Surgical Treatment of Lumbosacral Stenosis Caused by Bacterial Discospondylitis in a Great Dane Dog

  • A 6-year-old, 26 kg spayed female Great Dane presented with back pain and hindlimb paresis. On neurological examination, severe pain was detected on the lumbosacral joint displaying nerve-root signature. The animal presented with lower motor neuron paresis with normal deep pain perception. Radiographic examination revealed narrowing of the lumbosacral joint disc space with endplate destructive lysis. Magnetic resonance imaging of the lumbosacral joint revealed a cauda equina compression, especially on the left. On T2 and T1-weighted images, a mass sized 1 × 1 cm was identified laterally to the left of the lumbosacral joint with hyperintense signal. The lumbosacral joint was stabilized by applying the dorsal distraction fixation-fusion technique and dorsal laminectomy. The soft tissue mass was removed, and a bacterial culture was performed. Coagulase-negative Staphylococcus spp. were detected and discospondylitis was treated with clindamycin for 6 weeks. The patient showed clinical improvement without pain and hindlimb paresis until 6 months follow-up postoperatively.

Application of periostin peptide-decorated self-assembled protein cage nanoparticles for therapeutic angiogenesis

  • Peptides are gaining substantial attention as therapeutics for human diseases. However, they have limitations such as low bioavailability and poor pharmacokinetics. Periostin, a matricellular protein, can stimulate the repair of ischemic tissues by promoting angiogenesis. We have previously reported that a novel angiogenic peptide (amino acids 142-151) is responsible for the pro-angiogenic activity of periostin. To improve the in vivo delivery efficiency of periostin peptide (PP), we used proteins self-assembled into a hollow cage-like structure as a drug delivery nanoplatform in the present study. The periostin peptide was genetically inserted into lumazine synthase (isolated from Aquifex aeolicus) consisting of 60 identical subunits with an icosahedral capsid architecture. The periostin peptide-bearing lumazine synthase protein cage nanoparticle with 60 periostin peptides multivalently displayed was expressed in Escherichia coli and purified to homogeneity. Next, we examined angiogenic activities of this periostin peptide-bearing lumazine synthase protein cage nanoparticle. AaLS-periostin peptide (AaLS-PP), but not AaLS, promoted migration, proliferation, and tube formation of human endothelial colony-forming cells in vitro. Intramuscular injection of PP and AaLS-PP increased blood perfusion and attenuated severe limb loss in the ischemic hindlimb. However, AaLS did not increase blood perfusion or alleviate tissue necrosis. Moreover, in vivo administration of AaLS-PP, but not AaLS, stimulated angiogenesis in the ischemic hindlimb. These results suggest that AaLS is a highly useful nanoplatform for delivering pro-angiogenic peptides such as PP.

Synergetic effect of soluble whey protein hydrolysate and Panax ginseng berry extract on muscle atrophy in hindlimb-immobilized C57BL/6 mice

  • Background: Sarcopenia, defined as loss of muscle mass and strength with age, becomes a public health concern as the elderly population increases. This study aimed to determine whether the mixture of soluble whey protein hydrolysate (WPH) and Panax ginseng berry extract (GBE) has a synergetic effect on sarcopenia and, if so, to identify the relevant mechanisms and optimal mixing ratio. Methods: In the first experiment, C57BL/6 mice were hindlimb immobilized for one-week and then administered WPH 800 mg/kg, GBE 100 mg/kg, WPH 800 mg/kg+ GBE 100 mg/kg mixture, and Fructus Schisandrae extract (SFE) 200 mg/kg for two weeks. In the second experiment, experimental design was same, but mice were administered three different doses of WPH and GBE mixture (WPH 800 mg/kg+ GBE 100 mg/kg, WPH 800 mg/kg+ GBE 90 mg/kg, WPH 1000 mg/kg+ GBE 75 mg/kg). Results: In the first experiment, we confirmed the synergetic effect of WPH and GBE on muscle mass and identified that GBE was more effective on the protein synthesis side, and WPH tended to be slightly more effective for protein degradation. In the second experiment, among three different ratios, the WPH 800 mg/kg+ GBE 100 mg/kg was most effective for muscle mass and strength. The mixtures activated muscle protein synthesis via PI3K/Akt/mTORc1 pathway and inhibited muscle protein degradation via suppressing ubiquitin-proteasome system (UPS) and autophagy-lysosome system (ALS), and these effects were more GBE dose-dependent than WPH. Conclusion: The WPH and GBE mixture having a synergetic effect is a potential agent to prevent sarcopenia.

Computed Tomographic Findings in a Foal with Acute Hindlimb Lameness

  • A one-month-old Thoroughbred colt presented with left hindlimb lameness grade 5/5, according to the American Association of Equine Practitioners' lameness scale. The colt started showing signs of lameness two weeks earlier without being involved in an accident. A local veterinarian examined the foal; radiography revealed no significant findings under the hip joint. No improvement was noted after 15 days of non-steroidal anti-inflammatory drugs (NSAIDs) medication. On presentation at our hospital, ultrasonography was performed, which revealed no significant findings in the iliac wings. The foal underwent a computed tomography (CT) scan under general anesthesia. CT revealed bone cysts in the following that could have caused the lameness: the left transverse process of the 5th, 6th lumbar, and the 1st sacrum vertebrae; osteophytes in the auricular surface of the ilium, suggestive of sacroiliac arthritis. The foal recovered smoothly from anesthesia with assistance. The foal was treated with NSAIDs and rested for more than six months. The owner reported that the foal showed no lameness one year later. CT revealed bony changes in the lumbosacral region that were not detected by radiography and ultrasonography, suggesting that CT could be useful for detecting abnormalities in the pelvic region of horses.

Effects of ursolic acid on muscle mass and bone microstructure in rats with casting-induced muscle atrophy

  • [Purpose] Recent studies suggest that ursolic acid (UA) is a potential candidate for a resistance exercise mimetic that can increase muscle mass and alleviate the deleterious effect of skeletal muscle atrophy on bone health. However, these studies evaluated the effects of UA on skeletal muscle and bone tissues, and they have not verified whether such effect could occur concurrently on muscle and bone, as is the case with resistance exercise. Thus, the aim of this study was to analyze the effect of UA injection on muscle mass and bone microstructure using an animal model of atrophy to demonstrate the potential of UA as a resistance exercise mimetic. [Methods] The immobilization (IM) method was used on the left hindlimb of Sprague Dawley (SD) rats for 10 days to induce muscle atrophy, whereas the right hindlimb was used as an internal control (IC). The animal models were divided into two groups, SED (sedentary, n=6) and UA (n=6) to demonstrate the effect of UA on atrophic skeletal muscles. The UA group received a daily intraperitoneal injection of UA (5 mg/kg/day) for 8 weeks. After 10 days of IM, the data collected for the IC were compared with that of IM to determine whether muscle atrophy might occur. [Results] Muscle atrophy was induced and bone mineral density (BMD) decreased significantly. The 8-week UA treatment significantly increased the gastrocnemius muscle mass compared to the SED group. In regard to the effect of UA on bones, negative results such as a decrease in BMD, trabecular bone volume fraction, and trabecular number, and an increase in trabecular separation, were observed in the SED group, but no such difference was observed in the UA group. No significant difference was observed in atrophic hindlimbs between SED and UA groups. [Conclusion] These results alone are insufficient to suggest that UA is a potential resistance exercise mimetic for atrophic skeletal muscle and weakened bone. However, this study will help determine the potential of UA as a resistance exercise mimetic.

A Case of Cutaneous Fibroma in a Korean Indigenous Cattle

  • A case of cutaneous fibroma was diagnosed in the skin of the right hindlimb of a Korean indigenous cattle in Kyungpook province, Korea. Grossly, the protruding skin nodules consisted of a solitary mass $(1.5{\times}1.5{\sim}3.0{\times}3.0cm)$ above large grapelike warty nodules $(11.0{\times}11.0cm)$ on the leg. These masses were firm and rubbery, and the cut surface was gray to white. Histopathologically, the benign neoplastic nodules consisted of spindle-type fibroblasts with collagen. Melanoma, sarcoma, and fibropapilloma were excluded because there was no sign of melanin, muscle type cells, or epidermal proliferation. To our knowledge, this is the first report of a cutaneous fibroma in a Korean indigenous cattle in Korea.

Nailbed malignant melanoma in three dogs

  • Three 8-12-year-old male cocker spaniels presented with an appendicular mass accompanied by pain, inflammation, lameness, and loss of nail in the digits of the forelimb or hindlimb. A histological examination revealed dermal masses of the digit composed of numerous neoplastic cells with marked pleomorphism and high mitosis. The neoplastic cells showed a strong invasive tendency into the epidermis and adjacent bony tissues, such as distal phalanx. Immunohistochemistry of the neoplastic cells in three masses revealed positive reactions for Melan-A. These three dogs were diagnosed as nailbed malignant melanoma in the digit based on the history, clinical signs, and histopathologic features.

Bilateral Osteoarthritis of Coxofemoral Joint in a Thoroughbred Horse

  • A 3-year-old black Thoroughbred colt weighing 500 kg was admitted to the Equine Hospital of Korea Racing Authority (KRA) due to the chronic gait abnormality of the hindlimbs. The history revealed that the patient had run and fallen onto the fence of the farm about at the age of 1 year old and the lameness had been worsening insidiously since the accident. The degree of lameness was 2/5 in accordance with the AAEP lameness grading system in both hindlimbs at hand walking and trot. The lameness was exacerbated at lunging clockwise and counterclockwise on both hindlimbs. The definitive diagnosis of the bilateral osteoarthritis of coxofemoral joint was established using radiography under general inhalation anesthesia. The prognosis was considered to be poor with low expectancy for racing and the colt was expelled from the racecourse eventually. The clinicians are required to consider the possibility of osteoarthritis of coxofemoral joint presented with chronic hindlimb lameness although it is rare. This is the first report on the bilateral osteoarthritis coxofemoral joint in Republic of Korea.

당뇨병 이환견에서 발생한 부갑상샘기능저하증 증례 (Hypoparathyroidism in a diabetic dog)

  • A miniature schnauzer (8 years old, male castrated), diagnosed as diabetes mellitus 6 months ahead, were referred to the Veterinary Medical Teaching Hospital of our University with hindlimb stiffness, facial rubbing and tetany. Serum chemistry showed severe hypocalcemia, hypomagnesemia, and measured iPTH (intact pharathyroid hormone) concentration was low (< 3.0 pmol/L). Consistent rate injection of calcium gluconate (282 mg/kg over 30 min) resulted in prompt relief of the tetany. Then typical treatment for hypoparathyroidism (calcium gluconate and dihydrotachysterol per os) with magnesium administration could control the neuromuscular signs. Diabetes mellitus is being controlled by insulin therapy and prescription diet. Hypomagnesemia in diabetic dog may be induced hypoparathyrodism.

한냉이 흰쥐 대퇴사두근의 허혈 - 재관류 후 SOD 발현에 미치는 영향 (The Effect of Cold Application on Ischemia-Reperfusion Injury to Quadriceps Femoris Muscle of the Rats)

  • The purpose of this study was to investigate the effect of cold application on ischemia-reperfusion injury to quadriceps fomoris muscle of the hindlimbs of the rats. Nine weeks old male Sprague-Dawley white rats were divided into three groups : 1) control(only ischemia-reperfusion), 2) cold application before reperfusion(PreCold), 3) cold application after reperfusion(PostCold). All groups were 30 minute, 1 hour, 3 hours reperfusion after 2 hours ischemia with clamping abdominal artery, and investigate superoxide dismutase(SOD) immunohistochemical reaction for quadriceps femoris muscle of right hindlimb. SOD immunohistochemical reaction of experimental groups were more than the control group. Especially, SOD immunohistochemical reaction of PreCold were less than the PostCold.
