HBCAT has a neuroprotective effect against global cerebral ischemia.

뇌 MR영상 수동분할을 위한 VTK기반의 3차원 가시화 소프트웨어 툴 설계 (Design of 3D Visualization Software Tool Based on VTK for Manual Brain Segmentation of MRI)

  • Mild Cognitive Impairment(MCI) is a prior step to Alzheimer's Disease(AD). It is different from AD which is seriously affecting daily life. Particularly, the hippocampus could be charged a crucial function for forming memory. MCI has a high risk about progress to AD. Our investigated research for a relationship between hippocampus and AD has been studied. The measurement of hippocampus volumetric is one of the most commonly used method. The three dimensional reconstructed medical images could be passible to interpret and its examination in various aspects but the cost of brain research with the medical equipment is very high. In this study, 3D visualization was performed from a series of brain Magnetic Resonance Images(MRI) and we have designed and implemented a competitive software tool based on the open libraries of Visualization ToolKit(VTK). Consequently, our visualization software tool could be useful to various medical fields and specially prognosis and diagnosis for MCI patients.

Pre-ischemic Treatment with Ampicillin Reduces Neuronal Damage in the Mouse Hippocampus and Neostriatum after Transient Forebrain Ischemia

  • Ampicillin, a $\beta$-lactam antibiotic, has been reported to induce astrocytic glutamate transporter-l which plays a crucial role in protecting neurons against glutamate excitotoxicity. We investigated the effect of ampicillin on neuronal damage in the mouse hippocampus and neostriatum following transient global forebrain ischemia. Male C57BL/6 mice were anesthetized with halothane and subjected to bilateral occlusion of the common carotid artery for 40 min. Ampicillin was administered post-ischemically (for 3 days) and/or pre-ischemically (for $3{\sim}5$ days until one day before the onset of ischemia). Pre- and post-ischemic treatment with ampicillin (50 mg/kg/day or 200 mg/kg/day) prevented ischemic neuronal death in the medial CAI area of the hippocampus as well as the neostriatum in a dose-dependent manner. In addition, ischemic neuronal damage was reduced by pre-ischemic treatment with ampicillin (200 mg/kg/day). In summary, our results suggest that ampicillin plays a functional role as a chemical preconditioning agent that protects hippocampal neurons from ischemic insult.

남해안 동대만 잘피밭에서 서식하는 해마(Hippocampus coronatus)의 출현량 및 먹이습성 (Abundances and feeding habits of Hippocampus coronatus in an eelgrass (Zostera marina) bed of Dongdae Bay, Korea)

  • A total of 164 individuals of Hippocampus coronatus were collected from an eelgrass bed in Dongdae Bay, Korea from September 2006 to August 2007. The number of individuals of H. coronatus was higher in September 2006. The size of H. coronatus ranged from 2.4 to 9.3cm in height (Ht), and most of individuals were small size below 5cm (Ht). H. coronatus was a carnivore which consumed mainly gammarid amphipods and copepods. Its diets also included a small amount ofmysids, ostracods, brachiopods, caprellid amphipods, bathynellaceas, isopods, tanaids, and ascothoracids. The diets of H. coronatus underwent size-related changes; smaller individuals consumed copepods, while larger individuals ate gammarid amphipods and mysids. The dietary breadth index of H. coronatus was increased with increasing of their size.

식용 빅벨리 해마(Hippocampus abdominalis) 유래 단백질 가수분해물의 항산화와 항고혈압 효능 (Antioxidant and Antihypertension Effects of Enzyme Hydrolysate from Hippocampus abdominalis)

  • Seahorses have long been used as ornamental and medicinal products. The sea horse Hippocampus abdominalis has a beautiful color and unique shape and is also used for ornamental purposes and as a traditional medicine in China. This study examined the value of H. abdominalis as a health functional food or food additive. H. abdominalis was hydrolyzed using seven proteases: flavourzyme, neutrase, alcalase, trypsin, kojizyme, pepsin and protamex. The yields of all of the enzyme hydrolysates were higher than that of the aqueous extract. Of the enzymatic hydrolysates, seahorse Protamex hydrolysate (SHP) gave the highest yield and had excellent antioxidant and angiotensin-I converting enzyme inhibitory activities. It protected Vero cells against oxidative by 2,2-azobis-(2-amidinopropane) dihydrochloride (AAPH) and antihypertension in Spontaneously Hypertensive Rats. This study attempted to demonstrate H. abdominalis as a health functional food or food additive in the future.
